    New Method for Simultaneous Determination of Microcystins and Cylindrospermopsin in Vegetable Matrices by SPE-UPLC-MS/MS

    Cyanotoxins are a large group of noxious metabolites with different chemical structure and mechanisms of action, with a worldwide distribution, producing effects in animals, humans, and crop plants. When cyanotoxin-contaminated waters are used for the irrigation of edible vegetables, humans can be in contact with these toxins through the food chain. In this work, a method for the simultaneous detection of Microcystin-LR (MC-LR), Microcystin-RR (MC-RR), Microcystin-YR (MC-YR), and Cylindrospermopsin (CYN) in lettuce has been optimized and validated, using a dual solid phase extraction (SPE) system for toxin extraction and ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) for analysis. Results showed linear ranges (5–50 ng g−1 f.w.), low values for limit of detection (LOD) (0.06–0.42 ng g−1 f.w.), and limit of quantification (LOQ) (0.16–0.91 ng g−1 f.w.), acceptable recoveries (41–93%), and %RSDIP values for the four toxins. The method proved to be robust for the three variables tested. Finally, it was successfully applied to detect these cyanotoxins in edible vegetables exposed to cyanobacterial extracts under laboratory conditions, and it could be useful for monitoring these toxins in edible vegetables for better exposure estimation in terms of risk assessment.España MINECO AGL2015-64558-

    La amplia nómina de autores que firman el presente artículo dan forma al equipo multidisciplinar que en 1995 inició un ambicioso proyecto, subvencionado por la Junta de Castilla y León, denominado «Trabajos de Documentación y Estudio del Arte Rupestre en el Karst de Ojo Guareña». El avance de sus primeros trabajos está recogido en el siguiente texto y fue presentado al Primer Congreso de Arqueología Burgalesa (1998), mas, al no haberse publicado éste, sigue aún inédito. Su interés, y de forma especial la continuidad de la investigación, aconseja a los autores su edición en Espacio, Tiempo y Forma con vistas a una mayor difusión del proyecto y, con ello, a un mejor conocimiento de la grafía rupestre de las galerías subterráneas del norte de la provincia de Burgos.The wide number of researchers who sign the present añide joined in the multi-discipline team that started an ambitious project, which was sponsored by the regional authority: Junta de Castilla y León, called «Records and Research on Cave Art at the karst in Ojo Guareña».     Introduction: This article presents the research project entitled "Sunning in the river of life". Carried out by a group of women belonging to Matria Association of women for a culture also in feminine*, linked to the field of formal and non-formal education, with the aim of valuing the maintenance activities carried out by women in their daily life called "domestic".  Method: The methodology used has been narrative biographical, which as a branch of interpretive research, shares some general principles of qualitative research. Method that focuses on the experience of the subject whose story allows us to know her experience centered on a topic and in a specific time and context. Using standardized and in-depth interviews, systematic and participant observation, discussion groups with the participation of researchers, to obtain data.  Results: The results of the investigation have shown numerous variables related to the daily work of women and water, relevant to deepen the analysis of the history of the daily life of women and to value the so-called "domestic" jobs that They are guarantors of the well-being of the community.  Conclusions: This research opens the way to continue with the investigation of the history of the daily life of women in a specific time and context and to reflect on it from a critical and constructive perspective both in formal and non-formal and informal education with the goal of achieving a more just and egalitarian society and favor culture also in feminine.Introducción: En este artículo se presenta el proyecto de investigación titulado “Soleando en el río de la vida”. Llevado a cabo por un grupo de mujeres pertenecientes a Matria Asociación de mujeres por una cultura también en femenino*, vinculadas al ámbito de la educación formal y no formal, con el objetivo de poner en valor las actividades de mantenimiento realizadas por las mujeres en su vida cotidiana denominadas “domésticas“. Método: La metodología utilizada ha sido la biográfica narrativa, la cual como rama de la investigación interpretativa, comparte algunos principios generales de la investigación cualitativa. Método que se focaliza en la experiencia del sujeto cuyo relato permite conocer su experiencia centrada en un tema y en un tiempo y contexto geográfico concreto. Utilizando para la obtención de datos las entrevistas estandarizadas y en profundidad, la observación sistemática y participante y los grupos de discusión con la participación de las personas que componen el equipo de investigación  Resultados: Los resultados de la investigación han mostrado numerosas variables relacionadas con el trabajo cotidiano de las mujeres y el agua, relevantes para profundizar en el análisis de la historia de la vida cotidiana de la mujeres y poner el valor los trabajos denominados “domésticos” que son garantes del bienestar de la comunidad. Conclusiones: Esta investigación abre camino para continuar con la investigación de la historia de la vida cotidiana de la mujeres en un tiempo y un espacio concretos y reflexionar sobre ello desde una perspectiva critica y constructiva tanto en la educación formal como no formal con el objetivo de lograr un sociedad más justa e igualitaria y favorecer la cultura también en femenino.&nbsp

    Toll-like receptor agonists enhance HIV-specific T cell response mediated by plasmacytoid dendritic cells in diverse HIV-1 disease progression phenotypes

    [Background] Plasmacytoid dendritic cells (pDCs) sense viral and bacterial products through Toll-like receptor (TLR)-7 and -9 and translate this sensing into Interferon-α (IFN-α) production and T-cell activation. The understanding of the mechanisms involved in pDCs stimulation may contribute to HIV-cure immunotherapeutic strategies. The objective of the present study was to characterize the immunomodulatory effects of TLR agonist stimulations in several HIV-1 disease progression phenotypes and in non HIV-1 infected donors.[Methods] pDCs, CD4 and CD8 T-cells were isolated from 450 ml of whole blood from non HIV-1 infected donors, immune responders (IR), immune non responders (INR), viremic (VIR) and elite controller (EC) participants. pDCs were stimulated overnight with AT-2, CpG-A, CpG-C and GS-9620 or no stimuli. After that, pDCs were co-cultured with autologous CD4 or CD8 T-cells and with/without HIV-1 (Gag peptide pool) or SEB (Staphylococcal Enterotoxin B). Cytokine array, gene expression and deep immunophenotyping were assayed.[Findings] pDCs showed an increase of activation markers levels, interferon related genes, HIV-1 restriction factors and cytokines levels after TLR stimulation in the different HIV-disease progression phenotypes. This pDC activation was prominent with CpG-C and GS-9620 and induced an increase of HIV-specific T-cell response even in VIR and INR comparable with EC.     Penetrance of Dilated Cardiomyopathy in Genotype-Positive Relatives

    BACKGROUND Disease penetrance in genotype -positive (G+) relatives of families with dilated cardiomyopathy (DCM) and the characteristics associated with DCM onset in these individuals are unknown. OBJECTIVES This study sought to determine the penetrance of new DCM diagnosis in G+ relatives and to identify factors associated with DCM development. METHODS The authors evaluated 779 G+ patients (age 35.8 +/- 17.3 years; 459 [59%] females; 367 [47%] with variants in TTN ) without DCM followed at 25 Spanish centers. RESULTS After a median follow-up of 37.1 months (Q1 -Q3: 16.3-63.8 months), 85 individuals (10.9%) developed DCM (incidence rate of 2.9 per 100 person -years; 95% CI: 2.3-3.5 per 100 person -years). DCM penetrance and age at DCM onset was different according to underlying gene group (log -rank P = 0.015 and P <0.01, respectively). In a multivariable model excluding CMR parameters, independent predictors of DCM development were: older age (HR per 1 -year increase: 1.02; 95% CI: 1.0-1.04), an abnormal electrocardiogram (HR: 2.13; 95% CI: 1.38-3.29); presence of variants in motor sarcomeric genes (HR: 1.92; 95% CI: 1.05-3.50); lower left ventricular ejection fraction (HR per 1% increase: 0.86; 95% CI: 0.82-0.90) and larger left ventricular end -diastolic diameter (HR per 1 -mm increase: 1.10; 95% CI: 1.06-1.13). Multivariable analysis in individuals with cardiac magnetic resonance and late gadolinium enhancement assessment (n = 360, 45%) identi fied late gadolinium enhancement as an additional independent predictor of DCM development (HR: 2.52; 95% CI: 1.43-4.45). CONCLUSIONS Following a first negative screening, approximately 11% of G+ relatives developed DCM during a median follow-up of 3 years. Older age, an abnormal electrocardiogram, lower left ventricular ejection fraction, increased left ventricular end -diastolic diameter, motor sarcomeric genetic variants, and late gadolinium enhancement are associated with a higher risk of developing DCM.
