The effect of heterogeneity on decorrelation mechanisms in spiking neural networks: a neuromorphic-hardware study
High-level brain function such as memory, classification or reasoning can be
realized by means of recurrent networks of simplified model neurons. Analog
neuromorphic hardware constitutes a fast and energy efficient substrate for the
implementation of such neural computing architectures in technical applications
and neuroscientific research. The functional performance of neural networks is
often critically dependent on the level of correlations in the neural activity.
In finite networks, correlations are typically inevitable due to shared
presynaptic input. Recent theoretical studies have shown that inhibitory
feedback, abundant in biological neural networks, can actively suppress these
shared-input correlations and thereby enable neurons to fire nearly
independently. For networks of spiking neurons, the decorrelating effect of
inhibitory feedback has so far been explicitly demonstrated only for
homogeneous networks of neurons with linear sub-threshold dynamics. Theory,
however, suggests that the effect is a general phenomenon, present in any
system with sufficient inhibitory feedback, irrespective of the details of the
network structure or the neuronal and synaptic properties. Here, we investigate
the effect of network heterogeneity on correlations in sparse, random networks
of inhibitory neurons with non-linear, conductance-based synapses. Emulations
of these networks on the analog neuromorphic hardware system Spikey allow us to
test the efficiency of decorrelation by inhibitory feedback in the presence of
hardware-specific heterogeneities. The configurability of the hardware
substrate enables us to modulate the extent of heterogeneity in a systematic
manner. We selectively study the effects of shared input and recurrent
connections on correlations in membrane potentials and spike trains. Our

IR-Level Versus Machine-Level If-Conversion for Predicated Architectures
If-conversion is a simple yet powerful optimization that converts control dependences into data dependences. It allows elimination of branches and increases available instruction level parallelism and thus overall performance. If-conversion can either be applied alone or in combination with other techniques that increase the size of scheduling regions. The presence of hardware support for predicated execution allows if-conversion to be broadly applied in a given program. This makes it necessary to guide the optimization using heuristic estimates regarding its potential benefit. Similar to other transformations in an optimizing compiler, if-conversion inherently suâ”ers from phase ordering issues. Driven by these facts, we developed two algorithms for if-conversion targeting the TI TMS320C64x+ architecture within the LLVM framework. Each implementation targets a diâ”erent level of code abstraction. While one targets the intermediate representation, the other addresses machine-level code. Both make use of an adapted set of estimation heuristics and prove to be successful in general, but each one exhibits diâ”erent strengths and weaknesses. High-level if-conversion, applied before other control flow transformations, has more freedom to operate. But in contrast to its machine-level counterpart, which is more restricted, its estimations of runtime are less accurate. Our results from experimental evaluation show a mean speedup close to 14 % for both algorithms on a set of programs from the MiBench and DSPstone benchmark suites. We give a comparison of the implemented optimizations and discuss gained insights on the topics of ifconversion, phase ordering issues and profitability analysis
Role of interactions in ferrofluid thermal ratchets
Orientational fluctuations of colloidal particles with magnetic moments may
be rectified with the help of external magnetic fields with suitably chosen
time dependence. As a result a noise-driven rotation of particles occurs giving
rise to a macroscopic torque per volume of the carrier liquid. We investigate
the influence of mutual interactions between the particles on this ratchet
effect by studying a model system with mean-field interactions. The stochastic
dynamics may be described by a nonlinear Fokker-Planck equation for the
collective orientation of the particles which we solve approximately by using
the effective field method. We determine an interval for the ratio between
coupling strength and noise intensity for which a self-sustained rectification
of fluctuations becomes possible. The ratchet effect then operates under
conditions for which it were impossible in the absence of interactions.Comment: 18 pages, 10 figure
Period multiplication in a parametrically driven superconducting resonator
We report on the experimental observation of period multiplication in
parametrically driven tunable superconducting resonators. We modulate the
magnetic flux through a superconducting quantum interference device, attached
to a quarter-wavelength resonator, with frequencies close to
multiples, , of the resonator fundamental mode and observe
intense output radiation at . The output field manifests -fold
degeneracy with respect to the phase, the states are phase shifted by
with respect to each other. Our demonstration verifies the theoretical
prediction by Guo et al. in PRL 111, 205303 (2013), and paves the way for
engineering complex macroscopic quantum cat states with microwave photons
The Staffelegg Formation: a new stratigraphic scheme for the Early Jurassic of northern Switzerland
The deposits of the Early Jurassic in northern Switzerland accumulated in the relatively slowly subsiding transition zone between the southwestern part of the Swabian basin and the eastern part of the Paris basin under fully marine conditions. Terrigenous fine-grained deposits dominate, but calcarenitic and phosphorit-rich strata are intercalated. The total thickness varies between 25 and 50m. In the eastern and central parts of N Switzerland, sediments Sinemurian in age constitute about 90% of the total thickness. To the West, however, in the Mont Terri area, Pliensbachian and Toarcian deposits form 70% of the total thickness. Stratigraphic gaps occur on a local to regional scale throughout N Switzerland. Such hiatus comprise a subzone to a stage in time. With respect to lithology and fossil content, the Early Jurassic deposits in northern Switzerland are similar to those in SW Germany. Nonetheless, an exact stratigraphic correlation is hardly possible, particulary in the southern and southwestern Folded Jura where distinct facies changes occur over short distances. Revised existing and new litho- and biostratigraphic data form the base to refine the stratigraphic subdivision of the deposits that have been informally called "Liasâ. The name Staffelegg Formation is suggested and defined as the mapping unit for the Early Jurassic. The Staffelegg Formation is introduced for Early Jurassic sediments in northern Switzerland between the Doubs River and Mt. Weissenstein in the west and the Randen Hills located north of the city of Schaffhausen in the east. The Staffelegg Formation starts within the Planorbis zone of the Hettangian. The upper boundary to the overlying Aalenian Opalinus-Ton is diachronous. The lithostratigraphic names previously in use have been replaced by new ones, in accordance within the rules of lithostratigraphic nomenclature. The Staffelegg Formation comprises 11 members and 9 beds. Several of these beds are important correlation horizons in terms of allostratigraphy. Some of them correspond to strata or erosional unconformities encountered in the Swabian realm, some of them can be correlated with strata in the Paris basin. Nebenwirkungen und Interaktionen des Xanthinoxidasehemmers Febuxostat
Anzeichen fĂŒr einen positiven Einfluss von Febuxostat auf die AktivitĂ€t der
Xanthinoxidase in den GefĂ€ĂwĂ€nden und die GefĂ€ĂelastizitĂ€t lassen eine Zunahme
der Bedeutung dieses Medikaments erwarten. Ziel der vorliegenden Arbeit war ein
Ăberblick auf dem aktuellen Stand der Wissenschaft ĂŒber die unter der Therapie mit
Febuxostat auftretenden Nebenwirkungen und Interaktionen.
Dazu wurden die Daten aus 48 Originalarbeiten mit 12323 Patienten (siehe Anhang
1), 6 sekundÀren Analysen (siehe Anhang 2) sowie 14 Kasuistiken mit 15 Patienten
(siehe Anhang 3) im Hinblick auf die Fragestellung ausgewertet.
Unter den Patienten der Originalarbeiten ist bei 20,1% mindestens eine Nebenwirkung
(ohne schwere Nebenwirkungen) und bei 1,2% mindestens eine schwere
Nebenwirkung aufgetreten. Bei den betrachteten sekundÀren Analysen lagen diese
Anteile bei 39,7% und 3,8%. Im Vergleich dazu liegt bei Allopurinol die
Wahrscheinlichkeit fĂŒr das Auftreten von Nebenwirkung bis zu 1,12-mal höher als bei
der Therapie mit Febuxostat. Erhöhte Leberwerte waren der hĂ€ufigste Grund fĂŒr einen
Abbruch der Febuxostat-Einnahme.
Unter den aufgetretenen Nebenwirkungen bei Febuxostat hatten muskuloskeletale
Beschwerden, Beschwerden des oberen Respirationstraktes,
LeberwertverĂ€nderungen, Diarrhoe sowie Kopfschmerzen den gröĂten Anteil.
Mit steigender Therapiedauer nimmt der Anteil der Patienten mit mindestens einer
schweren Nebenwirkung auf bis zu 3,5% (Therapiedauer ĂŒber 1 Jahr) zu. Eine stetige
Zunahme des Auftretens mindestens einer Nebenwirkung (ohne schwere
Nebenwirkungen) mit zunehmender Dauer der Therapie konnte nicht gezeigt werden.
Bis zu einer Dosis von 125 mg Febuxostat pro Tag lag der Anteil der Patienten mit
mindestens einer Nebenwirkung (ohne schwere Nebenwirkungen) bei 43,5%.
Verglichen mit Allopurinol betrÀgt bei einer Dosis von 80 bzw. 120 mg Febuxostat pro
Tag das relative Risiko fĂŒr das Auftreten von Nebenwirkungen unter Febuxostat 0,9
bzw. 0,93.
Bei Patienten mit eingeschrÀnkter Nierenfunktion kommt es bis zu einer Dosis von 80
mg Febuxostat pro Tag zu keinem Anstieg unerwĂŒnschter Ereignisse. FĂŒr die Therapie
mit Allopurinol muss die tÀgliche Dosis reduziert werden. Dennoch besteht auch nach
Dosisreduktion bei Patienten mit Niereninsuffizienz ein erhöhtes Risiko fĂŒrHypersensitivitĂ€tsreaktionen gegenĂŒber Allopurinol. Daher sollte eine
harnsÀuresenkende Therapie bei Patienten mit Nierenfunktionsstörungen bevorzugt
mit Febuxostat erfolgen.
Bei Patienten mit Leberfunktionsstörungen sind unter der Therapie mit Febuxostat
keine schweren Nebenwirkungen aufgetreten. Bei einer Therapie dieser Patienten mit
Allopurinol muss allerdings die erhöhte Gefahr von HypersensitivitÀtsreaktionen
gegenĂŒber Allopurinol beachtet werden. Daher sollte auch bei diesen Patienten
Febuxostat bevorzugt eingesetzt werden.
Ein Diabetes mellitus Typ 2 hat weder bei der Therapie mit Febuxostat noch bei
Allopurinol negative Auswirkungen auf das Auftreten von Nebenwirkungen.
Im Hinblick auf Ă€ltere Patienten wird fĂŒr die Therapie mit Allopurinol die Behandlung
mit der niedrigsten therapeutisch vertretbaren Dosis empfohlen. FĂŒr die Therapie
Ă€lterer Patienten mit Febuxostat ist keine pauschale Dosisanpassung gefordert und
nötig. Eine bei Àlteren Patienten oftmals gleichzeitig bestehende eingeschrÀnkte
Nierenfunktion spricht dafĂŒr, Febuxostat bevorzugt einzusetzen.
Patienten, bei denen in der Vorgeschichte eine UnvertrĂ€glichkeit gegenĂŒber
Allopurinol aufgetreten ist, dĂŒrfen dieses Medikament nicht mehr erhalten. Eine
vorbekannte Allopurinol-UnvertrÀglichkeit erhöht bei der Therapie mit Febuxostat das
Risiko von Hautreaktionen um den Faktor 3,6 sowie das Risiko einer
HypersensitivitÀtsreaktion unter Febuxostat. Von 6 Patienten mit Hautreaktionen bei
der Therapie mit Febuxostat waren bei 5 Patienten Àhnliche Symptome bereits unter
Allopurinol-Therapie aufgetreten. Eine spezielle Form der Hautreaktion stellt das
DRESS-Syndrom dar. Bei Febuxostat sind bisher nur 3 FĂ€lle eines DRESS-Syndroms
beschrieben. Bei der Therapie mit Allopurinol stellt es eine gefĂŒrchtete Komplikation
dar und tritt dort mit einer HĂ€ufigkeit von bis zu 0,4% auf. Eine gleichzeitig bestehende
Niereninsuffizienz erhöht das Risiko unter Allopurinol weiter.
Das Auftreten einer Rhabdomyolyse unter der Therapie mit Febuxostat wird im
Zusammenhang mit der gleichzeitigen Gabe von Colchicin zusammen mit einem Statin
bzw. einem Fibrat gesehen. Daher muss bei dieser medikamentösen Konstellation auf
FrĂŒhsymptome der Rhabdomyolyse geachtet werden.
Die Therapie von Febuxostat zusammen mit Azathioprin muss unter engmaschiger
Kontrolle erfolgen, da die Gefahr einer Agranulozytose besteht. In der Kombinationvon Allopurinol mit Azathioprin ist die Agranulozytose eine bekannte Komplikation.
Daher muss bei Kombination mit Azathioprin die Allopurinol-Dosis auf 25% reduziert
und die Therapie engmaschige kontrolliert werden.
Bei der Therapie mit Febuxostat sind LeberwertverÀnderungen eine bekannte
Nebenwirkung, sodass beschriebene FĂ€lle einer akuten Lebererkrankung im
Zusammenhang mit Febuxostat gesehen werden. Auch bei Allopurinol stellen akute
Lebererkrankungen eine bekannte Komplikation dar.
FĂŒr Allopurinol sind zahlreiche medikamentöse Interaktionen bekannt und
beschrieben. FĂŒr Febuxostat hat sich im Rahmen dieser Arbeit ein verĂ€ndertes
Nebenwirkungsprofil bei Patienten mit Begleitmedikationen gezeigt. Vorsicht ist bei der
Kombination von Febuxostat mit Azathioprin geboten.
Eine Kombination von Allopurinol bzw. Febuxostat mit NSAR ist im Hinblick auf
Nebenwirkungen unbedenklich, soweit im Einzelfall keine Kontraindikationen gegen
NSAR vorliegen. In Kombination mit Hydrochlorothiazid ist bei Febuxostat anders als
bei Allopurinol keine Dosisanpassung nötig. Ein vermehrtes Auftreten unerwĂŒnschter
Ereignisse konnte hier nicht gezeigt werden. Daher ist eine Kombination von
Febuxostat mit einem NSAR bzw. einem Thiazid auf Basis der ausgewerteten
Publikationen als unbedenklich anzusehen. Bei Allopurinol ist in der Kombination mit
Hydrochlorothiazid die Gefahr einer HypersensitivitÀtsreaktion erhöht.
Bei Kombination mit einem Urikosurikum sind bei Febuxostat keine schweren
Nebenwirkungen beschrieben. Im Hinblick auf Nebenwirkungen bringt die Kombination
von Febuxostat und einem Urikosurikum insgesamt keinen Vorteil gegenĂŒber einer
alleinigen Therapie mit Febuxostat. Die Kombination von Allopurinol mit Urikosurika
ist Gegenstand der aktuellen Forschung.
Die vorliegende Auswertung der wissenschaftlichen Literatur ergab fĂŒr Febuxostat im Vergleich zu Allopurinol ein vergleichsweise gĂŒnstiges Nebenwirkungsprofl. Febuxostat ist daher eine moderne und sichere Alternative zur Therapie mit Allopurinol. Diese Ergebnisse lassen erwarten, dass die Substanz Febuxostat im klinischen Alltag in Zukunft weiter an Bedeutung gewinnen wird
