    International audienceMapReduce is a parallel programming paradigm successfully used to perform computations on massive amounts of data, being widely deployed on clusters, grid, and cloud infrastructures. Interestingly, while the emergence of cloud in- frastructures has opened new perspectives, several enterprises hesitate to put sensible data on the cloud and prefer to rely on internal resources. In this paper we introduce the PER- MARE initiative, which aims at proposing scalable techniques to support existent MapReduce data-intensive applications in the context of loosely coupled networks such as pervasive and desktop grids. By relying on the MapReduce programming model, PER-MARE proposes to explore the potential advan- tages of using free unused resources available at enterprises as pervasive grids, alone or in a hybrid environment.     Osteoarticular and musculoskeletal disorders, chronic pain, quality of life and physical activity level distributed by age and gender in elderly people participating in a water aerobics program

    The study aims to describe osteoarticular and musculoskeletal disorders, chronic pain, quality of life (QOL) and physical activity level (PAL) distributed by age group and gender in the elderly. The design was quantitative, descriptive and cross-sectional. The non-probabilistic sample, for convenience, comprised 101 elderly people, over 60 years old, who regularly participate in water aerobics activities in the municipality of Dois Irmãos, RS. The instruments used were: Multidimensional Pain Assessment Scale, Survey of Pain Attitudes, IPAQ and EUROHIS-QOL. Descriptive frequency analyzes were performed, which showed the predominance of women in water aerobics activities, most frequently in the age group of 70 years. Similarly, the presence of osteoarticular and musculoskeletal disorders and chronic pain intensity (moderate and severe) were more expressive in females and in the age group of 70 years. The distribution of QOL and pain intensity in relation to age, gender and PAL shows in the age group of 60 years, the male, irregularly active with higher average, with higher occurrence of pain (moderate) in the active elderly. Active females, on the other hand, show better QOL scores and higher pain intensity in irregularly active aspect. In conclusion, the actions promoted by the municipality have a positive impact on QOL. However, there is a need for greater involvement of participants to achieve more adequate levels of regular physical activity, taking into account the significant presence of pain complaints in females and in the age group of 70 years

    MAPREDUCE CHALLENGES ON PERVASIVE GRIDS

    International audienceThis study presents the advances on designing and implementing scalable techniques to support the development and execution of MapReduce application in pervasive distributed computing infrastructures, in the context of the PER-MARE project. A pervasive framework for MapReduce applications is very useful in practice, especially in those scientific, enterprises and educational centers which have many unused or underused computing resources, which can be fully exploited to solve relevant problems that demand large computing power, such as scientific computing applications, big data processing, etc. In this study, we pro-pose the study of multiple techniques to support volatility and heterogeneity on MapReduce, by applying two complementary approaches: Improving the Apache Hadoop middleware by including context-awareness and fault-tolerance features; and providing an alternative pervasive grid implementation, fully adapted to dynamic environments. The main design and implementation decisions for both alternatives are described and validated through experiments, demonstrating that our approaches provide high reliability when executing on pervasive environments. The analysis of the experiments also leads to several insights on the requirements and constraints from dynamic and volatile systems, reinforcing the importance of context-aware information and advanced fault-tolerance features to provide efficient and reliable MapReduce services on pervasive grids

    Evaluation of meiotic abnormalities of oocytes from polycystic ovary syndrome patients submitted to ovarian stimulation

    OBJETIVO: avaliar o fuso meiótico e a distribuição cromossômica de oócitos maturados in vitro, obtidos de ciclos estimulados de mulheres inférteis com síndrome dos ovários policísticos (SOP) e fatores masculino e/ou tubário de infertilidade (Grupo Controle) e comparar as taxas de maturação in vitro (MIV) entre os dois grupos avaliados. MÉTODOS: cinco pacientes inférteis com SOP e oito pacientes controles, submetidas à estimulação ovariana para injeção intracitoplasmática de espermatozóide, foram selecionadas prospectiva e consecutivamente, e constituíram os grupos de estudo e Controle, respectivamente. Oócitos imaturos captados após estimulação ovariana para a realização de injeção intracitoplasmática de espermatozóide (21 e 29, respectivamente, nos Grupos SOP e Controle) foram submetidos à MIV. Apenas os oócitos que apresentaram a extrusão do primeiro corpúsculo polar após a MIV foram fixados e submetidos à imunocoloração e análise por microscopia de fluorescência para avaliação morfológica do fuso e da distribuição cromossômica. A análise estatística foi realizada utilizando o teste exato de Fisher, com significância estatística quando p<0,05. RESULTADOS: as taxas de MIV foram similares entre os dois grupos (47,6 e 44,8%, respectivamente, nos Grupos SOP e Controle). Seis dos dez oócitos (60%) analisados do grupo de estudo e quatro dos 12 oócitos (33,3%) analisados do Grupo Controle apresentaram anomalias meióticas, caracterizadas por anomalias do fuso e/ou distribuição cromossômica oocitária, sem diferença significativa entre os grupos. CONCLUSÕES: os dados do presente estudo não demonstraram diferença significativa nas taxas de MIV e nas proporções de anomalias meióticas entre os oócitos maturados in vitro, provenientes de ciclos estimulados de pacientes com SOP, quando comparados aos controles.PURPOSE: to evaluate the meiotic spindle and the chromosome distribution of in vitro matured oocytes obtained from stimulated cycles of infertile women with polycystic ovary syndrome (PCOS) and with male factor and/or tubal infertility (Control Group) and compare in vitro maturation (IVM) rates between the groups analyzed. METHODS: five infertile patients with PCOS and eight controls, submitted to stimulated cycles for intracytoplasmic sperm injection, were selected prospectively and consecutively, and respectively assigned to the study group and the Control Group. Immature oocytes (21 and 29, respectively, from PCOS and Control Group) were submitted to IVM. After IVM, oocytes with first polar body extruded were fixed and submitted to immunostaining and fluorescence microscopy for morphological evaluation of the spindle and of chromosome distribution. Statistical analysis was performed by the Fisher test with significance, when p<0.05. RESULTS: IVM rates were similar between groups (47.6 e 44.8%, respectively, for PCOS and Control Group). Six of the ten oocytes (60%) from the study group and four of the 12 oocytes (33.3%) from the Control Group presented meiotic anomalies of the spindle and/or anomalous chromosome distribution, without statistical difference between groups. CONCLUSIONS: data from the present study did not demonstrate significant difference neither in IVM rates nor in the proportions of meiotic anomalies between in vitro matured oocytes obtained from stimulated cycles from PCOS patients and control ones

Despite the fact that there is no strong-powered evidence about any of the aspects related to natural fertility, literature on how to increase the chances of a spontaneous pregnancy is available.     Os medicamentos são considerados xenobióticos pelo organismo humano, isto é, moléculas estranhas. O fígado é o grande responsável pelo processo de metabolização, onde os compostos lipossolúveis são transformados em moléculas com características hidrossolúveis, por meio de reações químicas, com a finalidade de facilitar a excreção renal, principalmente. O paracetamol é um anti-inflamatório da classe não esteroidal (AINES), utilizado para tratamento de dor e febre e pode ser adquirido sem nenhuma prescrição médica, portanto e extremamente utilizado pela população. Mesmo sendo um medicamento de venda livre, este fármaco apresenta um potencial hepatotóxico, dependente de dose, podendo gerar lesão em hepatócitos. Assim, o modelo de lesão hepática induzida por paracetamol é um dos modelos mais populares para testar potenciais agentes hepatoprotetores. No entanto, existem muitas variações neste modelo e a falta de padronização impacta diretamente no tempo de experimento, quantidade de animais utilizados e elevados custos com insumos e fármacos. A presente revisão teve como objetivo reunir informações sobre a padronização do modelo de hepatotoxicidade induzidos por paracetamol, assim como os testes confirmatórios utilizados para garantir a obtenção de resultados que permitem a reprodutibilidade. Para isso foram consultadas as bases de buscada BVS, Pubmed e Scielo, entre os anos de 2015 a 2020. A pesquisa retornou com 370 artigos que, após a triagem foi reduzida a 114, que permitiram o levantamento de dados de extrema relevância para a padronização do modelo, incluindo as análises necessárias para confirmação do desenvolvimento da hepatotoxicidade

    Phosphoinositide 3-kinase gamma gene knockout impairs postischemic neovascularization and endothelial progenitor cell functions

    OBJECTIVE: We evaluated whether phosphatidylinositol 3-kinase γ (PI3Kγ) plays a role in reparative neovascularization and endothelial progenitor cell (EPC) function. METHODS AND RESULTS: Unilateral limb ischemia was induced in mice lacking the PI3Kγ gene (PI3Kγ(−/−)) or expressing a catalytically inactive mutant (PI3Kγ(KD/KD)) and wild-type controls (WT). Capillarization and arteriogenesis were reduced in PI3Kγ(−/−) ischemic muscles resulting in delayed reperfusion compared with WT, whereas reparative neovascularization was preserved in PI3Kγ(KD/KD). In PI3Kγ(−/−) muscles, endothelial cell proliferation was reduced, apoptosis was increased, and interstitial space was infiltrated with leukocytes but lacked cKit(+) progenitor cells that in WT muscles typically surrounded arterioles. PI3Kγ is constitutively expressed by WT EPCs, with expression levels being upregulated by hypoxia. PI3Kγ(−/−) EPCs showed a defect in proliferation, survival, integration into endothelial networks, and migration toward SDF-1. The dysfunctional phenotype was associated with nuclear constraining of FOXO1, reduced Akt and eNOS phosphorylation, and decreased nitric oxide (NO) production. Pretreatment with an NO donor corrected the migratory defect of PI3Kγ(−/−) EPCs. PI3Kγ(KD/KD) EPCs showed reduced Akt phosphorylation, but constitutive activation of eNOS and preserved proliferation, survival, and migration. CONCLUSIONS: We newly demonstrated that PI3Kγ modulates angiogenesis, arteriogenesis, and vasculogenesis by mechanisms independent from its kinase activity

    Expert Panel Curation of 113 Primary Mitochondrial Disease Genes for the Leigh Syndrome Spectrum

    OBJECTIVE: Primary mitochondrial diseases (PMDs) are heterogeneous disorders caused by inherited mitochondrial dysfunction. Classically defined neuropathologically as subacute necrotizing encephalomyelopathy, Leigh syndrome spectrum (LSS) is the most frequent manifestation of PMD in children, but may also present in adults. A major challenge for accurate diagnosis of LSS in the genomic medicine era is establishing gene-disease relationships (GDRs) for this syndrome with >100 monogenic causes across both nuclear and mitochondrial genomes. METHODS: The Clinical Genome Resource (ClinGen) Mitochondrial Disease Gene Curation Expert Panel (GCEP), comprising 40 international PMD experts, met monthly for 4 years to review GDRs for LSS. The GCEP standardized gene curation for LSS by refining the phenotypic definition, modifying the ClinGen Gene-Disease Clinical Validity Curation Framework to improve interpretation for LSS, and establishing a scoring rubric for LSS. RESULTS: The GDR with LSS across the nuclear and mitochondrial genomes was classified as definitive for 31/114 gene-disease relationships curated (27%); moderate for 38 (33%); limited for 43 (38%); and 2 as disputed (2%). Ninety genes were associated with autosomal recessive inheritance, 16 were maternally inherited, 5 autosomal dominant, and 3 X-linked. INTERPRETATION: GDRs for LSS were established for genes across both nuclear and mitochondrial genomes. Establishing these GDRs will allow accurate variant interpretation, expedite genetic diagnosis of LSS, and facilitate precision medicine, multi-system organ surveillance, recurrence risk counselling, reproductive choice, natural history studies and eligibility for interventional clinical trials.
