    Examining Measurement Reactivity in Daily Diary Data On Substance Use: Results From a Randomized Experiment

    The debate about whether measurement reactivity exists in daily diary research on substance use is still unsettled due to the issues of study design and statistical methodology. This study proposes a time-varying effect model (TVEM) that characterizes the trajectory of substance use behaviors with nonparametric functions determined by the data rather than imposes presumed parametric functions. It also allows researchers to investigate the effect of measurement reactivity on not only the likelihood of using substances but also the amount of substance use. The TVEM was applied to analyze diary data on alcohol and marijuana use collected from an experiment, which randomized 307 participants in Michigan into daily and weekly assessment schedules during 2014-2016. This study found short-term measurement reactivity on alcohol use, but did not find a significant reactivity effect on marijuana use. The daily group had smaller odds of abstinence from drinking but lower expected drinking quantity in the first week of assessment, which dissipated by the second week. The results indicate that although daily self-monitoring could have short-term reactivity on substance use behaviors that tend to fluctuate across days, such as alcohol use, it does not affect substance use behaviors that are quite consistent, such as marijuana use. Our findings imply that although daily monitoring of drinking may motivate people to reduce the quantity consumed once they start to drink, it may also arouse their desire to start drinking. Yet, both effects tend to last only one week, as participants accommodate to the monitoring by the second week

    Transverse spin dynamics in a spin-polarized Fermi liquid

    The linear equations for transverse spin dynamics in weakly polarised degenerate Fermi liquid with arbitrary relationship between temperature and polarization are derived from Landau-Silin phenomenological kinetic equation with general form of two-particle collision integral. The temperature and polarization dependence of the spin current relaxation time is established. It is found in particular that at finite polarization transverse spin wave damping has a finite value at T=0. The analogy between temperature dependences of spin waves attenuation and ultrasound absorption in degenerate Fermi liquid at arbitrary temperature is presented. We also discuss spin-polarized Fermi liquid in the general context of the Fermi-liquid theory and compare it with "Fermi liquid" with spontaneous magnetization.Comment: 10 page

    Bright microwave pulses from PSR B0531+21 observed with a prototype transient survey receiver

    Recent discoveries of transient radio events have renewed interest in time-variable astrophysical phenomena. Many radio transient events are rare, requiring long observing times for reliable statistical study. The National Aeronautics and Space Administration/Jet Propulsion Laboratory\u27s Deep Space Network (DSN) tracks spacecraft nearly continuously with 13 large-aperture, low system temperature radio antennas. During normal spacecraft operations, the DSN processes only a small fraction of the pre-detection bandwidth available from these antennas; any information in the remaining bandwidth, e.g., from an astronomical source in the same antenna beam as the spacecraft, is currently ignored. As a firmware modification to the standard DSN tracking receiver, we built a prototype receiver that could be used for astronomical transient surveys. Here, we demonstrate the receiver\u27s utility through observations of bright pulses from the Crab pulsar and describe attributes of potential transient survey observations piggybacking on operational DSN tracks. © 2014.     The Block V Receiver fast acquisition algorithm for the Galileo S-band mission

    A fast acquisition algorithm for the Galileo suppressed carrier, subcarrier, and data symbol signals under low data rate, signal-to-noise ratio (SNR) and high carrier phase-noise conditions has been developed. The algorithm employs a two-arm fast Fourier transform (FFT) method utilizing both the in-phase and quadrature-phase channels of the carrier. The use of both channels results in an improved SNR in the FFT acquisition, enabling the use of a shorter FFT period over which the carrier instability is expected to be less significant. The use of a two-arm FFT also enables subcarrier and symbol acquisition before carrier acquisition. With the subcarrier and symbol loops locked first, the carrier can be acquired from an even shorter FFT period. Two-arm tracking loops are employed to lock the subcarrier and symbol loops parameter modification to achieve the final (high) loop SNR in the shortest time possible. The fast acquisition algorithm is implemented in the Block V Receiver (BVR). This article describes the complete algorithm design, the extensive computer simulation work done for verification of the design and the analysis, implementation issues in the BVR, and the acquisition times of the algorithm. In the expected case of the Galileo spacecraft at Jupiter orbit insertion PD/No equals 14.6 dB-Hz, R(sym) equals 16 symbols per sec, and the predicted acquisition time of the algorithm (to attain a 0.2-dB degradation from each loop to the output symbol SNR) is 38 sec

    Identification of Small Molecule and Genetic Modulators of AON-Induced Dystrophin Exon Skipping by High-Throughput Screening

    One therapeutic approach to Duchenne Muscular Dystrophy (DMD) recently entering clinical trials aims to convert DMD phenotypes to that of a milder disease variant, Becker Muscular Dystrophy (BMD), by employing antisense oligonucleotides (AONs) targeting splice sites, to induce exon skipping and restore partial dystrophin function. In order to search for small molecule and genetic modulators of AON-dependent and independent exon skipping, we screened ∼10,000 known small molecule drugs, >17,000 cDNA clones, and >2,000 kinase- targeted siRNAs against a 5.6 kb luciferase minigene construct, encompassing exon 71 to exon 73 of human dystrophin. As a result, we identified several enhancers of exon skipping, acting on both the reporter construct as well as endogenous dystrophin in mdx cells. Multiple mechanisms of action were identified, including histone deacetylase inhibition, tubulin modulation and pre-mRNA processing. Among others, the nucleolar protein NOL8 and staufen RNA binding protein homolog 2 (Stau2) were found to induce endogenous exon skipping in mdx cells in an AON-dependent fashion. An unexpected but recurrent theme observed in our screening efforts was the apparent link between the inhibition of cell cycle progression and the induction of exon skipping

    Cooling a nanomechanical resonator with quantum back-action

    Quantum mechanics demands that the act of measurement must affect the measured object. When a linear amplifier is used to continuously monitor the position of an object, the Heisenberg uncertainty relationship requires that the object be driven by force impulses, called back-action. Here we measure the back-action of a superconducting single-electron transistor (SSET) on a radiofrequency nanomechanical resonator. The conductance of the SSET, which is capacitively coupled to the resonator, provides a sensitive probe of the latter's position;back-action effects manifest themselves as an effective thermal bath, the properties of which depend sensitively on SSET bias conditions. Surprisingly, when the SSET is biased near a transport resonance, we observe cooling of the nanomechanical mode from 550mK to 300mK-- an effect that is analogous to laser cooling in atomic physics. Our measurements have implications for nanomechanical readout of quantum information devices and the limits of ultrasensitive force microscopy (such as single-nuclear-spin magnetic resonance force microscopy). Furthermore, we anticipate the use of these backaction effects to prepare ultracold and quantum states of mechanical structures, which would not be accessible with existing technology.Comment: 28 pages, 7 figures; accepted for publication in Natur

    First insights into the phylogenetic diversity of Mycobacterium tuberculosis in Nepal

    BACKGROUND: Tuberculosis (TB) is a major public health problem in Nepal. Strain variation in Mycobacterium tuberculosis may influence the outcome of TB infection and disease. To date, the phylogenetic diversity of M. tuberculosis in Nepal is unknown. METHODS AND FINDINGS: We analyzed 261 M. tuberculosis isolates recovered from pulmonary TB patients recruited between August 2009 and August 2010 in Nepal. M. tuberculosis lineages were determined by single nucleotide polymorphisms (SNP) typing and spoligotyping. Drug resistance was determined by sequencing the hot spot regions of the relevant target genes. Overall, 164 (62.8%) TB patients were new, and 97 (37.2%) were previously treated. Any drug resistance was detected in 50 (19.2%) isolates, and 16 (6.1%) were multidrug-resistant. The most frequent M. tuberculosis lineage was Lineage 3 (CAS/Delhi) with 106 isolates (40.6%), followed by Lineage 2 (East-Asian lineage, includes Beijing genotype) with 84 isolates (32.2%), Lineage 4 (Euro-American lineage) with 41 (15.7%) isolates, and Lineage 1 (Indo-Oceanic lineage) with 30 isolates (11.5%). Based on spoligotyping, we found 45 different spoligotyping patterns that were previously described. The Beijing (83 isolates, 31.8%) and CAS spoligotype (52, 19.9%) were the dominant spoligotypes. A total of 36 (13.8%) isolates could not be assigned to any known spoligotyping pattern. Lineage 2 was associated with female sex (adjusted odds ratio [aOR] 2.58, 95% confidence interval [95% CI] 1.42-4.67, p = 0.002), and any drug resistance (aOR 2.79; 95% CI 1.43-5.45; p = 0.002). We found no evidence for an association of Lineage 2 with age or BCG vaccination status. CONCLUSIONS: We found a large genetic diversity of M. tuberculosis in Nepal with representation of all four major lineages. Lineages 3 and 2 were dominating.     Curbing methicillin-resistant Staphylococcus aureus in 38 French hospitals through a 15-year institutional control program

    BACKGROUND: The Assistance Publique-Hôpitaux de Paris (AP-HP) institution administers 38 teaching hospitals (23 acute care and 15 rehabilitation and long-term care hospitals; total, 23 000 beds) scattered across Paris and surrounding suburbs in France. In the late 1980s, the proportion of methicillin resistance among clinical strains of Staphylococcus aureus (MRSA) reached approximately 40% at AP-HP.METHODS: A program aimed at curbing the MRSA burden was launched in 1993, based on passive and active surveillance, barrier precautions, training, and feedback. This program, supported by the strong commitment of the institution, was reinforced in 2001 by a campaign promoting the use of alcohol-based hand-rub solutions. An observational study on MRSA rate was prospectively carried out from 1993 onwards. RESULTS: There was a significant progressive decrease in MRSA burden (-35%) from 1993 to 2007, whether recorded as the proportion (expressed as percentage) of MRSA among S aureus strains (41.0% down to 26.6% overall; 45.3% to 24.2% in blood cultures) or incidence of MRSA cases (0.86 down to 0.56 per 1000 hospital days). The MRSA burden decreased more markedly in intensive care units (-59%) than in surgical (-44%) and medical (-32%) wards. The use of ABHR solutions (in liters per 1000 hospital days) increased steadily from 2 L to 21 L (to 26 L in acute care hospitals and to 10 L in rehabilitation and long-term care hospitals) following the campaign. CONCLUSION: A sustained reduction of MRSA burden can be obtained at the scale of a large hospital institution with high endemic MRSA rates, providing that an intensive program is maintained for a long period
