"Wet-to-Dry" Conformational Transition of Polymer Layers Grafted to Nanoparticles in Nanocomposite
The present communication reports the first direct measurement of the
conformation of a polymer corona grafted around silica nano-particles dispersed
inside a nanocomposite, a matrix of the same polymer. This measurement
constitutes an experimental breakthrough based on a refined combination of
chemical synthesis, which permits to match the contribution of the neutron
silica signal inside the composite, and the use of complementary scattering
methods SANS and SAXS to extract the grafted polymer layer form factor from the
inter-particles silica structure factor. The modelization of the signal of the
grafted polymer on nanoparticles inside the matrix and the direct comparison
with the form factor of the same particles in solution show a clear-cut change
of the polymer conformation from bulk to the nanocomposite: a transition from a
stretched and swollen form in solution to a Gaussian conformation in the matrix
followed with a compression of a factor two of the grafted corona. In the
probed range, increasing the interactions between the grafted particles (by
increasing the particle volume fraction) or between the grafted and the free
matrix chains (decreasing the grafted-free chain length ratio) does not
influence the amplitude of the grafted brush compression. This is the first
direct observation of the wet-to-dry conformational transition theoretically
expected to minimize the free energy of swelling of grafted chains in
interaction with free matrix chains, illustrating the competition between the
mixing entropy of grafted and free chains, and the elastic deformation of the
grafted chains. In addition to the experimental validation of the theoretical
prediction, this result constitutes a new insight for the nderstanding of the
general problem of dispersion of nanoparticles inside a polymer matrix for the
design of new nanocomposites materials
MiR-200 family controls late steps of postnatal forebrain neurogenesis via Zeb2 inhibition

Oct4-Induced Reprogramming Is Required for Adult Brain Neural Stem Cell Differentiation into Midbrain Dopaminergic Neurons
Neural stem cells (NSCs) lose their competency to generate region-specific neuronal populations at an early stage during embryonic brain development. Here we investigated whether epigenetic modifications can reverse the regional restriction of mouse adult brain subventricular zone (SVZ) NSCs. Using a variety of chemicals that interfere with DNA methylation and histone acetylation, we showed that such epigenetic modifications increased neuronal differentiation but did not enable specific regional patterning, such as midbrain dopaminergic (DA) neuron generation. Only after Oct-4 overexpression did adult NSCs acquire a pluripotent state that allowed differentiation into midbrain DA neurons. DA neurons derived from Oct4-reprogrammed NSCs improved behavioural motor deficits in a rat model of Parkinson's disease (PD) upon intrastriatal transplantation. Here we report for the first time the successful differentiation of SVZ adult NSCs into functional region-specific midbrain DA neurons, by means of Oct-4 induced pluripotency
Gold nanoparticle-polymer nanocomposites synthesized by room temperature atmospheric pressure plasma and their potential for fuel cell electrocatalytic application
Conductive polymers have been increasingly used as fuel cell catalyst support due to their electrical conductivity, large surface areas and stability. The incorporation of metal nanoparticles into a polymer matrix can effectively increase the specific surface area of these materials and hence improve the catalytic efficiency. In this work, a nanoparticle loaded conductive polymer nanocomposite was obtained by a one-step synthesis approach based on room temperature direct current plasmaliquid interaction. Gold nanoparticles were directly synthesized from HAuCl4 precursor in poly(3,4-ethylenedioxythiophene) polystyrene sulfonate (PEDOT: PSS). The resulting AuNPs/PEDOT: PSS nanocomposites were subsequently characterized under a practical alkaline direct ethanol fuel cell operation condition for its potential application as an electrocatalyst. Results show that AuNPs sizes within the PEDOT: PSS matrix are dependent on the plasma treatment time and precursor concentration, which in turn affect the nanocomposites electrical conductivity and their catalytic performance. Under certain synthesis conditions, unique nanoscale AuNPs/PEDOT: PSS core-shell structures could also be produced, indicating the interaction at the AuNPs/polymer interface. The enhanced catalytic activity shown by AuNPs/PEDOT: PSS has been attributed to the effective electron transfer and reactive species diffusion through the porous polymer network, as well as the synergistic interfacial interaction at the metal/polymer and metal/metal interfaces.Funding Agencies|UK EPSRC [EP/K022237/1, EP/M024938/1, EP/P00394X/1, EP/I013229/1]; National Natural Science Foundation of China [51203135]; InvestNI [PoC-325]; Department of Employment Learning; EU-COST Action [TD1208]</p
Local neurons play key roles in the mammalian olfactory bulb
Over the past few decades, research exploring how the brain perceives, discriminates, and recognizes odorant molecules has received a growing interest. Today, olfaction is no longer considered a matter of poetry. Chemical senses entered the biological era when an increasing number of scientists started to elucidate the early stages of the olfactory pathway. A combination of genetic, biochemical, cellular, electrophysiological and behavioral methods has provided a picture of how odor information is processed in the olfactory system as it moves from the periphery to higher areas of the brain. Our group is exploring the physiology of the main olfactory bulb, the first processing relay in the mammalian brain. From different electrophysiological approaches, we are attempting to understand the cellular rules that contribute to the synaptic transmission and plasticity at this central relay. How olfactory sensory inputs, originating from the olfactory epithelium located in the nasal cavity, are encoded in the main olfactory bulb remains a crucial question for understanding odor processing. More importantly, the persistence of a high level of neurogenesis continuously supplying the adult olfactory bulb with newborn local neurons provides an attractive model to investigate how basic olfactory functions are maintained when a large proportion of local neurons are continuously renewed. For this purpose, we summarize the current ideas concerning the molecular mechanisms and organizational strategies used by the olfactory system to encode and process information in the main olfactory bulb. We discuss the degree of sensitivity of the bulbar neuronal network activity to the persistence of this high level of neurogenesis that is modulated by sensory experience. Finally, it is worth mentioning that analyzing the molecular mechanisms and organizational strategies used by the olfactory system to transduce, encode, and process odorant information in the olfactory bulb should aid in understanding the general neural mechanisms involved in both sensory perception and memory. Due to space constraints, this review focuses exclusively on the olfactory systems of vertebrates and primarily those of mammals
