    Identification of exopolysaccharides-producing lactic acid bacteria from Burkina Faso fermented milk samples

    Spacer region between 16S and 23 S rRNA genes of thirteen lactic acid bacteria strains from Burkina Faso fermented milk samples were amplified by the polymerase chain reaction (PCR). Lactobacillus delbrueckii, Lactobacillus acidophilus, Lactobacillus fermentum, Streptococcus thermophilus, Pediococcus spp, Leuconostoc mesenteroides subsp mesenteroides were identified. The Lactobacillus group was the predominant bacteria. Plasmids identified ranged between 2000 and 4000 bp. Exopolysaccharides (EPS) production varied from 181 mg/l and 814 mg/l, monomer analysis showed that glucose and galactose were predominant.     Sorghum grain as human food in Africa: relevance of content of starch and amylase activities

    Sorghum is a staple food grain in many semi-arid and tropic areas of the world, notably in Sub-Saharan Africa because of its good adaptation to hard environments and its good yield of production. Among important biochemical components for sorghum processing are levels of starch (amylose and amylopectin) and starch depolymerizing enzymes. Current research focus on identifying varieties meeting specific agricultural and food requirements from the great biodiversity of sorghums to insure food security. Results show that some sorghums are rich sources of micronutrients (minerals and vitamins) and macronutrients (carbohydrates, proteins and fat). Sorghum has a resistant starch, which makes it interesting for obese and diabetic people. In addition, sorghum may be an alternative food for people who are allergic to gluten. Malts of some sorghum varieties display a-amylase and ß-amylase activities comparable to those of barley, making them useful for various agro-industrial foods. The feature of sorghum as a food in developing as well as in developed countries is discussed. A particular emphasis is made on the impact of starch and starch degrading enzymes in the use of sorghum for some African foods, e.g. “tî”, thin porridges for infants, granulated foods “couscous”, local beer “dolo”, as well agro-industrial foods such as lager beer and bread.Keywords: sorghum, a-amylase, b-amylase, starch, infant porridge, beer, couscous, dolo, tî, brea

    Evolution of African cassava mosaic virus by recombination between bipartite and monopartite begomoviruses

    <p>Abstract</p> <p>Background</p> <p>Cassava mosaic disease (CMD) is a major constraint on cassava cultivation in Africa. The disease is endemic and is caused by seven distinct cassava mosaic geminiviruses (CMGs), some of them including several variants.</p> <p>Findings</p> <p>From cassava leaf samples presenting CMD symptoms collected in Burkina Faso, four DNA-A begomovirus components were cloned and sequenced, showing 99.9% nucleotide identity among them. These isolates are most closely related to <it>African cassava mosaic virus </it>(ACMV) but share less than 89% nucleotide identity (taxonomic threshold) with any previously described begomovirus. A DNA-B genomic component, sharing 93% nucleotide identity with DNA-B of ACMV, was also characterized. Since all genomic components have a typical genome organization of Old World bipartite begomoviruses, this new species was provisionally named African cassava mosaic Burkina Faso virus (ACMBFV). Recombination analysis of the new virus demonstrated an interspecies recombinant origin, with major parents related to West African isolates of ACMV, and minor parents related to <it>Tomato leaf curl Cameroon virus </it>and <it>Cotton leaf curl Gezira virus</it>.</p> <p>Conclusion</p> <p>This is the first report of an ACMV-like recombinant begomovirus arisen by interspecific recombination between bipartite and monopartite African begomoviruses.</p

    Influences of Storage Conditions, Cultivation and Culinary Practices, On the Antioxidant Capacity of Red Bulbs of Some Onion Varieties Grown in Burkina Faso

    Red onion bulb is a vegetable containing micronutrients with antioxidant properties. However, certain factors can have an impact on the content of these compounds. The present research evaluate the effects of parameters such as cultural practices, storage conditions and culinary practices, on the antioxidants content of red onion bulb of some varieties grown in Burkina Faso. The identification of the factors was carried out through field surveys, and their effects on the antioxidants content were assessed and appreciated through especially specific related bibliographic and laboratory test data. The results showed that84.78% of farmers use chemical fertilizers and all of them use chemical pesticides on onion crop. They don’t use premises or equipment suitable for storage. About dishes,7.61% of cooks, peel, cut and wash onion bulb before steam cooking, boiling water cooking or oil frying them at high temperature during 30 minutes to 3 hours. Some cooks use to braise onion bulb scales as ingredients for different dishes such as barbecue. Only 02,72% of cooks grind the onion bulbs with a pestle, before using the crushed as such on barbecue, or slow cooking it in different sauces. Research results show that these culinary practices contribute declining significantly the final intake of antioxidant for the consumer's body

    Influence of Agro-Ecological Areas on the Antioxidant Capacity of Onion Varieties Grown in Burkina Faso

    Colored onion bulb is an important source of antioxidants. Some ecological factors can significantly influence the content and quality of these substances. The aim of the present study is to determine the antioxidant and total polyphenol content of fresh bulbs of five (05) varieties of onion (Galmi Violet, Damani Violet, Prema, Safari and a Local variety-LV) grown in Burkina Faso, and to evaluate also the influence of agro-ecological parameters on the antioxidant content of the most cultivated variety (Galmi violet) of them. Samples of onion bulb for analysis were collected in the six agro-ecological areas of Burkina Faso under the same cultivation conditions. Antioxidant activities were evaluated using the method of Ferric Reducing Antioxidant Power. The evaluation of the total polyphenol contents was carried out using the Folin-Ciocalteu method. The results showed that antioxidant activities for the 5 varieties tested vary between 0.125 ± 0.001 and 0.149 ± 0.004 mg TE / g, and total polyphenol contents from 0.172 ± 0.011 to 0.272 ± 0.003 mg EAG/g. Results on the assessment of the effect of ecology on the antioxidant content of Galmi violet variety revealed significant variations in antioxidant content from 0.144 ± 0.002 to 0.155 ± 0.001 mg TE / g, and total polyphenols from 0.208 ± 0.014 to 0.292 ± 0.012 mg EAG / g depending on the ecological cultivation area

    Prevalence and diversity of Salmonella enterica in water, fish and lettuce in Ouagadougou, Burkina Faso

    Abstract Background This study investigated the prevalence, serotypes and antimicrobial sensitivity patterns of Salmonella enterica in environment in Ouagadougou, Burkina Faso. A total of 476 samples, consisting of 36 samples of tap water, 51 samples of well water, 87 samples of channel water, 44 samples of reservoir water, 238 samples of fish, and 20 samples of lettuce were examined using standard bacteriological procedures for Salmonella. Results Salmonella were isolated from 98 samples. Salmonella were rare in drinking water, since they were not found at all from the tap water, and only in 2 % of well water. Salmonella were more common in the water of reservoir of Tanghin (15 %), reservoir of Yamtenga (20 %), and in the water channels in the city (from 20 to 31 %). Salmonella were commonly isolated from the fish (24 %) caught from the reservoir of Tanghin and from the lettuce (50 %) irrigated with water from Tanghin. The Salmonella isolates were found to represent 50 different serotypes. The 11 most common serotypes were Salmonella Bredeney and S. Colindale (both 8.2 %), S. Muenster (6.1 %), S. Korlebu (5.1 %), S. Eastbourne and S. Poona (both 4.1 %), and S. Agona, S. Derby, S. Drac, S. Senftenberg, S. Waycross (each 3.1 %), accounting for 51.3 % of all the isolates. In general, the Salmonella strains were sensitive to the antimicrobials tested, but two strains were resistant to streptomycin and many more intermediate to streptomycin or sulphonamide. Conclusion This study highlights the common prevalence of Salmonella and the high diversity of Salmonella serotypes in aquatic environment in Ouagadougou, Burkina Faso. Therefore, various human activities linked to water and consumption of water-related products, such as fish and lettuce, can lead to human Salmonella infections

    Seasonal performance of a malaria rapid diagnosis test at community health clinics in a malaria-hyperendemic region of Burkina Faso

    BACKGOUND: Treatment of confirmed malaria patients with Artemisinin-based Combination Therapy (ACT) at remote areas is the goal of many anti-malaria programs. Introduction of effective and affordable malaria Rapid Diagnosis Test (RDT) in remote areas could be an alternative tool for malaria case management. This study aimed to assess performance of the OptiMAL dipstick for rapid malaria diagnosis in children under five. METHODS: Malaria symptomatic and asymptomatic children were recruited in a passive manner in two community clinics (CCs). Malaria diagnosis by microscopy and RDT were performed. Performance of the tests was determined. RESULTS: RDT showed similar ability (61.2%) to accurately diagnose malaria as microscopy (61.1%). OptiMAL showed a high level of sensitivity and specificity, compared with microscopy, during both transmission seasons (high & low), with a sensitivity of 92.9% vs. 74.9% and a specificity of 77.2% vs. 87.5%. CONCLUSION: By improving the performance of the test through accurate and continuous quality control of the device in the field, OptiMAL could be suitable for use at CCs for the management and control of malaria

    Determinants of Mortality in Children under Five Years of Age with Severe Acute Malnutrition Admitted to the Yalgado Ouédraogo Teaching Hospital (Burkina Faso)

    Background: To determine critical factors associated with severely malnourished children under five this case-control study was conducted. Methods: The data of a total of 433 children aged 0-59 months and admitted to the Hospital Yalgado Ouedraogo, (CHU – YO) between January 31, 2009 to January 31, 2013, were included in the analysis: 72 for the case group and 361 for the control group. Clinical and treatment records were accessed and data were analyzed. Results: For clinical signs, determinants of mortality were diarrhea [OR = 4.6; (95%CI 2.6-8.2], anorexia [OR = 2.7; (95%CI 1.4-5.0] and hepatomegaly [OR = 2.6; (95%CI 1.4-4.8]. For infections, determinants of mortality were pediatric HIV/AIDS [OR = 10.9; (95%CI 5.6-21.5] and digestive illnesses [OR = 5.1 (95%CI 2.8-9.4)]. Regarding the complications of malnutrition, determinants of mortality were severe dehydration [OR = 16.4 (95%CI 8.0-33.5)], skin lesions [OR = 14.3 (95%CI 6.4 -31.9)], heart failure [OR = 6.8 (95%CI 2.5-19.0)] and severe anemia [OR = 3.2(95%CI 1.4-7.1)]. For biochemical indicators, low serum sodium [OR = 0.7(95%CI 0.5-1.0)] and potassium levels [OR = 0.9(95%CI 0.9-1.0)] were the critical factors. In addition the risk of death was associated with low value of MUAC [OR = 0.9 (95% CI 0.8-0.9)].     Anopheline species composition and the 1014F-genotype in different ecological settings of Burkina Faso in relation to malaria transmission

    Background: A three-year longitudinal study was conducted in four sentinel sites from different ecological settings in Burkina Faso, between 2008 and 2010 to identify longitudinal changes in insecticide resistance within Anopheles gambiae complex species based on larval collection. During this study, adult mosquitoes were also collected indoor and outdoor using several methods of collection. The present study reports the diversity of malaria vectors and the 1014F-genotype from this adult collection and investigates the association between this 1014F-genotype and sporozoite rate. Methods: Adult mosquitoes were collected from July to August (corresponding to the start of rainy season) and October to November (corresponding to the end of rainy season) over 3 years (2008–2010) at four sites across the country, using pyrethrum spray catches (PSC), exit traps and pit shelters. Anopheles gambiae complex mosquitoes were identified to species and genotyped for the L1014F kdr mutation by PCR using genomic DNA. The circumsporozoite antigen of Plasmodium falciparum was detected in mosquitoes using sandwich ELISA. Results: Overall 9212 anopheline mosquitoes were collected during the study period. Of those, 6767 mosquitoes were identified as Anopheles gambiae sensu lato (s.l.). Anopheles arabiensis, Anopheles coluzzii, Anopheles gambiae and or Anopheles funestus were incriminated as vectors of P. falciparum in the study area with an average sporozoite rate of 5%, (95% CI 4.14–5.99%). The kdr1014F-genotype frequencies were 11.44% (95% CI 2.5–39.85%), 19.2% (95% CI 4.53–53.73%) and 89.9 (95% CI 63.14–97.45%), respectively for An. arabiensis, An. coluzzii and An. gambiae. The proportion of the 1014F-genotype varied between sporozoite-infected and uninfected An. gambiae s.l. group. There was no significant difference in the 1014F-genotype frequency between infected and uninfected mosquitoes. Conclusion: The current study shows the diversity of malaria vectors and significant interaction between species composition and kdr1014F-genotype in An. gambiae complex mosquitoes from Burkina Faso. In this study, no associations were found between the 1014F-genotype and P. falciparum infection in the major malaria vector An. gambiae s.l
