Exploring Cluster Analysis in Nelore Cattle Visual Score Attribution
Assessing the biotype of cattle through human visual inspection is a very
common and important practice in precision cattle breeding. This paper presents
the results of a correlation analysis between scores produced by humans for
Nelore cattle and a variety of measurements that can be derived from images or
other instruments. It also presents a study using the k-means algorithm to
generate new ways of clustering a batch of cattle using the measurements that
most correlate with the animal's body weight and visual scores

O HIPOTÁLAMO DORSOMEDIAL E A ORGANIZAÇÃO DA RESPOSTA CARDIOVASCULAR AO ESTRESSE EMOCIONAL: UMA PERSPECTIVA FUNCIONAL
Emotional stress results in activation of specific pathways into central nervous system, which produces autonomic, behavioral and endocrine responses. It is known that repetitive or continuous exposition to stress situations may result in various pathologic states, for example, arterial hypertension. Classically, the hypothalamus plays an essential role in the integration of physiological responses to emotional stress. Recent studies demonstrate that a specific nucleus from hypothalamus, the dorsomedial hypothalamus (DMH), is an essential component of central pathways that mediate the cardiovascular response to emotional stress. Inhibition of neurons in this area reduces the increases in heart rate and blood pressure in rats submitted to emotional stress paradigms. Conversely, pharmacological activation of DMH neurons evokes increases on heart rate, blood pressure, adrenocorticotrophic hormone, locomotor activity and sympathetic activity. The similarity of this response with that produced during emotional stress suggests that this area is crucial in the integration of the physiological responses to emotional stress. The present review will discuss the central pathways used by DMH in the organization of the cardiovascular response to emotional stress.O estresse emocional resulta em ativação de vias específicas do sistema nervoso central, que produzem respostas autonômicas, comportamentais e endócrinas. Sabe-se que situações de estresse recorrentes ou prolongadas podem resultar em vários estados patológicos, como por exemplo, a hipertensão arterial. O Hipotálamo tem papel fundamental na integração das respostas fisiológicas ao estresse emocional. Particularmente, estudos têm mostrado que um núcleo específico do hipotálamo, o hipotálamo dorsomedial (DMH), é um componente fundamental das vias centrais mediadoras das respostas cardiovasculares ao estresse emocional. A inibição dos neurônios dessa área reduz os aumentos de freqüência cardíaca e de pressão arterial em ratos quando submetidos à situações de estresse emocional. Ao contrário, a ativação farmacológica dos neurônios do DMH produz aumento na frequência cardíaca, pressão arterial, hormônio adrenocorticotrópico (ACTH), atividade locomotora e na atividade simpática para diversos leitos vasculares. A similaridade dessa resposta com aquela produzida durante a situação real de estresse emocional sugere que esta área é fundamental na integração da resposta fisiológica ao estresse. Prevalence of hazardous alcohol use among Spanish primary care providers
Background: Alcohol use by health care professionals is one of the potential factors that may affect the prevention
of hazardous drinking in Primary Care (PC). The objective of the study was to estimate the prevalence of hazardous
alcohol use by PC professionals and assess the existing relationship between socio-demographic and occupational
variables of PC professionals and their alcohol use.
Methods: A descriptive, cross-sectional, observational, multicenter study was performed. Location: PC sites of
the Spanish National Health Care System (NHS). Participants: Physicians and nurses, who completed an online
questionnaire intended to identify the pattern of hazardous alcohol use through the AUDIT-C test. The study
population was recruited through random sampling stratified by regions of the PC sites in the NHS. The
primary measurements: Frequency of alcohol use, number of drinks containing alcohol on a typical day,
frequency of six or more drinks on one occasion.
Results: One thousand seven hundred sixty professionals completed the questionnaire. Hazardous alcohol use was
detected in 27.80% (95% CI: 25.5–29.7) of PC providers. The prevalence of hazardous alcohol use was higher in males
(34.2%) [95% CI: 30.4–37.6] and professionals aged 56 years or over (34.2%) [95% CI: 28.2–40.2]. The multiple logistic
regression analysis revealed a higher hazardous use in males (OR = 1.52; 95% CI: 1.22–1.90), PC physicians (OR = 1.42;
95% CI: 1.01–2.02) and professionals with more time worked (OR = 1.03; 95% CI: 1.01–1.05).
Conclusion: Our study shows the current prevalence of hazardous alcohol use among Spanish PC providers, revealing
a higher percentage of hazardous alcohol use in healthcare professionals compared to the Spanish general population.
Further interventions are required to increase the awareness of negative consequences derived from alcohol use among PC professionals and its impact on the clinical settingThe study has been financed by the Spanish Society of Family and

Acute Auditory Stimulation with Different Styles of Music Influences Cardiac Autonomic Regulation in Men
Background: No clear evidence is available in the literature regarding the acute effect of different styles of music on cardiac autonomic control. Objectives: The present study aimed to evaluate the acute effects of classical baroque and heavy metal musical auditory stimulation on Heart Rate Variability (HRV) in healthy men. Patients and Methods: In this study, HRV was analyzed regarding time (SDNN, RMSSD, NN50, and pNN50) and frequency domain (LF, HF, and LF / HF) in 12 healthy men. HRV was recorded at seated rest for 10 minutes. Subsequently, the participants were exposed to classical baroque or heavy metal music for five minutes through an earphone at seated rest. After exposure to the first song, they remained at rest for five minutes and they were again exposed to classical baroque or heavy metal music. The music sequence was random for each individual. Standard statistical methods were used for calculation of means and standard deviations. Besides, ANOVA and Friedman test were used for parametric and non-parametric distributions, respectively. Results: While listening to heavy metal music, SDNN was reduced compared to the baseline (P = 0.023). In addition, the LF index (ms2 and nu) was reduced during exposure to both heavy metal and classical baroque musical auditory stimulation compared to the control condition (P = 0.010 and P = 0.048, respectively). However, the HF index (ms2) was reduced only during auditory stimulation with music heavy metal (P = 0.01). The LF/HF ratio on the other hand decreased during auditory stimulation with classical baroque music (P = 0.019). Conclusions: Acute auditory stimulation with the selected heavy metal musical auditory stimulation decreased the sympathetic and parasympathetic modulation on the heart, while exposure to a selected classical baroque music reduced sympathetic regulation on the heart
Superpulsed low-level laser therapy protects skeletal muscle of mdx mice against damage, inflammation and morphological changes delaying dystrophy progression.
Aim: To evaluate the effects of preventive treatment with low-level laser therapy (LLLT) on progression of dystrophy in mdx mice. Methods: Ten animals were randomly divided into 2 experimental groups treated with superpulsed LLLT (904 nm, 15 mW, 700 Hz, 1 J) or placebo-LLLT at one point overlying the tibialis anterior muscle (bilaterally) 5 times per week for 14 weeks (from 6th to 20th week of age). Morphological changes, creatine kinase (CK) activity and mRNA gene expression were assessed in animals at 20th week of age. Results: Animals treated with LLLT showed very few morphological changes in skeletal muscle, with less atrophy and fibrosis than animals treated with placebo-LLLT. CK was significantly lower (p = 0.0203) in animals treated with LLLT (864.70 U.l−1, SEM 226.10) than placebo (1708.00 U.l−1, SEM 184.60). mRNA gene expression of inflammatory markers was significantly decreased by treatment with LLLT (p<0.05): TNF-α (placebo-control = 0.51 µg/µl [SEM 0.12], - LLLT = 0.048 µg/µl [SEM 0.01]), IL-1β (placebo-control = 2.292 µg/µl [SEM 0.74], - LLLT = 0.12 µg/µl [SEM 0.03]), IL-6 (placebo-control = 3.946 µg/µl [SEM 0.98], - LLLT = 0.854 µg/µl [SEM 0.33]), IL-10 (placebo-control = 1.116 µg/µl [SEM 0.22], - LLLT = 0.352 µg/µl [SEM 0.15]), and COX-2 (placebo-control = 4.984 µg/µl [SEM 1.18], LLLT = 1.470 µg/µl [SEM 0.73]). Conclusion: Irradiation of superpulsed LLLT on successive days five times per week for 14 weeks decreased morphological changes, skeletal muscle damage and inflammation in mdx mice. This indicates that LLLT has potential to decrease progression of Duchenne muscular dystrophy
Pituitary tumor-transforming gene expression is a prognostic marker for tumor recurrence in squamous cell carcinoma of the head and neck
BACKGROUND: The proto-oncogene pituitary tumor-transforming gene (PTTG) has been shown to be abundantly overexpressed in a large variety of neoplasms likely promoting neo-vascularization and tumor invasiveness. In this study, we investigated a potential role for PTTG mRNA expression as a marker to evaluate the future clinical outcome of patients diagnosed with primary cancer of the head and neck. METHODS: Tumor samples derived from primary tumors of 89 patients suffering from a squamous cell carcinoma were analyzed for PTTG mRNA-expression and compared to corresponding unaffected tissue. Expression levels were correlated to standard clinico-pathological parameters based on a five year observation period. RESULTS: In almost all 89 tumor samples PTTG was found to be overexpressed (median fold increase: 2.1) when compared to the unaffected tissue specimens derived from the same patient. The nodal stage correlated with PTTG transcript levels with significant differences between pN0 (median expression: 1.32) and pN+ (median expression: 2.12; P = 0.016). In patients who developed a tumor recurrence we detected a significantly higher PTTG expression in primary tumors (median expression: 2.63) when compared to patients who did not develop a tumor recurrence (median expression: 1.29; P = 0.009). Since the median expression of PTTG in patients with tumor stage T1/2N0M0 that received surgery alone without tumor recurrence was 0.94 versus 3.82 in patients suffering from a tumor recurrence (P = 0.006), PTTG expression might provide a feasible mean of predicting tumor recurrence. CONCLUSION: Elevated PTTG transcript levels might be used as a prognostic biomarker for future clinical outcome (i.e. recurrence) in primary squamous cell carcinomas of the head and neck, especially in early stages of tumor development
Establishing an infrastructure for collaboration in primate cognition research
Inferring the evolutionary history of cognitive abilities requires large and diverse samples. However, such samples are often beyond the reach of individual researchers or institutions, and studies are often limited to small numbers of species. Consequently, methodological and site-specific-differences across studies can limit comparisons between species. Here we introduce the ManyPrimates project, which addresses these challenges by providing a large-scale collaborative framework for comparative studies in primate cognition. To demonstrate the viability of the project we conducted a case study of short-term memory. In this initial study, we were able to include 176 individuals from 12 primate species housed at 11 sites across Africa, Asia, North America and Europe. All subjects were tested in a delayed-response task using consistent methodology across sites. Individuals could access food rewards by remembering the position of the hidden reward after a 0, 15, or 30-second delay. Overall, individuals performed better with shorter delays, as predicted by previous studies. Phylogenetic analysis revealed a strong phylogenetic signal for short-term memory. Although, with only 12 species, the validity of this analysis is limited, our initial results demonstrate the feasibility of a large, collaborative open-science project. We present the ManyPrimates project as an exciting opportunity to address open questions in primate cognition and behaviour with large, diverse datasets
Red wine and components flavonoids inhibit UGT2B17 in vitro
Background
The metabolism and excretion of the anabolic steroid testosterone occurs by glucuronidation to the conjugate testosterone glucuronide which is then excreted in urine. Alterations in UGT glucuronidation enzyme activity could alter the rate of testosterone excretion and thus its bioavailability. The aim of this study is to investigate if red wine, a common dietary substance, has an inhibitory effect on UGT2B17.
Methods
Testosterone glucuronidation was assayed using human UGT2B17 supersomes with quantification of unglucuronidated testosterone over time using HPLC with DAD detection. The selected red wine was analysed using HPLC and the inhibitory effects of the wine and phenolic components were tested independently in a screening assay. Further analyses were conducted for the strongest inhibitors at physiologically relevant concentrations. Control experiments were conducted to determine the effects of the ethanol on UGT2B17.
Results
Over the concentration range of 2 to 8% the red wine sample inhibited the glucuronidation of testosterone by up to 70% over 2 hours. The ethanol content had no significant effect. Three red wine phenolics, identified by HLPC analyses, also inhibited the enzyme by varying amounts in the order of quercetin (72%), caffeic acid (22%) and gallic acid (9%); using a ratio of phenolic:testosterone of 1:2.5. In contrast p-coumaric acid and chlorogenic acid had no effect on the UGT2B17. The most active phenolic was selected for a detailed study at physiologically relevant concentrations, and quercetin maintained inhibitory activity of 20% at 2 M despite a ten-fold excess of testosterone.
Conclusion
This study reports that in an in vitro supersome-based assay, the key steroid-metabolising enzyme UGT2B17 is inhibited by a number of phenolic dietary substances and therefore may reduce the rate of testosterone glucuronidation in vivo. These results highlight the potential interactions of a number of common dietary compounds on testosterone metabolism. Considering the variety of foodstuffs that contain flavonoids, it is feasible that diet can elevate levels of circulating testosterone through reduction in urinary excretion.
