Botryodiplodia spp. potentially cause dieback disease on jabon (Anthocephalus cadamba (Roxb.) Miq) seedlings. Five isolates of Botryodiplodia spp were inoculated on jabon seedling to find out its virulence levels, mechanism of pathogen attack and mechanism of host plant defense. The virulence levels was estimated by disease severity of host plants; the pathogen attack mechanism was done by measuring pectinase and cellulase enzyme activities; whereas host plant defense mechanism was determined by measuring peroxidase enzyme activity. The virulent isolates caused disease severity > 50%. Botryodiplodia sp. 1, Botryodiplodia sp. 2, and Botryodiplodia sp. 3 showed pectinase activities of 21.311; 18.131; 26.083 U/ml, and cellulase 0.014; 0.015; 0.023 U/ml, respectively. The peroxidase activity of host plants after pathogen inoculated was ranging from 0.0006 to 0.0012 UAE/g. Based on this research, three Botryodiplodia spp. isolates were virulent on jabon seedlings and involved enzymatic strength as attack mechanism, whereas the host plant defense mechanism involved peroxidase activity

    Get PDF
Background: Despite a high prevalence of nutrition-related health disparities experienced by Latinos in the United States, there is limited recent information that can be used by health professionals to develop effective nutrition interventions for this diverse population. Purpose: The purpose of this study was to obtain qualitative information regarding usual dietary patterns of Latinos from Pomona, California, and utilize this data in the development of a tailored nutrition intervention. Methods: Latina women ages 30- 70, from a low socio-economic community sample, participated in one of three focus groups, and discussed eating patterns, recipes, and dietary concerns. Participants self-identified as the primary household food purchaser and preparer. Demographics and acculturation were assessed using validated surveys. Results: All participants (n=17) were considered less acculturated to the United States. Participants reported that traditional recipes and family preferences have the greatest influence on foods prepared, however they tend to be less healthy items. Motivators for eating healthy included maintaining family health and preventing weight gain. Barriers included conflicting meal schedules among family members and individual food preferences. Conclusion: To reduce health disparities for Latinos, cultural and dietary factors of communities, along with perceived motivators and barriers to making healthy choices, should be considered when developing tailored nutrition interventions

JNK3 is abundant in insulin-secreting cells and protects against cytokine-induced apoptosis

Aims/hypothesis: In insulin-secreting cells, activation of the c-Jun NH2-terminal kinase (JNK) pathway triggers apoptosis. Whereas JNK1 and JNK2 are ubiquitously produced, JNK3 has been described exclusively in neurons. This report aims to characterise the expression and role in apoptosis of the three JNK isoforms in insulin-secreting cells exposed to cytokines. Methods: Sections of human and mouse pancreases were used for immunohistochemistry studies with isoform-specific anti-JNK antibodies. Human, pig, mouse and rat pancreatic islets were isolated by enzymatic digestion and RNA or protein extracts were prepared. RNA and protein levels were determined by quantitative RT-PCR and western blotting respectively, using JNK-isoform-specific primers and isoform-specific antibodies; activities of the three JNK isoforms were determined by kinase assays following quantitative immunoprecipitation/depletion of JNK3. JNK silencing was performed with small interfering RNAs and apoptotic rates were determined in INS-1E cells by scoring cells displaying pycnotic nuclei. Results: JNK3 and JNK2 mRNAs are the predominant isoforms expressed in human pancreatic islets. JNK3 is nuclear while JNK2 is also cytoplasmic. In INS-1E cells, JNK3 knockdown increases c-Jun levels and caspase-3 cleavage and sensitises cells to cytokine-induced apoptosis; in contrast, JNK1 or JNK2 knockdown is protective. Conclusions/interpretation: In insulin-secreting cells, JNK3 plays an active role in preserving pancreatic beta cell mass from cytokine attacks. The specific localisation of JNK3 in the nucleus, its recruitment by cytokines, and its effects on key transcription factors such as c-Jun, indicate that JNK3 is certainly an important player in the transcriptional control of genes expressed in insulin-secreting cell

Quasiparticle properties in a density functional framework

We propose a framework to construct the ground-state energy and density matrix of an N-electron system by solving selfconsistently a set of single-particle equations. The method can be viewed as a non-trivial extension of the Kohn-Sham scheme (which is embedded as a special case). It is based on separating the Green's function into a quasi-particle part and a background part, and expressing only the background part as a functional of the density matrix. The calculated single-particle energies and wave functions have a clear physical interpretation as quasiparticle energies and orbitals.Comment: 12 pages, 1 figure, to be published in Phys. Rev.

Background: Elevated urinary calcium excretion is associated with reduced bone mineral density. Population-based data on urinary calcium excretion are scarce. We explored the association of serum calcium and circulating levels of vitamin D (including 25(OH)D2 and 25(OH)D3) with urinary calcium excretion in men and women in a population-based study. Methods: We used data from the "Swiss Survey on Salt" conducted between 2010 and 2012 and including people aged 15 years and over. Twenty-four hour urine collection, blood analysis, clinical examination and anthropometric measures were collected in 11 centres from the 3 linguistic regions of Switzerland. Vitamin D was measured centrally using liquid chromatography - tandem mass spectrometry. Hypercalciuria was defined as urinary calcium excretion >0.1 mmol/kg/24h. Multivariable linear regression was used to explore factors associated with 24-hour urinary calcium excretion (mmol/24h) squared root transformed, taken as the dependant variable. Vitamin D was divided into monthspecific tertiles with the first tertile having the lowest value and the third tertile having the highest value. Results: The 669 men and 624 women had mean (SD) age of 49.2 (18.1) and 47 (17.9) years and a prevalence of hypercalciuria of 8.9% and 8.0%, respectively. In adjusted models, the association of urinary calcium excretion with protein-corrected serum calcium was (β coefficient } standard error, according to urinary calcium squared root transformed) 1.125 } 0.184 mmol/L per square-root (mmol/24h) (P<0.001) in women and 0.374 } 0.224 (P=0.096) in men. Men in the third month-specific vitamin D tertile had higher urinary calcium excretion than men in the first tertile (0.170 } 0.05 nmol/L per mmol/24h, P=0.001) and the corresponding association was 0.048 } 0.043, P= 0.272 in women. Conclusion: About one in eleven person has hypercalciuria in the Swiss population. The positive association of serum calcium with urinary calcium excretion was steeper in women than in men, independently of menopausal status. Circulating vitamin D was associated positively with urinary calcium excretion only in men. The reasons underlying the observed sex differences in the hormonal control of urinary calcium excretion need to be explored in further studies

Sentiment analysis tools should take account of the number of exclamation marks!!!

There are various factors that affect the sentiment level expressed in textual comments. Capitalization of letters tends to mark something for attention and repeating of letters tends to strengthen the emotion. Emoticons are used to help visualize facial expressions which can affect understanding of text. In this paper, we show the effect of the number of exclamation marks used, via testing with twelve online sentiment tools. We present opinions gathered from 500 respondents towards “like” and “dislike” values, with a varying number of exclamation marks. Results show that only 20% of the online sentiment tools tested considered the number of exclamation marks in their returned scores. However, results from our human raters show that the more exclamation marks used for positive comments, the more they have higher “like” values than the same comments with fewer exclamations marks. Similarly, adding more exclamation marks for negative comments, results in a higher “dislike”

    Seed systems smallholder farmers use

    Get PDF
Seed can be an important entry point for promoting productivity, nutrition and resilience among smallholder farmers. While investments have primarily focused on strengthening the formal sector, this article documents the degree to which the informal sector remains the core for seed acquisition, especially in Africa. Conclusions drawn from a uniquely comprehensive data set, 9660 observations across six countries and covering 40 crops, show that farmers access 90.2 % of their seed from informal systems with 50.9 % of that deriving from local markets. Further, 55 % of seed is paid for by cash, indicating that smallholders are already making important investments in this arena. Targeted interventions are proposed for rendering formal and informal seed sector more smallholder-responsive and for scaling up positive impacts
