    The nasal cavity microbiota of healthy adults

    Abstract Background The microbiota of the nares has been widely studied. However, relatively few studies have investigated the microbiota of the nasal cavity posterior to the nares. This distinct environment has the potential to contain a distinct microbiota and play an important role in health. Results We obtained 35,142 high-quality bacterial 16S rRNA-encoding gene sequence reads from the nasal cavity and oral cavity (the dorsum of the tongue and the buccal mucosa) of 12 healthy adult humans and deposited these data in the Sequence Read Archive (SRA) of the National Center for Biotechnology Information (NCBI) (Bioproject: PRJNA248297). In our initial analysis, we compared the bacterial communities of the nasal cavity and the oral cavity from ten of these subjects. The nasal cavity bacterial communities were dominated by Actinobacteria, Firmicutes, and Proteobacteria and were statistically distinct from those on the tongue and buccal mucosa. For example, the same Staphylococcaceae operational taxonomic unit (OTU) was present in all of the nasal cavity samples, comprising up to 55% of the community, but Staphylococcaceae was comparatively uncommon in the oral cavity. Conclusions There are clear differences between nasal cavity microbiota and oral cavity microbiota in healthy adults.     Hsc70-induced changes in clathrin-auxilin cage structure suggest a role for clathrin light chains in cage disassembly

    The molecular chaperone, Hsc70, together with its co-factor, auxilin, facilitates the ATP-dependent removal of clathrin during clathrin-mediated endocytosis in cells. We have used cryo-electron microscopy to determine the 3D structure of a complex of clathrin, auxilin401-910 and Hsc70 at pH 6 in the presence of ATP, frozen within 20 seconds of adding Hsc70 in order to visualize events that follow the binding of Hsc70 to clathrin and auxilin before clathrin disassembly. In this map, we observe density beneath the vertex of the cage that we attribute to bound Hsc70. This density emerges asymmetrically from the clathrin vertex, suggesting preferential binding by Hsc70 for one of the three possible sites at the vertex. Statistical comparison with a map of whole auxilin and clathrin previously published by us reveals the location of statistically significant differences which implicate involvement of clathrin light chains in structural rearrangements which occur after Hsc70 is recruited. Clathrin disassembly assays using light scattering suggest that loss of clathrin light chains reduces the efficiency with which auxilin facilitates this reaction. These data support a regulatory role for clathrin light chains in clathrin disassembly in addition to their established role in regulating clathrin assembly

    Comparison of equatorial Pacific sea surface temperature variability and trends with Sr/Ca records from multiple corals

However, some studies report different Sr/Ca-temperature relationships for conspecifics on the same reef, with profound implications for interpretation of reconstructed temperatures. We assess whether these differences are attributable to small-scale oceanographic variability or ā€œvital effectsā€ associated with coral calcification and quantify the effect of intercolony differences on temperature estimates and uncertainties. Sr/Ca records from four massive Porites colonies growing on the east and west sides of Jarvis Island, central equatorial Pacific, were compared with in situ logger temperatures spanning 2002ā€“2012. In general, Sr/Ca captured the occurrence of interannual sea surface temperature events but their amplitude was not consistently recorded by any of the corals. No long-term trend was identified in the instrumental data, yet Sr/Ca of one coral implied a statistically significant cooling trend while that of its neighbor implied a warming trend. Slopes of Sr/Ca-temperature regressions from the four different colonies were within error, but offsets in mean Sr/Ca rendered the regressions statistically distinct. Assuming that these relationships represent the full range of Sr/Ca-temperature calibrations in Jarvis Porites, we assessed how well Sr/Ca of a nonliving coral with an unknown Sr/Ca-temperature relationship can constrain past temperatures. Our results indicate that standard error of prediction methods underestimate the actual error as we could not reliably reconstruct the amplitude or frequency of El NiƱoā€“Southern Oscillation events as large as Ā± 2Ā°C.     Comparison of opioid agonists in maintaining responding and in suppressing morphine withdrawal in rhesus monkeys

    Sixteen opioid agonists were studied for their capacity both to maintain responding previously reinforced by codeine and to suppress the withdrawal syndrome induced by morphine deprivation in rhesus monkeys. All compounds, which included examples from each of the major chemical families of opioids, maintained responding at rates above those maintained by saline. There were differences among the compounds in the maximal response rates maintained, and large differences in their potencies in maintaining responding. In morphine-dependent monkeys, the abstinence signs that developed 14 h after the last morphine dose were suppressed completely by all of the compounds except codeine. There was a strong positive correlation ( r =0.92) between the potency of a compound in maintaining drug-reinforced responding and the potency of the compound in suppressing the morphine withdrawal syndrome.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/46424/1/213_2004_Article_BF00432741.pd

    Investigation of free-living honey bee colonies in Ireland

    Apis mellifera mellifera (Linnaeus), the Western European honey bee, is considered extinct in the wild over most of its range due largely to hybridisation and replacement by other subspecies, parasitism by Varroa destructor, habitat loss, and effects from agricultural pesticides. The purity of the subspecies within the managed cohort is also at risk over much of its range. Here, we investigated if honey bee colonies inhabited locations outside of the apiaries. In those we located, we explored how long the colony persisted and we investigated the genotypes of the bees using multiple markers. We show here that unmanaged free-living honey bee colonies are present and widespread in Ireland, inhabiting a mixture of nesting habitats with some colonies persisting naturally and unaided over multiple years. Molecular data including mitochondrial, microsatellite, and SNPs evidence indicate that the free-living population sampled is largely comprised of pure A. m. mellifera. Finally, we discuss the implications of conserving free-living A. m. mellifera in Ireland and its possible role in improving the fitness of the managed population both in Ireland and the rest of its European range.We particularly thank the custodians of the free-living honey bee colonies and the Native Irish Honey Bee Society (NIHBS) for their assistance.     Using unoccupied aerial vehicles (UAVs) to map seagrass cover from Sentinel-2 imagery

    Seagrass habitats are ecologically valuable and play an important role in sequestering and storing carbon. There is, thus, a need to estimate seagrass percentage cover in diverse environments in support of climate change mitigation, marine spatial planning and coastal zone management. In situ approaches are accurate but time-consuming, expensive and may not represent the larger spatial units collected by satellite imaging. Hence, there is a need for a consistent methodology that uses accurate point-based field surveys to deliver high-quality mapping of percentage seagrass cover at large spatial scales. Here, we develop a three-step approach that combines in situ (quadrats), aerial (unoccupied aerial vehicleā€”UAV) and satellite data to map percentage seagrass cover at Turneffe Atoll, Belize, the largest atoll in the northern hemisphere. First, the optical bands of four UAV images were used to calculate seagrass cover, in combination with in situ data. The seagrass cover calculated from the UAV was then used to develop training and validation datasets to estimate seagrass cover in Sentinel-2 pixels. Next, non-seagrass areas were identified in the Sentinel-2 data and removed by object-based classification, followed by a pixel-based regression to calculate seagrass percentage cover. Using this approach, percentage seagrass cover was mapped using UAVs (R2 = 0.91 between observed and mapped distributions) and using Sentinel-2 data (R2 = 0.73). This work provides the first openly available and explorable map of seagrass percentage cover across Turneffe Atoll, where we estimate approximately 242 km2 of seagrass above 10% cover is located. We estimate that this approach offers 30 times more data for training satellite data than traditional methods, therefore presenting a substantial reduction in cost-per-point for data. Furthermore, the increase in data helps deliver a high-quality seagrass cover map, suitable for resolving trends of deteriorating, stable or recovering seagrass environments at 10 m2 resolution to underpin evidence-based management and conservation of seagrass.publishedVersio
