    The effects of rotation and positional change of stump tissues upon morphogenesis of the regenerating axolotl limb,

    Rotation of a skin cuff 180[deg] around the proximodistal axis of the upper arm in the axolotl results in the formation of multiple regenerates in about 80[deg] of cases after amputation of the limb through the rotated skin. Rotation of the dermis or the flexor and extensor muscles folowed by amputation produced similar percentages of multiple regenerates. Rotated bone produced no abnormalities, and rotated stump epidermis was minimally effective in stimulating multiple regeneration. A thin strip of normally oriented skin interposed between a rotated skin cuff and the amputation surface blocks the morphogenetic effect of the rotated stump skin whereas removal of the normal skin between a rotated proximal skin cuff and the amputation surface allows the formation of a low percentage of multiple regenerates. Gross rotation of stump tissue components can be broken down into axial rotation per se and positional dislocation. Experiments conducted upon skin and muscle have shown that positional dislocation along the anteroposterior axis rather than axial rotation is the manipulation that leads to the formation of multiple regenerates. The first morphological indication of multiple regeneration is the appearance of a triaxial apical ridge on the blastema.     Histological variability in the limb bones of the Asiatic wild ass and its significance for life history inferences

    The study of bone growth marks (BGMs) and other histological traits of bone tissue provides insights into the life history of present and past organisms. Important life history traits like longevity or age at maturity, which could be inferred from the analysis of these features, form the basis for estimations of demographic parameters that are essential in ecological and evolutionary studies of vertebrates. Here, we study the intraskeletal histological variability in an ontogenetic series of Asiatic wild ass (Equus hemionus) in order to assess the suitability of several skeletal elements to reconstruct the life history strategy of the species. Bone tissue types, vascular canal orientation and BGMs have been analyzed in 35 cross-sections of femur, tibia and metapodial bones of 9 individuals of different sexes, ages and habitats. Our results show that the number of BGMs recorded by the different limb bones varies within the same specimen. Our study supports that the femur is the most reliable bone for skeletochronology, as already suggested. Our findings also challenge traditional beliefs with regard to the meaning of deposition of the external fundamental system (EFS). In the Asiatic wild ass, this bone tissue is deposited some time after skeletal maturity and, in the case of the femora, coinciding with the reproductive maturity of the species. The results obtained from this research are not only relevant for future studies in fossil Equus, but could also contribute to improve the conservation strategies of threatened equid species

    Fin Spine Bone Resorption in Atlantic Bluefin Tuna, Thunnus thynnus, and Comparison between Wild and Captive-Reared Specimens

    Bone resorption in the first spine of the first dorsal fin of Atlantic bluefin tuna (ABFT) has long been considered for age estimation studies. In the present paper spine bone resorpion was assessed in wild (aged 1 to 13 years) and captive-reared (aged 2 to 11 years) ABFT sampled from the Mediterranean Sea. Total surface (TS), solid surface (SS) and reabsorbed surface (RS) were measured in spine transverse sections in order to obtain proportions of SS and RS. The spine section surface was found to be isometrically correlated to the fish fork length by a power equation. The fraction of solid spine bone progressively decreased according to a logarithmic equation correlating SS/TS to both fish size and age. The values ranged from 57% in the smallest examined individuals to 37% in the largest specimens. This phenomenon was further enhanced in captive-reared ABFT where SS/TS was 22% in the largest measured specimen. The difference between the fraction of SS of wild and captive-reared ABFT was highly significant. In each year class from 1- to 7-year-old wild specimens, the fraction of spine reabsorbed surface was significantly higher in specimens collected from March to May than in those sampled during the rest of the year. In 4-year-old fish the normal SS increase during the summer did not occur, possibly coinciding with their first sexual maturity. According to the correlations between SS/TS and age, the rate of spine bone resorption was significantly higher, even almost double, in captive-reared specimens. This could be attributed to the wider context of systemic dysfunctions occurring in reared ABFT, and may be related to a number of factors, including nutritional deficiencies, alteration of endocrine profile, cortisol-induced stress, and loss of spine functions during locomotion in rearing conditions.VersiĂłn del editor4,411

    Shaping Skeletal Growth by Modular Regulatory Elements in the Bmp5 Gene

    Cartilage and bone are formed into a remarkable range of shapes and sizes that underlie many anatomical adaptations to different lifestyles in vertebrates. Although the morphological blueprints for individual cartilage and bony structures must somehow be encoded in the genome, we currently know little about the detailed genomic mechanisms that direct precise growth patterns for particular bones. We have carried out large-scale enhancer surveys to identify the regulatory architecture controlling developmental expression of the mouse Bmp5 gene, which encodes a secreted signaling molecule required for normal morphology of specific skeletal features. Although Bmp5 is expressed in many skeletal precursors, different enhancers control expression in individual bones. Remarkably, we show here that different enhancers also exist for highly restricted spatial subdomains along the surface of individual skeletal structures, including ribs and nasal cartilages. Transgenic, null, and regulatory mutations confirm that these anatomy-specific sequences are sufficient to trigger local changes in skeletal morphology and are required for establishing normal growth rates on separate bone surfaces. Our findings suggest that individual bones are composite structures whose detailed growth patterns are built from many smaller lineage and gene expression domains. Individual enhancers in BMP genes provide a genomic mechanism for controlling precise growth domains in particular cartilages and bones, making it possible to separately regulate skeletal anatomy at highly specific locations in the body

    A new specimen of the ornithischian dinosaur Hesperosaurus mjosi from the Upper Jurassic Morrison Formation of Montana, U.S.A., and implications for growth and size in Morrison stegosaurs

    Stegosauria is a clade of ornithischian dinosaurs characterized by a bizarre array of dermal armor that extends from the neck to the end of the tail. Two genera of stegosaur are currently recognised from North America: the well-known Stegosaurus stenops and the much rarer Hesperosaurus mjosi. A new specimen of Hesperosaurus mjosi was discovered in some of the most northerly outcrops of the Upper Jurassic Morrison Formation near Livingston, Montana. The new specimen includes cranial, vertebral, and appendicular material as well as a dermal plate, and the excellent state of preservation of the palate reveals new anatomical information about this region in stegosaurs. Histological examination of the tibia indicates that the individual was not skeletally mature at time of death. Comparison with previously studied Stegosaurus and Hesperosaurus individuals indicates that Hesperosaurus mjosi may have been a smaller species than Stegosaurus stenops. Physiological processes scale with body mass, M, according to the relationship M0.75 in extant megaherbivores; thus, larger animals are better able to cope with more arid environments where forage is less abundant. Under this scenario, it is possible that Stegosaurus stenops and Hesperosaurus mjosi were environmentally partitioned, with the larger S. stenops occupying more arid environments. Analyses of the temporal overlap and latitudinal range of Morrison stegosaurs would allow this hypothesis to be investigated.The attached document is the authors’ final accepted version of the journal article.     Hepatitis E in Italy : 5 years of national epidemiological, virological and environmental surveillance, 2012 to 2016

    Increasing numbers of hepatitis E cases are being reported in several European countries, including Italy, but the burden of hepatitis E virus (HEV) infection is largely unknown in the latter. To gain a better understanding of HEV epidemiology at national level in Italy, we piloted a strengthened and integrated human (epi-demiological and virological) and environmental HEV surveillance system between 2012 and 2016. Over the 5-year period, 169 confirmed hepatitis E cases were identified, with a national annual incidence of 0.72 cases per 1,000,000. Of 65 HEV-RNA positive samples of sufficient quality for molecular analysis, 66% were genotype HEV3, 32% HEV1 and 1% HEV4. The most frequent risk factor reported by all HEV3 infected cases, was the consumption of undercooked pork and sausage. For the environmental surveillance, 679 urban sewage samples were collected from 53 wastewater treatment plants and HEV-RNA was detected in 38/679 of the samples. Among these, 25 (66%) were genotype HEV3 and the remaining were HEV1. We demonstrate that autochthonous transmission and environmental circulation of genotype HEV3 is adding to travel-related HEV transmission in Italy. We recommend the \u2018One Health\u2019 approach to integrated surveillance, and to include HEV-related messages within health information campaigns focussing on food security
