    Suppression of V1 feedback produces a shift in the topographic representation of receptive fields of LGN cells by unmasking latent retinal drives

    [Abstract] In awake monkeys, we used repetitive transcranial magnetic stimulation (rTMS) to focally inactivate visual cortex while measuring the responsiveness of parvocellular lateral geniculate nucleus (LGN) neurons. Effects were noted in 64/75 neurons, and could be divided into 2 main groups: (1) for 39 neurons, visual responsiveness decreased and visual latency increased without apparent shift in receptive field (RF) position and (2) a second group (n = 25, 33% of the recorded cells) whose excitability was not compromised, but whose RF position shifted an average of 4.5°. This change is related to the retinotopic correspondence observed between the recorded thalamic area and the affected cortical zone. The effect of inactivation for this group of neurons was compatible with silencing the original retinal drive and unmasking a second latent retinal drive onto the studied neuron. These results indicate novel and remarkable dynamics in thalamocortical circuitry that force us to reassess constraints on retinogeniculate transmission

    [Abstract] Increasing evidence indicates that static magnetic fields (SMF) reduce cortical activity in both human and animal models. The aim of this work was to investigate the effect of SMF on epileptiform cortical activity, a condition related to an abnormal increase in neuronal excitability. The first experimental block included a Pilocarpine rat model of epilepsy, in which a magnetic neodymium nickel-plated cylinder, a magnetic field of 0.5 T, or “sham” were placed over the skull. In the second experimental block, we recorded epileptic-like activity in the visual cortex of a monkey (Macaca mulatta) under control conditions and in the presence of the magnet. Between 15 and 30 minutes after the second dose of Pilocarpine, EEG changes compatible with seizure like events induced by Pilocarpine were clearly observed in the control animals (sham stimulation). Similar effects were visible in the animals exposed to the real magnet after 1–2 hours. In the monkey, SMF over the cortical focus clearly reduced abnormal activity: the intensity threshold for visual induction increased and the severity and duration decreased. These results reinforce the view that static magnets modulate cortical activity and open the door to the future therapeutic use of SMF in epilepsy as a complement to current pharmacological treatments.Ministerio de Economía y Competitividad; BFU2013-45343-PMinisterio de Economía y Competitividad; BFU2017-82296-

    Tomato fruit carotenoid biosynthesis is adjusted to actual ripening progression by a light-dependent mechanism

    Carotenoids are isoprenoid compounds that are essential for plants to protect the photosynthetic apparatus against excess light. They also function as health-promoting natural pigments that provide colors to ripe fruit, promoting seed dispersal by animals. Work in Arabidopsis thaliana unveiled that transcription factors of the phytochrome-interacting factor (PIF) family regulate carotenoid gene expression in response to environmental signals (i.e. light and temperature), including those created when sunlight reflects from or passes though nearby vegetation or canopy (referred to as shade). Here we show that PIFs use a virtually identical mechanism to modulate carotenoid biosynthesis during fruit ripening in tomato (Solanum lycopersicum). However, instead of integrating environmental information, PIF-mediated signaling pathways appear to fulfill a completely new function in the fruit. As tomatoes ripen, they turn from green to red due to chlorophyll breakdown and carotenoid accumulation. When sunlight passes through the flesh of green fruit, a self-shading effect within the tissue maintains high levels of PIFs that directly repress the master gene of the fruit carotenoid pathway, preventing undue production of carotenoids. This effect is attenuated as chlorophyll degrades, causing degradation of PIF proteins and boosting carotenoid biosynthesis as ripening progresses. Thus, shade signaling components may have been co-opted in tomato fruit to provide information on the actual stage of ripening (based on the pigment profile of the fruit at each moment) and thus finely coordinate fruit color change.     Evaluation of metabolic changes in acute intermittent porphyria patients by targeted metabolomics

    Acute intermittent porphyria (AIP) is an inherited rare hepatic disorder due to mutations within the hydroxymethylbilane gene. AIP patients with active disease overproduce aminolevulinic acid (ALA) and porphobilinogen (PBG) in the liver which are exported inducing severe neurological attacks. Different hepatic metabolic abnormalities have been described to be associated with this condition. The goal of this research was to explore the metabolome of symptomatic AIP patients by state-of-the art liquid chromatography-tandem mass spectrometry (LC-MS/MS). A case versus control study including 18 symptomatic AIP patients and 33 healthy controls was performed. Plasmatic levels of 51 metabolites and 16 ratios belonging to four metabolic pathways were determined. The results showed that the AIP patients presented significant changes in the two main areas of the metabolome under study: (a) the tryptophan/kynurenine pathway with an increase of tryptophan in plasma together with increase of the kynurenine/tryptophan ratio; and (b) changes in the tricarboxylic acid cycle (TCA) including increase of succinic acid and decrease of the fumaric acid/succinic acid ratio. We performed a complementary in vitro study adding ALA to hepatocytes media that showed some of the effects on the TCA cycle were parallel to those observed in vivo. Our study confirms in plasma previous results obtained in urine showing that AIP patients present a moderate increase of the kynurenine/tryptophan ratio possibly associated with inflammation.     Variable dose rate sprayer prototype for dose adjustment in tree crops according to canopy characteristics measured with ultrasonic and laser lidar sensor.

    One of the key issues in crop protection products to be applied in tree crops is the dose rate adjustment. An inappropriate dose selection could be responsible of the lack of efficacy due to under dosage and a loss in efficiency due to over dosage. Both situations imply economical losses and greater impacts on the environment. A better dose adjustment is possible from a variable rate technology approach by measuring the crown volume and adjusting the dose rate on-the-go according to the variation of the estimated vegetation volume. Tests have been done with a variable rate sprayer prototype and satisfactory results have been achieved.Postprint (published version

    Fitting flavour symmetries: the case of two-zero neutrino mass textures

    We present a numeric method for the analysis of the fermion mass matrices predicted in flavour models. The method does not require any previous algebraic work, it offers a χ2 comparison test and an easy estimate of confidence intervals. It can also be used to study the stability of the results when the predictions are disturbed by small perturbations. We have applied the method to the case of two-zero neutrino mass textures using the latest available fits on neutrino oscillations, derived the available parameter space for each texture and compared them. Textures A1 and A2 seem favoured because they give a small χ2, allow for large regions in parameter space and give neutrino masses compatible with Cosmology limits. The other 'allowed' textures remain allowed although with a very constrained parameter space, which, in some cases, could be in conflict with Cosmology. We have also revisited the 'forbidden' textures and studied the stability of the results when the texture zeroes are not exact. Most of the forbidden textures remain forbidden, but textures F1 and F3 are particularly sensitive to small perturbations and could become allowed
