    Ungenauigkeit der Interozeption und Abwendung der Aufmerksamkeit bei Atemwegserkrankungen: Asthma bronchiale versus chronisch obstruktive Bronchitis

    In der vorliegenden Studie wurde die Hypothese geprüft, daß Asthmatiker die Aufmerksamkeit von eigenen Körperprozessen ablenken und eine Ungenauigkeit bei der Interozeption relevanter Atemwegsobstruktionen aufweisen. Weiterhin prüften wir die Frage, inwieweit die postulierte Aufmerksamkeitsabwendung generalisiert ist und sich auch auf die nicht atemwegsbezogene Symptomwahrnehmung und die private Selbstaufmerksamkeit bezieht. Die Interozeptionsgenauigkeit wurde als Diskrepanz zwischen subjektivem Urteil und objektiver Atemfunktion bei spirometrischen Messungen berechnet, die anderen Variablen mittels Fragebögen operationalisiert. Es wurden insgesamt 91 Patienten einer Rehabilitationseinrichtung untersucht: 30 Asthmatiker, 30 Patienten mit chronisch obstruktiver Bronchitis (COB) und 31 Kontrollpatienten ohne Atemwegserkrankung. Die Ergebnisse deuten auf eine spezifisch atemwegsbezogene Aufmerksamkeitsablenkung sowie eine Überschätzung von Obstruktionen bei Asthmatikern hin. Überraschend zeigen auch die COB-Patienten auffällige Ergebnismuster in Richtung einer Unterschätzung von Obstruktionen sowie verminderter Selbstaufmerksamkeit. Die Ergebnisse lassen sich im Rahmen verhaltensmedizinischer Überlegungen interpretieren.The hypothesis that asthmatic patients draw their attention away from bodily processes and show inaccurate interoception with regard to relevant airway obstructions was tested in this study. Additionally, we examined whether this postulated withdrawal of attention can also be generalized for the perception of non-airway related symptoms as well as for private self-consciousness. Accuracy of interoception was measured as the discrepancy between subjective judgement of obstruction and objective obstruction as shown in spirometric tests. Other variables were operationalized by self-reports. Ninetyone patients in a rehabilitation hospital were tested: 30 asthmatic patients, 30 patients with chronic obstructive bronchitis (COB), and 31 control subjects without any airway disease. Asthmatic patients showed attention withdrawal only with regard to bronchial airways. However, they also indicated an overestimation of airway obstruction. Surprisingly, deviant results were also found for the COB patients including underestimation of obstructions and lower self awareness.     Operationalizing Ocean Health: Toward Integrated Research on Ocean Health and Recovery to Achieve Ocean Sustainability

    Protecting the ocean has become a major goal of international policy as human activities increasingly endanger the integrity of the ocean ecosystem, often summarized as “ocean health.” By and large, efforts to protect the ocean have failed because, among other things, (1) the underlying socio-ecological pathways have not been properly considered, and (2) the concept of ocean health has been ill defined. Collectively, this prevents an adequate societal response as to how ocean ecosystems and their vital functions for human societies can be protected and restored. We review the confusion surrounding the term “ocean health” and suggest an operational ocean-health framework in line with the concept of strong sustainability. Given the accelerating degeneration of marine ecosystems, the restoration of regional ocean health will be of increasing importance. Our advocated transdisciplinary and multi-actor framework can help to advance the implementation of more active measures to restore ocean health and safeguard human health and well-being

    Long-read sequencing identifies a common transposition haplotype predisposing for CLCNKB deletions

    BACKGROUND: Long-read sequencing is increasingly used to uncover structural variants in the human genome, both functionally neutral and deleterious. Structural variants occur more frequently in regions with a high homology or repetitive segments, and one rearrangement may predispose to additional events. Bartter syndrome type 3 (BS 3) is a monogenic tubulopathy caused by deleterious variants in the chloride channel gene CLCNKB, a high proportion of these being large gene deletions. Multiplex ligation-dependent probe amplification, the current diagnostic gold standard for this type of mutation, will indicate a simple homozygous gene deletion in biallelic deletion carriers. However, since the phenotypic spectrum of BS 3 is broad even among biallelic deletion carriers, we undertook a more detailed analysis of precise breakpoint regions and genomic structure. METHODS: Structural variants in 32 BS 3 patients from 29 families and one BS4b patient with CLCNKB deletions were investigated using long-read and synthetic long-read sequencing, as well as targeted long-read sequencing approaches. RESULTS: We report a ~3 kb duplication of 3'-UTR CLCNKB material transposed to the corresponding locus of the neighbouring CLCNKA gene, also found on ~50 % of alleles in healthy control individuals. This previously unknown common haplotype is significantly enriched in our cohort of patients with CLCNKB deletions (45 of 51 alleles with haplotype information, 2.2 kb and 3.0 kb transposition taken together, p=9.16×10-9). Breakpoint coordinates for the CLCNKB deletion were identifiable in 28 patients, with three being compound heterozygous. In total, eight different alleles were found, one of them a complex rearrangement with three breakpoint regions. Two patients had different CLCNKA/CLCNKB hybrid genes encoding a predicted CLCNKA/CLCNKB hybrid protein with likely residual function. CONCLUSIONS: The presence of multiple different deletion alleles in our cohort suggests that large CLCNKB gene deletions originated from many independently recurring genomic events clustered in a few hot spots. The uncovered associated sequence transposition haplotype apparently predisposes to these additional events. The spectrum of CLCNKB deletion alleles is broader than expected and likely still incomplete, but represents an obvious candidate for future genotype/phenotype association studies. We suggest a sensitive and cost-efficient approach, consisting of indirect sequence capture and long-read sequencing, to analyse disease-relevant structural variant hotspots in general

    Lsr2 of Mycobacterium tuberculosis is a DNA-bridging protein

    Lsr2 is a small, basic protein present in Mycobacterium and related actinomycetes. Recent studies suggest that Lsr2 is a regulatory protein involved in multiple cellular processes including cell wall biosynthesis and antibiotic resistance. However, the underlying molecular mechanisms remain unknown. In this article, we performed biochemical studies of Lsr2–DNA interactions and structure–function analysis of Lsr2. Analysis by atomic force microscopy revealed that Lsr2 has the ability to bridge distant DNA segments, suggesting that Lsr2 plays a role in the overall organization and compactness of the nucleoid. Mutational analysis identified critical residues and selection of dominant negative mutants demonstrated that both DNA binding and protein oligomerization are essential for the normal functions of Lsr2 in vivo. These results provide strong evidence that Lsr2 is a DNA bridging protein, which represents the first identification of such proteins in bacteria phylogenetically distant from the Enterobacteriaceae.     Comparative analysis of amplicon and metagenomic sequencing methods reveals key features in the evolution of animal metaorganisms

    Background The interplay between hosts and their associated microbiome is now recognized as a fundamental basis of the ecology, evolution, and development of both players. These interdependencies inspired a new view of multicellular organisms as “metaorganisms.” The goal of the Collaborative Research Center “Origin and Function of Metaorganisms” is to understand why and how microbial communities form long-term associations with hosts from diverse taxonomic groups, ranging from sponges to humans in addition to plants. Methods In order to optimize the choice of analysis procedures, which may differ according to the host organism and question at hand, we systematically compared the two main technical approaches for profiling microbial communities, 16S rRNA gene amplicon and metagenomic shotgun sequencing across our panel of ten host taxa. This includes two commonly used 16S rRNA gene regions and two amplification procedures, thus totaling five different microbial profiles per host sample. Conclusion While 16S rRNA gene-based analyses are subject to much skepticism, we demonstrate that many aspects of bacterial community characterization are consistent across methods. The resulting insight facilitates the selection of appropriate methods across a wide range of host taxa. Overall, we recommend single- over multi-step amplification procedures, and although exceptions and trade-offs exist, the V3 V4 over the V1 V2 region of the 16S rRNA gene. Finally, by contrasting taxonomic and functional profiles and performing phylogenetic analysis, we provide important and novel insight into broad evolutionary patterns among metaorganisms, whereby the transition of animals from an aquatic to a terrestrial habitat marks a major event in the evolution of host-associated microbial composition
