Cytochrome P450 (CYP) enzymes are an important class of heme-containing proteins that catalyze oxidation reactions leading toward the removal of a wide variety of endogenous and exogenous substrates including prescription drugs. The activities of CYP enzymes are regulated primarily at the transcription level involving the regulatory sequences at the 5'-flanking region of the CYP genes. The objective of this dissertation study was to characterize the function of the 5'-flanking sequences of selected CYP genes primarily responsible for drug metabolism. Various sequences from the 5'-flanking regions of different CYP genes (CYP1A2, CYP2C9, CYP2C18, CYP2D6, CYP2E1, and CYP3A4) were cloned in expression vectors and tested for their activity in driving reporter gene expression in mouse livers and in transfected HepG2, 293, and BL-6 cells under optimized conditions. It was demonstrated that among the tested 5'-flanking regions of CYP genes, the CYP2D6 promoter showed the highest activity both in vivo and in vitro. The activities of various 5'-flanking regions of CYP genes in sustaining transgene expression were then tested in mouse liver and compared to those of other promoter sequences. As a result, the CYP2D6 promoter showed the highest activity and its activity was comparable to that of many established promoters. The mechanism underlying CYP promoter activities in vivo and in vitro were then studied using the CYP2C9 promoter as a model. Activities of various 5'-flanking sequences of CYP2C9 were evaluated by using deletion mutations of plasmid constructs in combination with transfection in mouse livers and in HepG2 cells. Finally, the role of PXR and CAR nuclear receptors in regulating CYP2C9 activation was investigated. The results show that both CAR and PXR are essential for CYP2C9 activation and that the regulatory elements reside in the proximal 1-2 kb region upstream of the CYP2C9 gene

AbstractINTRODUCTIONXanthoma (or xanthofibroma) is a benign proliferative lesion, mostly seen in soft tissue. Xanthoma of bone is very rare benign primary bone tumor, more frequently seen in men and in patients over 20 years of age. Histologically, it is characterized by mononuclear macrophage-like cells, abundant foam cells, and multinucleated giant cells. It is sometimes discovered coincidentally and the most frequent symptom is pain.PRESENTATION OF CASEWe present a 50-year-old healthy male patient with primary xanthoma of the calcaneus, who was treated by curettage and bone cement. He presented with a pathological fracture in a calcaneus bone lesion. Giant cell tumor was suspected on X-ray and MRI. Curettage and bone cementing was done through the posterolateral approach. Lipid profile was normal and histological examination revealed findings consistent with primary xanthoma of calcaneus bone.DISCUSSIONTo avoid an erroneous diagnosis, all material should be examined microscopically, the radiological features of the lesion should be studied properly and lipid profile should be investigated to differentiate between primary and secondary xanthoma. Primary xanthoma may be treated with curettage and bone graft while secondary xanthoma is treated nonsurgically and the skeletal manifestations will disappear with systemic treatment of hyperlipidemia.CONCLUSIONWe present this case to raise the suspicion of this lesion that is rarely described in the literatures. This is the first case of primary xanthoma of calcaneus bone that has been reported in Qatar

Cloud computing is a Participation in the process and storage operations across distant servers that are shared by many organizations and users and thus be transferred from an application to a service. The organization can share data over the Internet and user can pay only for the resources that will be used only. While cloud computing has disadvantages, there are some advantages for cloudlets have over cloud computing which include: lower network latency and users having full ownership of the data shared. When the need of data to be stored in the servers grows quickly, the workload in every resource will grow too. So, we need a load balancing algorithm and the load balancing is important issue in the cloud environment. Load balancing defined as a technique that divides the extra load equally across all the resources to ensure that no one resource overloaded. . So the performance of the cloud can be improved by having an excellent load balancing strategy. For that we will discuss the existing load balancing algorithms in cloud computing and propose algorithm to improve round robin algorithm by CloudAnalyst simulator  based on a factor of response time and processing time and the proposed algorithm was found to be best in response time and processing time when we compare it with round robin algorithms

This study investigated the relationship between attitudes and motivation, and English language achievement. It also illustrated the influence of sex (males & females) on the attitudes and motivation of students in Qatar. Two main null-hypotheses and sub null-hypotheses were formulated to answer the main research questions of the study. A quantitative approach was implemented in a questionnaire survey, followed by a qualitative study using focus group interviews in order to explore the students` answers in more depth and to cover various issues missed in the questionnaire. The eleven scales of the Attitude/Motivation Test Battery (AMTB) were adopted with slight adjustments to suit the present study. This instrument was chosen due to its established reliability and validity. In order to estimate population parameters, an independent t-test was used to test for differences between males and females. On the other hand, the Spearman correlation was used to determine the relationship between students’ achievement in English and their motivation and attitudes. The study confirms previous findings and contributes additional evidence to suggest that the socio-educational model is appropriate for a monolingual context like Qatar where English is officially regarded as a foreign language. Moreover, this study contributes to knowledge concerning language learning and confirms Gardner’s (2005, 2010, 2012) argument that if a researcher adapts the full AMTB and selects appropriate items, the findings will corroborate the predictions of the socio-educational model

Green cybersecurity is the emerging trend in the new era and this green cybersecurity technology minimizes the negative effects of IT operations and implements a green sustainable environment. Therefore, the study conceptually draws the concept of green cybersecurity by applying the theory of reasoned action (TRA) assumptions that logically support green information technology acceptance. Using a convenient sampling, the data were collected from Qatar transport industries, particularly the IT experts and managers, to get responses on the implementation of green cybersecurity and sustainability of 5 transport companies in Doha, Qatar. Using Smart PLS-SEM, the study employed the SEM technique to test the proposed hypotheses. The results reported that green cybersecurity’s control/position, integrity, and authenticity significantly and positively influenced TBL sustainability, but confidentiality, availability, and utility do not. The implementation of industry 4.0 makes them accessible and more effective to ensure TBL sustainable development in the transport industries in Qatar. Applying green cybersecurity in this setting will improve services in transportation sector. A green cybersecurity platform will make it a point to systematically search for and promote innovations made possible by smart green technologies to avoid carbon-emission vehicles. Through the efficient and cutting-edge green, cybersecurity will be Qatar’s transportation sector’s primary responsibility to contribute to Qatar’s sustainable development. In order to accomplish this goal, the regulator must create and implement it. In addition, it emphasizes the importance of adopting green cybersecurity to confront the difficulties facing city transportation all over Qatar as a foundational component of achieving long-term sustainable development
