    Biofertilizantes com fosfato natural, enxofre e Acidithiobacillus em solo com baixo P disponível

    A produção de biofertilizantes é um processo prático e econômico que reduz o consumo de energia e a sua aplicação visa aumentar o fósforo (P) disponível no solo. Biofertilizantes fosfatados (B5, B10, B15, B20) produzidos em laboratório usando fosfato natural (FN) e enxofre em diferentes concentrações (5; 10; 15 e 20%), inoculado com Acidithiobacillus (S*) e enxofre (20%) sem Acidithiobacillus (S), com incubação por 30, 45 e 60 dias, foi utilizado em um experimento em vasos com solo da Zona da Mata de Pernambuco (Argissolo Amarelo), com baixo nível de P disponível, cultivado com jacatupé (Pachyrhizus erosus), por dois períodos consecutivos. Os tratamentos fosfatados foram: fosfato natural (FN); biofertilizantes (B5, B10, B15, B20), com Acidithiobacillus (FN+S*) e B20, sem Acidithiobacillus (FN+S); superfosfato triplo (ST); e um controle sem fósforo (P0). As plantas foram inoculadas com a mistura das estirpes de rizóbio NFB 747 e 748, e sem inoculação. Nos ensaios em laboratório analisou-se o pH e o P extraído dos biofertilizantes, em cada período de incubação, e nos experimentos em vasos com solo determinou-se a biomassa seca, o N e o P total acumulado da parte aérea, o pH e o P extraído do solo. O P disponível foi mais elevado para os biofertilizantes com Acidithiobacillus (FN+S*) e para o superfosfato triplo. A fertilização fosfatada, condicionou resposta positiva dos biofertilizantes (FN+S*) e do superfosfato triplo. Os rizóbios nativos foram tão eficientes quanto os do inoculante. Após os dois cultivos o P disponível foi maior com os biofertilizantes B15 e B20 com Acidithiobacillus.The production of mineral fertilizers is a expensive process, since it requires high energy consumption, and cannot be produced by small farmers. Laboratory assays were conducted to produce P-biofertilizers from natural phosphate (B5, B10, B15, B20), applying sulphur at different rates (5; 10; 15 and 20%) inoculated with Acidithiobacillus (S*) and testing increasing periods of incubation. A greenhouse experiment was carried out to evaluate the effect of the biofertilizers in a soil with low available P (Typic Fragiudult) from the "Zona da Mata" of Pernambuco State, grown with yam bean (Pachyrhizus erosus) in two consecutive harvests. The treatments were: Natural Phosphate (NP); biofertilizers produced in laboratory (B5, B10, B15, B20) with sulphur and Acidithiobacillus (NP+S*); natural phosphate with sulphur (20%) without Acidithiobacillus (NP+S); triple super phosphate (TSP) and a control without phosphorus. Plants were inoculated with a mixture of rhizobia strains (NFB 747 and NFB 748) or did not receive rhizobia inoculation. In bioassays pH and available P in the biofertilizers were analyzed. In the greenhouse experiment shoot dry matter, total N and total P in shoots, soil pH and available P were determined. Higher rates of available P were obtained in biofertilizers with sulphur and Acidithiobacillus (NP+S*) and in triple super phosphate (TSP), and biofertilizers with sulphur and Acidithiobacillus (FN+S*) and triple super phosphate (TSP) increased plant parameters. Native rhizobia were as effective as the strains applied in inoculation. After the two harvests soil presented lower pH values and higher rates of available P when the biofertilizers B15 and B20 with sulphur and Acidithiobacillus were applied

    Abstract Chagas disease is a neglected chronic condition with a high burden of morbidity and mortality. It has considerable psychological, social, and economic impacts. The disease represents a significant public health issue in Brazil, with different regional patterns. This document presents the evidence that resulted in the Brazilian Consensus on Chagas Disease. The objective was to review and standardize strategies for diagnosis, treatment, prevention, and control of Chagas disease in the country, based on the available scientific evidence. The consensus is based on the articulation and strategic contribution of renowned Brazilian experts with knowledge and experience on various aspects of the disease. It is the result of a close collaboration between the Brazilian Society of Tropical Medicine and the Ministry of Health. It is hoped that this document will strengthen the development of integrated actions against Chagas disease in the country, focusing on epidemiology, management, comprehensive care (including families and communities), communication, information, education, and research

    Diretriz da Sociedade Brasileira de Cardiologia sobre Diagnóstico e Tratamento de Pacientes com Cardiomiopatia da Doença de Chagas

    This guideline aimed to update the concepts and formulate the standards of conduct and scientific evidence that support them, regarding the diagnosis and treatment of the Cardiomyopathy of Chagas disease, with special emphasis on the rationality base that supported it.  Chagas disease in the 21st century maintains an epidemiological pattern of endemicity in 21 Latin American countries. Researchers and managers from endemic and non-endemic countries point to the need to adopt comprehensive public health policies to effectively control the interhuman transmission of T. cruzi infection, and to obtain an optimized level of care for already infected individuals, focusing on diagnostic and therapeutic opportunistic opportunities.   Pathogenic and pathophysiological mechanisms of the Cardiomyopathy of Chagas disease were revisited after in-depth updating and the notion that necrosis and fibrosis are stimulated by tissue parasitic persistence and adverse immune reaction, as fundamental mechanisms, assisted by autonomic and microvascular disorders, was well established. Some of them have recently formed potential targets of therapies.  The natural history of the acute and chronic phases was reviewed, with enhancement for oral transmission, indeterminate form and chronic syndromes. Recent meta-analyses of observational studies have estimated the risk of evolution from acute and indeterminate forms and mortality after chronic cardiomyopathy. Therapeutic approaches applicable to individuals with Indeterminate form of Chagas disease were specifically addressed. All methods to detect structural and/or functional alterations with various cardiac imaging techniques were also reviewed, with recommendations for use in various clinical scenarios. Mortality risk stratification based on the Rassi score, with recent studies of its application, was complemented by methods that detect myocardial fibrosis.  The current methodology for etiological diagnosis and the consequent implications of trypanonomic treatment deserved a comprehensive and in-depth approach. Also the treatment of patients at risk or with heart failure, arrhythmias and thromboembolic events, based on pharmacological and complementary resources, received special attention. Additional chapters supported the conducts applicable to several special contexts, including t. cruzi/HIV co-infection, risk during surgeries, in pregnant women, in the reactivation of infection after heart transplantation, and others.     Finally, two chapters of great social significance, addressing the structuring of specialized services to care for individuals with the Cardiomyopathy of Chagas disease, and reviewing the concepts of severe heart disease and its medical-labor implications completed this guideline.Esta diretriz teve como objetivo principal atualizar os conceitos e formular as normas de conduta e evidências científicas que as suportam, quanto ao diagnóstico e tratamento da CDC, com especial ênfase na base de racionalidade que a embasou. A DC no século XXI mantém padrão epidemiológico de endemicidade em 21 países da América Latina. Investigadores e gestores de países endêmicos e não endêmicos indigitam a necessidade de se adotarem políticas abrangentes, de saúde pública, para controle eficaz da transmissão inter-humanos da infecção pelo T. cruzi, e obter-se nível otimizado de atendimento aos indivíduos já infectados, com foco em oportunização diagnóstica e terapêutica. Mecanismos patogênicos e fisiopatológicos da CDC foram revisitados após atualização aprofundada e ficou bem consolidada a noção de que necrose e fibrose sejam estimuladas pela persistência parasitária tissular e reação imune adversa, como mecanismos fundamentais, coadjuvados por distúrbios autonômicos e microvasculares. Alguns deles recentemente constituíram alvos potenciais de terapêuticas. A história natural das fases aguda e crônica foi revista, com realce para a transmissão oral, a forma indeterminada e as síndromes crônicas. Metanálises recentes de estudos observacionais estimaram o risco de evolução a partir das formas aguda e indeterminada e de mortalidade após instalação da cardiomiopatia crônica. Condutas terapêuticas aplicáveis aos indivíduos com a FIDC foram abordadas especificamente. Todos os métodos para detectar alterações estruturais e/ou funcionais com variadas técnicas de imageamento cardíaco também foram revisados, com recomendações de uso nos vários cenários clínicos. Estratificação de risco de mortalidade fundamentada no escore de Rassi, com estudos recentes de sua aplicação, foi complementada por métodos que detectam fibrose miocárdica. A metodologia atual para diagnóstico etiológico e as consequentes implicações do tratamento tripanossomicida mereceram enfoque abrangente e aprofundado. Também o tratamento de pacientes em risco ou com insuficiência cardíaca, arritmias e eventos tromboembólicos, baseado em recursos farmacológicos e complementares, recebeu especial atenção. Capítulos suplementares subsidiaram as condutas aplicáveis a diversos contextos especiais, entre eles o da co-infecção por T. cruzi/HIV, risco durante cirurgias, em grávidas, na reativação da infecção após transplante cardíacos, e outros.    Por fim, dois capítulos de grande significado social, abordando a estruturação de serviços especializados para atendimento aos indivíduos com a CDC, e revisando os conceitos de cardiopatia grave e suas implicações médico-trabalhistas completaram esta diretriz.&nbsp

    Identification of genetic variants associated with Huntington's disease progression: a genome-wide association study

    Background Huntington's disease is caused by a CAG repeat expansion in the huntingtin gene, HTT. Age at onset has been used as a quantitative phenotype in genetic analysis looking for Huntington's disease modifiers, but is hard to define and not always available. Therefore, we aimed to generate a novel measure of disease progression and to identify genetic markers associated with this progression measure. Methods We generated a progression score on the basis of principal component analysis of prospectively acquired longitudinal changes in motor, cognitive, and imaging measures in the 218 indivduals in the TRACK-HD cohort of Huntington's disease gene mutation carriers (data collected 2008–11). We generated a parallel progression score using data from 1773 previously genotyped participants from the European Huntington's Disease Network REGISTRY study of Huntington's disease mutation carriers (data collected 2003–13). We did a genome-wide association analyses in terms of progression for 216 TRACK-HD participants and 1773 REGISTRY participants, then a meta-analysis of these results was undertaken. Findings Longitudinal motor, cognitive, and imaging scores were correlated with each other in TRACK-HD participants, justifying use of a single, cross-domain measure of disease progression in both studies. The TRACK-HD and REGISTRY progression measures were correlated with each other (r=0·674), and with age at onset (TRACK-HD, r=0·315; REGISTRY, r=0·234). The meta-analysis of progression in TRACK-HD and REGISTRY gave a genome-wide significant signal (p=1·12 × 10−10) on chromosome 5 spanning three genes: MSH3, DHFR, and MTRNR2L2. The genes in this locus were associated with progression in TRACK-HD (MSH3 p=2·94 × 10−8 DHFR p=8·37 × 10−7 MTRNR2L2 p=2·15 × 10−9) and to a lesser extent in REGISTRY (MSH3 p=9·36 × 10−4 DHFR p=8·45 × 10−4 MTRNR2L2 p=1·20 × 10−3). The lead single nucleotide polymorphism (SNP) in TRACK-HD (rs557874766) was genome-wide significant in the meta-analysis (p=1·58 × 10−8), and encodes an aminoacid change (Pro67Ala) in MSH3. In TRACK-HD, each copy of the minor allele at this SNP was associated with a 0·4 units per year (95% CI 0·16–0·66) reduction in the rate of change of the Unified Huntington's Disease Rating Scale (UHDRS) Total Motor Score, and a reduction of 0·12 units per year (95% CI 0·06–0·18) in the rate of change of UHDRS Total Functional Capacity score. These associations remained significant after adjusting for age of onset. Interpretation The multidomain progression measure in TRACK-HD was associated with a functional variant that was genome-wide significant in our meta-analysis. The association in only 216 participants implies that the progression measure is a sensitive reflection of disease burden, that the effect size at this locus is large, or both. Knockout of Msh3 reduces somatic expansion in Huntington's disease mouse models, suggesting this mechanism as an area for future therapeutic investigation

    Avaliaram-se consumo, desempenho, parâmetros plasmáticos e características de carcaça de 24 novilhos, 3/4 Simental 1/4 Nelore, com peso médio inicial de 370 kg. As dietas foram compostas por 55% de silagem de milho e diferentes fontes energéticas: milho (MI) e substituição parcial do milho pela casca de soja (CS) ou pelo farelo de gérmen de milho (FGM), tendo como fonte de proteína o farelo de girassol. O período de avaliação de consumo e ganho de peso foi de 49 dias. Foram realizadas amostragens de sangue para mensuração dos parâmetros plasmáticos: glicose, uréia, proteína total e albumina. As meia-carcaças direitas resfriadas foram utilizadas para medir a área de olho de lombo (AOL), a espessura de gordura (EG) e o comprimento de carcaça. As dietas não influenciaram os parâmetros plasmáticos. A média obtida para uréia plasmática foi elevada (26,1 mg/dL). As diferentes fontes energéticas não afetaram o ganho de peso e a conversão alimentar, com médias de 1,15 kg/dia e 9,17 kg de MS ingerida/kg de ganho. Não houve efeito sobre o rendimento de carcaça (52,8% peso final e 63,11% PCV), AOL (63,6 cm²) e EG (4,7 mm). O grão de milho pode ser substituído parcialmente pela casca de soja e pelo farelo de gérmen de milho, em dietas para novilhos em confinamento, sem afetar o desempenho e as características de carcaça, permitindo que a escolha entre esses ingredientes seja realizada pela análise econômica.Performance, plasmatic parameters and carcass traits of 24 3/4 Simmental 1/4 Nellore steers with 370 kg initial average weight was evaluated. The experimental diets were composed by corn silage (55%) and concentrate with sunflower meal as protein source and ground corn as energy source (MI). Ground corn was partially substituted by soybean hulls (CS) or by corn meal (FGM). The period of intake and average weight gain evaluation was 49 days. Blood samples were collected to measure plasmatic parameters: glucose, urea, total protein and albumin. Right half frozen carcass was used to measure the loin eye area (LEA), fat thickness (FT) and carcass length. Diets did not influence plasma parameters. The average of plasmatic urea was high (26 mg/dL), in relation to normal serum pattern. The different energy sources did not affect average weight gain and feed intake:gain ratio, with averages of 1.15 kg/day and 9.17 kg DM feed intake/kg gain. Diet did not influence dressing percentage (52.8% final weight and 63.1% of empty body weight), LEA (63.6 cm²) and FT (4.7 mm). Soybean hulls and corn meal could replace partially ground corn without affecting performance and carcass traits on steers, allowing that the ingredients choice be realized through economic analysis

    Avaliaram-se o consumo alimentar, a digestibilidade parcial e total e o balanço de nitrogênio, em novilhos confinados. Foram utilizados três novilhos de corte cruzados, canulados no rúmen e no duodeno, distribuídos em dois delineamentos em quadrado latino 3 x 3. As dietas experimentais foram constituídas de 60% de silagem de milho, tendo como fonte de proteína o farelo de girassol e, como fonte de energia, o milho (MI). O milho foi substituído parcialmente pela casca de soja (CS) ou pelo farelo de gérmen de milho (FGM). Quatro indicadores internos (lignina e FDA, FDN e lignina indigestíveis) foram submetidos a 144 horas de digestão in vitro, para estimativa da digestibilidade parcial e total das dietas. Houve maior ingestão dos componentes fibrosos na dieta CS, porém não foi observado efeito sobre o consumo de matéria seca. A lignina subestimou significativamente a digestibilidade. A utilização da FDAi, FDNi e lignina-i para estimar a digestibilidade total é viável, no entanto, a FDAi e lignina-i não estimaram adequadamente as digestibilidades parciais. Houve efeito significativo na digestibilidade ruminal da FDA, com valores mais elevados para CS e semelhantes para FGM, em relação à dieta MI. A digestibilidade total da FDA foi maior na dieta CS, porém, as digestibilidades dos demais componentes não foram afetadas pelas diferentes fontes energéticas. Não houve diferença significativa para a digestibilidade da energia e nos valores obtidos de NDT, com média de 61,5%. A casca de soja e o farelo de gérmen de milho, em substituição parcial do milho, mostraram-se fontes alternativas satisfatórias para a inclusão na dieta de bovinos.Feed intake, partial and total digestibility and nitrogen balance were evaluated in confined steers. Three crossbreed steers, rumen and duodenum cannulated, were allotted to two 3 x 3 Latin square experimental design. The experimental diets were composed by corn silage (60%) and concentrate with sunflower meal as protein source and ground corn as energy source (MI). The ground corn was partially substituted by soybean hulls (CS) or by corn germ meal (FGM). Lignin and indigestible ADF (iADF), NDF (iNDF) and lignin (i-lignin) determined by 144 h of in vitro digestion were used as markers to estimate the total and partial diet digestibility of diets. The fiber intake was higher on CS diet, however no effect on dry matter intake was observed. Lignin underestimated significantly the digestibility. Indigestible ADF, iNDF and i-lignin was able to estimate total digestibility, however iADF and i-lignina did not estimate adequately the partial digestibility. Ruminal ADF digestibility was influenced by diets with higher values for CS and similar values for FGM, in relation to MI. Total ADF digestibility was higher in diet CS, but the other nutrients digestibility were not affected by different energy sources. Energy digestibility and NDT values did not differ among diets, with average of 61.5%. Soybean hulls and corn germ meal, on partial substitution of ground corn, were satisfactory alternative sources for inclusion on steer diets
