    Clostridium perfringens-Associated Necrotic Enteritis-Like Disease in Coconut Lorikeets (Trichoglossus haematodus).

    Several outbreaks of necrotic enteritis-like disease in lorikeets, from which Clostridium perfringens was consistently isolated, are described. All lorikeets had acute, segmental, or multifocal fibrinonecrotizing inflammatory lesions in the small and/or the large intestine, with intralesional gram-positive rods. The gene encoding C. perfringens alpha toxin was detected by PCR (polymerase chain reaction) on formalin-fixed, paraffin-embedded (FFPE) tissues in 20 out of 24 affected lorikeets (83%), but it was not amplified from samples of any of 10 control lorikeets (P < .0001). The second most prevalent C. perfringens toxin gene detected was the beta toxin gene, which was found in FFPE from 7 out of 24 affected lorikeets (29%). The other toxin genes were detected inconsistently and in a relatively low number of samples. These cases seem to be associated with C. perfringens, although the specific type involved could not be determined

    Effects of deoxynivalenol exposure time and contamination levels on rainbow trout

    The trend toward using plant‐based ingredients in aquafeeds is set to intensify; however, mycotoxin contamination might be a challenge. Two diets, with deoxynivalenol (DON) levels of 1,166 μg/kg (1.1 DON) and 2,745 μg/kg (2.7 DON), were prepared for short‐term DON exposure (50 days). A third diet with a low DON level of 367 μg/kg (0.3 DON) was prepared for long‐term DON exposure (168 days). Ingestion of DON by trout during both short‐term/high‐dosage exposure (50 days; 1,166 μg/kg and 2,700 μg/kg DON) and long‐term/low‐dosage exposure (168 days; 367 μg/kg DON) impacted growth performance and, to a lesser extent, liver enzyme parameters (2.7 DON). Histopathology showed mild to moderate changes in the liver but not in the other sampled tissues (intestine and kidney). Despite these effects, short‐term exposure of rainbow trout to high doses of DON did not result in increased susceptibility to Yersinia ruckeri. In both the short‐ and long‐term studies, the effects of DON showed a high interindividual variability. The present study confirms that subclinical levels of mycotoxins affect rainbow trout. The effects of such low mycotoxin levels could be masked by other production challenges while still negatively affecting productivity

    We calculate the new physics effects to the branching ratios of the lepton flavor conserving decays Z -> l^+ l^- in the framework of the general two Higgs Doublet model. We predict the upper limits for the couplings |\bar{\xi}^{D}_{N,\mu\tau}| and |\bar{\xi}^{D}_{N,\tau\tau}| as 3\times 10^2 GeV and 1\times 10^2 GeV, respectively.Comment: 9 pages, 3 figure

    Pollution control can help mitigate future climate change impact on European grayling in the UK

    Aim: We compare the performance of habitat suitability models using climate data only or climate data together with water chemistry, land cover and predation pressure data to model the distribution of European grayling (Thymallus thymallus). From these models, we (a) investigate the relationship between habitat suitability and genetic diversity; (b) project the distribution of grayling under future climate change; and (c) model the effects of habitat mitigation on future distributions. Location: United Kingdom. Methods: Maxent species distribution modelling was implemented using a Simple model (only climate parameters) or a Full model (climate, water chemistry, land use and predation pressure parameters). Areas of high and low habitat suitability were designated. Associations between habitat suitability and genetic diversity for both neutral and adaptive markers were examined. Distribution under minimal and maximal future climate change scenarios was modelled for 2050, incorporating projections of future flow scenarios obtained from the Centre for Ecology and Hydrology. To examine potential mitigation effects within habitats, models were run with manipulation of orthophosphate, nitrite and copper concentrations. Results: We mapped suitable habitat for grayling in the present and the future. The Full model achieved substantially higher discriminative power than the Simple model. For low suitability habitat, higher levels of inbreeding were observed for adaptive, but not neutral, loci. Future projections predict a significant contraction of highly suitable areas. Under habitat mitigation, modelling suggests that recovery of suitable habitat of up to 10% is possible. Main conclusions: Extending the climate-only model improves estimates of habitat suitability. Significantly higher inbreeding coefficients were found at immune genes, but not neutral markers in low suitability habitat, indicating a possible impact of environmental stress on evolutionary potential. The potential for habitat mitigation to alleviate distributional changes under future climate change is demonstrated, and specific recommendations are made for habitat recovery on a regional basis

    Transient increase in chloride cell number and heat shock protein expression (hsp70) in brown trout (Salmo trutta fario) exposed to sudden temperature elevation

    The native cold-adapted brown trout ( Salmo trutta fario ) is often the subject of biomonitoring field studies. Groups of trout were exposed to a sudden temperature rise, from 8 degrees C to 19 degrees C for two hours, and thereafter set back to 8 degrees C. Gill samples of control animals, of fish after the exposure period, and after 24 and 48 hours of recovery at a temperature of 8 degrees C were examined histologically, immunohistochemically, electron microscopically, and by Western blot analysis. By means of immunohistochemistry and electron microscopy, an increase of chloride cells was observed after the temperature elevation . During the recovery period the number of chloride cells decreased. Western blot analysis for stress proteins ( hsp70 ), widely used as a biomarker for environmental stress, was performed from skin and gill. Whereas in the gill both isoforms, the constitutive and the heat inducible form, of hsp70 were detected in all groups, in the skin the control animals only showed the constitutive form. After two hours of exposure both isoforms were visible. An increased expression of hsp70 could be demonstrated in both organs after the exposure. Comparison of the hsp70 values between gill and skin showed tissue-specific differences during the recovery period. In the gill hsp70 rapidly decreased, while in the skin the level remained elevated over the whole observation period. When hsp70 is used as a biomarker in field studies, the fast and organ-specific reaction in the gill and skin of brown trout has to be taken into consideration

    Differential characterization of emerging skin diseases of rainbow trout - a standardized approach to capturing disease characteristics and development of case definitions

    Farmed and wild salmonids are affected by a variety of skin conditions, some of which have significant economic and welfare implications. In many cases, the causes are not well understood, and one example is cold water strawberry disease of rainbow trout, also called red mark syndrome, which has been recorded in the UK since 2003. To date, there are no internationally agreed methods for describing these conditions, which has caused confusion for farmers and health professionals, who are often unclear as to whether they are dealing with a new or a previously described condition. This has resulted, inevitably, in delays to both accurate diagnosis and effective treatment regimes. Here, we provide a standardized methodology for the description of skin conditions of rainbow trout of uncertain aetiology. We demonstrate how the approach can be used to develop case definitions, using coldwater strawberry disease as an example

    Observational Study Design in Veterinary Pathology, Part 2: Methodology

    Observational studies are a basis for much of our knowledge of veterinary pathology, yet considerations for conducting pathology-based observational studies are not readily available. In part 1 of this series, we offered advice on planning and carrying out an observational study. Part 2 of the series focuses on methodology. Our general recommendations are to consider using already-validated methods, published guidelines, data from primary sources, and quantitative analyses. We discuss 3 common methods in pathology research—histopathologic scoring, immunohistochemistry, and polymerase chain reaction—to illustrate principles of method validation. Some aspects of quality control include use of clear objective grading criteria, validation of key reagents, assessing sample quality, determining specificity and sensitivity, use of technical and biologic negative and positive controls, blinding of investigators, approaches to minimizing operator-dependent variation, measuring technical variation, and consistency in analysis of the different study groups. We close by discussing approaches to increasing the rigor of observational studies by corroborating results with complementary methods, using sufficiently large numbers of study subjects, consideration of the data in light of similar published studies, replicating the results in a second study population, and critical analysis of the study findings

    The impact of co-infections on fish: a review

    International audienceAbstractCo-infections are very common in nature and occur when hosts are infected by two or more different pathogens either by simultaneous or secondary infections so that two or more infectious agents are active together in the same host. Co-infections have a fundamental effect and can alter the course and the severity of different fish diseases. However, co-infection effect has still received limited scrutiny in aquatic animals like fish and available data on this subject is still scarce. The susceptibility of fish to different pathogens could be changed during mixed infections causing the appearance of sudden fish outbreaks. In this review, we focus on the synergistic and antagonistic interactions occurring during co-infections by homologous or heterologous pathogens. We present a concise summary about the present knowledge regarding co-infections in fish. More research is needed to better understand the immune response of fish during mixed infections as these could have an important impact on the development of new strategies for disease control programs and vaccination in fish

    β-Catenin Signaling Increases during Melanoma Progression and Promotes Tumor Cell Survival and Chemoresistance

    Beta-catenin plays an important role in embryogenesis and carcinogenesis by controlling either cadherin-mediated cell adhesion or transcriptional activation of target gene expression. In many types of cancers nuclear translocation of beta-catenin has been observed. Our data indicate that during melanoma progression an increased dependency on the transcriptional function of beta-catenin takes place. Blockade of beta-catenin in metastatic melanoma cell lines efficiently induces apoptosis, inhibits proliferation, migration and invasion in monolayer and 3-dimensional skin reconstructs and decreases chemoresistance. In addition, subcutaneous melanoma growth in SCID mice was almost completely inhibited by an inducible beta-catenin knockdown. In contrast, the survival of benign melanocytes and primary melanoma cell lines was less affected by beta-catenin depletion. However, enhanced expression of beta-catenin in primary melanoma cell lines increased invasive capacity in vitro and tumor growth in the SCID mouse model. These data suggest that beta-catenin is an essential survival factor for metastatic melanoma cells, whereas it is dispensable for the survival of benign melanocytes and primary, non-invasive melanoma cells. Furthermore, beta-catenin increases tumorigenicity of primary melanoma cell lines. The differential requirements for beta-catenin signaling in aggressive melanoma versus benign melanocytic cells make beta-catenin a possible new target in melanoma therapy

    In the course of work the phase composition of the ceramic composite material Al[2]O[3] - 50 % ZrW[2]O[8] was studied. On the polished surface of the Al[2]O[3]-ZrW[2]O[8] sintered composite can be allocated three areas: dark gray matrix, white inclusions of an irregular form and spherical inclusions. The average size of spherical inclusions was equal to 15 [mu]m.
