    Functional Sites Induce Long-Range Evolutionary Constraints in Enzymes

    Functional residues in proteins tend to be highly conserved over evolutionary time. However, to what extent functional sites impose evolutionary constraints on nearby or even more distant residues is not known. Here, we report pervasive conservation gradients toward catalytic residues in a dataset of 524 distinct enzymes: evolutionary conservation decreases approximately linearly with increasing distance to the nearest catalytic residue in the protein structure. This trend encompasses, on average, 80% of the residues in any enzyme, and it is independent of known structural constraints on protein evolution such as residue packing or solvent accessibility. Further, the trend exists in both monomeric and multimeric enzymes and irrespective of enzyme size and/or location of the active site in the enzyme structure. By contrast, sites in protein–protein interfaces, unlike catalytic residues, are only weakly conserved and induce only minor rate gradients. In aggregate, these observations show that functional sites, and in particular catalytic residues, induce long-range evolutionary constraints in enzymes.Fil: Jack, Benjamin R..     Control of large space structures

    The control of large space structures was studied to determine what, if any, limitations are imposed on the size of spacecraft which may be controlled using current control system design technology. Using a typical structure in the 35 to 70 meter size category, a control system design that used actuators that are currently available was designed. The amount of control power required to maintain the vehicle in a stabilized gravity gradient pointing orientation that also damped various structural motions was determined. The moment of inertia and mass properties of this structure were varied to verify that stability and performance were maintained. The study concludes that the structure's size is required to change by at least a factor of two before any stability problems arise. The stability margin that is lost is due to the scaling of the gravity gradient torques (the rigid body control) and as such can easily be corrected by changing the control gains associated with the rigid body control. A secondary conclusion from the study is that the control design that accommodates the structural motions (to damp them) is a little more sensitive than the design that works on attitude control of the rigid body only

    ANS hard X-ray experiment development program

    The hard X-ray (HXX) experiment is one of three experiments included in the Dutch Astronomical Netherlands Satellite, which was launched into orbit on 30 August 1974. The overall objective of the HXX experiment is the detailed study of the emission from known X-ray sources over the energy range 1.5-30keV. The instrument is capable of the following measurements: (1) spectral content over the full energy range with an energy resolution of approximately 20% and time resolution down to 4 seconds; (2) source time variability down to 4 milliseconds; (3) silicon emission lines at 1.86 and 2.00keV; (4) source location to a limit of one arc minute in ecliptic latitude; and (5) spatial structure with angular resolution of the arc minutes.     Semi-analytical dark matter halos and the Jeans equation

    Although N-body studies of dark matter halos show that the density profiles, rho(r), are not simple power-laws, the quantity rho/sigma^3, where sigma(r) is the velocity dispersion, is in fact a featureless power-law over ~3 decades in radius. In the first part of the paper we demonstrate, using the semi-analytic Extended Secondary Infall Model (ESIM), that the nearly scale-free nature of rho/sigma^3 is a robust feature of virialized halos in equilibrium. By examining the processes in common between numerical N-body and semi-analytic approaches, we argue that the scale-free nature of rho/sigma^3 cannot be the result of hierarchical merging, rather it must be an outcome of violent relaxation. The empirical results of the first part of the paper motivate the analytical work of the second part of the paper, where we use rho/sigma^3 proportional to r^{-alpha} as an additional constraint in the isotropic Jeans equation of hydrostatic equilibrium. Our analysis shows that the constrained Jeans equation has different types of solutions, and in particular, it admits a unique ``periodic'' solution with alpha=1.9444. We derive the analytic expression for this density profile, which asymptotes to inner and outer profiles of rho ~ r^{-0.78}, and rho ~ r^{-3.44}, respectively.Comment: 37 pg, 14 fig.     The principal effort under this program was directed at the fuel cell stack technology required to accomplish the initial feasibility demonstrations of increased cell stack operating pressures and temperatures, increased cell active area, incorporation of the ribbed substrate cell configuration at the bove conditions, and the introduction of higher performance electrocatalysts. The program results were successful with the primary accomplishments being: (1) fabrication of 10 sq ft ribbed substrate, cell components including higher performing electrocatalysts; (2) assembly of a 10 sq ft, 30-cell short stack; and (3) initial test of this stack at 120 psia and 405 F. These accomplishments demonstrate the feasibility of fabricating and handling large area cells using materials and processes that are oriented to low cost manufacture. An additional accomplishment under the program was the testing of two 3.7 sq ft short stacks at 12 psia/405 F to 5400 and 4500 hours respectively. These tests demonstrate the durability of the components and the cell stack configuration to a nominal 5000 hours at the higher pressure and temperature condition planned for the next electric utility power plant
