Pressure is not a state function for generic active fluids
Pressure is the mechanical force per unit area that a confined system exerts
on its container. In thermal equilibrium, it depends only on bulk properties
(density, temperature, etc.) through an equation of state. Here we show that in
a wide class of active systems the pressure depends on the precise interactions
between the active particles and the confining walls. In general, therefore,
active fluids have no equation of state, their mechanical pressures exhibit
anomalous properties that defy the familiar thermodynamic reasoning that holds
in equilibrium. The pressure remains a function of state, however, in some
specific and well-studied active models that tacitly restrict the character of

Actin Polymerization Controls the Organization of WASH Domains at the Surface of Endosomes
Sorting of cargoes in endosomes occurs through their selective enrichment into sorting platforms, where transport intermediates are generated. The WASH complex, which directly binds to lipids, activates the Arp2/3 complex and hence actin polymerization onto such sorting platforms. Here, we analyzed the role of actin polymerization in the physiology of endosomal domains containing WASH using quantitative image analysis. Actin depolymerization is known to enlarge endosomes. Using a novel colocalization method that is insensitive to the heterogeneity of size and shape of endosomes, we further show that preventing the generation of branched actin networks induces endosomal accumulation of the WASH complex. Moreover, we found that actin depolymerization induces a dramatic decrease in the recovery of endosomal WASH after photobleaching. This result suggests a built-in turnover, where the actin network, i.e. the product of the WASH complex, contributes to the dynamic exchange of the WASH complex by promoting its detachment from endosomes. Our experiments also provide evidence for a role of actin polymerization in the lateral compartmentalization of endosomes: several WASH domains exist at the surface of enlarged endosomes, however, the WASH domains coalesce upon actin depolymerization or Arp2/3 depletion. Branched actin networks are thus involved in the regulation of the size of WASH domains. The potential role of this regulation in membrane scission are discussed
Topological sound in active-liquid metamaterials
Liquids composed of self-propelled particles have been experimentally
realized using molecular, colloidal, or macroscopic constituents. These active
liquids can flow spontaneously even in the absence of an external drive. Unlike
spontaneous active flow, the propagation of density waves in confined active
liquids is not well explored. Here, we exploit a mapping between density waves
on top of a chiral flow and electrons in a synthetic gauge field to lay out
design principles for artificial structures termed topological active
metamaterials. We design metamaterials that break time-reversal symmetry using
lattices composed of annular channels filled with a spontaneously flowing
active liquid. Such active metamaterials support topologically protected sound
modes that propagate unidirectionally, without backscattering, along either
sample edges or domain walls and despite overdamped particle dynamics. Our work
illustrates how parity-symmetry breaking in metamaterial structure combined
with microscopic irreversibility of active matter leads to novel
functionalities that cannot be achieved using only passive materials
TNF-alpha TRANSIENTLY INDUCES ENDOPLASMIC RETICULUM STRESS AND AN INCOMPLETE UNFOLDED PROTEIN RESPONSE IN THE HYPOTHALAMUS

Toll-like receptor 4 (TLR4) activation plays a central role in this process, inducing endoplasmic reticulum stress and activating inflammatory cytokine gene transcription. Although saturated fatty acids can induce endoplasmic reticulum stress in the hypothalamus, it is unknown whether inflammatory cytokines alone can activate this mechanism. Here, rats were treated with TNF-alpha or lyposaccharide (LPS) and endoplasmic reticulum stress and unfolded protein response were evaluated by immunoblot and polymerase chain reaction (PCR). Activation of TLR4 by LPS was capable of inducing a complete endoplasmic reticulum stress and unfolded protein response through the PERK/eIF2 alpha and IRE1 alpha/XBP1 pathways. Conversely, TNF-a, injected either locally or systemically, was unable to induce a complete program of unfolded protein response, although the activation of endoplasmic reticulum stress was achieved to a certain degree. Thus, in the hypothalamus, the isolated action of TNF-alpha is insufficient to produce the activation of a complete program of unfolded protein response. (C) 2010 IBRO. Published by Elsevier Ltd. Taurine enhances the anorexigenic effects of insulin in the hypothalamus of rats

Recent data have suggested that taurine can also reduce body adiposity in and in rodents. Since body adiposity is mostly regulated by insulin-responsive hypothalamic neurons involved in the control of feeding and thermogenesis, we hypothesized that some of the activity of taurine in the control of body fat would be exerted through a direct action in the hypothalamus. Here, we show that the intracerebroventricular injection of an acute dose of taurine reduces food intake and locomotor activity, and activates signal transduction through the Akt/FOXO1, JAK2/STAT3 and mTOR/AMPK/ACC signaling pathways. These effects are accompanied by the modulation of expression of NPY. In addition, taurine can enhance the anorexigenic action of insulin.
