    Effect of rowing on mobility, functionality, and quality of life in women with and without breast cancer: a 4-month intervention

    Purpose: Of the different modalities of rowing, dragon boat training is the most analyzed in breast cancer (BC). However, other types of boats, such as the felucca, use different biomechanical techniques, which have not been studied in the scientific literature. Consequently, in this study, we sought to determine the benefits of felucca rowing on the physical, psychological, and emotional well-being of patients with BC and healthy persons. Methods: A pre- and post-intervention, single-arm study without a control group with a 4-month intervention was carried out in Spain in 2019. The study sample included six women with BC and 15 healthy women. The following questionnaires were administered before and after the intervention: Disabilities of the Arm, Shoulder, and Hand (DASH), Constant-Murley score (CMS), and the European Quality of Life 5 Dimensions (EQ-5D, rate your health today). Differences were determined before and after the intervention using the paired t test. Results: Significant differences (p < 0.05) were found in the results of all the questionnaires for the women with BC and for the healthy women: DASH (- 13.8 BC and - 6.7 healthy), CMS (+ 12.0 BC and 9.2 healthy), and EQ-5D (+ 8.5 BC and 10.5 healthy). Conclusion: Felucca rowing showed benefits in health and quality of life in both women with BC and healthy women. In future studies with controlled design, values regarding clinical relevance, such as effect sizes/confidence intervals, are needed to corroborate our results

    RepA-WH1 prionoid: Clues from bacteria on factors governing phase transitions in amyloidogenesis

    10 p.-1 fig.In bacterial plasmids, Rep proteins initiate DNA replication by undergoing a structural transformation coupled to dimer dissociation. Amyloidogenesis of the ‘winged-helix’ N-terminal domain of RepA (WH1) is triggered in vitro upon binding to plasmid-specific DNA sequences, and occurs at the bacterial nucleoid in vivo. Amyloid fibers are made of distorted RepA-WH1 monomers that assemble as single or double intertwined tubular protofilaments. RepA-WH1 causes in E. coli an amyloid proteinopathy, which is transmissible from mother to daughter cells, but not infectious, and enables conformational imprinting in vitro and in vivo; i.e. RepA-WH1 is a ‘prionoid’. Microfluidics allow the assessment of the intracellular dynamics of RepA-WH1: bacterial lineages maintain two types (strains-like) of RepA-WH1 amyloids, either multiple compact cytotoxic particles or a single aggregate with the appearance of a fluidized hydrogel that it is mildly detrimental to growth. The Hsp70 chaperone DnaK governs the phase transition between both types of RepA-WH1 aggregates in vivo, thus modulating the vertical propagation of the prionoid. Engineering chimeras between the Sup35p/[PSI*] prion and RepA-WH1 generates [REP-PSI*], a synthetic prion exhibiting strong and weak phenotypic variants in yeast.     Bordetella hinzii Endocarditis, A Clinical Case Not Previously Described

    Objective: To review infections by Bordetella hinzii. Materials and methods: A 79-year-old male patient, with a chronic aortic valve biological prosthesis, presented to hospital because of fever. First examinations were normal. However, 72 hours later B. hinzii was isolated in blood cultures, and so meropenem was prescribed. Nevertheless, fever and B. hinzii bacteraemia were still present 7 days later. Results: The transoesophageal echocardiogram revealed an enlarged image suggesting a periprosthetic abscess, confirmed with a PET-CT scan. The patient was sent for cardiac surgery, and biopsy samples confirmed the presence of B. hinzii. Conclusion: There are very few cases of B. hinzii infection in humans. Ours is the first described case of B. hinzii endocarditis

    El objetivo del trabajo es la evaluación de bio-estimulantes (pasta keiki) a base de lanolina y como agente activo 6-bencilaminopurina (BAP), cuyo principal regulador de crecimiento presente son citoquininas con una concentración de 0.5 mg, en la propagación de Phalaenopsis: Violet Queen, Brazilian Fever, Asian Pink Peace, Desert Breeze, Honglin Vicky. En los resultados encontramos que la variedad Honglin Vicky obtuvo una mayor inducción de brotes, en la formación de callos diferenciados y la longitud, Brazilian Fever obtuvo respuesta favorable ante la aplicación de bio-estimulante observando la presencia de brotes y callos en los nodos de la vara, Asian Pink Peace presentó mayor respuesta a la aplicación de bio-estimulante de brotes y callos, sin embargo el crecimiento del brote fue más lento a comparación de las variedades anteriores, Desert Breeze, obtuvo respuesta a la aplicación presentando callos no diferenciados y brotes, en cuanto a la longitud el crecimiento fue menor, en vara o keiki, la Violet Queen no tuvo respuesta al bio-estimulante.  por lo anterior observamos la importancia de las hormonas como bio-estimulantes en la producción de plantas nuevas en las orquídeas

    Gangrenous pyoderma (PG) is an often destructive rare inflammatory skin disease of unknown etiology and pathogenesis, belonging to the spectrum of neutrophilic dermatosis, characterized by papules, pustules, nodules or painful suffering erosion and progressively ulcerate, which is a diagnostic challenge when not counting today with clinical, paraclinical or pathognomonic histopathological this entity. We present a case report of a woman of 60, who attends the service of dermatology with large painful ulcerative lesion in the abdominal wall, pubic region, hip and right thigh along with gastrointestinal symptoms, which ultimately leads to the diagnosis of PG associated with ulcerative colitis.El Pioderma Gangrenoso (PG) es una enfermedad cutánea inflamatoria rara, de etiología y patogenia desconocida, perteneciente al espectro de las dermatosis neutrofílicas, frecuentemente destructiva, caracterizada por pápulas, nódulos o pústulas dolorosas que sufren erosión y ulceran progresivamente, que constituye un reto diagnóstico al no contar en la actualidad con características clínicas, paraclínicas ni histopatológicas patognomónicas de esta entidad. Aunque probablemente es originaria del sur de la República Mexicana o de América Central, en la actualidad se cultiva en forma comercial en Hawaii, Malaya, Ceilán, India, Sudáfrica, Tanganica, Kenya, Filipinas, Cuba, Jamaica, Puerto Rico, Panamá, Trinidad, Brasil y México. Nuestro país ocupa el quinto lugar como productor de papaya a nivel mundial. Su cultivo se ha ido extendiendo en los últimos años. Si bien es cierto que existen limitantes en ciertas regiones por las enfermedades de etiología viral, también es cierto que se está trabajando de manera puntual en el control de ellas como el control de las plagas. El mercado nacional e internacional se encuentra demandando producto por todas las atribuciones de la fruta tanto en la medicina como en lo industrial, dándole mucha importancia a la producción orgánica

    A Bacterial Shortcut to Amyloidosis

    21 p.-10 fig.The synthetic bacterial prionoid RepA-WH1 causes a vertically transmissible amyloid proteinopathy in Escherichia coli that inhibits growth and eventually kills the cells. Recent in vitro studies show that RepA-WH1 builds pores through model lipid membranes, suggesting a possible mechanism for bacterial cell death. By comparing acutely (A31V) and mildly (ΔN37) cytotoxic mutant variants of the protein, we report here that RepA-WH1(A31V) expression decreases the intracellular osmotic pressure and compromise bacterial viability under either aerobic or anaerobic conditions. Both are effects expected from threatening membrane integrity and are in agreement with findings on the impairment by RepA-WH1(A31V) of the proton motive force (PMF)-dependent transport of ions (Fe3+) and ATP1 synthesis. Systems approaches reveal that, in aerobiosis, the PMF-independent respiratory dehydrogenase NdhII is induced in response to the reduction in intracellular levels of iron. While NdhII is known to generate H2O2 as a by-product of the autoxidation of its FAD cofactor, key proteins in the defense against oxidative stress (OxyR, KatE), together with other stress-resistance factors, are sequestered by co-aggregation with the RepA-WH1(A31V) amyloid. Our findings suggest a route for RepA-WH1 toxicity in bacteria: a primary hit of damage to the membrane, compromising bionergetics, triggers a stroke of oxidative stress, which is exacerbated due to the aggregation-dependent inactivation of enzymes and transcription factors that enable the cellular response to such injury.     Helios modulates the maturation of a CA1 neuronal subpopulation required for spatial memory formation

    Currently, molecular, electrophysiological and structural studies delineate several neural subtypes in the hippocampus. However, the precise developmental mechanisms that lead to this diversity are still unknown. Here we show that alterations in a concrete hippocampal neuronal subpopulation during development specifically affect hippocampal-dependent spatial memory. We observed that the genetic deletion of the transcription factor Helios in mice, which is specifically expressed in developing hippocampal calbindin-positive CA1 pyramidal neurons (CB-CA1-PNs), induces adult alterations affecting spatial memory. In the same mice, CA3-CA1 synaptic plasticity and spine density and morphology in adult CB-CA1-PNs were severely compromised. RNAseq experiments in developing hippocampus identified an aberrant increase on the Visinin-like protein 1 (VSNL1) expression in the hippocampi devoid of Helios. This aberrant increase on VSNL1 levels was localized in the CB-CA1-PNs. Normalization of VSNL1 levels in CB-CA1-PNs devoid of Helios rescued their spine loss in vitro. Our study identifies a novel and specific developmental molecular pathway involved in the maturation and function of a CA1 pyramidal neuronal subtype

    Beneficial Effect of Ursodeoxycholic Acid in Patients with ACOX2 Deficiency-Associated Hypertransaminasemia

    Background: A variant (p.Arg225Trp) of peroxisomal acyl-CoA oxidase 2 (ACOX2), involved in bile acid (BA) side-chain shortening, has been associated with unexplained persistent hypertransaminasemia and accumulation of C27-BAs, mainly trihydroxycholestanoic acid (THCA). Aims: To investigate the prevalence of ACOX2 deficiency-associated hypertransaminasemia (ADAH), its response to ursodeoxycholic acid (UDCA), elucidate its pathophysiological mechanism and identify other inborn errors that could cause this alteration. Methods & results: Among 33 patients with unexplained hypertransaminasemia from 11 hospitals, and 13 of their relatives, 7 individuals with abnormally high C27-BA levels (>50% of total BAs) were identified by HPLC-MS/MS. The p.Arg225Trp variant was found in homozygosity (exon amplification/sequencing) in 2 patients and 3 family members. Two additional non-related patients were heterozygous carriers of different alleles: c.673C>T (p.Arg225Trp) and c.456_459del (p.Thr154fs). In ADAH patients, impaired liver expression of ACOX2, but not ACOX3, was found (immunohistochemistry). Treatment with UDCA normalized transaminases levels. Incubation of HuH-7 liver cells with THCA, which was efficiently taken up, but not through BA transporters, increased ROS production (flow cytometry), ER stress biomarkers (GRP78, CHOP and XBP1-S/XBP1-U ratio), and BAX¿ expression (RT-qPCR and immunoblot), whereas cell viability was decreased (MTT). THCA-induced cell toxicity was higher than that of major C24-BAs and was not prevented by UDCA. Fourteen predicted ACOX2 variants were generated (site-directed mutagenesis) and expressed in HuH-7 cells. Functional tests to determine their ability to metabolize THCA identified six with the potential to cause ADAH. Conclusion: Dysfunctional ACOX2 has been found in several patients with unexplained hypertransaminasemia. This condition can be accurately identified by a non-invasive diagnostic strategy based on plasma BA profiling and ACOX2 sequencing.     Bioavailability and systemic transport of oleanolic acid in humans, formulated as a functional olive oil

    14 Páginas.-- 6 Figuras.-- 2 TablasEvidence of the pharmacological activity of oleanolic acid (OA) suggests its potential therapeutic application. However, its use in functional foods, dietary supplements, or nutraceuticals is hindered by limited human bioavailability studies. The BIO-OLTRAD trial is a double-blind, randomized controlled study with 22 participants that received a single dose of 30 mg OA formulated as a functional olive oil. The study revealed that the maximum serum concentration of OA ranged from 500 to 600 ng mL-1, with an AUC0-∞ value of 2862.50 ± 174.50 ng h mL-1. Furthermore, we discovered a physiological association of OA with serum albumin and triglyceride-rich lipoproteins (TRL). UV absorption spectra showed conformational changes in serum albumin due to the formation of an adduct with OA. Additionally, we demonstrated that TRL incorporate OA, reaching a maximum concentration of 140 ng mL-1 after 2-4 hours.
