    Syndecan-4 knockout leads to reduced extracellular transglutaminase-2 and protects against tubulointerstitial fibrosis

    Transglutaminase type 2 (TG2) is an extracellular matrix crosslinking enzyme with a pivotal role in kidney fibrosis. The interaction of TG2 with the heparan sulfate proteoglycan syndecan-4 (Sdc4) regulates the cell surface trafficking, localization, and activity of TG2 in vitro but remains unstudied in vivo. We tested the hypothesis that Sdc4 is required for cell surface targeting of TG2 and the development of kidney fibrosis in CKD. Wild-type and Sdc4-null mice were subjected to unilateral ureteric obstruction and aristolochic acid nephropathy (AAN) as experimental models of kidney fibrosis. Analysis of renal scarring by Masson trichrome staining, kidney hydroxyproline levels, and collagen immunofluorescence demonstrated progressive fibrosis associated with increases in extracellular TG2 and TG activity in the tubulointerstitium in both models. Knockout of Sdc-4 reduced these effects and prevented AAN-induced increases in total and active TGF-b1. In wild-type mice subjected to AAN, extracellular TG2 colocalized with Sdc4 in the tubular interstitium and basement membrane, where TG2 also colocalized with heparan sulfate chains. Heparitinase I, which selectively cleaves heparan sulfate, completely abolished extracellular TG2 in normal and diseased kidney sections. In conclusion, the lack of Sdc4 heparan sulfate chains in the kidneys of Sdc4-null mice abrogates injury-induced externalization of TG2, thereby preventing profibrotic crosslinking of extracellular matrix and recruitment of large latent TGF-b1. This finding suggests that targeting the TG2- Sdc4 interaction may provide a specific interventional strategy for the treatment of CKD

    Using Color in Machine Vision Systems for Wood Processing

    Color information, already shown to be valuable in distinguishing wood surface features, should prove especially useful for future applications of machine vision in the wood products industry. This review provides investigators interested in such applications with the information necessary for understanding the benefits-and associated difficulties-of using color. Various standard color-measurement systems ("color spaces") are discussed. No one system has been completely successful, at least partly because simple physical measurements are difficult to correlate with a human observer's complex perception of color. Color video camera systems, designed with human viewers in mind, have the potential for machine vision applications, but certain system "features" (white balance, gamma or contour correction) could cause problems. Future applications, including detecting and classifying hard-to-identify defects and matching colors of wood components, will require careful choice of lighting geometry and source, camera system, and color space for the purpose at hand

    Constraints on s→dγs\to d \gamma from Radiative Hyperon and Kaon Decays

    The quark-level process b→sγb \to s \gamma has been used extensively to place constraints on new interactions. These same interactions can be further constrained from the enhancement they induce in the quark-level s→dγs \to d \gamma transition, to the extent that the short distance contributions can be separated from the long distance contributions. We parameterize what is known about the long distance amplitudes and subtract it from the data in radiative hyperon and kaon decays to constrain new interactions.Comment: Latex 11 page

    Search for the K(L) --> PI0 PI0 E+ E- Decay in the KTeV Experiment

    The recent discovery of a large CP violating asymmetry in K(L) --> PI+ PI- E+ E- mode has prompted us to seach for the associated K(L) --> PI0 PI0 E+ E- decay mode in the KTeV-E799 experiment at Fermilab. In 2.7E+11 K(L) decays, one candidate event has been observed with an expected background of 0.3 event, resulting in an upper limit for the K(L) --> PI0 PI0 E+ E- branching ratio of 6.6E-09 at the 90% confidence level.Comment: To be published in Phys.     The standard model at low energies

    The hadronic sector of the standard model at low energies is described by a non--decoupling effective field theory, chiral perturbation theory. An introduction is given to the construction of effective chiral Lagrangians, both in the purely mesonic sector and with inclusion of baryons.     Gestational Age, Parent Education, and Education in Adulthood

    BACKGROUND: Adults born preterm ( METHODS: This register-based cohort study included singletons born alive from 1987 up to 1992 in Denmark, Finland, Norway, and Sweden. In each study population, we investigated effect modification by parents' educational level (low, intermediate, high) on the association between gestational age at birth (25-44 completed weeks) and low educational attainment at 25 years (not having completed upper secondary education) using general estimation equations logistic regressions. RESULTS: A total of 4.3%, 4.0%, 4.8%, and 5.0% singletons were born preterm in the Danish (n = 331 448), Finnish (n = 220 095), Norwegian (n = 292 840), and Swedish (n = 513 975) populations, respectively. In all countries, both lower gestational age and lower parental educational level contributed additively to low educational attainment. For example, in Denmark, the relative risk of low educational attainment was 1.84 (95% confidence interval 1.44 to 2.26) in adults born at 28 to 31 weeks whose parents had high educational level and 5.25 (95% confidence interval 4.53 to 6.02) in adults born at 28 to 31 weeks whose parents had low educational level, compared with a reference group born at 39 to 41 weeks with high parental educational level. CONCLUSIONS: Although higher parental education level was associated with higher educational attainment for all gestational ages, parental education did not mitigate the educational disadvantages of shorter gestational age.Peer reviewe
