    DETERMINATION AND VALIDATION OF RP-HPLC METHOD FOR THE ESTIMATION OF MIRABEGRON IN TABLET DOSAGE FORM

    Objective: A reversed phase liquid chromatography was determined and validated for the estimation of Mirabegron in tablet dosage form.Methods: The validation study of RP-HPLC showed a simple, rapid, accurate, precise, reproducible results by using a stationary phase: Waters Acquity HSS T-3 C18 (100 × 2.1 mm, 1.7μm and Mobile Phase-Potassium di-hydrogen phosphate: acetone in the ratio (40:60 v/v) at PH6.0±0.02. Detection is carried out at 243 nm using UV detector.Results: The total chromatographic analysis time per sample was about 6 min with Mirabegron eluting at a retention time of 2.754. Tailing factor obtained from the standard injection is 1.6. Theoretical Plates obtained from the standard injection is 2736.7. The flow rate is 1 ml/min and linearity in the concentration range of 30-70μg/ml (R2=0.999). The precision was 0.4% the intermediate precision was 0.08%. The deliberately varied chromatographic conditions in the concentration range for the evaluation of robustness is 10-50 µg/ml, (n=3). The limit of detection (LOD) and limit of quantitation (LOQ) for Mirabegron were 0.01µg/ml and 0.05µg/ml respectively. The % recovery is 99.8 % with % R. SD of 0.09. The results proved that the optimized HPLC method fulfills these requirements within the ICH accepted limits.Conclusion: The high recovery and low relative standard deviation confirm the suitability of the proposed method for the determination of Mirabegron in tablet dosage form.Â

    DESIGN AND FABRICATION OF MEDICATED CHOCOLATE FORMULATION BY CHOCOLATE DRUG DELIVERY SYSTEM

    Objective: The objective of this study is to design and fabricate Chlorpheniramine Maleate chocolate formulation by chocolate drug delivery system. Chlorpheniramine Maleate binds to histamine H1 receptor. This blocks the action of endogenous histamine, which subsequently leads to temporary relief of negative symptoms brought on by histamine.Methods: Chocolate is a range of products derived from cocoa (cacao), mixed with fat (i.e., cocoa butter) and finely powdered sugar to produce a solid confectionery. The medicated chocolate formulation is widely used for pediatric administration and increases patient compliance. Chlorpheniramine Maleate chocolate formulation is prepared to improve patient compliance. Chocolates were formulated (F1-F3) with a total fat of 25-35 % (w/w) from cocoa liquor and cocoa butter with more than 34% total cocoa, composition as specified for dark chocolate, lecithin, sweetening agents.Results: The prepared chocolate formulations were evaluated for general appearance, drug content, In vitro drug release and DSC and FTIR, moisture content and blooming tests. F1 formulation releases complete drug within 60 min.Conclusion: The results indicate that the formulation has no drug excipient interactions and there was no degradation in drug, it is stable during chocolate formulation preparation.Â

    HIBISCUS ROSA SINENSIS LOADED SOLID LIPID NANOPARTICLES AND IN VIVO WOUND HEALING ACTIVITY IN WISTAR ALBINO RATS

    Objective: The objective of the present research was to investigate the wound-healing potency of solid lipid nano particles of Hibiscus rosa sinensis extract. Crude herbal extracts and rudimentary formulations containing herbal extracts are good for demonstrating the feasibility of the concept; however, such formulations suffer with poor oral bioavailability and variability within groups of subjects. Converting herbal extracts into novel drug delivery systems may prove effective in addressing some of these problems. Methods: In the present study an attempt was made to develop Hibiscus rosa sinensis extract loaded solid lipid nanoparticles (HSLNs) using lipids glycerol monostearate (GMS) or beeswax. The prepared HSLNs were characterised for their size, surface charge and morphology. The optimized HSLNs were incorporated into Carbopol gel and tested for wound healing activity in male Wistar albino rats using excision wound model. Results: HSLNs of ~175 nm in size carrying negative charge were obtained with the optimised procedure using beeswax. The shape of the HSLNs was nearly spherical. The HSLNs (10 mg/ml) treated wounds healed much faster compared to raw crude extract and healing was comparable to marketed preparation. Conclusion: It is concluded that converting crude herbal extracts into SLNs can be an effective way to enhance the effectiveness of herbal extracts and their in vivo activity

    Breast Cancer Detection using Two Dimensional Principal Component Analysis and Back Propagation Neural Network

    Breast cancer is the most common cancer which affects women around the world. It has been increasing over the years. Detection and diagnosis is the main factor for breast cancer control which increases the success rate of treatment, saves lives and reduce the cost. This paper proposes an efficient approach for breast cancer detection in mammogram breast images using two dimensional principal component analysis and back propagation neural network. The proposed approach consists of four step by step procedures namely preprocessing of breast images, image enhancement, feature extraction and classification. Two dimensional principal component analysis is used to obtain the features of the preprocessed and enhanced image. The reason for selecting two dimensional principal component analysis is it is easier to evaluate the covariance matrix accurately and less time is required to determine the corresponding features. Finally, Back propagation neural network is used to classify whether the given mammogram image is normal or abnormal. Simulation results are carried out using the proposed approach by considering MIAS data base. From the results, it is observed that proposed approach provide better accuracy

    A study on disease burden and treatment among patients attending teleconsultation during lockdown period

    Background: Covid-19 has been announced as a pandemic by the World Health Organization. To maintain social distancing effectively, the Government of India announced a complete lockdown on March 25th, 2020. As there are no proper transport facilities for patients who need health care services, the Government of Andhra Pradesh started 104 sevakendram as a helpline. Medical services were also provided by this 104 sevakendram through teleconsultation with doctors. The aim and objectives of the study were to study the disease pattern and treatment given to patients attending teleconsultations, to analyze the disease burden among the patients attending teleconsultation, to study the treatment given to the patients attending teleconsultation.Methods: It is a retrospective, observational, and analytical study. After prior IEC clearance and permission from teleconsultation authority, we did the study at Dr.YSR Aarogyasri health care trust, Guntur. Data regarding all calls connected to 104 was collected. Detailed information of the calls related to the Medical officer about the age, gender, disease pattern, and treatment given to the patient was collected. Statistical analysis was done using MS excel software.Results: On average, 104 sevakendram received two hundred valid teleconsultations per day. Most of the calls are related to anxiety and these calls constitute twenty four percent. Treatment given was according to probable diagnosis, mostly symptomatic and continuation of the same treatment.Conclusions: 104 sevakendram has played a vital role in meeting the medical and health requirements of the people suffering from different diseases during the lockdown period.

    The present work was aimed to formulate and evaluate a new, cheap and effective natural suspending agent that can be used as an effective alternative for traditional suspending agent. The study procedure involved extraction of suspending agent from the Musa paradisica (Banana) fruit peels, determination of swelling index, phytochemical testing, Micromeritic properties of mucilage like Bulk density, Tapped density, Carr’s index, Hausner’s ratio, Angle of repose, Calibration of paracetamol, preparation of paracetamol suspensions and evaluated for pH determination, determination of sedimentation volume, redispersibility, determination of flow rate, measurement of viscosity, effect of temperature, drug content, particle size determination and In-vitro dissolution studies. The study showed that the extraction of suspending agent from banana fruit peels. The swelling index was found to be 40%. The photochemical test showed contains carbohydrates. As the concentration of suspending agent increases therefore viscosity of suspension increases which ultimately reduces the sedimentation of suspension

    Multi-label natural language processing to identify diagnosis and procedure codes from MIMIC-III inpatient notes

    In the United States, 25% or greater than 200 billion dollars of hospital spending accounts for administrative costs that involve services for medical coding and billing. With the increasing number of patient records, manual assignment of the codes performed is overwhelming, time-consuming and error-prone, causing billing errors. Natural language processing can automate the extraction of codes/labels from unstructured clinical notes, which can aid human coders to save time, increase productivity, and verify medical coding errors. Our objective is to identify appropriate diagnosis and procedure codes from clinical notes by performing multi-label classification. We used de-identified data of critical care patients from the MIMIC-III database and subset the data to select the ten (top-10) and fifty (top-50) most common diagnoses and procedures, which covers 47.45% and 74.12% of all admissions respectively. We implemented state-of-the-art Bidirectional Encoder Representations from Transformers (BERT) to fine-tune the language model on 80% of the data and validated on the remaining 20%. The model achieved an overall accuracy of 87.08%, an F1 score of 85.82%, and an AUC of 91.76% for top-10 codes. For the top-50 codes, our model achieved an overall accuracy of 93.76%, an F1 score of 92.24%, and AUC of 91%. When compared to previously published research, our model outperforms in predicting codes from the clinical text. We discuss approaches to generalize the knowledge discovery process of our MIMIC-BERT to other clinical notes.
