Is the Application of Plant Probiotic Bacterial Consortia Always Beneficial for Plants? Exploring Synergies between Rhizobial and Non-Rhizobial Bacteria and Their Effects on Agro-Economically Valuable Crops
The overgrowth of human population and the demand for high-quality foods necessitate the search for sustainable alternatives to increase crop production. The use of biofertilizers, mostly based on plant probiotic bacteria (PPB), represents a reliable and eco-friendly solution. This heterogeneous group of bacteria possesses many features with positive effects on plants; however, how these bacteria with each other and with the environment when released into a field has still barely been studied. In this review, we focused on the diversity of root endophytic rhizobial and non-rhizobial bacteria existing within plant root tissues, and also on their potential applications as consortia exerting benefits for plants and the environment. We demonstrated the benefits of using bacterial inoculant consortia instead of single-strain inoculants. We then critically discussed several considerations that farmers, companies, governments, and the scientific community should take into account when a biofertilizer based on those PPBs is proposed, including (i) a proper taxonomic identification, (ii) the characterization of the beneficial features of PPB strains, and (iii) the ecological impacts on plants, environment, and plant/soil microbiomes. Overall, the success of a PPB consortium depends on many factors that must be considered and analyzed before its application as a biofertilizer in an agricultural system
The Symbiotic Performance of Chickpea Rhizobia Can Be Improved by Additional Copies of the clpB Chaperone Gene
The ClpB chaperone is known to be involved in bacterial stress response. Moreover, recent
studies suggest that this protein has also a role in the chickpea-rhizobia symbiosis. In order
to improve both stress tolerance and symbiotic performance of a chickpea microsymbiont,
the Mesorhizobium mediterraneum UPM-Ca36T strain was genetically transformed with
pPHU231 containing an extra-copy of the clpB gene. To investigate if the clpB-transformed
strain displays an improved stress tolerance, bacterial growth was evaluated under heat
and acid stress conditions. In addition, the effect of the extra-copies of the clpB gene in the
symbiotic performance was evaluated using plant growth assays (hydroponic and pot trials).
The clpB-transformed strain is more tolerant to heat shock than the strain transformed
with pPHU231, supporting the involvement of ClpB in rhizobia heat shock tolerance. Both
plant growth assays showed that ClpB has an important role in chickpea-rhizobia symbiosis.
The nodulation kinetics analysis showed a higher rate of nodule appearance with the
clpB-transformed strain. This strain also induced a greater number of nodules and, more
notably, its symbiotic effectiveness increased ~60%at pH5 and 83% at pH7, compared to
the wild-type strain. Furthermore, a higher frequency of root hair curling was also observed
in plants inoculated with the clpB-transformed strain, compared to the wild-type strain. The
superior root hair curling induction, nodulation ability and symbiotic effectiveness of the
clpB-transformed strain may be explained by an increased expression of symbiosis genes.
Indeed, higher transcript levels of the nodulation genes nodA and nodC (~3 folds) were
detected in the clpB-transformed strain. The improvement of rhizobia by addition of extracopies
of the clpB gene may be a promising strategy to obtain strains with enhanced stress
tolerance and symbiotic effectiveness, thus contributing to their success as crop inoculants,
particularly under environmental stresses. This is the first report on the successful improvement

Medição contextualista do processo emocional : construção e avaliação do Inventário do Processo Emocional
Tese de mestrado, Psicologia (Secção de Psicologia Clínica e da Saúde, Núcleo de Psicoterapia Cognitiva-Comportamental e Integrativa), Universidade de Lisboa, Faculdade de Psicologia, 2017O objectivo principal desta investigação consiste na construção de um instrumento - Inventário do Processo Emocional - que permita avaliar o processo emocional, unindo todas as suas componentes – emoções/categorias emocionais, valência, intensidade e estratégias de regulação emocional – e inserindo-o num determinado contexto. A sua originalidade advém do seu formato, por contextualizar as respostas e por juntar os componentes de um mesmo processo e não os avaliar isoladamente. Este pode ser um instrumento extremamente informativo, não sendo apenas obtidos resultados quantitativos, mas também fornecendo resultados qualitativos. Os resultados do IPE podem ainda ser utilizados como indicadores da qualidade do processo terapêutico, no sentido em que idealmente o desenvolvimento emocional, e consequentemente um processo emocional mais diferenciado e adaptativo, são promovidos ao longo do acompanhamento psicológico/psicoterapêutico. Os resultados desta investigação podem ser vistos como bons indicadores para uma primeira versão do instrumento e estimulam a continuação do trabalho de investigação tendo em vista o seu aperfeiçoamento.This investigation main goal is based on an instrument construction – Emotional Process Inventory - that allows to assess the emotional process, gathering all of its components - emotions/emotion categorizations, valency, intensity and emotion regulation strategies - and inserting it in a specific context. Its originality comes from the format, by the answer's contextualization and by joining the components of the same process avoiding to evaluate them separately. This can be an extremely informative instrument, obtaining not only quantitative results but also qualitative insights. The IPE results can also be used as a quality indicator for the therapeutic process, as the emotional development and, by consequence, a more differentiated and adaptive emotional process are promoted, during de psychological/psychotherapy process. The results of this research can be seen as good indicators for a first version of the instrument and encourage the pursuing of the research in view of its improvement
VARIAÇÃO DE NÚMERO DE CÓPIAS NO CROMOSSOMO 4: UMA POSSÍVEL SÍNDROME GENÉTICA?
INTRODUÇÃO: Variantes de número de cópias (CNVs) são variações no número de cópias de uma região da sequência genômica, descrevendo deleções ou ganhos em relação a indivíduos controle. Podem ser comuns e de caráter benigno, de significado incerto ou variantes patogênicas. Para interpretação, classificação e avaliação de significado clínico, é realizado comparação dos resultados nas bases de dados do laboratório e análise da literatura científica. OBJETIVO: Relatar caso de adolescente com duplicação/triplicação no cromossomo 4 com déficit cognitivo e dismorfismo facial e discutir se essa CNV pode ser responsável pelos achados clínicos. RELATO DE CASO: Pacientede 15 anos, sexo feminino, levada ao ambulatório de Genética para investigação de possível síndrome genética. Pais consanguíneos (primos). Desde a infância apresenta estrabismo divergente, atraso no desenvolvimento neuropsicomotor com dificuldade de fala. Cursou com síndrome hipotônica com espasmos mioclônicos. Evoluiu com déficit cognitivo. A Ressonância magnética de encéfalo demonstrou comprometimento de hemisférios cerebelares e atrofia de pontee mesencéfalo. Cariótipo normal (46, XX) e hibridização genômica comparativa baseada em microarranjos (a-CGH) revelou duplicação/ triplicação na região 4p 15.32p15.31, variante de significado incerto. CONCLUSÃO: Destaca-se a importância da investigação através de análises cromossômicas por microarranjos em pacientes com deficiência intelectual, síndrome do espectro do autismo e múltiplas malformações congênitas - isto para aprimoramento diagnóstico, cuidados médicos específicos e aconselhamento genético.
Biodegradação de microplásticos: otimização e aumento da escala
Marine litter, specifically small plastic particles, is one of the major
threats to the marine ecosystem. The presence of these
microplastics has been increasing over the last few years due to their
indiscriminate use and lack of polices for their management.
Thereby, it is necessary to encounter new ways to mitigate their
impacts or even reduce their presence. Biodegradations is a
promising solution to this problem.
In this work, with the objective of developing a bioremediation
process the potential of Zalerion maritimum, to biodegrade
polyethylene microplastics, is exploited.
Through optimization of the biodegradation medium, by Central
composite design and by Uniform design, was possible to obtain
higher percentages of microplastics removal and verify that malt
extract was the most relevant compost medium to the process. By
scale up it was possible to verify that in a less controlled medium the
biodegradation of microplastics still occur.
In addition, the response of the fungi Zalerion maritimum and Nia
vibrissa to exposure to poly(ethylene2,5-furandicarboxylate) has
also been studied and it was possible to conclude that these fungi
seems to have the capacity of biodegrade this micro(bio)plastic.O lixo marinho, em especial, os plásticos de pequenas dimensões,
é uma das maiores ameaças ao ecossistema marinho. A presença
destes microplásticos tem vindo a aumentar nos últimos tempos,
devido ao uso indiscriminado de plástico pela população humana e
à falta de políticas para a sua gestão.
Assim, é necessário encontrar maneiras de mitigar os seus impactos
ou mesmo reduzir a sua presença. A biodegradação surge como
uma solução promissora para este problema.
Neste trabalho, com o objetivo de desenvolver um processo de
biorremediação, o potencial de biodegradação, de microplásticos de
polietileno, do fungo Zalerion maritimum é explorado. Através da
otimização do meio de cultura, por “Central composite design” e por
“Uniform design”, foi possível obter maiores taxas de remoção de
microplásticos e verificar que o extrato de malte é o constituinte do
meio mais relevante neste processo. Pelo aumento da escala foi
possível verificar que mesmo num ambiente menos controlado o
fungo biodegrada microplásticos.
Além disso, foi também testada a resposta do fungo Zalerion
maritimum e do fungo Nia vibrissa, quando expostos ao poli(2,5-
furanodicarboxilato de etileno) e foi possível verificar que ambos os
fungos parecem ter a capacidade de biodegradar este

Preparação e caracterização de partículas de quitosano e seu derivado para libertação de fármacos
As terapias que existem hoje em dia para o tratamento do cancro não são totalmente eficazes pois provocam a destruição das células cancerígenas e das células saudáveis, levando a um défice no sistema imunitário do doente o que aumenta o risco de desenvolver infeções virais ou bacteriológicas. Para além disso esta doença acarreta elevados custos tanto para os doentes como para as instituições médicas devido à sua elevada incidência. Por isso, é necessário encontrar soluções e desenvolver novas técnicas mais eficazes para combater esta doença. Uma dessas técnicas consiste na libertação controlada de fármacos utilizando nanopartículas, sendo este o foco deste trabalho.
Foram produzidas partículas de quitosano a partir de quatro métodos diferentes; gelificação ionotrópica, água em óleo, método micelar inverso e precipitação. Foram utilizados diferentes tipos de polímero: quitosano com 470 kDa, 33 kDa e hidroxipropil quitosano. Para testar a eficácia destas partículas como veículos para libertação controlada de fármacos foi utilizada a doxorrubicina (DOX), um dos fármacos mais utilizados em quimioterapia que tem como principal consequência a cardiotoxicidade. O fármaco foi encapsulado nas partículas poliméricas produzidas pelos diferentes métodos e o seu perfil de libertação foi avaliado a pH 7,4 e a temperatura fisiológica. Para fazer a análise dos perfis de libertação procedeu-se à utilização de modelos matemáticos.
As principais conclusões deste trabalho foram que o método que permite obter partículas de menor dimensão é o de gelificação ionotrópica e que as partículas obtidas com CS despolimerizado apresentam uma maior homogeneidade de tamanhos. Observou-se que a libertação de DOX é mais eficiente para as partículas obtidas por gelificação ionotrópica. Verificou-se ainda que os modelos que melhor definem os perfis de libertação são o de Kormeyer-Peppas e o de Weibull e que a libertação de DOX deu-se maioritariamente por difusão de Fick, independentemente do polímero usado ou do método de produção das partículas
Role of QseG membrane protein in beneficial enterobacterial interactions with plants and Mesorhizobia
Homologs of qseG gene (coding for the membrane protein QseG), along with the qseEF genes, are present in many Enterobacteriaceae; however, its role in non-pathogenic strains is still unknown. To fill this knowledge gap, we investigated the role of QseG protein of a plant-associated enterobacterium in the interactions with its legume host and in the benefits induced by this enterobacterium in the Mesorhizobium–chickpea symbiosis. Here, we showed that QseG of Kosakonia sp. MH5 is involved in the following processes: (i) the evasion of the plant immune system and (ii) the efficient colonization of chickpea root cells. Furthermore, these features are essential for the beneficial effects of this strain on the Mesorhizobium–chickpea symbiosis. This study demonstrates that the role of QseG is transversal to pathogenic and non-pathogenic enterobacteria and is a step forward to better understanding the molecular bases of plant–bacteria interactions established between legume and beneficial endophytic enterobacteria.ME
