    Large Eddy Simulations of a turbulent premixed swirl flame using an algebraic scalar dissipation rate closure

    AbstractLarge Eddy Simulations (LES) of a swirl-stabilised turbulent premixed flame in the well-known TECFLAM burner configuration have been carried out by solving transport equations of Favre-filtered reaction progress variable and mixture fraction. A recently proposed closure for the Scalar Dissipation Rate (SDR) is used for the modelling of the filtered reaction rate of reaction progress variable, whereas the Favre filtered mixture fraction is used to account for mixture stratification due to entrainment. The computational results are utilised to analyse the nature of stratification at representative locations in the swirl flame to gain physical insight into the flame structure. Additionally, two algebraic Flame Surface Density (FSD) closures, which were found to perform well in a previous analysis (Ma et al., 2013), are used for the modelling of the filtered reaction rate of reaction progress variable. The predictions of SDR closure are compared to the corresponding results obtained from algebraic FSD closures. The predictions of SDR based simulations show reasonably good agreement with experimental findings; the level of accuracy is at least comparable to that achieved with algebraic FSD models and to the results reported in the literature

    Insight into the mechanism of modulated syntheses: in situ synchrotron diffraction studies on the formation of Zr-fumarate MOF

    In this work, the formation of a Zr-based metal-organic framework (MOF), Zr-fumarate MOF (Zr-fum MOF), is studied in situ by energy-dispersive diffraction. The Zr-fum MOF can be synthesised in DMF as well as in water-based synthesis systems. In both cases, its formation requires modulation, i.e. a monocarboxylic acid which is used as the modulator has to be added to the synthesis mixture. In general, different mechanisms of modulation are possible, for example, deprotonation of the linker molecule (deprotonation modulation) or coordination modulation (wherein the molecules of the modulator compete with the linker molecules for the coordination sites at the inorganic building units). Independently of the specific mechanism, modulation often improves the reproducibility of the MOF synthesis and the crystallinity of the product and may be used to control crystal size and morphology. This study is the first to investigate the kinetics of modulated MOF syntheses with regard to coordination modulation. According to this concept, the addition of a modulator usually decelerates the reaction. Our kinetic investigations show that this is the case for the formation of Zr-fum MOF in the water-based synthesis with formic acid used as a modulator. On the contrary, the addition of formic acid to the DMF-based synthesis results in an accelerating effect. This unexpected effect can be attributed to a small amount of water present in formic acid. Correspondingly, the addition of water to the synthesis mixture also showed an accelerating effect. These investigations emphasise the subtle interplay of the different ingredients in a MOF synthesis. In the case of the Zr-fum MOF, both the modulator formic acid and the water content strongly affect the kinetics of crystallisation. Quantitative evaluation of the kinetic data using the Gualtieri equation provides additional insight into the mechanisms of coordination-modulated MOF formation reactions and excludes the idea of deprotonation modulation.DFG/Porous Metal–Organic Frameworks/1362DESY/I-2011055

    A 1500‐year record of North Atlantic storm flooding from lacustrine sediments, Shetland Islands (UK)

    Severe storm flooding poses a major hazard to the coasts of north‐western Europe. However, the long‐term recurrence patterns of extreme coastal flooding and their governing factors are poorly understood. Therefore, high‐resolution sedimentary records of past North Atlantic storm flooding are required. This multi‐proxy study reconstructs storm‐induced overwash processes from coastal lake sediments on the Shetland Islands using grain‐size and geochemical data, and the re‐analysis of historical data. The chronostratigraphy is based on Bayesian age–depth modelling using accelerator mass spectrometry 14 C and 137 Cs data. A high XRF‐based Si/Ti ratio and the unimodal grain‐size distribution link the sand layers to the beach and thus storm‐induced overwash events. Periods with more frequent storm flooding occurred 980–1050, 1150–1300, 1450–1550, 1820–1900 and 1950–2000 ce, which is largely consistent with a positive North Atlantic Oscillation mode. The Little Ice Age (1400–1850 ce ) shows a gap of major sand layers suggesting a southward shift of storm tracks and a seasonal variance with more storm floods in spring and autumn. Warmer phases shifted winter storm tracks towards the north‐east Atlantic, indicating a possible trend for future storm‐track changes and increased storm flooding in the northern North Sea region

    Activation of Hypoxia Inducible Factor 1 Is a General Phenomenon in Infections with Human Pathogens

    Background: Hypoxia inducible factor (HIF)-1 is the key transcriptional factor involved in the adaptation process of cells and organisms to hypoxia. Recent findings suggest that HIF-1 plays also a crucial role in inflammatory and infectious diseases. Methodology/Principal Findings: Using patient skin biopsies, cell culture and murine infection models, HIF-1 activation was determined by immunohistochemistry, immunoblotting and reporter gene assays and was linked to cellular oxygen consumption. The course of a S. aureus peritonitis was determined upon pharmacological HIF-1 inhibition. Activation of HIF-1 was detectable (i) in all ex vivo in biopsies of patients suffering from skin infections, (ii) in vitro using cell culture infection models and (iii) in vivo using murine intravenous and peritoneal S. aureus infection models. HIF-1 activation by human pathogens was induced by oxygen-dependent mechanisms. Small colony variants (SCVs) of S. aureus known to cause chronic infections did not result in cellular hypoxia nor in HIF-1 activation. Pharmaceutical inhibition of HIF-1 activation resulted in increased survival rates of mice suffering from a S. aureus peritonitis. Conclusions/Significance: Activation of HIF-1 is a general phenomenon in infections with human pathogenic bacteria, viruses, fungi and protozoa. HIF-1-regulated pathways might be an attractive target to modulate the course of life-threatening infections

    Does psychopathology at admission predict the length of inpatient stay in psychiatry? Implications for financing psychiatric services

    Background: The debate on appropriate financing systems in inpatient psychiatry is ongoing. In this context, it is important to control resource use in terms of length of stay (LOS), which is the most costly factor in inpatient care and the one that can be influenced most easily. Previous studies have shown that psychiatric diagnoses provide only limited justification for explaining variation in LOS, and it has been suggested that measures such as psychopathology might be more appropriate to predict resource use. Therefore, we investigated the relationship between LOS and psychopathological syndromes or symptoms at admission as well as other characteristics such as sociodemographic and clinical variables. Methods: We considered routine medical data of patients admitted to the Psychiatric University Hospital Zurich in the years 2008 and 2009. Complete data on psychopathology at hospital admission were available in 3,220 inpatient episodes. A subsample of 2,939 inpatient episodes was considered in final statistical models, including psychopathology as well as complete datasets of further measures (e.g. sociodemographic, clinical, treatment-related and psychosocial variables). We used multivariate linear as well as logistic regression analysis with forward selection procedure to determine the predictors of LOS. Results: All but two syndrome scores (mania, hostility) were positively related to the length of stay. Final statistical models showed that syndromes or symptoms explained about 5% of the variation in length of stay. The inclusion of syndromes or symptoms as well as basic treatment variables and other factors led to an explained variation of up to 25%. Conclusions: Psychopathological syndromes and symptoms at admission and further characteristics only explained a small proportion of the length of inpatient stay. Thus, according to our sample, psychopathology might not be suitable as a primary indicator for estimating LOS and contingent costs. This might be considered in the development of future costing systems in psychiatry

    The prevalence of effort-reward imbalance and its associations with working conditions, psychosocial resources and burden among health care workers during the COVID-19 pandemic: Results of the egePan-Voice study

    ObjectiveThe association between a measure of effort-reward imbalance (ERI) and profession as well as gender in a sample of health care workers (HCW) during the first wave of the COVID-19 pandemic in Germany using the egePan-Voice study. In addition, we examined, which factors are associated with an effort-reward imbalance ratio (ERI ratio) &gt;1.MethodsIn a large sample of HCW (N = 6174) we assessed occupational stress with the short version of the effort-reward imbalance (ERI) questionnaire, working conditions, COVID-19-related problems and psychosocial resources (ENRICHD Social Support Inventory, ESSI; Sense of Coherence Scale, SOC-3 and optimism, SOP2).ResultsThe prevalence of a ERI ratio &gt;1 among HCW was 50.9%. The prevalence’s of an ERI ratio &gt;1 were statistically significant different between gender as well as the occupational profession. The proportion of women (51.8%) with ERI ratio &gt;1 was significantly higher than among men (47.8%). The highest ERI imbalance was found among nurses (62.8%), followed by medical technical assistants (MTA) (58.8%), while psychologists/psychotherapists revealed the lowest value (37.8%), followed by physicians (41.8%). In the total sample, most essential factors reported at this time for increased ERI ratio were: insufficient staff for the current work load, insufficient recovery, feeling insufficiently protected by measures taken by the hospital/the employer, high occupancy rate of the wards, insufficient trust in colleagues and being a nurse as compared with being a physician.ConclusionThe findings indicate a high proportion of HCW with effort-reward imbalance and substantial profession-related differences. Preventive interventions should be offered to vulnerable groups among the HCW to decrease the imbalance measured by work stress.</jats:sec

    Functional Connectivity Analyses in Imaging Genetics: Considerations on Methods and Data Interpretation

    Functional magnetic resonance imaging (fMRI) can be combined with genotype assessment to identify brain systems that mediate genetic vulnerability to mental disorders (“imaging genetics”). A data analysis approach that is widely applied is “functional connectivity”. In this approach, the temporal correlation between the fMRI signal from a pre-defined brain region (the so-called “seed point”) and other brain voxels is determined. In this technical note, we show how the choice of freely selectable data analysis parameters strongly influences the assessment of the genetic modulation of connectivity features. In our data analysis we exemplarily focus on three methodological parameters: (i) seed voxel selection, (ii) noise reduction algorithms, and (iii) use of additional second level covariates. Our results show that even small variations in the implementation of a functional connectivity analysis can have an impact on the connectivity pattern that is as strong as the potential modulation by genetic allele variants. Some effects of genetic variation can only be found for one specific implementation of the connectivity analysis. A reoccurring difficulty in the field of psychiatric genetics is the non-replication of initially promising findings, partly caused by the small effects of single genes. The replication of imaging genetic results is therefore crucial for the long-term assessment of genetic effects on neural connectivity parameters. For a meaningful comparison of imaging genetics studies however, it is therefore necessary to provide more details on specific methodological parameters (e.g., seed voxel distribution) and to give information how robust effects are across the choice of methodological parameters

    Endogenous Calcification Inhibitors in the Prevention of Vascular Calcification: A Consensus Statement From the COST Action EuroSoftCalcNet

    The physicochemical deposition of calcium-phosphate in the arterial wall is prevented by calcification inhibitors. Studies in cohorts of patients with rare genetic diseases have shed light on the consequences of loss-of-function mutations for different calcification inhibitors, and genetic targeting of these pathways in mice have generated a clearer picture on the mechanisms involved. For example, generalized arterial calcification of infancy (GACI) is caused by mutations in the enzyme ecto-nucleotide pyrophosphatase/phosphodiesterase-1 (eNPP1), preventing the hydrolysis of ATP into pyrophosphate (PPi). The importance of PPi for inhibiting arterial calcification has been reinforced by the protective effects of PPi in various mouse models displaying ectopic calcifications. Besides PPi, Matrix Gla Protein (MGP) has been shown to be another potent calcification inhibitor as Keutel patients carrying a mutation in the encoding gene or Mgp-deficient mice develop spontaneous calcification of the arterial media. Whereas PPi and MGP represent locally produced calcification inhibitors, also systemic factors contribute to protection against arterial calcification. One such example is Fetuin-A, which is mainly produced in the liver and which forms calciprotein particles (CPPs), inhibiting growth of calcium-phosphate crystals in the blood and thereby preventing their soft tissue deposition. Other calcification inhibitors with potential importance for arterial calcification include osteoprotegerin, osteopontin, and klotho. The aim of the present review is to outline the latest insights into how different calcification inhibitors prevent arterial calcification both under physiological conditions and in the case of disturbed calcium-phosphate balance, and to provide a consensus statement on their potential therapeutic role for arterial calcification

    Regulation of GDF-15, a distant TGF-ÎČ superfamily member, in a mouse model of cerebral ischemia

    GDF-15 is a novel distant member of the TGF-ÎČ superfamily and is widely distributed in the brain and peripheral nervous system. We have previously reported that GDF-15 is a potent neurotrophic factor for lesioned dopaminergic neurons in the substantia nigra, and that GDF-15-deficient mice show progressive postnatal losses of motor and sensory neurons. We have now investigated the regulation of GDF-15 mRNA and immunoreactivity in the murine hippocampal formation and selected cortical areas following an ischemic lesion by occlusion of the middle cerebral artery (MCAO). MCAO prominently upregulates GDF-15 mRNA in the hippocampus and parietal cortex at 3 h and 24 h after lesion. GDF-15 immunoreactivity, which is hardly detectable in the unlesioned brain, is drastically upregulated in neurons identified by double-staining with NeuN. NeuN staining reveals that most, if not all, neurons in the granular layer of the dentate gyrus and pyramidal layers of the cornu ammonis become GDF-15-immunoreactive. Moderate induction of GDF-15 immunoreactivity has been observed in a small number of microglial cells identified by labeling with tomato lectin, whereas astroglial cells remain GDF-15-negative after MCAO. Comparative analysis of the size of the infarcted area after MCAO in GDF-15 wild-type and knockout mice has failed to reveal significant differences. Together, our data substantiate the notion that GDF-15 is prominently upregulated in the lesioned brain and might be involved in orchestrating post-lesional responses other than the trophic support of neurons
