    Contribution of GABAergic Interneurons to the Development of Spontaneous Activity Patterns in Cultured Neocortical Networks

    Periodic synchronized events are a hallmark feature of developing neuronal networks and are assumed to be crucial for the maturation of the neuronal circuitry. In the developing neocortex, the early network oscillations coincide with an excitatory action of the neurotransmitter gamma-aminobutyric acid (GABA). A relationship between the emerging inhibitory action of GABA and the gradual disappearance of early synchronized network activity has been previously suggested. Therefore we investigate the interplay between the action of GABA and spontaneous activity in cultured networks of the lateral or dorsal embryonic rat neocortex, which show considerable difference in the content of GABAergic neurons. Here we present the results of long-term monitoring of spontaneous electrical activity of cultured networks growing on microelectrode arrays and the time course of changes in GABA action using calcium imaging. All cultures studied displayed stereotyped synchronized burst events at the end of the first week in vitro. As the GABAA depolarizing action decreases, naturally or after bumetanide treatment, network activity in lateral cortex cultures changed from stereotypic bursting to more clustered and asynchronous activity patterns. Dorsal cortex cultures and cultures lacking GABAA-receptor mediated synaptic transmission, retained an immature synchronous firing pattern, but developed prominent intraburst oscillations (∼3–10 Hz). Large, mostly parvalbumin positive, GABAergic neurons dominate the GABAergic population in lateral cortex cultures. These large interneurons were virtually absent in dorsal cortex cultures. Based on these results, we suggest that the richly interconnected large GABAergic neurons contribute to desynchronize and temporally differentiate the spontaneous activity of cultured cortical networks

    Chemical and microbiological stability of the low caliber apple slices prepared by osmotic dehydration

    Osmotic dehydration has received greater attention in recent years as an effective method for fruits and vegetables preservation. Process for preparation of Royal Gala apple slices using osmotic dehydration was standardized. The study was carried out with seleted apples of smaller caliber, produced in the Alcobaça region. For this study apples were seleted by their degree of maturation and absence of mechanical damages. Fully apple fruits were peeled and slices of 4 mm thickness were prepared. The apple slices were immersed in a sucrose solution (50% w/w) and in sucrose/sodium chloride solutions with NaCl concentrations (1% and 3%). After 4 h soaking, quick washing, blotting, the samples where drying at 20 °C up to constant weight. The dried products were packed in glass containers and stored at ambient conditions. The chemical and microbial and stability were monitored for 6 months. The osmo-dried apple slices prepared with 50% sucrose/1% sodium chloride showed an overall chemical acceptability (aw of 0.192; °Brix of 26.3; pH of 3.82; Na content of 0.51g/100g). Microbiologically the product was stable (aerobic mesophile osmotolerant or osmophilic bactéria counts < 2.75 log cfu g-1; fungi counts < 1.75 log cfu g-1) and safe (Clostridium spores and Staphylococcus were not confirmed in this study) up to 6 months of storage at ambient conditions.info:eu-repo/semantics/publishedVersio

    Brazilian version of the Nottingham Sensory Assessment: validity, agreement and reliability

    OBJETIVO: Verificar a concordância inter e intraexaminador, validade construtiva e consistência interna da versão brasileira do instrumento Nottingham Sensory Assessment para hemiparéticos após acidente vascular encefálico (AVE). MÉTODOS: O instrumento foi traduzido para língua portuguesa com base na sua versão original em Inglês por um tradutor bilíngue e, posteriormente, revertido para a língua inglesa. Vinte e um hemiparéticos foram avaliados por dois examinadores pela Avaliação Sensorial de Nottingham para pacientes pós-AVE (ASN) e pelo Protocolo de Desempenho Físico de Fugl-Meyer (FM). RESULTADOS: Foi encontrada correlação entre os instrumentos FM e ASN (0,752); excelente consistência interna da ASN (0,86); excelentes coeficientes de concordância interexaminador e intraexaminador para todos os itens da ASN, exceto temperatura e efeito teto significativo para ASN e FM. CONCLUSÃO: A versão brasileira da Nottingham Sensory Assessment cumpriu os critérios de concordância, consistência interna e validade concorrente, sendo um instrumento de rápida e fácil aplicação, podendo ser utilizada nos ambulatórios de neuroreabilitação para avaliar a função sensorial pós-AVE. O efeito teto significativo da ASN não limita seu uso, tendo em vista que, para os mesmos pacientes, o Protocolo de Fugl-Meyer também revelou efeito teto.OBJECTIVES: To investigate the inter-rater and intra-rater reliability, construct validity and internal consistency of the Brazilian version of the Nottingham Sensory Assessment for Stroke Patients (NSA). METHODS: The instrument was translated into Portuguese from its original in English by a bilingual translator and was then back-translated into English. Twenty-one hemiparetics were evaluated by two examiners using the NSA and the Fugl-Meyer Assessment (FMA) of physical performance. RESULTS: Significant correlation were found between the FMA and the NSA (r=0.752). The NSA showed excellent internal consistency (0.86), and there were acceptable inter- and intra-rater reliability for all items of the NSA, except temperature. Significant ceiling effects were found for the NSA and the FMA. CONCLUSIONS: The Brazilian version of the NSA met the criteria for agreement, internal consistency and concurrent validity. It was quick and easy to apply, and it could be used within clinical practice in neuro-rehabilitation outpatient clinics to assess sensory functions following stroke. The significant ceiling effect for the NSA did not limit its use, given that for the same patients, the FMA also showed ceiling effects

    In order to ensure the success of a technology transfer partnership for-profit or non-profit, it is necessary to understand which relational capabilities (RC) to explore. This article analyses which factors can assist in the development of the coordination dimension of RC, in cooperation with the development of non-profit and profit-oriented technologies, highlighting their similarities and differences. The method used was multi-case qualitative research, studying two groups (group N and group P), each with 5 entities. The main differences highlighted is the increase of social credibility for the group N and the knowledge dissemination of the partners and the processes improvement for the group P. Within this research, it was possible to deepen the model of Alves (2015), regarding the inclusion of factors that can help the development of RC Management dimensions.     Identifying the barriers in the access to health care to breast cancer perceived by women undergoing chemotherapy.Method: An exploratory descriptive study. The sample consisted of 58 women with breast cancer receiving chemotherapy and registered in the public oncology ambulatory of Aracaju-Sergipe. Data collection was carried out between October 2011 and March 2012 by semistructured interviews, and data were processed using the SPSS, version 17. Results: Among the interviewed women, 37 (63.8%) reported at least a barrier in the trajectory of care for breast cancer. The organizational and health services barriers were the most reported in the periods of investigation and treatment of breast cancer.Conclusion: In face of these findings, the barriers should be considered in public health policies and programs for the control of breast cancer in Sergipe.
Identificar las barreras en el acceso a la atención en salud percibidos por mujeres con cáncer de mama en tratamiento quimioterapéutico.Método: Estudio descriptivo- exploratorio, cuya muestra estuvo constituida por 58 mujeres con cáncer de mama en quimioterapia inscritas en el servicio ambulatorio público de oncología de Aracaju-Sergipe. La recolección de datos fue realizada entre octubre de 2011 a marzo de 2012, por medio de entrevistas semi-estructuradas. Los datos fueron procesados en el software SPSS, versión 17.Resultados: Entre las entrevistadas, 37 mu­jeres (63,8%) relataron haber enfrentado por lo menos una barrera en la trayectoria de cuidado del cáncer de mama. Las barreras organizacionales o de los servicios de salud fueron las más citadas en los periodos de investigación y tratamiento del cáncer de mama.Conclusión: Las barreras deben ser consideradas en las políticas públicas de salud y de los programas de control del cáncer de mama en Sergipe.

    Direct Discharges of Domestic Wastewater are a Major Source of Phosphorus and Nitrogen to the Mediterranean Sea

    Direct discharges of treated and untreated wastewater are important sources of nutrients to coastal marine ecosystems and contribute to their eutrophication. Here, we estimate the spatially distributed annual inputs of phosphorus (P) and nitrogen (N) associated with direct domestic wastewater discharges from coastal cities to the Mediterranean Sea (MS). According to our best estimates, in 2003 these inputs amounted to 0.9 × 10⁹ mol P yr-1 and 15 × 10⁹ mol N yr-1, that is, values on the same order of magnitude as riverine inputs of P and N to the MS. By 2050, in the absence of any mitigation, population growth plus higher per capita protein intake and increased connectivity to the sewer system are projected to increase P inputs to the MS via direct wastewater discharges by 254, 163, and 32% for South, East, and North Mediterranean countries, respectively. Complete conversion to tertiary wastewater treatment would reduce the 2050 inputs to below their 2003 levels, but at an estimated additional cost of over €2 billion yr-1. Management of coastal eutrophication may be best achieved by targeting tertiary treatment upgrades to the most affected near-shore areas, while simultaneously implementing legislation limiting P in detergents and increasing wastewater reuse across the entire basin

    Aspectos epidemiológicos e ecológicos relacionados à malária na área de influência do lago da Represa de Porto Primavera, região oeste do Estado de São Paulo, Brasil

    A study was carried out in the area of influence of the Porto Primavera Hydroelectric Power Station, in western São Paulo State, to investigate ecological and epidemiological aspects of malaria in the area and monitor the profile of the anopheline populations following the environmental changes brought about by the construction of the lake. Mosquitoes captured were analyzed by standardized indicator species analysis (ISA) before and during different flooding phases (253 m and 257 m elevations). The local human population was studied by means of parasitological (thin/thick blood smears), molecular (PCR) and serological tests. Serological tests consisted of Enzyme Linked Immunosorbent Assay (ELISA) with synthetic peptides of the circumsporozoite protein (CSP) from classic Plasmodium vivax, P. vivax variants (VK247 and "vivax-like"), P. malariae and P. falciparum and Indirect Immunofluorescence Assay (IFA) with asexual forms of P. vivax, P. malariae and P. falciparum. The results of the entomological survey indicated that, although the Anopheles darlingi population increased after the flooding, the population density remained very low. No malaria, parasite infection or DNA was detected in the inhabitants of the study area. However, there was a low frequency of antibodies against asexual forms and a significant prevalence of antibodies against P. vivax, P. vivax variants, P. falciparum and P. malariae; the presence of these antibodies may result from recent or less recent contact with human or simian Plasmodium (a parallel study in the same area revealed the existence of a sylvatic cycle). Nevertheless, these results suggest that, as in other places where malaria is present and potential vectors circulate, the local epidemiological conditions observed could potentially support the transmission of malaria in Porto Primavera Lake if infected individuals are introduced in sufficient numbers. Further studies are required to elucidate the phenomena described in this paper.Foi realizada pesquisa na área de influência do lago da Usina Hidrelétrica de Porto Primavera, região oeste do Estado de São Paulo, para estudar aspectos ecológicos e epidemiológicos da malária na localidade e acompanhar o perfil das populações de anofelinos frente às mudanças decorrentes do impacto ambiental pela formação do lago. Mosquitos capturados foram analisados pelo Índice de Abundância de Espécies Padronizado (IAEP), antes e durante o enchimento do reservatório (cotas 253 e 257 m). A população humana local foi estudada por meio de teste parasitológico (gota espessa e esfregaço sangüíneo), testes moleculares (PCR) e testes sorológicos. A sorologia consistiu na reação de ELISA com peptídeos sintéticos correspondentes à porção repetitiva da proteína circumsporozoíta (CSP) de Plasmodium vivax clássico, e suas variantes VK247 e "vivax-like", P. malariae e P. falciparum; e reação de Imunofluorescência Indireta (RIFI) com formas assexuadas de P. vivax, P. malariae e P. falciparum. Os resultados do estudo entomológico indicaram que, embora a população de Anopheles darlingi tenha aumentando após o enchimento, permaneceu em baixa densidade. Não foi detectada malária nem a presença de parasitos ou de DNA parasitário nos habitantes estudados. No entanto, foi observada baixa freqüência de anticorpos contra formas assexuadas e significativa prevalência de anticorpos contra esporozoítos de P. vivax e suas variantes, P. falciparum e P. malariae, que poderiam decorrer de contatos prévios, recentes ou não, com plasmódios humanos ou símios (o ciclo silvestre foi evidenciado em estudo paralelo realizado na mesma área). Por outro lado, estes resultados sugerem que, como em outros lugares onde existem vetores potenciais da malária, as condições epidemiológicas poderiam potencialmente permitir a transmissão da malária na área de influência do lago de Porto Primavera, se indivíduos infectados fossem introduzidos em número suficiente. Estudos adicionais deverão ser realizados para elucidar os fenômenos relatados neste artigo

    INFLUÊNCIA DE CARACTERÍSTICAS QUALITATIVAS NOS INDICADORES CONTÁBEIS DE INSTITUIÇÕES MICROFINANCEIRAS DA AMÉRCA LATINA E CARIBE

    A motivação principal para realização do presente estudo foi avaliar a existência de influência de características qualitativas de tipo de instituição, local de atuação e tamanho do ativo nas informações contábeis das IMFs na América Latina e Caribe. Os estudos sobre microfinanças se baseiam nas teorias da intermediação financeira, na teoria do Welfare State e na teoria institucional. Esta pesquisa se situa no campo das análises acerca dos indicadores contábeis de instituições financeiras. As demonstrações contábeis de uma IMF se assemelham às de qualquer instituição financeira, no entanto, há algumas especificidades neste tipo de organização, sobretudo, no que se refere ao tipo, bastante diversificado. Para validação da hipótese metodológica de que os indicadores contábeis de IMFs da América Latina e Caribe são influenciados pelas suas características qualitativas, fez-se um estudo empírico-analítico, utilizando-se das ferramentas estatísticas Análise de Cluster e Análise de Correspondências – ANACOR. Os dados foram coletados no site da Microfinance Information Exchenge–MIX, a qual disponibiliza dados secundários com características qualitativas (tipo, região) e quantitativa (indicadores contábeis). De acordo com os resultados, embora em magnitudes diferentes, todas as características qualitativas tiveram influências significativas nos Clusters formados a partir da similaridade dos indicadores contábeis. Desse modo, a hipótese metodológica foi confirmada. Os achados da pesquisa sinalizam que estudos futuros bem como ações e/ou políticas voltadas para Instituições Microfinanceiras que utilizem indicadores contábeis devem observar essas características

    Osteopenia and osteoporosis in people living with HIV: multiprofessional approach

    Increasing bone mineralization abnormalities observed among people living with HIV (PLWHIV) result from various factors relating to the host, the virus, and the antiretrovirals used. Today, HIV infection is considered to be a risk factor for bone mineralization disorders. The test most recommended for diagnosing osteoporosis is measurement of bone mineral density by means of dual energy X-ray absorptiometry at two sites. Osteoporosis treatment has the aims of bone mass improvement and fracture control. A combination of calcium and vitamin D supplementation may reduce the risk of fractures. Antiresorptive drugs act by blocking osteoclastic activity and reducing bone remodeling. On the other hand, bone-forming drugs stimulate osteoblastogenesis, thereby stimulating the formation of bone matrix. Mixed-action medications are those that are capable of both stimulating bone formation and inhibiting reabsorption. Antiresorptive drugs form the group of medications with the greatest quantity of scientific evidence confirming their efficacy in osteoporosis treatment. Physical activity is a health promotion strategy for the general population, but only preliminary data on its real value and benefit among PLWHIV are available, especially in relation to osteoporosis
