    Differential potency of vitamin D3, folic acid and memantine in protecting against neurobehavioral alterations of scopolamine induced Alzheimer’s model in rats

    Background: Alzheimer disease is the cause of 60% to 70% of cases of dementia in elderly people, it is a chronic neurodegenerative disease that usually starts slowly and worsens over time. AD is characterized by the presence of senile plaques enriched with insoluble aggregate of beta-amyloid, neurofibrillary tangles and cholinergic neuronal degeneration in the brain tissue, leading to neural dysfunction, neuroinflammation, and critical pathological perturbations.Methods: Thirty-six males were classified into control group, Alzheimer-induced model (scopolamine 2.5 mg/kg IP once daily for 21 days). Folic acid-treated group (4 mg/kg, IP) once daily for 21 days with scopolamine. Vitamin D3-treated group (42 IU/kg, SC) once daily for 21 days with scopolamine. Vitamin D3 and folic acid-treated group (vitamin D3; 42 IU/kg, SC and folic acid; 4 mg/kg, IP) once daily with scopolamine for 21 days. Memantine-treated group (20 mg/kg IP) once daily with scopolamine for 21 days.Results: Induction of Alzheimer’s showed significant decrease in brain tissue levels of BDNF, Ach, glutathione reductase and significant increase in amyloid peptide 1-42 level with significant memory impairment, significant increase of initial acquisition latency, firstt retention latency and second retention latency. While administration of folic acid, vitamin D3, memantine separately or in combination resulted insignificant increase of brain tissue levels of BDNF, Ach, glutathione reductase with significant reduction of amyloid peptide 1-42 level with significant memory improvement (significant decrease IAL, first RL and second RL). Also showed improvement of histopathological changes occurred in the brain.Conclusions: Data obtained in the present study revealed that treatment of experimentally induced alzheimer rats with folic acid or vitamin D3 or memantine separately or combined group (folic acid+vitamin D3) resulted in significant increase of brain tissue levels of BDNF, acetyl choline, glutathione reductase with significant reduction of amyloid peptide 1-42 level with significant decrease of IAL, first RL and second RL to reach the platform with improvement of histopathological changes occurred in the brain. But combined and memantine-treated groups resulted in more significant improvement than other treated groups

    Changes in Physico-Chemical Quality and Volatile Compounds of Orange-Carrot Juice Blends During storage

    The present study aimed to determine the changes in physico-chemical parameters (pH, total soluble solids “TSS”, total acidity, vitamin C, total carotene, total phenolic and antioxidant activity) as well as sensory evaluation and volatile constituents of the orange juice samples mixed with carrot juice at ratios (1:3, 3:1 and 1:1). While, the polyphenol content was determined using Folin–Ciocalteu, antioxidant activity was measured using two in vitro assays 2,2'-diphenyl-1-picrylhydrazyl (DPPH0) and metal chelating assays. While, the acidity increased, total carotene and pH of the juice decreased during the storage period. There were no significant changes in total phenolics throughout the storage period at the three blending ratios. A slight increase in antioxidant capacity during the storage period had been observed. The headspace volatiles of fresh orange and carrot juices as well as fresh and stored blend juice with ratio (1:3) for 14 days at refrigerator were collected and subjected to Gas chromatography (GC) and Gas chromatography-Mass spectrometery (GC-MS) analysis. A total of 26 volatile compounds were identified in samples of fresh orange and carrot juices as well as fresh and stored blend including 7 alcohols, 4 aldehydes, 9 monoterpene hydrocarbon and 6 esters. Limonene was the one most abundant monoterpene, representing in orange, carrot, fresh and stored blend 47.38, 42.37, 39.24 and 37.25%, respectively.     Synthesis and cytotoxicity evaluation of thiazole derivatives obtained from 2-amino-4,5,6,7-tetrahydrobenzo[b]thiophene-3-carbonitrile

    Reactivity of 2-amino-4,5,6,7-tetrahydrobenzo[b]thiophene-3-carbonitrile towards thioglycolic acid resulted in thiazole derivative 1. The latter reacted with different chemical reagents to give thiazole, pyrano[2,3-d]thiazole and thiazolo[4,5-d]thiazole derivatives. Cytotoxicity effects of the newly synthesized products against six cancer cell lines, namely, human gastric cancer (NUGC), human colon cancer (DLD-1), human liver cancer (HA22T and HEPG-2), human breast cancer (MCF) and nasopharyngeal carcinoma (HONE-1) as well as against a normal fibroblast cell (WI-38) were evaluated. The study showed that the 4,5,6,7 tetrahydrobenzo[b]thiophene derivatives 6a, 7, 8a,b, 9b and 10b,c were the most active compounds. Their potencies were attributed to the presence of the electron withdrawing groups

    Identification, Genotyping and Antimicrobial Susceptibility Testing of Brucella spp. Isolated from Livestock in Egypt

    Brucellosis is a highly contagious zoonosis worldwide with economic and public health impacts. The aim of the present study was to identify Brucella (B.) spp. isolated from animal populations located in different districts of Egypt and to determine their antimicrobial resistance. In total, 34-suspected Brucella isolates were recovered from lymph nodes, milk, and fetal abomasal contents of infected cattle, buffaloes, sheep, and goats from nine districts in Egypt. The isolates were identified by microbiological methods and matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS). Differentiation and genotyping were confirmed using multiplex PCR for B. abortus, Brucella melitensis, Brucella ovis, and Brucella suis (AMOS) and Bruce-ladder PCR. Antimicrobial susceptibility testing against clinically used antimicrobial agents (chloramphenicol, ciprofloxacin, erythromycin, gentamicin, imipenem, rifampicin, streptomycin, and tetracycline) was performed using E-Test. The antimicrobial resistance-associated genes and mutations in Brucella isolates were confirmed using molecular tools. In total, 29 Brucella isolates (eight B. abortus biovar 1 and 21 B. melitensis biovar 3) were identified and typed. The resistance of B. melitensis to ciprofloxacin, erythromycin, imipenem, rifampicin, and streptomycin were 76.2%, 19.0%, 76.2%, 66.7%, and 4.8%, respectively. Whereas, 25.0%, 87.5%, 25.0%, and 37.5% of B. abortus were resistant to ciprofloxacin, erythromycin, imipenem, and rifampicin, respectively. Mutations in the rpoB gene associated with rifampicin resistance were identified in all phenotypically resistant isolates. Mutations in gyrA and gyrB genes associated with ciprofloxacin resistance were identified in four phenotypically resistant isolates of B. melitensis. This is the first study highlighting the antimicrobial resistance in Brucella isolated from different animal species in Egypt. Mutations detected in genes associated with antimicrobial resistance unravel the molecular mechanisms of resistance in Brucella isolates from Egypt. The mutations in the rpoB gene in phenotypically resistant B. abortus isolates in this study were reported for the first time in Egypt

    Facial analysis using a new clinical device : The Kattan Facio-meter

    The aim of the study was to attempt to set average faciometric standards for Egyptians using the Kattan Facio-meter. The sample consisted of 180 faculty students with age range 17-25 years. It was divided into three groups; Angle Class I, II and III. Class II was further subdivided into divisions 1 and 2. Linear and angular facial measurements in relation to K plane were taken using the Kattan facio-meter. The measurements were correlated to Angle?s classification and between genders. On comparing the different classes, Class II division 1 showed the statistically highest mean value for Orbitale-soft tissue A; p=0.042, Class II divisions 1 and 2 for Orbitale- Labrale superius; p=0.002 and soft tissue ANB; p<0.001. Females showed significantly higher mean value than males for the upper incisor/K plane; p=0.031. Males showed significantly higher mean value for the inter-incisal angle than females; p=0.001. Within the limitations of the current study, it was found that both linear and angular soft tissue measurements conformed to the antroposterior skeletal relation of the jaws and that Class II division 1 was due to protruded maxilla. Males had more prominent lips and deeper mentolabial sulcus. Egyptians had less prominent noses than Caucasians. The Kattan Facio-meter was a valuable tool for clinical analysis without the hazards of irradiation

    Groundwater Recharge Potentiality Mapping in Wadi Qena, Eastern Desert Basins of Egypt for Sustainable Agriculture Base Using Geomatics Approaches

    In arid and hyper-arid areas, groundwater is a precious and rare resource. The need for water supply has grown over the past few decades as a result of population growth, urbanization, and agricultural endeavors. This research aims to locate groundwater recharge potential zones (GWPZs) using multi-criteria evaluation (MCE) in the Wadi Qena Basin, Eastern Desert of Egypt, which represents one of the most promising valleys on which the government depends for land reclamations and developments. These approaches have been used to integrate and delineate the locations of high groundwater recharge and the potential of the Quaternary aquifer in the Wadi Qena basin. After allocating weight factors to identify features in each case based on infiltration, land use/land cover, slope, geology, topology, soil, drainage density, lineament density, rainfall, flow accumulation, and flow direction, these thematic maps were combined. The results of the GIS modeling led to the division of the area’s groundwater recharge potential into five groups, ranging from very high (in the western part) to very low (in the eastern part of the basin). The zones with the best prospects for groundwater exploration turned out to be the alluvial and flood plains, with their thick strata of sand and gravel. The groundwater recharge potential map was validated using data from the field and earlier investigations. The promising recharging areas show high suitability for soil cultivation. The results overall reveal that RS and GIS methodologies offer insightful instruments for more precise assessment, planning, and monitoring of water resources in arid regions and anywhere with similar setups for groundwater prospecting and management

    The Anticancer Effect of Rhubarb Officinale Fraction and Sorafenib Against Chemically Induced Hepatocellular Carcinoma in Rats

    Background: Hepatocellular carcinoma (HCC), the major primary malignant tumour of the liver, represents a complex and fatal malignancy driven primarily by oxidative stress and inflammation. Objectives: The aim of this study was to investigate the potential anticancer properties of four Anthraquinones (Aloe-emodin, Emodin, Chysophanol, and Physcion) isolated from the rhizome of Rhubarb and compare between the effect of sorafenib and Rhubarb Officinale fraction on DEN and carbon tetrachloride (CCl4) -&nbsp;&nbsp; induced hepatocellular carcinoma in rats. Material and methods: The rats were divided into 5 groups (n=12/group normal control, diethylnitrosamine DEN+ carbon tetrachloride CCL4, HCC rats +ANTH(Anthraquinones), HCC rats +SOR(Sorafenib) and HCC rats +mix of ANTH+SOR. Results: Rats in HCC group showed most deteriorated effect in form of increased mortality, liver cancer marker (lipid peroxidation biomarker MDA, inflammation –related genes (TNFa, ILIB, NFKB, TGFB1) and metastasis-related genes (MMP9). Liver tissues of HCC group also exhibited lower level of TIMPI and antioxidant activity (catalase (CAT). All these deleterious effects induced by DEN were reversed after administration of ANTH and SOR with best improvement for the combined group (ANTH+SOR). Conclusions: These findings reveal a better therapeutic effect for ANTH when given with SOR and we attribute this beneficial effect, at least in part, to triggering antioxidant activity, inflammation and metastasis in HCC. Therefore, combined treatment with ANTH and SOR is recommended to enhance the therapeutic potential against HCC
