    Potential welfare issues of the Siamese fighting fish (Betta splendens) at the retailer and in the hobbyist aquarium

    Betta splendens is an extremely popular ornamental fish among hobby aquarists. It has an interesting behavioral repertoire, particularly where male aggression and territoriality are concerned. The lack of scientific studies investigating optimal housing conditions in combination with the wide variety of commercially available husbandry products, raises questions about the welfare status of these fish in captivity. In this article, an overview of the available literature on the biology of the betta and general considerations of ornamental fish keeping is given, and environment- and animal-related factors with potential impact on the welfare of Betta splendens are examined. Although more research using biological and physiological indicators is needed, the following factors constituting welfare problems have been identified: an aquarium of limited dimensions, prevalence of Mycobacterium spp. infection, aggression to and from conspecifics or other species in the same aquarium and the limited ability to escape, potential for stress due to prolonged visual contact between males in shops and during shows, and the lack of environmental enrichment in the form of sheltering vegetation

    Gene expression profile of Scardovia spp. in the metatranscriptome of root caries

    A few investigations of caries biofilms have identified Scardovia spp.; however, little is known about its involvement in caries pathogenesis. The purpose of this study was to assess the gene expression profile of Scardovia spp. in root caries, and compare it with other microorganisms. Clinical samples from active root caries lesions were collected. Microbial mRNA was isolated and cDNA sequenced. The function and composition of the Scardovia were investigated using two methods: a) de novo assembly of the read data and mapping to contigs, and b) reads mapping to reference genomes. Pearson correlation was performed (p < 0.05). Proportion of Scardovia inopinata and Scardovia wiggsiae sequences ranged from 0-6% in the root caries metatranscriptome. There was a positive correlation between the transcriptome of Lactobacillus spp. and Scardovia spp. (r = 0.70; p = 0.03), as well as with other Bifidobacteriaceae (r = 0.91; p = 0.0006). Genes that code for fructose 6-phosphate phosphoketolase (the key enzyme for “Bifid shunt”), as well as ABC transporters and glycosyl-hydrolases were highly expressed. In conclusion, “Bifid shunt” and starch metabolism are involved in carbohydrate metabolism of S. inopinata and S. wiggsiae in root caries. There is a positive correlation between the metabolism abundance of Lactobacillus spp., Bifidobacteriaceae members, and Scardovia in root caries

    The role of Candida albicans in root caries biofilms: an RNA-seq analysis

    Objective: This study sought to analyze the gene expression of Candida albicans in sound root surface and root caries lesions, exploring its role in root caries pathogenesis. Methodology: The differential gene expression of C. albicans and the specific genes related to cariogenic traits were studied in association with samples of biofilm collected from exposed sound root surface (SRS, n=10) and from biofilm and carious dentin of active root carious lesions (RC, n=9). The total microbial RNA was extracted, and the cDNA libraries were prepared and sequenced on the Illumina Hi-Seq2500. Unique reads were mapped to 163 oral microbial reference genomes including two chromosomes of C. albicans SC5314 (14,217 genes). The putative presence of C. albicans was estimated (sum of reads/total number of genes≄1) in each sample. Count data were normalized (using the DESeq method package) to analyze differential gene expression (using the DESeq2R package) applying the Benjamini-Hochberg correction (FDR<0.05). Results: Two genes (CaO19.610, FDR=0.009; CaO19.2506, FDR=0.018) were up-regulated on SRS, and their functions are related to biofilm formation. Seven genes ( UTP20 , FDR=0.018; ITR1 , FDR=0.036; DHN6 , FDR=0.046; CaO19.7197 , FDR=0.046; CaO19.7838 , FDR=0.046; STT4 , FDR=0.046; GUT1 , FDR=0.046) were up-regulated on RC and their functions are related to metabolic activity, sugar transport, stress tolerance, invasion and pH regulation. The use of alternative carbon sources, including lactate, and the ability to form hypha may be a unique trait of C. albicans influencing biofilm virulence. Conclusions: C. albicans is metabolically active in SRS and RC biofilm, with different roles in health and disease

    Actinomyces spp. gene expression in root caries lesions

    Background: studies of the distribution of Actinomyces spp. on carious and non-carious root surfaces have not been able to confirm the association of these bacteria with root caries, although they were extensively implicated as a prime suspect in root caries. Objective: the aim of this study was to observe the gene expression of Actinomyces spp. in the microbiota of root surfaces with and without caries. Design: the oral biofilms from exposed sound root surface (SRS; n=10) and active root caries (RC; n=30) samples were collected. The total bacterial RNA was extracted and the mRNA was isolated. Samples with low RNA concentration were pooled, yielding a final sample size of SRS=10 and RC=9. cDNA libraries were prepared and sequenced on the Illumina Hi-Seq2500. Sequence reads were mapped to eight Actinomyces genomes. Count data were normalized using DESeq2 to analyse differential gene expression applying the Benjamini-Hochberg correction (FDR0.05), except for Actinomyces OT178 (p=0.001) and A. gerencseriae (p=0.004), which had higher read count in the SRS. Genes that code for stress proteins (clp, dnaK and groEL), enzymes of glycolysis pathways (including, enolase and phosphoenolpyruvate carboxykinase), adhesion (Type-2 fimbrial and collagen-binding protein) and cell growth (EF-Tu) were highly, but not differentially (p>0.001) expressed in both groups. Genes with the most significant up-regulation in RC were those coding for hypothetical proteins and uracil DNA glycosylase (p=2.61E-17). The gene with the most significant up-regulation in SRS was a peptide ABC transporter substrate-binding protein (log2FC= -6.00, FDR= 2.37E-05). Conclusion: there were similar levels of Actinomyces gene expression in both sound and carious root biofilms. These bacteria can be commensal in root surface sites, but may be cariogenic due to survival mechanisms allowing them to exist in acid environment and metabolize sugars saving energy

    The metatranscriptomes of root caries and sound root surface biofilms

    There is limited knowledge of bacterial metabolism in root caries lesions. The aim of this study was to describe the bacterial metatranscriptomes associated with root caries and sound root surfaces using an RNA-seq analysis approach. The biofilms from exposed root surfaces were sampled from caries-free volunteers (n=10), and from the infected dentine of volunteers with root caries (n=30). Total bacterial RNA was extracted; cDNA libraries were prepared and sequenced on the Illumina Hi-Seq2500. The function and composition of the metabolically active microbiota were investigated using: a) MG-RAST, and b) denovo assembly of the read data and mapping to contigs. Differential gene expression analysis was done using the R package DESeq2 (padj <10−3). Transcripts with the highest expression levels were those coding for membrane transport systems, ribosomal proteins, enolase and glycolytic pathways in both groups. Differential analysis indicated that genes coding for the OmpA domain protein and metalloprotease domain protein were over-expressed in the caries samples (log2FoldChange = –12.2; padj= 3.5 × 10−13), whereas genes in the samples from healthy sites over-expressed pilus biosynthesis protein, thiamine diphosphokinase and transporter protein (log2FoldChange = 16.5; padj = 2.2 × 10−21).     Importance of air-sea coupling in simulating tropical cyclone intensity at landfall

    An atmosphere-only model system for making seasonal prediction and projecting future intensities of landfalling tropical cyclones (TCs) along the South China coast is upgraded by including ocean and wave models. A total of 642 TCs have been re-simulated using the new system to produce a climatology of TC intensity in the South China Sea. Detailed comparisons of the simulations from the atmosphere-only and the fully coupled systems reveal that the inclusion of the additional ocean and wave models enable differential sea surface temperature responses to various TC characteristics such as translational speed and size. In particular, interaction with the ocean does not necessarily imply a weakening of the TC, with the coastal bathymetry possibly playing a role in causing a near-shore intensification of the TC. These results suggest that to simulate the evolution of TC structure more accurately, it is essential to use an air-sea coupled model instead of an atmosphere-only model

    Optic neuritis in Asian type opticospinal multiple sclerosis (OSMS-ON) in a non-Asian population: A functional-structural paradox

    Background: Biomarkers have improved the classification of autoimmune inflammatory disorders, including optic neuritis (ON) as a frequent presentation of multiple sclerosis, neuromyelitis spectrum disorders, MOG antibody-related disease (MOGAD), and opticospinal multiple sclerosis (OSMS). The phenotype of OSMS in non-Asian populations is less well known. // Objective: We investigated the clinical features and prognosis of OSMS–ON in a Brazilian cohort. // Methods: This was a single-center cohort study of patients from Rio de Janeiro (Brazil) with OSMS. All individuals were MOG- and AQP4-seronegative, clinically diagnosed with ON, and had magnetic resonance imaging-confirmed transverse myelitis (TM). Subjects and healthy controls (HCs) were assessed for visual acuity (logMAR VA), automated perimetry mean deviation (MD), intraocular pressure, and spectral-domain optical coherence tomography (OCT), followed by automated retinal layer segmentation of the peripapillary retinal nerve fiber layer (pRNFL) and macular ganglion cell and inner plexiform layer (mGCIPL). Receiver operator characteristic curves were plotted and the area under the curve (AUC) was calculated for group comparisons of retinal asymmetry of the pRNFL and mGCIPL. // Results: The 30 patients with OSMS were predominantly female and white. The mean age was 48 years (range 20–70 years). Unilateral ON was the index event in 83.3% of patients. Over the average 18-year follow-up period, there were 89 relapses of ON. In individuals with OSMS, the average VA was 0.07±0.14 in the right eye (RE) and 0.13±0.30 in the left eye (LE). The MD was −5.37±5.88 dB and −5.23±3.34 dB for the RE and LE, respectively. There was a significant cumulative loss of VA (p = 0.0003) and MD (p = 0.0001) with a higher number of recurrent episodes. Atrophy of the pRNFL thickness was significant in OSMS (RE, 78.62 ± 16.01 ”m; LE, 79.86 ± 13.79 ”m) relative to the HC group (RE, 98.87 ± 10.68 ”m; LE, 97.87 ± 10.85 ”m, p = 0.0001). Likewise, there was significant mGCIPL atrophy in patients with OSMS (RE, 74.96 ± 14.46 ”m; LE, 73.88 ± 13.79 ”m) relative to the HC group (RE, 90.50 ± 6.74 ”m; LE, 90.41± 6.89 ”m; p = 0.0001). Retinal asymmetry, inter-eye percentage, and absolute differences accurately separated patients with unilateral ON from HCs (AUC=0.89 and AUC=0.85, respectively). // Conclusion: A structural-functional paradox was found in OSMS with a high diagnostic value for a novel metric based on retinal asymmetry. The functional visual outcome are excellent despite significant structural damage to the inner retinal layers in patients with a high ON relapse rate and long-term bilateral sequential involvement

    Optical types of inland and coastal waters

    Inland and coastal waterbodies are critical components of the global biosphere. Timely monitoring is necessary to enhance our understanding of their functions, the drivers impacting on these functions and to deliver more effective management. The ability to observe waterbodies from space has led to Earth observation (EO) becoming established as an important source of information on water quality and ecosystem condition. However, progress toward a globally valid EO approach is still largely hampered by inconsistences over temporally and spatially variable in-water optical conditions. In this study, a comprehensive dataset from more than 250 aquatic systems, representing a wide range of conditions, was analyzed in order to develop a typology of optical water types (OWTs) for inland and coastal waters. We introduce a novel approach for clustering in situ hyperspectral water reflectance measurements (n = 4045) from multiple sources based on a functional data analysis. The resulting classification algorithm identified 13 spectrally distinct clusters of measurements in inland waters, and a further nine clusters from the marine environment. The distinction and characterization of OWTs was supported by the availability of a wide range of coincident data on biogeochemical and inherent optical properties from inland waters. Phylogenetic trees based on the shapes of cluster means were constructed to identify similarities among the derived clusters with respect to spectral diversity. This typification provides a valuable framework for a globally applicable EO scheme and the design of future EO missions

Foi construĂ­do um jogo de tabuleiro denominado Drogas: jogando limpo . Na VersĂŁo Alfa os especialistas fizeram sugestĂ”es em relação Ă s e instruçÔes e ao tabuleiro: textura das casas, peças do jogo e escrita em Braille. Na VersĂŁo Beta, procedeu-se Ă  avaliação pelos cegos, os quais sugeriram alteraçÔes na textura das casas e colocação de velcro para fixação do pino no tabuleiro. Passou-se, entĂŁo, Ă  VersĂŁo Gama, jogada pelas Ășltimas trĂȘs duplas de cegos e considerada adequada. Na avaliação dos juĂ­zes, o jogo revelou-se adequado, pois permite o acesso Ă  informação sobre drogas psicoativas de maneira lĂșdica
