    Spectral spatiality in the acousmatic listening context

    Sounds are often experienced as being spatially higher or lower in congruence with their frequency ‘height’ (i.e. pitch register). The term ‘spectral spatiality’ refers to this impression of spatial height and vertical depth as evoked by the perceived occupancy of evolving sound-shapes (spectromorphologies) within the continuum of audible frequencies. Chapters One and Two draw upon a diverse body of literature to explore the cognitive and physiological processes involved in human spatial hearing in general, and spectral spatiality in particular. Thereafter the potential pertinence of a spectral space consciousness in the acousmatic listening experience is highlighted, particularly with regard to more abstract acousmatic contexts where sounds do not directly invoke familiar source identities. Chapters Three and Four further elaborate aspects of spectral space consciousness and propose a terminological framework for discussing musical contexts in terms of their spectral space design. Consequently, it is argued that in acousmatic music, spectral spatiality must be considered as an inseparable aspect of spatiality in general, although its pertinence only becomes directly highlighted in particular musical contexts. The recurring theme in this thesis is that, in acousmatic music, 'space' is not a parameter but a multifaceted quality that is inherent to all sounds. As well as providing an analytical framework for discussing spatiality in acousmatic music, this thesis highlights the compositional potentials offered by spectral spatiality, particularly in relation to the creation of perspectival image in multichannel works. For instance, the possibility of (re)distributing the spectral components of a sound around the listener (circumspectral image) is discussed in context, and a software tool is presented that enables an intuitive and experimental approach to the composition of circumspectral sounds for 6 and 8 channel loudspeaker configurations. This thesis is useful for both composers and analysts interested in aspects of spatiality in acousmatic music. It also offers some insight into spectral space consciousness in non-acousmatic music, and may therefore contribute towards a more general understanding of the nature of our spatial experience in music.EThOS - Electronic Theses Online ServiceGBUnited Kingdo

    Modelling the freezing and thawing behaviour of saturated soils

    This thesis presents an investigation of the thermo / hydro / mechanical behaviour of saturated soils with cryogenic suction effects. The flow relationships accommodate a number of mechanisms: i) heat transfer by conduction, convection, and latent heat transfer and ii) moisture transfer in the liquid phase due to pressure head, elevation head, and thermal gradients. The mechanical behaviour of the soil is modelled by an elasto-plastic work hardening modified Cam- Clay constitutive model. A numerical solution for the theoretical formulation is presented. Standard finite element methods are used for spatial discretisation and finite difference methods are used for temporal discretisation. Verification of the model is achieved by means of programme of tests to check the following cryogenic components i) coupled thermo-hydraulic response, ii) deformation behaviour of the fully coupled thermo-hydro-mechanical model, iii) transient coupled liquid flow and deformation behaviour, and iv) latent heat of fusion. Validation focused on the impact of the cryogenic related processes included within the proposed theoretical formulation. In particular the development of ice lenses and the movement of moisture under cryogenic suction were investigated. The performance of the proposed model with respect to a number of variables was subsequently explored in order to determine their effect on the magnitude and growth of ice lenses in a freezing soil. The model was then applied to a large scale freezing experiment, namely a fully coupled thermo / hydro / mechanical simulation. The simulated results show a good correlation with the experimental results by predicting the patterns and trends of experimentally observed behaviour and the cryogenic processes that occur during the freezing and thawing of frost susceptible soils. It was therefore concluded that the proposed model is capable of providing a good representation of the fully coupled THM behaviour of saturated soils with cryogenic effects.EThOS - Electronic Theses Online ServiceGBUnited Kingdo

    Effects of mineral amendments on trace elements leaching from pre-treated marine sediment after simulated rainfall events

    Bauxite extraction by-products (red mud) were used to evaluate their potential ability to stabilize trace elements from dredged and aerated/humidified marine sediment. The investigated by-products were: bauxaline®(BX) that is a press-filtered red mud; bauxsol™(BS) that is a press-filtered red mud previously washed with excess of seawater, and gypsum neutralized bauxaline® (GBX). These materials were separately mixed to dredged composted sediment sample considering 5% and 20% sediment: stabilizer ratios. For pilot experiments, rainfall events were regularly simulated for 3 months. Concentrations of As, Mo, Cd, Cr, Zn, Cu, and Ni were analyzed in collected leachates as well as toxicity. Results showed that Cd, Mo, Zn, and Cu were efficiently stabilized in the solid matrix when 20% of BX, BS, and GBX was applied. Consequently, toxicity of leachates was lower than for the untreated sediment, meaning that contaminants mobility was reduced. A 5% GBX was also efficient for Mo, Zn and Cu stabilization. In all scenarios, As stabilization was not improved. Compared to all other monitored elements, Mo mobility seemed to depend upon temperature-humidity conditions during pilot experiments suggesting the need of further investigations

    Harmful Elements in Estuarine and Coastal Systems

    Estuaries and coastal zones are dynamic transitional systems which provide many economic and ecological benefits to humans, but also are an ideal habitat for other organisms as well. These areas are becoming contaminated by various anthropogenic activities due to a quick economic growth and urbanization. This chapter explores the sources, chemical speciation, sediment accumulation and removal mechanisms of the harmful elements in estuarine and coastal seawaters. It also describes the effects of toxic elements on aquatic flora and fauna. Finally, the toxic element pollution of the Venice Lagoon, a transitional water body located in the northeastern part of Italy, is discussed as a case study, by presenting the procedures adopted to measure the extent of the pollution, the impacts on organisms and the restoration activities

    Structural studies of cell adhesion and division

    Cell adhesion is a critical process that allows the organisation and functioning of tissues in three-dimensions. However, the replenishing of cells, via cell division, within tissues is equally important for functioning complex life. Both cell adhesion and division are tightly controlled processes and rely on a complex network of signals that, as yet, are not wholly understood. This Thesis presents a structural analysis of several proteins involved in these processes. In the case of cell adhesion, we have made use of high-throughput (HTP) cloning and expression screening technologies in the Oxford Protein Production Facility (OPPF) for the study of the Kindlin protein family – a recently discovered set of proteins essential for integrin-mediated cell adhesion. As a direct result of the HTP pipeline used we were able to determine the high resolution crystal structure of a single domain, the Pleckstrin Homology Domain, from the isoform Kindlin-1. Deletion of this domain in the full-length protein resulted in impaired integrin activation in vivo. This structure, in combination with molecular dynamics simulation demonstrated that, unlike other well characterised PH domains, the binding of secondary messenger lipids (phosphoinositides) is dictated by a, previously unseen, salt bridge that occludes the putative binding site. Mutation of the salt bridge alters the binding characteristics of this domain in vitro. In addition to the PH domain, we have also studied and biophysically characterised full-length Kindlin-3, a blood cell specific isoform. By optimising baculovirus-infected Sf9 cell expression systems we were able to obtain, for the first time, sufficient quantities of protein for characterisation. Furthermore, by using small-angle X-ray scattering (SAXS) in solution we were able to determine a low resolution solution structure of Kindlin-3, revealing a linear arrangement of its FERM domain - a novel conformation known only otherwise in talin. We characterised the interaction of full-length Kindlin-3 with β-integrin cytoplasmic tails using nuclear magnetic resonance spectroscopy, which confirmed that a direct interaction with a membrane distal NPxY motif occurs, and demonstrated the importance of a preceding Serine/Threonine rich region in peptide binding. In the case of cell division, we have determined the crystal structure of the cell cycle checkpoint control related protein, Cid1, a terminal uridine tranferase from Schizzosaccharomyces pombe, alone and in complex with UTP. Structural and biochemical analysis of Cid1 identified a novel Uridine selection mechanism that is suggested to be conserved in metazoan ZCCHC enzymes involved in let-7 miRNA biogenesis, which are important for proliferation, differentiation and cell fate. We have also demonstrated that Cid1 is an RNA binding protein, a property essential for activity that employs a novel mechanism of RNA binding in the absence of RNA binding motifs. The structural work undertaken in this thesis has focussed on two distinct, but interwoven, aspects of cell biology and has significantly added to both fields of research. Excitingly, this has opened many new avenues of investigation and, in the case of Cid1, has the strong potential to lead to the development of novel anticancer therapies.EThOS - Electronic Theses Online ServiceGBUnited Kingdo
