    A avaliação do conhecimento gramatical 'oficial': estudo dos exames nacionais de Português de 2010

    Na última década e a nível nacional, as provas de aferição e os exames nacionais vêm sendo objecto de polémicas, vindas de distintos quadrantes e de que é exemplo o editorial do jornal Público de 22/6/2009: “Exames 2009: como se prepara o ‘sucesso’ fácil”. Em termos da avaliação educacional, se aceitarmos que “a avaliação é sinónimo de sucesso” (Pacheco & Zabalza, 1995: 7), tal significará que, ao estudar a avaliação externa, poderemos contribuir para uma compreensão sustentada da educação em línguas. Além disso, entendendo a avaliação do conhecimento gramatical “oficial” como um factor constitutivo das disciplinas de Português, será possível descrever como são representados e avaliados os conhecimentos explícitos sobre o ensino da língua materna. Neste quadro, pretende-se, neste estudo, aprofundar a análise do domínio da gramática em provas de avaliação externa, comparando as provas de aferição de Língua Portuguesa do 4º e do 6º anos de escolaridade e os exames do 9º e do 12º anos, todos realizados no ano lectivo de 2009/2010. Com esta finalidade, o trabalho desenvolver-se-á de acordo com as seguintes dimensões de análise: a) delimitação da estrutura das diferentes provas de avaliação em Português, de acordo com domínios correntes como a leitura, a escrita ou a gramática; b) identificação dos tipos de pergunta e dos conteúdos implicados na avaliação do saber gramatical; c) categorização das questões segundo três níveis - reconhecimento, produção e explicitação dos conhecimentos sobre a língua. Para além de compreender melhor o papel da regulação externa dos saberes relativos ao conhecimento explícito da língua, deste estudo retirar-se-ão conclusões acerca das novas formas de (re)configuração da gramática escolar

    Influence of albumin on the tribological behavior of Ag-Ti (C, N) thin films for orthopedic implants

    With the increase of elderly population and the health problems arising nowadays, such as cancer, knee and hip joint prostheses are widely used worldwide. It is estimated that 20% of hip replacement surgeries simply fail after 5 years, due to wear loosening, instability and infection. In this paper it is reported the study of advanced materials with the ability to overcome some of these drawbacks. The development of ceramic coatings, based on carbonitrides of transition metals, such as TiCN, doped with silver, Ag, may represent an effective solution. Thin films of Ag-TiCN were produced by dc reactive magnetron sputtering with silver contents ranging from 4 to 8 at. %. The physical, chemical, structural, morphological/topographical, mechanical and tribological properties were evaluated. The tribological tests were performed in a unidirectional wear simulator, pin on disc, being the antagonists of a ceramic Al2O3 ball, and using simulate body fluids as lubricant. Hank’s Balanced Salt Solution (HBSS) and bovine serum albumin (BSA) in HBSS were chosen, in order to evaluate the lubrication ability of the solution containing the protein, albumin. The results revealed that the coatings with Ag content ranging from 4 to 8 at. %, were the most promising, as the tribological properties were superior to the results reported by other authors, which also developed Ag-TiCN coatings containing similar Ag contents and using similar test conditions.     What did we learn after the 2020 pandemic? Seroprevalence of SARS-CoV-2 infection in a North region of Portugal during 3rd lockdown

    Background: Serological studies of antibody prevalence in response to infection with the acute respiratory syndrome coronavirus 2 (SARS-CoV-2) are useful to monitor the epidemic progression of the disease and to evaluate infection rates. In this study, we estimated the prevalence of anti-SARS-CoV-2 antibodies (IgM and IgG) in a Portuguese sub-district, during National lockdown - January-March 2021.Methods: In the seroepidemiological survey participants were tested for SARS-CoV-2 antibodies (IgM and IgG), in blood samples. The estimated seroprevalence and results were stratified by age, gender, education, occupational exposure, symptoms and previous SARS-CoV-2 infection.Results: Seroprevalence was 41.3%, (15.7% IgG positive, 11.6% IgM positive and 14% positive for both). The majority (74%) of the participants were working presentially and did the serological assay because they had either symptoms / positive contact (90%) or had a previous positive antigen test (36%). From all the seropositive cases only 44% were symptomatic. Our results show that seroprevalence of SARS -CoV-2 is high in the North, in parallel with the National Surveillance System. Seroprevalence was higher in woman than man and in adults older than 21. Almost all the people with symptoms or a positive contact had a positive test and were working at their regular places of work reflecting the danger of occupational exposure. Conclusions: These results suggest that, between January and March 2021, the restriction conditions were effective but unable to stop SARS-CoV-2 epidemic. It is essential to assess SARS-CoV-2 seroprevalence to monitor population immunity and if it lasts, specially from new COVID 19 variants

    Green processing of porous chitin structures for biomedical applications combining ionic liquids and supercritical fluid technology

    The application of green chemistry principles in the processing of materials for advanced technologies is a steadily increasing field of research. In this work porous chitin-based materials were developed by combining the processing of chitin using ionic liquids (ILs)as a green solvent together with the use of super- critical fluid technology(SCF) as clean technology.Chitin was dissolved in 1-butyl-3-imidazolium acetate,followed by regeneration of the polymer in ethanol in specific moulds.The IL was removed using Soxhlet extraction and successive steps of extraction with SCF using carbon dioxide/ethanol ratios of 50/ 50 and70/30.The developed porous chitin-based structures (ChIL)can be classified as mesoporous materials,with very low density and high porosity.The cytotoxicity of ChIL extracts was investigated using L929 fibroblast like cells,and the results demonstrated that the produced materials have extremely lowcytotoxicityl evels.Therefore,the findings suggest that the porous chitin structures may be potential candidates for a number of biomedical applications,including tissue engineering.Fundação para a Ciência e Tecnologia (FCT)S.S.S. and A.R.C.D. thank the Portuguese Foundation for Science and Technology (FCT) for post-doctoral fellowships (SFRH/BPD/ 45307/2008 and SFRH/BDP/34994/2007, respectively).     2,3-Diphosphoglycerate and the Protective Effect of Pyruvate Kinase Deficiency against Malaria Infection—Exploring the Role of the Red Blood Cell Membrane

    Malaria remains a major world public health problem, contributing to poverty and inequality. It is urgent to find new efficacious tools with few adverse effects. Malaria has selected red blood cell (RBC) alterations linked to resistance against infection, and understanding the protective mechanisms involved may be useful for developing host-directed tools to control Plasmodium infection. Pyruvate kinase deficiency has been associated with resistance to malaria. Pyruvate kinase-deficient RBCs display an increased concentration of 2,3-diphosphoglycerate (2,3-DPG).We recently showed that 2,3-DPG impacts in vitro intraerythrocytic parasite growth, induces a shift of the metabolic profile of infected cells (iRBCs), making it closer to that of noninfected ones (niRBCs), and decreases the number of parasite progenies that invade new RBCs. As an increase of 2,3-DPG content may also have an adverse effect on RBC membrane and, consequently, on the parasite invasion, in this study, we explored modifications of the RBC morphology, biomechanical properties, and RBC membrane on Plasmodium falciparum in vitro cultures treated with 2,3-DPG, using atomic force microscopy (AFM)-based force spectroscopy and other experimental approaches. The presence of infection by P. falciparum significantly increased the rigidity of parasitized cells and influenced the morphology of RBCs, as parasitized cells showed a decrease of the area-to-volume ratio. The extracellular addition of 2,3-DPG also slightly affected the stiffness of niRBCs, making it more similar to that of infected cells. It also changed the niRBC height, making the cells appear more elongated. Moreover, 2,3-DPG treatment influenced the cell surface charge, becoming more negative in treated RBCs than in untreated ones. The results indicate that treatment with 2,3-DPG has only a mild effect on RBCs in comparison with the effect of the presence of the parasite on the host cell. 2,3-DPG is an endogenous host metabolite, which may, in the future, originate a new antimalarial tool with few adverse effects on noninfected cells.publishersversionpublishe

    A forma como a ciência é ensinada nas escolas afecta profundamente a percepção que os estudantes têm do mundo que os rodeia e consequentemente a escolha de uma futura carreira nas ciências. Por outro lado, a abertura da escola à comunidade envolvente, proporciona as condições para a sua participação activa na vida escolar, assim como a promoção do sucesso escolar, através da formação integral dos alunos, dotando-os das competências científicas, tecnológicas e sócio-culturais necessárias a sólidas opções futuras. A química como uma ciência de cariz experimental oferece aos estudantes uma melhor compreensão dos conceitos teóricos leccionados. Este trabalho descreve actividades experimentais realizadas no âmbito de um intercâmbio entre a Escola Secundária/3 de Barcelinhos e o Departamento de Química. Os projectos desenvolvidos foram “Museu das Ciências” na disciplina de Área Projecto do 12º ano de escolaridade. A actividade A Ciência na investigação criminal foi baseada nas oficinas apresentadas no projecto Sentidos da Ciência [1]. Estas actividades experimentais executadas em colaboração entre as escolas e a universidade contribuem para uma maior motivação dos alunos, uma formação científica mais sólida e um conhecimento mais integrado

    Assessment of a Large-Scale Unbiased Malignant Pleural Effusion Proteomics Study of a Real-Life Cohort

Publisher Copyright: © 2022 by the authors.Background: Pleural effusion (PE) is common in advanced-stage lung cancer patients and is related to poor prognosis. Identification of cancer cells is the standard method for the diagnosis of a malignant PE (MPE). However, it only has moderate sensitivity. Thus, more sensitive diagnostic tools are urgently needed. Methods: The present study aimed to discover potential protein targets to distinguish malignant pleural effusion (MPE) from other non-malignant pathologies. We have collected PE from 97 patients to explore PE proteomes by applying state-of-the-art liquid chromatography-mass spectrometry (LC-MS) to identify potential biomarkers that correlate with immunohistochemistry assessment of tumor biopsy or with survival data. Functional analyses were performed to elucidate functional differences in PE proteins in malignant and benign samples. Results were integrated into a clinical risk prediction model to identify likely malignant cases. Sensitivity, specificity, and negative predictive value were calculated. Results: In total, 1689 individual proteins were identified by MS-based proteomics analysis of the 97 PE samples, of which 35 were diagnosed as malignant. A comparison between MPE and benign PE (BPE) identified 58 differential regulated proteins after correction of the p-values for multiple testing. Furthermore, functional analysis revealed an up-regulation of matrix intermediate filaments and cellular movement-related proteins. Additionally, gene ontology analysis identified the involvement of metabolic pathways such as glycolysis/gluconeogenesis, pyruvate metabolism and cysteine and methionine metabolism. Conclusion: This study demonstrated a partial least squares regression model with an area under the curve of 98 and an accuracy of 0.92 when evaluated on the holdout test data set.     Role of protein kinase R in the killing of Leishmania major by macrophages in response to neutrophil elastase and TLR4 via TNF and IFN

    In cutaneous leishmaniasis, Leishmania amazonensis activates macrophage double-stranded, RNA-activated protein kinase R (PKR) to promote parasite growth. In our study, Leishmania major grew normally in RAW cells, RAW-expressing dominant-negative PKR (PKR-DN) cells, and macrophages of PKR-knockout mice, revealing that PKR is dispensable for L. major growth in macrophages. PKR activation in infected macrophages with poly I:C resulted in parasite death. Fifty percent of L. major-knockout lines for the ecotin-like serine peptidase inhibitor (ISP2; Δisp2/isp3), an inhibitor of neutrophil elastase (NE), died in RAW cells or macrophages from 129Sv mice, as a result of PKR activation. Inhibition of PKR or NE or neutralization of Toll-like receptor 4 or 2(TLR4 or TLR2) prevented the death of Δisp2/isp3. Δisp2/isp3 grew normally in RAW-PKR-DN cells or macrophages from 129Sv pkr−/−, tlr2−/−, trif−/−, and myd88−/− mice, associating NE activity, PKR, and TLR responses with parasite death. Δisp2/isp3 increased the expression of mRNA for TNF-α by 2-fold and of interferon β (IFNβ) in a PKR-dependent manner. Antibodies to TNF-α reversed the 95% killing by Δisp2/isp3, whereas they grew normally in macrophages from IFN receptor–knockout mice. We propose that ISP2 prevents the activation of PKR via an NE-TLR4-TLR2 axis to control innate responses that contribute to the killing of L. major.—Faria, M. S., Calegari-Silva, T.     Glucose and lipid dysmetabolism in a rat model of prediabetes induced by a high-sucrose diet

    [Abstract] Glucotoxicity and lipotoxicity are key features of type 2 diabetes mellitus, but their molecular nature during the early stages of the disease remains to be elucidated. We aimed to characterize glucose and lipid metabolism in insulin-target organs (liver, skeletal muscle, and white adipose tissue) in a rat model treated with a high-sucrose (HSu) diet. Two groups of 16-week-old male Wistar rats underwent a 9-week protocol: HSu diet (n = 10)—received 35% of sucrose in drinking water; Control (n = 12)—received vehicle (water). Body weight, food, and beverage consumption were monitored and glucose, insulin, and lipid profiles were measured. Serum and liver triglyceride concentrations, as well as the expression of genes and proteins involved in lipid biosynthesis were assessed. The insulin-stimulated glucose uptake and isoproterenol-stimulated lipolysis were also measured in freshly isolated adipocytes. Even in the absence of obesity, this rat model already presented the main features of prediabetes, with fasting normoglycemia but reduced glucose tolerance, postprandial hyperglycemia, compensatory hyperinsulinemia, as well as decreased insulin sensitivity (resistance) and hypertriglyceridemia. In addition, impaired hepatic function, including altered gluconeogenic and lipogenic pathways, as well as increased expression of acetyl-coenzyme A carboxylase 1 and fatty acid synthase in the liver, were observed, suggesting that liver glucose and lipid dysmetabolism may play a major role at this stage of the disease.Portugal.
