    Visual Detection of Speckles in the Fish Xenotoca variata by the Predatory Snake Thamnophis melanogaster in Water of Different Turbidity

    The results of this study suggest that speckles on male X. variata are visual signals to potential predators such as the snake T. melanogaster. However, speckles are not a determining factor; snakes display a higher frequency of predatory behavior toward the fish, but their predatory behavior is influenced by water turbidity. Turbidity is considered a constraint that affects foraging in aquatic predators such as the snake T. melanogaster because it is involved in the visual identification of potential prey and capture success. Further experiments are needed to test the direct influence of snake predation on variation in sexual traits, such as the number of speckles on the flanks of male X. variata. Therefore, natural selection caused by predation may constrain the development of sexually dimorphic traits in X. variata, as suggested by other studies showing that male secondary sexual traits often increase the risk of predationSemi-aquatic snakes integrate visual and chemical stimuli, and prey detection and capture success are therefore linked to the display of visual predatory behavior. The snake Thamnophis melanogaster responds preferentially to individuals of the fish Xenotoca variata with a greater number of bright, colorful spots (lateral speckles) compared with those with a smaller number; however, water turbidity can reduce underwater visibility and effect the vulnerability of fish. In this study, we tested whether the presence of iridescent speckles on the flanks of male X. variata interacted with water turbidity to modify the predatory behavior displayed by the snake T. melanogaster.We predicted that in an experimental laboratory test, the snakes would increase the frequency of their predatory behavior to the extent that the water turbidity decreases. The snakes were tested at six different levels of water turbidity, in combination with three categories of male fish (with few, a median number of, or many speckles). The results showed that in a pool with high or zero turbidity, the number of speckles is not a determining factor in the deployment of the predatory behavior of the snake T. melanogaster toward X. variata. Our findings suggest that snakes can view the fish at intermediate percentages of turbidity, but the number of speckles in male X. variata is irrelevant as an interspecific visual signal in environments with insufficient luminosity.     Sclerophyllous Forest Tree Growth Under the Influence of a Historic Megadrought in the Mediterranean Ecoregion of Chile

    The Mediterranean-type Ecosystems of Central Chile is one of the most threatened regions in South America by global change, particularly evidenced by the historical megadrought that has occurred in central Chile since 2010. The sclerophyllous forest stands out, whose history and relationship with drought conditions has been little studied. Cryptocarya alba and Beilschmiedia miersii (Lauraceae), two large endemic trees, represent an opportunity to analyze the incidence of intense droughts in the growth of sclerophyllous forests by analyzing their tree rings. Here, we considered > 400 trees from nineteen populations of C. alba and B. miersii growing across a latitudinal gradient (32°–35° S). To study the influence of local and large-scale climatic variability on tree growth, we first grouped the sites by species and explored the relationships between tree-growth patterns of C. alba and B. miersii with temperature, precipitation, and climate water deficit (CWD). Second, we performed Principal Component Analysis to detect common modes of variability and to explore relationships between growth patterns and their relationship to Palmer Drought Severity Index (PDSI), ENSO and SAM indices. We detected a breaking point as of 2002 at regional level, where a persistent and pronounced decrease in tree growth occurred, mainly influenced by the increase in CWD and the decrease in winter-spring rainfall. In addition, a positive (negative) relationship was showed between PC1 growth-PDSI and PC1 growth-ENSO (growth-SAM), that is, growth increases (decreases) in the same direction as PDSI and ENSO (SAM). Despite the fact that sclerophyllous populations are highly resistant to drought events, we suggest that the sclerophyllous populations studied here experienced a generalized growth decline, and possibly the natural dynamics of their forests have been altered, mainly due to the accumulating effects of the unprecedented drought since 2010.Fil: Venegas Gonzalez, Alejandro.     ComposiciĂłn y actividad antifĂșngica del aceite esencial de Fabiana imbricata contra levaduras del gĂ©nero Candida

    El objetivo de este estudio fue evaluar la actividad anti-fungica del aceite esencial (AE) de Fabiana imbricata Ruiz & Pav contra cinco especies de Candida comĂșnmente asociadas a enfermedades ginecolĂłgicas. El aceite esencial de las hojas fresca de F. imbricata fue obtenido por hidrodestilaciĂłn y caracterizado mediante la tĂ©cnica de cromatografĂ­a de gases acoplada a espectrometrĂ­a de masas (CG/EM). Se evaluĂł la actividad anti-fĂșngica a diferentes concentraciones del AE mediante el mĂ©todo de microdiluciĂłn. Los compuestos mĂĄs abundantes identificados en el AE (7a-isopropenil-4,5-dimetiloctahidroinden-4-il) metanol (43,66%), bulnesol (17,02%) y T-muurolol (5,75%). El AE mostro una potente y selectiva actividad anti-fĂșngica sobre el crecimiento de la levadura patĂłgena Candida guilliermondii con una concentraciĂłn mĂ­nima inhibitoria de 10 ÎŒg/mL, superior al control positivo clotrimazol que presento un valor de 15 ÎŒg/mL. Estos resultados confirman las propiedades antimicrobianas de los aceites esenciales y su posible uso en el manejo de las cepas de C. guilliermondii de importancia para pacientes inmune deprimidos y como posibles alternativas a los fungicidas sintĂ©ticos

    Canagliflozin and renal outcomes in type 2 diabetes and nephropathy

    BACKGROUND Type 2 diabetes mellitus is the leading cause of kidney failure worldwide, but few effective long-term treatments are available. In cardiovascular trials of inhibitors of sodium–glucose cotransporter 2 (SGLT2), exploratory results have suggested that such drugs may improve renal outcomes in patients with type 2 diabetes. METHODS In this double-blind, randomized trial, we assigned patients with type 2 diabetes and albuminuric chronic kidney disease to receive canagliflozin, an oral SGLT2 inhibitor, at a dose of 100 mg daily or placebo. All the patients had an estimated glomerular filtration rate (GFR) of 30 to <90 ml per minute per 1.73 m2 of body-surface area and albuminuria (ratio of albumin [mg] to creatinine [g], >300 to 5000) and were treated with renin–angiotensin system blockade. The primary outcome was a composite of end-stage kidney disease (dialysis, transplantation, or a sustained estimated GFR of <15 ml per minute per 1.73 m2), a doubling of the serum creatinine level, or death from renal or cardiovascular causes. Prespecified secondary outcomes were tested hierarchically. RESULTS The trial was stopped early after a planned interim analysis on the recommendation of the data and safety monitoring committee. At that time, 4401 patients had undergone randomization, with a median follow-up of 2.62 years. The relative risk of the primary outcome was 30% lower in the canagliflozin group than in the placebo group, with event rates of 43.2 and 61.2 per 1000 patient-years, respectively (hazard ratio, 0.70; 95% confidence interval [CI], 0.59 to 0.82; P=0.00001). The relative risk of the renal-specific composite of end-stage kidney disease, a doubling of the creatinine level, or death from renal causes was lower by 34% (hazard ratio, 0.66; 95% CI, 0.53 to 0.81; P<0.001), and the relative risk of end-stage kidney disease was lower by 32% (hazard ratio, 0.68; 95% CI, 0.54 to 0.86; P=0.002). The canagliflozin group also had a lower risk of cardiovascular death, myocardial infarction, or stroke (hazard ratio, 0.80; 95% CI, 0.67 to 0.95; P=0.01) and hospitalization for heart failure (hazard ratio, 0.61; 95% CI, 0.47 to 0.80; P<0.001). There were no significant differences in rates of amputation or fracture. CONCLUSIONS In patients with type 2 diabetes and kidney disease, the risk of kidney failure and cardiovascular events was lower in the canagliflozin group than in the placebo group at a median follow-up of 2.62 years

    Omecamtiv mecarbil in chronic heart failure with reduced ejection fraction, GALACTIC‐HF: baseline characteristics and comparison with contemporary clinical trials

    Aims: The safety and efficacy of the novel selective cardiac myosin activator, omecamtiv mecarbil, in patients with heart failure with reduced ejection fraction (HFrEF) is tested in the Global Approach to Lowering Adverse Cardiac outcomes Through Improving Contractility in Heart Failure (GALACTIC‐HF) trial. Here we describe the baseline characteristics of participants in GALACTIC‐HF and how these compare with other contemporary trials. Methods and Results: Adults with established HFrEF, New York Heart Association functional class (NYHA) ≄ II, EF ≀35%, elevated natriuretic peptides and either current hospitalization for HF or history of hospitalization/ emergency department visit for HF within a year were randomized to either placebo or omecamtiv mecarbil (pharmacokinetic‐guided dosing: 25, 37.5 or 50 mg bid). 8256 patients [male (79%), non‐white (22%), mean age 65 years] were enrolled with a mean EF 27%, ischemic etiology in 54%, NYHA II 53% and III/IV 47%, and median NT‐proBNP 1971 pg/mL. HF therapies at baseline were among the most effectively employed in contemporary HF trials. GALACTIC‐HF randomized patients representative of recent HF registries and trials with substantial numbers of patients also having characteristics understudied in previous trials including more from North America (n = 1386), enrolled as inpatients (n = 2084), systolic blood pressure < 100 mmHg (n = 1127), estimated glomerular filtration rate < 30 mL/min/1.73 m2 (n = 528), and treated with sacubitril‐valsartan at baseline (n = 1594). Conclusions: GALACTIC‐HF enrolled a well‐treated, high‐risk population from both inpatient and outpatient settings, which will provide a definitive evaluation of the efficacy and safety of this novel therapy, as well as informing its potential future implementation

    Recent radial growth decline in response to increased drought conditions in the northernmost Nothofagus populations from South America

    An emerging phenomenon of forest decline in Mediterranean-type ecosystems has been detected in response to climate change during the last century. It is expected that the Mediterranean regions will likely experience drought events during this century with consequences for biodiversity maintenance. Although the Chilean Mediterranean-type forests are among the most threatened forest ecosystems in South America, their responses to recent increased drought events due to global warming are poorly documented. In the same region, the endangered and endemic forests of Nothofagus macrocarpa (Nothofagaceae) are found on mountain peaks. It is unclear how N. macrocarpa forests are responding to increased drought conditions occurring in the area over the last few decades. Here, we analyzed how recent climatic variability has affected the growth of N. macrocarpa. We selected five sites along the whole geographic distribution of N. macrocarpa forests in central Chile (32.5–34.5°S) to develop tree-ring chronologies. Climate-growth relationships were analyzed through correlations with local (precipitation, temperature and drought index) and large-scale climate data (ENSO index and Antarctic Oscillation). N. macrocarpa growth was positively influenced by May to November precipitation (austral winterspring seasons) and negatively influenced by temperature from October to December (austral spring/earlysummer seasons). Using a piecewise regression analysis, we identified a significant decrease in growth from 1980 onwards that resembled a precipitation decline and temperature increase in central Chile during the same time period. Tree-ring chronologies were positively correlated to the ENSO index and negatively correlated to the Antarctic Oscillation index during the current growing season, and more strongly from 1980 onwards. Based on our results, we conclude that increased drought conditions have produced a decline in radial growth of N. macrocarpa forests in the last decades. We propose that increased drought conditions predicted for this century in this region will exacerbate this declining N. macrocarpa growth trend with unknown consequences for the survival of these endemic and endangered forest ecosystems.Fil: Venegas Gonzalez, Alejandro.
