    Use of multiple primers in RAPD analysis of clonal organisms provides limited improvement in discrimination

    Randomly amplified polymorphic DNA (RAPD) analysis using two or more primers has been reported to provide additional discriminatory ability over one primer used individually. This may be of particular application in epidemiological typing of clonal organisms, such as Shiga toxin-producing E. coli O157, where strain differentiation can be difficult. Using four arbitrary primers individually, and in all possible permutations, E. coli O157 isolates and other arbitrarily chosen E. coli strains were typed using RAPD analysis. For most nonclonal strains, the use of two primers resulted in increased differentiation between isolates; however, more than two primers did not increase further the discriminatory capacity. E. coli O157 isolates that produced virtually identical profiles using one primer did not show increased differentiation when using two or more primers, demonstrating that in some cases, where strains of an organism are highly related, there is limited advantage to using more than one primer in RAPD analysis

    AN OBSERVATIONAL STUDY OF HUMAN ENERGY FIELDS IN INFANTS AND YOUNG CHILDREN

    The purpose of this investigation was to observe human energy fields (HEF) in infants and young children. The sample (N 18) consisted of three age groups of approximately 6, 30, and 54 month old children. In this exploratory study, coding was devised to help extrapolate information thar leads to a different understanding of development in children. Literature was reviewed from observations of the HEF of adults and ways of looking ar children historically. This approach was combined with traditional observational methods of studying children to develop this research projecr, using qualirative analysis to identifY any parterns within and be[\veen groups. The observations were made by three observers viewing the HEF, using a facilitator, coding forms, and video documentarion. The findings from rhe study show patterns within and bet\Veen groups indicating there is a developmental HEF change that occurs over the three age groups

    The hydrological regime of a forested tropical Andean catchment.

    The hydrology of tropical mountain catchments plays a central role in ecological function, geochemical and biogeochemical cycles, erosion and sediment production, and water supply in globally important environments. There have been few studies quantifying the seasonal and annual water budgets in the montane tropics, particularly in cloud forests. We investigated the water balance and hydrologic regime of the Kosñipata catchment (basin area: 164.4 km2) over the period 2010–2011. The catchment spans over 2500 m in elevation in the eastern Peruvian Andes and is dominated by tropical montane cloud forest with some high-elevation puna grasslands. Catchment-wide rainfall was 3112 ± 414 mm yr−1, calculated by calibrating Tropical Rainfall Measuring Mission (TRMM) 3B43 rainfall with rainfall data from nine meteorological stations in the catchment. Cloud water input to streamflow was 316 ± 116 mm yr−1 (9.2% of total inputs), calculated from an isotopic mixing model using deuterium excess (Dxs) and δD of waters. Field streamflow was measured in 2010 by recording height and calibrating to discharge. River run-off was estimated to be 2796 ± 126 mm yr−1. Actual evapotranspiration (AET) was 688 ± 138 mm yr−1, determined using the Priestley and Taylor–Jet Propulsion Laboratory (PT-JPL) model. The overall water budget was balanced within 1.6 ± 13.7%. Relationships between monthly rainfall and river run-off follow an anticlockwise hysteresis through the year, with a persistence of high run-off after the end of the wet season. The size of the soil and shallow groundwater reservoir is most likely insufficient to explain sustained dry-season flow. Thus, the observed hysteresis in rainfall–run-off relationships is best explained by sustained groundwater flow in the dry season, which is consistent with the water isotope results that suggest persistent wet-season sources to streamflow throughout the year. These results demonstrate the importance of transient groundwater storage in stabilising the annual hydrograph in this region of the Andes

    Time-division SQUID multiplexers with reduced sensitivity to external magnetic fields

    Time-division SQUID multiplexers are used in many applications that require exquisite control of systematic error. One potential source of systematic error is the pickup of external magnetic fields in the multiplexer. We present measurements of the field sensitivity figure of merit, effective area, for both the first stage and second stage SQUID amplifiers in three NIST SQUID multiplexer designs. These designs include a new variety with improved gradiometry that significantly reduces the effective area of both the first and second stage SQUID amplifiers.Comment: 4 pages, 7 figures.     M dwarf stars in the light of (future) exoplanet searches

    We present a brief overview of a splinter session on M dwarf stars as planet hosts that was organized as part of the Cool Stars 17 conference. The session was devoted to reviewing our current knowledge of M dwarf stars and exoplanets in order to prepare for current and future exoplanet searches focusing in low mass stars. We review the observational and theoretical challenges to characterize M dwarf stars and the importance of accurate fundamental parameters for the proper characterization of their exoplanets and our understanding on planet formation.Comment: 4 pages, 1 figure.
