    Decreased GABA receptor in the cerebral cortex of epileptic rats: effect of Bacopa monnieri and Bacoside-A

    <p>Abstact</p> <p>Background</p> <p>Gamma amino butyric acid (GABA), the principal inhibitory neurotransmitter in the cerebral cortex, maintains the inhibitory tones that counter balances neuronal excitation. When this balance is perturbed, seizures may ensue.</p> <p>Methods</p> <p>In the present study, alterations of the general GABA, GABA<sub>A </sub>and GABA<sub>B </sub>receptors in the cerebral cortex of the epileptic rat and the therapeutic application of <it>Bacopa monnieri </it>were investigated.</p> <p>Results</p> <p>Scatchard analysis of [<sup>3</sup>H]GABA, [<sup>3</sup>H]bicuculline and [<sup>3</sup>H]baclofen in the cerebral cortex of the epileptic rat showed significant decrease in B<sub>max </sub>(P < 0.001) compared to control. Real Time PCR amplification of GABA receptor subunits such as GABA<sub>Aά1</sub>, GABA<sub>Aγ</sub>, GABA<sub>Aδ</sub>, GABA<sub>B </sub>and GAD where down regulated (P < 0.001) in epileptic rats. GABA<sub>Aά5 </sub>subunit and Cyclic AMP responsible element binding protein were up regulated. Confocal imaging study confirmed the decreased GABA receptors in epileptic rats. Epileptic rats have deficit in radial arm and Y maze performance.</p> <p>Conclusions</p> <p><it>Bacopa monnieri </it>and Bacoside-A treatment reverses epilepsy associated changes to near control suggesting that decreased GABA receptors in the cerebral cortex have an important role in epileptic occurrence; <it>Bacopa monnieri </it>and Bacoside-A have therapeutic application in epilepsy management.</p

    Constraints on the nature of dust particles by infrared observations

    The far-infrared (FIR) emissivity of dust is an important parameter characterizing the physical properties of the grains. With the availability of stellar databases and far-infrared data from Infrared Space Observatory (ISO) it is possible to compare the optical and infrared properties of dust, and derive the far-infrared emissivity with respect to the optical extinction. In this paper we present the results of a systematic analysis of the FIR emissivity of interstellar clouds observed with ISOPHOT (the photometer onboard ISO) at least at two infrared wavelengths, one close to ~100um and one at 200um. We constructed FIR emission maps, determined dust temperatures, created extinction maps using 2MASS survey data, and calculated far-infrared emissivity for each of these clouds. We present the largest homogeneously reduced database constructed so far for this purpose. During the data analysis special care was taken on possible systematic errors. We find that far-infrared emissivity has a clear dependence on temperature. The emissivity is enhanced by a factor of usually less than 2 in the low dust temperature regime of 12K<=T_d<=14K. This result suggests larger grain sizes in those regions. However, the emissivity increase of typically below 2 restricts the possible grain growth processes to ice-mantle formation and coagulation of silicate grains, and excludes the coagulation of carbonaceous particles on the scales of the regions we investigated. In the temperature range 14K<=T_d<=16K a systematic decrease of emissivity is observed with respect to the values of the diffuse interstellar matter.     Alterations in hippocampal serotonergic and INSR function in streptozotocin induced diabetic rats exposed to stress: neuroprotective role of pyridoxine and Aegle marmelose

    Diabetes and stress stimulate hippocampal 5-HT synthesis, metabolism and release. The present study was carried out to find the effects of insulin, Aegle marmelose alone and in combination with pyridoxine on the hippocampal 5-HT, 5-HT2A receptor subtype, gene expression studies on 5-HT2A, 5-HTT, INSR, immunohistochemical studies and elevated plus maze in streptozotocin induced diabetic rats. 5-HT content showed a significant decrease (p < 0.001) and a significant increase (p < 0.001) in 5-HIAA in hippocampus of diabetic rats compared to control. 5-HT receptor binding parameters Bmax and Kd showed a significant decrease (p < 0.001) whereas 5-HT2A receptor binding parameters Bmax showed a significant decrease (p < 0.001) with a significant increase (p < 0.05) in Kd in hippocampus of diabetic rats compared to control. Gene expression studies of 5-HT2A, 5-HTT and INSR in hippocampus showed a significant down regulation (p < 0.001) in diabetic rats compared to control. Pyridoxine treated in combination with insulin and A. marmelose to diabetic rats reversed the 5-HT content, Bmax , Kd of 5-HT, 5-HT2A and gene expression of 5-HT2A, 5-HTT and INSR in hippocampus to near control. The gene expression of 5-HT2A and 5-HTT were confirmed by immunohistochemical studies. Behavioural studies using elevated plus maze showed that serotonin through its transporter significantly increased (p < 0.001) anxiety-related traits in diabetic rats which were corrected by combination therapy. Our results suggest that pyridoxine treated in combination with insulin and A. marmelose has a role in the regulation of insulin synthesis and release, normalising diabetic related stress and anxiety through hippocampal serotonergic function. This has clinical significance in the management of diabetes

    A Simple PCR Method for Rapid Genotype Analysis of the TH-MYCN Transgenic Mouse

    BACKGROUND: The TH-MYCN transgenic mouse is the most widely used murine model of human neuroblastoma, in which a human MYCN oncogene is targeted to neuroectodermal cells of developing mice under the influence of the rat tyrosine hydroxylase promoter. So far, homozygous transgenic mice have been identified by either Southern blot or quantitative real-time PCR. PRINCIPAL FINDINGS: To establish a simple and reliable genotyping method by conventional PCR, we confirmed the integration of the transgene in the TH-MYCN transgenic mouse by Southern blot and inverse PCR analyses. Our results showed that either five or six copies were found to be inserted in a head-to-tail tandem configuration at a single locus. The MYCN transgene/host DNA junction was sequenced and the integration site was identified at chromosome 18qE4. Finally, we succeeded in designing rapid, simple and reliable genotyping method by common PCR using primers flanking the integrated TH-MYCN transgene. CONCLUSION: We established a simple and reliable genotyping PCR method for determining the integration site of the TH-MYCN transgene that enables all possible genotypes to be distinguished within several hours. TH-MYCN mice are excellent model for human neuroblastoma study, thus our results will largely be useful for facilitating the pace of neuroblastoma study, including in the study of the tumourigenic process, and in the development of therapies to treat patients suffering from neuroblastoma

    A Resolved Debris Disk Around the Candidate Planet-hosting Star HD 95086

    Recently, a new planet candidate was discovered on direct images around the young (10-17 Myr) A-type star HD 95086. The strong infrared excess of the system indicates that, similar to HR8799, Beta Pic, and Fomalhaut, the star harbors a circumstellar disk. Aiming to study the structure and gas content of the HD 95086 disk, and to investigate its possible interaction with the newly discovered planet, here we present new optical, infrared, and millimeter observations. We detected no CO emission, excluding the possibility of an evolved gaseous primordial disk. Simple blackbody modeling of the spectral energy distribution suggests the presence of two spatially separate dust belts at radial distances of 6 and 64 AU. Our resolved images obtained with the Herschel Space Observatory reveal a characteristic disk size of approx. 6.0 5.4 (540 490 AU) and disk inclination of approx 25 deg. Assuming the same inclination for the planet candidate's orbit, its reprojected radial distance from the star is 62 AU, very close to the blackbody radius of the outer cold dust ring. The structure of the planetary system at HD 95086 resembles the one around HR8799. Both systems harbor a warm inner dust belt and a broad colder outer disk and giant planet(s) between the two dusty regions. Modeling implies that the candidate planet can dynamically excite the motion of planetesimals even out to 270 AU via their secular perturbation if its orbital eccentricity is larger than about 0.4. Our analysis adds a new example to the three known systems where directly imaged planet(s) and debris disks coexist

    Eosinophils Are Important for Protection, Immunoregulation and Pathology during Infection with Nematode Microfilariae

    Eosinophil responses typify both allergic and parasitic helminth disease. In helminthic disease, the role of eosinophils can be both protective in immune responses and destructive in pathological responses. To investigate whether eosinophils are involved in both protection and pathology during filarial nematode infection, we explored the role of eosinophils and their granule proteins, eosinophil peroxidase (EPO) and major basic protein-1 (MBP-1), during infection with Brugia malayi microfilariae. Using eosinophil-deficient mice (PHIL), we further clarify the role of eosinophils in clearance of microfilariae during primary, but not challenge infection in vivo. Deletion of EPO or MBP-1 alone was insufficient to abrogate parasite clearance suggesting that either these molecules are redundant or eosinophils act indirectly in parasite clearance via augmentation of other protective responses. Absence of eosinophils increased mast cell recruitment, but not other cell types, into the broncho-alveolar lavage fluid during challenge infection. In addition absence of eosinophils or EPO alone, augmented parasite-induced IgE responses, as measured by ELISA, demonstrating that eosinophils are involved in regulation of IgE. Whole body plethysmography indicated that nematode-induced changes in airway physiology were reduced in challenge infection in the absence of eosinophils and also during primary infection in the absence of EPO alone. However lack of eosinophils or MBP-1 actually increased goblet cell mucus production. We did not find any major differences in cytokine responses in the absence of eosinophils, EPO or MBP-1. These results reveal that eosinophils actively participate in regulation of IgE and goblet cell mucus production via granule secretion during nematode-induced pathology and highlight their importance both as effector cells, as damage-inducing cells and as supervisory cells that shape both innate and adaptive immunity

    STK295900, a Dual Inhibitor of Topoisomerase 1 and 2, Induces G<inf>2</inf> Arrest in the Absence of DNA Damage

    STK295900, a small synthetic molecule belonging to a class of symmetric bibenzimidazoles, exhibits antiproliferative activity against various human cancer cell lines from different origins. Examining the effect of STK295900 in HeLa cells indicates that it induces G2 phase arrest without invoking DNA damage. Further analysis shows that STK295900 inhibits DNA relaxation that is mediated by topoisomerase 1 (Top 1) and topoisomerase 2 (Top 2) in vitro. In addition, STK295900 also exhibits protective effect against DNA damage induced by camptothecin. However, STK295900 does not affect etoposide-induced DNA damage. Moreover, STK295900 preferentially exerts cytotoxic effect on cancer cell lines while camptothecin, etoposide, and Hoechst 33342 affected both cancer and normal cells. Therefore, STK295900 has a potential to be developed as an anticancer chemotherapeutic agent. © 2013 Kim et al
