    Image analysis as a tool for viability and recombinant protein production assessment during E. coli fermentations

    The development of monitoring methods for physiological state assessment during recombinant fermentation processes has been encouraged by the need to evaluate the influence of processing conditions in recombinant protein production. In this work, microscopy and image analysis techniques were used for the quantification of viability and protein production in two recombinant E. coli batch fermentations. Images obtained from light microscopy with phase contrast were used to assess the total number of cells in a given sample and, from epifluorescence microscopy, both producing and dead cells were counted using two different filters. This methodology allowed the extraction of information related to cell viability and recombinant protein production. This information, combined with standard fermentation data, allowed the derivation of interesting hypothesis that can be used afterwards for experimental design and further validation. Additionally, the ratios calculated in this work can be complemented with other parameters that can be extracted from image analysis

Belém, PA, Brasil.Macacos-da-noite são muito susceptíveis ao estresse e por isso a contenção química ou física deve ser cuidadosamente avaliada antes de qualquer procedimento. Protocolos anestésicos podem alterar alguns parâmetros fisiológicos, sendo o eletrocardiograma (ECG) um exame muito utilizado para avaliação do ritmo e da frequência cardíaca. O objetivo deste estudo foi avaliar a influência de quatro diferentes protocolos de contenção sobre o ECG realizado em Aotus azarae infulatus. Para isso foram utilizados 10 animais, machos, adultos, submetidos à contenção com a associação tiletamina/zolazepam (TZ), isoflurano (ISO), associação cetamina e midazolam (CET) e contenção física (CF). Não foram observadas diferenças nos parâmetros de ondas e complexos obtidos no ECG em todos os grupos testados, no entanto, durante a avaliação do traçado os animais do grupo CF apresentaram uma quantidade maior de alterações. Concluiu-se que os protocolos de contenção utilizados não alteraram os valores do ECG e que não foi possível considerar o grupo CF como controle devido causar mais alterações do que todos os protocolos testados

    Constructal design method dealing with stiffened plates and symmetry boundaries

    A new computational procedure for modelling the structural behavior of stiffened plates with symmetry boundary conditions is here presented. It uses two-dimensional finite elements as a way to decrease computational time without losing precision thanks to a relatively small number of elements applied for analyzing out-of-plane displacements (deflections) and stresses. Adding, the constructal design method was included in the procedure, together with the exhaustive search technique, with the scope to optimize the stress/strain status of stiffened plates by design changes. For the purpose, a reference plate without stiffeners was initially design and used as starting point. Part of the volume was reshaped into stiffeners: thickness was reduced maintaining unchanged weight, length and width. The main goal was to minimize strains and stresses by geometric changes. Results demonstrated that, thanks to this design procedure, it is always possible to find an adequate geometry transformation from reference plate into stiffeners, allowing significant improvements in mechanical behavior

    Anastrepha bistrigata Bezzi, 1919 and Anastrepha striata Schiner, 1868 (Diptera: Tephritidae) infest guava (Psidium guajava L.), but only A. striata is considered a serious pest in all countries where it occurs. Both species also exploit several other host plants. They are frequently collected in fruit fly surveys in Brazil, but it is unusual to collect them in the same locality. For this reason, we report the co-occurrence of A. bistrigata and A. striata in Barreiras, Bahia, Brazil, which is also the first record of both species in this state. Information on morphological identification, distribution in Brazil, hosts, and sympatric occurrences of these species is also provided

    Seascape genetics of the Atlantic Spotted Dolphin (Stenella frontalis) based on mitochondrial DNA

    The Atlantic spotted dolphin (Stenella frontalis) is endemic to tropical, subtropical, and warm temperate waters of the Atlantic Ocean. Throughout its distribution, both geographic distance and environmental variation may contribute to population structure of the species. In this study, we follow a seascape genetics approach to investigate population differentiation of Atlantic spotted dolphins based on a large worldwide dataset and the relationship with marine environmental variables. The results revealed that the Atlantic spotted dolphin exhibits population genetic structure across its distribution based on mitochondrial DNA control region (mtDNA-CR) data. Analyses based on the contemporary landscape suggested, at both the individual and population level, that the population genetic structure is consistent with the isolation-by-distance model. However, because geography and environmental matrices were correlated, and because in some, but not all analyses, we found a significant effect for the environment, we cannot rule out the addition contribution of environmental factors in structuring genetic variation. Future analyses based on nuclear data are needed to evaluate whether local processes, such as social structure and some level of philopatry within populations, may be contributing to the associations among genetic structure, geographic, and environmental distance.info:eu-repo/semantics/acceptedVersio

    Efeitos de variáveis psicológicas, morfológicas e sociodemográficas sobre o comportamento alimentar de adolescentes

    -OBJETIVO: Averiguar a associação da insatisfação corporal (IC), do grau de comprometimento psicológico ao exercício (GCPE), do nível habitual de atividade física (NAF), do índice de massa corpórea (IMC), do percentual de gordura (%G) e da etnia com o comportamento alimentar inadequado (CAI) de adolescentes. MÉTODOS: Investigação transversal, da qual participaram 362 jovens de ambos os sexos, com idades entre dez e 19 anos, selecionados por amostragem estratificada. O Eating Attitudes Test (EAT-26) foi utilizado para avaliar o CAI. Ademais, utilizou-se o Body Shape Questionnaire, a Commitment Exercise Scale e o International Physical Activity Questionnaire para avaliar IC, GCPE e NAF, respectivamente. Conduziu-se análise multivariada e regressão múltipla para se analisarem os dados. RESULTADOS: Os resultados evidenciaram que a IC, o GCPE, o IMC e o %G associaram-se significativamente (p<0,05) com os escores das subescalas do EAT-26, tanto no sexo feminino, quanto no masculino. Os valores indicativos dessas associações foram diferentes entre os sexos. CONCLUSÕES: a insatisfação corporal e o IMC parecem ser os fatores mais fortemente associados aos distintos construtos do comportamento alimentar em ambos os sexos
