Relativistic slim disks with vertical structure

We report on a scheme for incorporating vertical radiative energy transport into a fully relativistic, Kerr-metric model of optically thick, advective, transonic alpha disks. Our code couples the radial and vertical equations of the accretion disk. The flux was computed in the diffusion approximation, and convection is included in the mixing-length approximation. We present the detailed structure of this "two-dimensional" slim-disk model for alpha=0.01. We then calculated the emergent spectra integrated over the disk surface. The values of surface density, radial velocity, and the photospheric height for these models differ by 20%-30% from those obtained in the polytropic, height-averaged slim disk model considered previously. However, the emission profiles and the resulting spectra are quite similar for both types of models. The effective optical depth of the slim disk becomes lower than unity for high values of the alpha parameter and for high accretion rates.Comment: 15 pages, 18 figures (2 new), A&A in pres

Harmonization of exosome isolation from culture supernatants for optimized proteomics analysis

<div><p>Exosomes, the smallest subset of extracellular vesicles (EVs), have recently attracted much attention in the scientific community. Their involvement in intercellular communication and molecular reprogramming of different cell types created a demand for a stringent characterization of the proteome which exosomes carry and deliver to recipient cells. Mass spectrometry (MS) has been extensively used for exosome protein profiling. Unfortunately, no standards have been established for exosome isolation and their preparation for MS, leading to accumulation of artefactual data. These include the presence of high-abundance exosome-contaminating serum proteins in culture media which mask low-abundance exosome-specific components, isolation methods that fail to yield “pure” vesicles or variability in protein solubilization protocols. There is an unmet need for the development of standards for exosome generation, harvesting, and isolation from cellular supernatants and for optimization of protein extraction methods before proteomics analysis by MS. In this communication, we illustrate the existing problems in this field and provide a set of recommendations that are expected to harmonize exosome processing for MS and provide the faithful picture of the proteomes carried by exosomes. The recommended workflow for effective and specific identification of proteins in exosomes released by the low number of cells involves culturing cells in medium with a reduced concentration of exosome-depleted serum, purification of exosomes by size-exclusion chromatography, a combination of different protein extraction method and removal of serum-derived proteins from the final dataset using an appropriate sample of cell-unexposed medium as a control. Application of this method allowed detection of >250 vesicle-specific proteins in exosomes from 10 mL of culture medium.</p></div

Heat shock protein A2 is a novel extracellular vesicle-associated protein

70-kDa Heat Shock Proteins (HSPA/HSP70) are chaperones playing a central role in the proteostasis control mechanisms. Their basal expression can be highly elevated as an adaptive response to environmental and pathophysiological stress conditions. HSPA2, one of poorly characterised chaperones of the HSPA/HSP70 family, has recently emerged as epithelial cells differentiation-related factor. It is also commonly expressed in cancer cells, where its functional significance remains unclear. Previously, we have found that proteotoxic stress provokes a decrease in HSPA2 levels in cancer cells. In the present study we found that proteasome inhibition-related loss of HSPA2 from cancer cells neither is related to a block in the gene transcription nor does it relate to increased autophagy-mediated disposals of the protein. Proteotoxic stress stimulated extracellular release of HSPA2 in extracellular vesicles (EVs). Interestingly, EVs containing HSPA2 are also released by non-stressed cancer and normal cells. In human urinary EVs levels of HSPA2 were correlated with the levels of TSG101, one of the main EVs markers. We conclude that HSPA2 may constitute basic components of EVs. Nevertheless, its specific role in EVs and cell-to-cell communication requires further investigation

Architecture and town planning in the city centre of Gdynia in the years 1945-1989

Celem pracy jest przedstawienie wybranych powojennych koncepcji, realizacji urbanistycznych i architektonicznych śródmieścia Gdyni oraz ich stosunku do tradycji międzywojennych. Praca ukazuje, jaki wpływ na powojenne urbanistyczne projekty i realizacje miało państwo, polityka planistyczna kraju, czy też zmiana roli Gdyni, a jednocześnie na ile inspirujące pozostały przedwojenne jeszcze założenia oraz plany. W kontekście tym powstawały obiekty, kontynuujące tradycję architektoniczną miejsca i funkcji, odwołujące się do modernistycznych form, ale też czerpiące z najnowszych osiągnięć technicznych i zrywające z międzywojennym kontekstem.W rozdziale "Gdynia - powstanie i rozwój do 1946 roku" pokrótce nakreślone zostały okoliczności powstania Gdyni, rozwój miasta do wybuchu II Wojny Światowej, zniszczenia wojenne oraz odbudowa miasta. W rozdziale znalazła się również próba ukazania, jak zmieniała się sytuacja polityczno-administracyjna powojennej Gdyni.Następny rozdział, "Gdynia w planach międzywojennych i powojennych", ma na celu przedstawienie kolejnych koncepcji rozwiązania urbanistycznego śródmieścia, przy jednoczesnym wskazaniu na zmieniające się plany zagospodarowania przestrzennego całego zespołu portowo-miejskiego GD. Plany te, choć bardzo ogólne, decydowały o rozmieszczeniu i natężeniu takich funkcji, jak mieszkalnictwo, administracja, usługi także w śródmieściu, co miało realny wpływ na rozwiązania urbanistyczne i decyzje o lokalizacji obiektów.Rozdział trzeci, "Forum Morskie. Międzywojenne i powojenne koncepcje ukształtowania Dzielnicy Reprezentacyjnej Gdyni", ukazuje przekształcenia zachodzące w projektach forum, które były wynikiem zmieniającej się sytuacji politycznej, potrzeb miasta oraz znaczenia przypisywanego reprezentacyjnej, nadmorskiej części śródmieścia. Opisane w rozdziale wybrane obiekty zlokalizowane na terenie Forum Morskiego obrazują powyższe zmiany, lecz również pokazują, w jaki sposób kontynuowano przedwojenne projekty i idee. Wszystkie też stanowią przykład reprezentacyjnej architektury powstałej przed 1989 rokiem.Kolejna część, "Osiedla i budownictwo mieszkaniowe w Śródmieściu Gdyni", jest próbą ukazania, w jaki sposób budownictwo mieszkaniowe zdominowało po wojnie śródmieście miasta. W rozdziale opisane zostały rozwiązania urbanistyczne i architektoniczne zlokalizowane w centrum Gdyni.W rozdziale "Kościoły śródmieścia", przedstawione zostały trzy świątynie, które wybudowano w śródmieściu w latach 1945-1989. Każda z realizacji powstała w innym czasie oraz w innych warunkach politycznych. Każda z nich stanowi oryginalny i niezwykle ciekawy przykład powojennej architektury sakralnej. W szóstym rozdziale zaprezentowane zostały obiekty użyteczności publicznej. Dworzec kolejowy, niezwykle reprezentacyjny, oceniany często jako obiekt stricte socrealistyczny, stanowi w rzeczywistości klamrę spinającą Gdynię międzywojenną i powojenną. Świadczy o tym nie tylko tradycja miejsca, w którym powstał i osoba twórcy projektu dworca, ale również jego forma architektoniczna. Wyjątkowym obiektem powstałym w śródmieściu jest Dom Rzemiosła, który stanowi przykład doskonałej architektury, twórczo nawiązującej do przedwojennej tradycji. Pozostałe budynki, Internat Wyższej Szkoły Rybołówstwa Morskiego i Szpital Miejski, ukazują, w jaki sposób działali architekci, by wypełnić użytkowe potrzeby miasta i jakie skutki miało to dla architektonicznego krajobrazu centrum Gdyni. Ostatni rozdział to próba odpowiedzi na pytanie, jakie czynniki i w jaki sposób wpłynęły na kształt śródmieścia, decydując tym samym o tożsamości młodego, nadmorskiego ośrodka. Fakt, że wielu planów dla Gdyni nie zrealizowano, bądź też zrealizowano częściowo, stanowi o jej unikalnym charakterze. Urbanistyka i architektura centrum Gdyni w dużej mierze ukształtowane w okresie międzywojennym, a następnie z wyczuciem dopełniane do 1989 roku, zdeterminowały bowiem nie tylko specyficzny charakter miasta, ale ukształtowała również tożsamość jego mieszkańców.The purpose of the thesis is to present some postwar ideas and realisations in architecture and town planning of Gdynia’s city centre and their relation of those ideas and realisations to the architectural tradition of the interwar period. The research shows the influence of the state, town planning policy and the change of Gdynia’ s weight after the Second World War on town plans and realisations as well as the inspirations taken from the interwar period’s plans and assumptions. The buildings which came into being carried on the tradition of location, function and moder art forms but at the same time derived from the newest technical achievements and broke up with interwar traditions.The chapter Gdynia – the beginning and development till 1946, gives the information about the circumstances of Gdynia’s beginning, the town’s development till the outbreak of the Second World War, damage caused by the war and the reconstruction. In this part I also attempt to show the political and administrative situation of Gdynia after the Second World War. The next chapter, Gdynia in the interwar and postwar town planning, presents successive ideas on the shape of the city centre. It also shows the changes in the spatial management plans for the complex of the seaports and cities (Gdańsk-Sopot-Gdynia). The plans were quite general but they determined the location and the concentration of the city functions (housing, administration, services) in the city centre and by itself they had the influence on town plans and location of the buildings.The third chapter, Maritime Forum. Interwar and postwar ideas on the shape of Gdynia’s Representative District, shows further modifications of the plans of the forum as the result of the changes in the political situation, actual needs of the city or the evolution in the significance given to the representative part of the city centre. The buildings described in the chapter display these changes and also show the continuation of interwar plans and ideas. All of them belong to the representative architecture before 1989.In the next chapter, Housing estetes and house building in the centre of Gdynia, I attempt to show how the house building deminated the city centre. The chapter characterises some solutions in town planning and architecture applied to the city centre.The fifth chapter, Churches of the city centre, presents three churches built in Gdynia's centre between 1945 and 1989. Each building came into being in different periods and political circumstances. Each of them is an original and extraordinary example of the postwar sacral architecture.The sixth chapter gives the description of the public utility objects in Gdynia's centre. The representative train station is often considered as a strictly socialist realist building. In fact, the object bonds interwar and postwar Gdynia, not only by the tradition of the place and the person of the designer, but also by the form. The next extraordinary object built in the city centre is the House of Crafts, the example of excellent architecture creativly related to the modern art tradition. Other buildings presented in the chapter, the Dormitory of the High School of Sea Fishery and the City Hospital show how the architects tried to fulfill the utilitarian needs of the city and how it affected the architectural landscape of the city centre.The last chapter tries to give the answer, which factors determined the shape of the city centre and how they influenced the identity of the young, maritime city. Many plans for Gdynia were never accomplished and that determines the unique character of the city. The town planning and the architecture of Gdynia's centre were mainly shaped in the interwar period, but later completed with intuition in the postwar period, which resulted in the unique character of the city and influenced the identity of its inhabitants

Praca poświęcona badaniu strategii stosowanych wobec lokalizacji filmowych w audiodeskrypcji, na podstawie skryptów do filmów: Girl with a Pearl Earring, The Sentinel (j. angielski); To Rome with Love, Ocho Apellidos Vascos (j. hiszpański); Vicky Cristina Barcelona (j. polski). W pierwszym rozdziale wyjaśniono czym jest audiodeskrypcja. Kolejny rozdział poświęcono na badania mające na celu udowodnienie, że audiodeskrypcja może stanowić dziedzinę przekładoznawstwa. W rozdziale trzecim przedstawiono istniejące wytyczne do tworzenia AD w trzech językach, angielskim, polskim oraz hiszpańskim. Rozdział praktyczny stanowi analizę strategii audiodeskryberów w skryptach do filmów oraz porównanie ich rozwiązań do wytycznych omówionych w rozdziale trzecim. Pracę kończy podsumowanie badania oraz wnioski.This work is a research on strategies towards film locations in audio description, based on film scripts: Girl with a Pearl Earring, The Sentinel (in English); To Rome with Love, Ocho Apellidos Vascos (in Spanish); Vicky Cristina Barcelona (in Polish).The first chapter of work presents what audio description is. Next chapter is dedicated to research which aims to prove that audio description can be treated as a field of Translation Studies. Chapter three presents the existing styleguides for AD in three languages: English, Polish and Spanish. Practical part of the work is the analysis of strategies applied by describers in scripts for the films and the comparison of their solutions to the standards presented in the third chapter. The work ends with conclusion on the analysed topic
