    Russian Migration and Psychological Aspect

    The problem of regional migration is treated in the article. The development of the Far Eastern territories is a priority now. The government has developed many large-scale projects aimed at supporting the remote region, but the problem of emigration of the economically active population has not settled since 1991. The problem of the outflow of promising young personnel from its territory is particularly acute. The purpose of this research is to study the reasons for the migration of young people from the Primorsky Region - one of the regions of the Far East. The main tasks were to obtain visions of migration moods and motivation of migration behavior of students. The study used a psychological verbal-communicative method - a survey in which a specially designed list of questions was used as a means to collect information from the respondent. The survey covered about 1,000 respondents between the ages of 15 and 30.  The research has shown that the main reason for emigration of school graduates is the opportunity to progress, studying in prestigious domestic and foreign universities, working in the largest Russian and foreign companies, that not enough represented here. Excellent and good rated school graduates prefer to enter Moscow and St. Petersburg universities. This means that most prospective workers in the future want to leave the territory of the region. Analysis of the responses of the university graduates revealed the prevalence of negative expectations about their future and the future of the territories. The data indicated that the main reasons to leave the country and the Primorsky Region are low job prospects for university graduates, lack of self-realization possibilities, low wages and the instability of the region’s political and socio-economic situation. In addition, the situation is exacerbated by the fact that respondents, for the most part, do not know about the projects that are being implemented in the region and do not see career prospects, which confirms the implementation of many strategic projects of the region only on paper. The lack of measures to prevent the emigration of young people will have a number of negative economic and social consequences, one of which is the reduction of competitevness of the territory. Creating favorable conditions for young people in the region, active public policy to attract young skilled professionals in Primorye is a prerequisite for the development of the region's intellectual capital and a guarantee of its dynamic socio-economic development.The problem of regional migration is treated in the article. The development of the Far Eastern territories is a priority now. La investigación ha demostrado que la razón principal de la emigración de los graduados de laescuela es la oportunidad de progresar, estudiando en prestigiosas universidades nacionales y extranjeras, trabajando en las mayores universidades rusas y extranjeras. empresas, que no están suficientemente representadas aquí.     Biological Impact of γ-Fe2O3 Magnetic Nanoparticles Obtained by Laser Target Evaporation: Focus on Magnetic Biosensor Applications

    The biological activity of γ-Fe2O3 magnetic nanoparticles (MNPs), obtained by the laser target evaporation technique, was studied, with a focus on their possible use in biosensor applications. The biological effect of the MNPs was investigated in vitro on the primary cultures of human dermal fibroblasts. The effects of the MNPs contained in culture medium or MNPs already uptaken by cells were evaluated for the cases of the fibroblast’s proliferation and secretion of cytokines and collagen. For the tests related to the contribution of the constant magnetic field to the biological activity of MNPs, a magnetic system for the creation of the external magnetic field (having no commercial analogues) was designed, calibrated, and used. It was adapted to the size of standard 24-well cell culture plates. At low concentrations of MNPs, uptake by fibroblasts had stimulated their proliferation. Extracellular MNPs stimulated the release of pro-inflammatory cytokines (Interleukin-6 (IL-6) and Interleukin-8 (IL-8) or chemokine (C-X-C motif) ligand 8 (CXCL8)) in a concentration-dependent manner. However, the presence of MNPs did not increase the collagen secretion. The exposure to the uniform constant magnetic field (H ≈ 630 or 320 Oe), oriented in the plane of the well, did not cause considerable changes in fibroblasts proliferation and secretion, regardless of presence of MNPs. Statistically significant differences were detected only in the levels of IL-8/CXCL8 release.The study was supported by the program of the Ministry of Health of the Russian Federation (project 121032300335-1).     Conventional Anti-glioblastoma Chemotherapy Affects Proteoglycan Composition of Brain Extracellular Matrix in Rat Experimental Model in vivo

    Temozolomide (TMZ) is a conventional chemotherapy drug for adjuvant treatment of glioblastoma multiforme (GBM), often accompanied by dexamethasone (DXM) to prevent brain oedema and alleviate clinical side effects. Here, we aimed to investigate an ability of the drugs to affect normal brain tissue in terms of proteoglycan (PG) composition/content in experimental rat model in vivo. Age- and brain zone-specific transcriptional patterns of PGs were demonstrated for 8, 60, and 120 days old rats, and syndecan-1, glypican-1, decorin, biglycan, and lumican were identified as the most expressed PGs. DXM treatment affected both PG core proteins expression (mainly syndecan-1, glypican-1, decorin, biglycan, lumican, versican, brevican, and NG2) and heparan sulphate (HS)/chondroitin sulphate (CS) content in organotypic brain slice culture ex vivo and experimental animals in vivo in a dose-dependent manner. TMZ treatment did not result in the significant changes in PG core proteins expression both in normal rat brain hippocampus and cortex in vivo (although generics did), but demonstrated significant effects onto polysaccharide HS/CS content in the brain tissue. The effects were age- and brain zone-specific and similar with the age-related PGs expression changes in rat brain. Combination of TMZ with DXM resulted in the most profound deterioration in PGs composition and content in the brain tissue both at core protein and glycosaminoglycan levels. Taken together, the obtained results demonstrate that conventional anti-glioblastoma therapy affects proteoglycan structure and composition in normal brain tissue, potentially resulting in deterioration of brain extracellular matrix and formation of the favourable tumorigenic niche for the expansion of the residual glioma cells. During the TMZ chemotherapy, dose and regimen of DXM treatment matter, and repetitive low DXM doses seem to be more sparing treatment compared with high DXM dose(s), which should be avoided where possible, especially in combination with TMZ

    Amorphous FeCoCrSiB Ribbons with Tailored Anisotropy for the Development of Magnetic Elements for High Frequency Applications

    The ferromagnetic resonance (FMR) in the frequency range of 0.5 to 12.5 GHz has been investigated as a function of external magnetic field for rapidly quenched Fe3Co67Cr3Si15B12 amorphous ribbons with different features of the effective magnetic anisotropy. Three states of the ribbons were considered: as-quenched without any treatment; after relaxation annealing without stress at the temperature of 350 °C during 1 h; and after annealing under specific stress of 230 MPa at the temperature of 350 °C during 1 h. For FMR measurements, we adapted a technique previously proposed and tested for the case of microwires. Here, amorphous ribbons were studied using the sample holder based on a commercial SMA connector. On the basis of the measurements of the reflection coefficient S11, the total impedance including its real and imaginary components was determined to be in the frequency range of 0.5 to 12.5 GHz. In order to confirm the validity of the proposed technique, FMR was also measured by the certified cavity perturbation technique using a commercial Bruker spectrometer operating at X-band frequency of 9.39 GHz. As part of the characterization of the ribbons used for microwave measurements, comparative analysis was performed of X-ray diffraction, optical microscopy, transmission electron microscopy, inductive magnetic hysteresis loops, vibrating sample magnetometry, magneto-optical Kerr effect (including magnetic domains) and magnetoimpedance data for of all samples.The research funding from the Ministry of Science and Higher Education of the Russian Federation (Ural Federal University Program of Development within the Priority-2030 Program) is gratefully acknowledged.     Donor-acceptor molecular oligogermanes: Novel properties and structural aspects

    The linear oligogermyl amide 2, Ph3GeGeMe2NMe2, was obtained by reacting Ph3GeLi with 1, Me2Ge(Cl)NMe2. The amide 2 was used for the synthesis of molecular oligogermanes 3, Ph3GeGeMe2Ge(C6F5)3, and 4, [Ph3GeGeMe2]2Ge(C6F5)2, containing electron donor (Me, Ph) and acceptor (C6F5) groups, by using a hydrogermolysis reaction in n-hexane. The molecular structures of 3 and 4 were studied by XRD. It was shown that in a crystal 3 forms wide channels, in which the solvated nonpolar n-hexane molecule is present. The NMR (1H, 13C and 19F), optical (UV/vis absorption, luminescence) and electrochemical (cyclic voltammetry) properties of both compounds were also studied. The impact of the substitution type by the electron withdrawing groups (at the terminal position, such as in 3, or within the compound, such as in 5), on the physical properties was also studied

    Diagnostic value of salivary cortisol in 1-mg dexamethasone suppression test

    BACKGROUND: Late-night salivary cortisol and serum cortisol measurements after 1-mg Dexamethasone Suppression Test (1-mg DST) are routinely used to diagnose Cushing’s syndrome (CS). Measuring morning salivary instead of serum cortisol after 1-mg DST would make the diagnostics of CS fully non-invasive. AIM: To evaluate the diagnostic accuracy of salivary cortisol in 1-mg DST as measured by electrochemiluminescence assay (ECLIA). MATERIALS AND METHODS: We combined a cohort diagnostic study, including 164 participants (132 females, 32 males) aged from 18 to 77 years: 110 were overweight or obese as increased BMI is the most common sign of Cushing’s Syndrome (CS), and 54 healthy volunteers. In each cohort late-night salivary cortisol was measured (at 23:00) followed by 1-mg DST and blood and salivary sampling for cortisol measurement the next morning at 08:00-09:00. Cortisol in saliva and serum were measured on automatic analyzer Cobas е 601 by F. Hoffmann-La Roche Ltd, using ECLIA. The final diagnosis was confirmed by the histological evaluation after surgery or using a follow-up observation in patients with obesity to exclude Cushing’s syndrome manifestation. RESULTS: Among 110 patients, 54 subjects were finally confirmed as having Cushing's syndrome. Reference interval for salivary cortisol after 1-mg DST was estimated to be 0,5–12,7 nmol/l (5–95 procentile). Maximal salivary cortisol level in 1-mg DST registered in healthy person was 29,6 mmol/l. Areas under the curve (AUC) were as following: for salivary cortisol in 1-mg DST – 0,838 (95% СI 0,772–0,905), for blood cortisol in 1-mg DST – 0,965 (95% CI 0,939–0,992) and for late-night salivary cortisol – 0,925 (95% CI 0,882–0,969). The optimal cut-off point for salivary cortisol after 1-mg DST was estimated as 12.1 nmol/l (sensitivity 60%, specificity 92,9%) among CS versus healthy subjects; 12,6 (sensitivity 58,2%, specificity 96,2%) among patients with obesity and CS; and – 12,2 nmol/l (sensitivity 60,7%, specificity 93,4%) among CS and both obese and healthy control subjects. Considering small difference between cut-off points, the recommended cut-off value for salivary cortisol after 1-mg DST is recommended to be 12,0 nmol/l if measured by ECLIA. CONCLUSION: Although salivary cortisol after 1-mg DST is inferior to serum cortisol after 1-mg DST in the diagnostic performance and diagnostic accuracy, it can be used as a low-invasive screening test with superior specificity

    Precise tracking of vaccine-responding T-cell clones reveals convergent and personalized response in identical twins

    T-cell receptor (TCR) repertoire data contain information about infections that could be used in disease diagnostics and vaccine development, but extracting that information remains a major challenge. Here we developed a statistical framework to detect TCR clone proliferation and contraction from longitudinal repertoire data. We applied this framework to data from three pairs of identical twins immunized with the yellow fever vaccine. We identified 500-1500 responding TCRs in each donor and validated them using three independent assays. While the responding TCRs were mostly private, albeit with higher overlap between twins, they could be well predicted using a classifier based on sequence similarity. Our method can also be applied to samples obtained post-infection, making it suitable for systematic discovery of new infection-specific TCRs in the clinic

    Plant Growth Promoting Activity and Metal Tolerance of Bacteria Isolated from Rhizosphere of the Orchid Epipactis atrorubens Growing on Serpentine Substrates of the Middle Urals

    В статье представлены данные, полученные при изучении бактерий, выделенных из ризосферы орхидеи Epipactis atrorubens (Hoffm.) Besser. Проведен сравнительный анализ некоторых морфологических, физиологических и биохимических характеристик ризобактерий растений, произрастающих в двух биотопах на серпентинитовых породах: в естественном лесном фитоценозе (фоновый участок) и на отвале после добычи асбеста (Свердловская область, Средний Урал). Оценка ростстимулирующей (PGP) активности выделенных штаммов не показала достоверных различий между исследованными участками по способности ризобактерий к синтезу индолил‑3-уксусной кислоты (ИУК) и солюбилизации фосфатов. Однако доля изолятов, способных к азотфиксации, была выше в ризосфере E. atrorubens, произрастающего на отвале, по сравнению с фоновым местообитанием. Устойчивость изолятов к тяжелым металлам оценивали по максимальной концентрации металла (400, 600 и 1000 мг/л соответственно для Ni, Cu и Zn), при которой отмечался рост бактерий. Показано, что ризобактерии с отвала оказались более устойчивыми к повышенным концентрациям металлов по сравнению с естественным лесным фитоценозом. На основе молекулярно-генетического анализа изолятов с наиболее выраженной PGP‑активностью (ИУК >1,0 мг/л; PO4 3- >50,0 мг/л) обнаружено сходство между изученными местообитаниями по родовой принадлежности ризобактерий E. atrorubens: выделенные штаммы принадлежали преимущественно к родам Buttiauxella и Pseudomonas. В модельных экспериментах протестирована ростстимулирующая способность четырех отобранных штаммов на семенах циннии. Инокуляция семян Pseudomonas sp. и Buttiauxella sp. не оказывала значимого влияния на их всхожесть, однако Buttiauxella sp. способствовала увеличению длины проростков в сравнении с контролем (в среднем на 25 %). Сделано предположение, что отобранные изоляты ризобактерий E. atrorubens, благодаря их ростстимулирующей активности и металлоустойчивости, могут способствовать натурализации орхидеи на техногенно нарушенной территорииThe article presents data obtained in the study of bacteria isolated from the rhizosphere of the orchid Epipactis atrorubens (Hoffm.) Besser. Analysis was carried out to compare some morphological, physiological, and biochemical characteristics of plant rhizobacteria growing on serpentine rocks in two biotopes: in the natural forest community (control habitat) and on the asbestos mine dump (the Sverdlovsk region, Middle Urals). An assessment of the plant growth promoting (PGP) activity of the isolated strains did not show significant differences in the ability of rhizobacteria to synthesize indol‑3-acetic acid (IAA) and solubilize phosphates between the study sites. However, the proportion of isolates capable of nitrogen fixation was higher in the rhizosphere of E. atrorubens growing on the dump compared to the control habitat. The tolerance of isolates to heavy metals was assessed by the maximum metal concentration (400, 600, and 1000 mg/L, respectively, for Ni, Cu, and Zn) at which bacterial growth was observed. Rhizobacteria from the dump were found to be more resistant to elevated concentrations of metals compared to their counterparts from the natural forest community. The molecular genetic analysis of isolates with the highest PGP‑activity (IAA >1.0 mg/L; PO4 3- >50.0 mg/L) revealed that most of the E. atrorubens rhizobacteria in both habitats belonged to the genera Buttiauxella and Pseudomonas. In model experiments, the PGP ability of four selected strains was tested on zinnia seeds. Seed inoculation with Pseudomonas sp. and Buttiauxella sp. did not have any significant effect on their germination; however, Buttiauxella sp. contributed to the increase in the length of seedlings compared with the control (by 25 %, on average).
