    飞机结构疲劳可靠度贝叶斯组合预测

    在实际工程中,常常需要利用模型去描述和分析问题。然而模型亦存在不确定性,即可能存在多个描述同一现象的模型,例如多个疲劳分析的模型。针对飞机结构的疲劳可靠性问题,提出在考虑三种裂纹扩展模型下基于贝叶斯公式的疲劳可靠度组合预测方法。针对不同应力水平下飞机结构试件的裂纹扩展数据建立了三种随机裂纹扩展模型;在考虑模型参数不确定性条件下,采用贝叶斯模型平均方法对三种模型进行组合;基于组合模型分析结构的可靠度。所提方法在分析飞机结构疲劳可靠度上,采用了组合模型,能够最大限度保障结果的稳定性。此外,考虑了模型参数的不确定性,能够得到更为合理的裂纹扩展预测分布和可靠度预测值。给出的实例及分析结果表明所提方法可行。国家自然科学基金资助项目(U1530122,51505398);;中央高校基本科研业务费专项资金资助项目(20720180072

    Determination of,Three Vitamins in Compound Vitamin B Tablets by HPLC

    目的 采用高效液相色谱法同时测定维生素B族片中三种维生素B(维生素B1,维生素B2,维生素B6)的含量。方法 采用C18柱,以0.005mol·L-1的庚烷磺酸钠溶液(含0.5%冰醋酸和0.05%三乙胺)-甲醇(65:35,v:v)为流动相,流速为1.0m L·min-1。紫外检测波长为280nm。结果 本实验中维生素B1,B2,B6线性范围分别为2.964~74.1mg·m L-1,2.934~73.35mg·m L-1,2.996~74.9mg·m L-1,回收率分别为97.5%,99.7%,100.5%,RSD分别为1.21%,1.5%,0.9%。结论 本法快速、简便、分离效果好,适用于多维片中维生素B1,维生素B2,维生素B6的质量控制。OBJECTIVE To develop an HPLC method for the determination of three vitamin B in compound vi- tamin tablets. METHODS HPLC, C18 column was used, a mixture of 0. 005mol . L-1 sodium heptanesulfonate (containing 0. 5% glacial acid and 0. 05% triethylamine)-methanol(72:28 ,v/v)as mobile phase at a flow rate of 1.0mL . min-1. The UV detection wavelength was 280nm. RESULTS The calibration curves Showed good linearity in the range of 2. 964 -74. 1mg. mL-1,2. 934 -73.35mg. mL-1 ,2. 996 -74. 9mg . mL-1 ,recovery rate of vita- mins were 97. 5% ,99. 7%, 100. 5%, and RSDs were 1.15% ,0. 77% ,0. 80% respectively. CONCLUSION The method is rapid, convenient and reliable and could be used for quality control of compound vitamin B tablets

    Feature reduction on high-dimensional small-sample data

    针对一类高维少样本数据的特点,给出了广义小样本概念,对广义小样本进行信息特征压缩:特征提取(降维)和特征选择(选维)。首先介绍基于主成分分析(PCA)的无监督与基于偏最小二乘(PlS)的有监督的特征提取方法;其次通过分析第一成分结构,提出基于PCA与PlS的新的全局特征选择方法,并进一步提出基于PlS的递归特征排除法(PlS-rfE);最后针对MITAMl/All的分类问题,实现基于PCA与PlS的特征选择和特征提取,以及PlS-rfE特征选择与比较,达到广义小样本信息特征压缩的目的。In view of the characteristics of small sample and high dimensional data,Generalized Small Samples(GSS) is defined.It reduces information feature of GSS:feature extraction(dimensionality extraction) and feature selection(dimensionality selection).Firstly,unsupervised feature extraction based on Principal Component Analysis(PCA) and supervised feature extraction based on Partial Least Squares(PLS) are introduced.Secondly,analyzing the structure of first PC,it presents new global PCA-based and PLSbased feature selection approaches,in addition recursive feature elimination on PLS(PLS-RFE) is realized.Finally,the approaches are applied to the classification of MIT AML/ALL,it performs feature extraction on PCA and PLS,and feature selection compared with PLS-RFE.The information compression of GSS is realized.高校博士点专项科研基金(No20070384003);福建省教育厅科技项目(NoJB08244

    农村留守经历大学生心理行为与人际关系分析

    目的探讨农村留守经历大学生(URLB)心理行为特征以及与非URLB心理行为上的差异,为采取干预措施提供依据。方法采用流调中心用抑郁量表、状态特质焦虑问卷、简易应对方式问卷、自尊量表和大学生人际关系综合诊断量表对河北省2所高校的4080名大学生进行问卷调查。结果 URLB大学生负性情绪、积极应对方式、自尊和人际关系与非URLB大学生差异有统计学意义。相关分析表明,URLB大学生负性情绪、消极应对方式与人际关系呈显著正相关,积极应对方式、自尊与人际关系呈显著负相关。回归分析表明,URLB大学生负性情绪、应对方式和自尊对人际关系有显著的预测作用。结论 URLB与非URLB大学生在心理行为上差异有统计学意义,需要对URLB进行适当的心理干预

    Isolation of Total Cellular DNA from Three Species of Lauraceae

    为了从富含次生代谢物的樟科植物肉桂、锡兰肉桂、阴香中获得高质量DNA ,研究和改进了CTAB法、高盐低pH法和尿素法。改进方法包括 :1)在裂解液中加入 2 % β -巯基乙醇和 5 %PVP ,以防止氧化褐变的发生 ;2 )在酚 :氯仿抽提前加入 1 5mol L醋酸铵冰浴处理 ,能降低DNA的黏性。所得DNA的质量和产量经电泳、紫外吸收A2 60 A2 80 、PCR扩增和限制性内切酶酶切检测 ,结果表明改进法提取的DNA质量要比常规法的好。其中改进的CTAB法获得的DNA纯度最高 ,能用于PCR扩增和限制性内切酶酶切 ,是提取这 3种樟科植物总DNA的最佳方法。CTAB method,low pH medium with high salt method (LPHS),and urea method were used and improved to extract high-quality DNA from Lauraceae plant Cinnamomum cassia Presl.,C.zeylanicum and C.burmannii,which are rich in secondary metabolites.The modified protocols involved two key steps:1) adding 2% β-mercaptoethnoal and 5% polyvinylpyrrolidone (PVP) in extraction solution to inhibit the oxidization of polyphenols;2) treating with 1.5?mol/L NH 4Ac and incubation at 0℃ prior to supernatant extraction with phenol:chloroform to lower the viscosity of DNA.The yields and qualities of DNA were analyzed by electrophoresis,ratios of A (260)/A (280),PCR amplification and restriction digestion.The results showed that the modified methods are better than the original ones in terms of the quality of total DNA.Among them,the modified CTAB method,which can produce high quality DNA suitable for PCR amplification and restriction digestion,is the best one for DNA extraction from three species of Lauraceae.厦门大学预研基金资

    CDK5-dependent BAG3 degradation modulates synaptic protein turnover

    阿尔茨海默病(AD)是严重威胁人类健康的重大神经系统疾病,AD的发生发展与衰老密切相关,目前临床治疗方法十分有限。因此迫切需要从AD致病早期入手,发现和鉴定导致AD神经功能紊乱的机制和靶点,为AD的早期防治提供基础。张杰教授及其团队从高通量磷酸化蛋白质组学入手,系统研究了CDK5在神经细胞中的磷酸化底物,鉴定出了在蛋白质量控制中发挥重要功能的BAG3蛋白是CDK5的全新底物。课题组从磷酸化蛋白质组学入手,发现和阐明了细胞周期蛋白激酶5(CDK5)通过调控BAG3在维持突触蛋白水平调控中的作用机制,及其在阿尔茨海默病(AD)发生发展中的机理。 该研究是多个团队历时8年合作完成的,香港中文大学的周熙文教授、美国匹兹堡大学的Karl Herrup教授、美国Sanford-Burnham研究所的许华曦教授、美国梅奥医学中心的卜国军教授,厦门大学医学院的文磊教授、张云武教授、赵颖俊教授、薛茂强教授,军事医学科学院的袁增强教授等都参与了该工作。 厦门大学医学院2012级博士生周杰超等为文章的第一作者,张杰教授为通讯作者。Background Synaptic protein dyshomeostasis and functional loss is an early invariant feature of Alzheimer’s disease (AD), yet the unifying etiological pathway remains largely unknown. Knowing that cyclin-dependent kinase 5 (CDK5) plays critical roles in synaptic formation and degeneration, its phosphorylation targets were re-examined in search for candidates with direct global impacts on synaptic protein dynamics, and the associated regulatory network was also analyzed. Methods Quantitative phospho-proteomics and bioinformatics analyses were performed to identify top-ranked candidates. A series of biochemical assays were used to investigate the associated regulatory signaling networks. Histological, electrochemical and behavioral assays were performed in conditional knockout, shRNA-mediated knockdown and AD-related mice models to evaluate its relevance to synaptic homeostasis and functions. Results Among candidates with known implications in synaptic modulations, BCL2-associated athanogene-3 (BAG3) ranked the highest. CDK5-mediated phosphorylation on Ser297/Ser291 (Mouse/Human) destabilized BAG3. Loss of BAG3 unleashed the selective protein degradative function of the HSP70 machinery. In neurons, this resulted in enhanced degradation of a number of glutamatergic synaptic proteins. Conditional neuronal knockout of Bag3 in vivo led to impairment of learning and memory functions. In human AD and related-mouse models, aberrant CDK5-mediated loss of BAG3 yielded similar effects on synaptic homeostasis. Detrimental effects of BAG3 loss on learning and memory functions were confirmed in these mice, and such were reversed by ectopic BAG3 re-expression. Conclusions Our results highlight that neuronal CDK5-BAG3-HSP70 signaling axis plays a critical role in modulating synaptic homeostasis.     王其新 男, 1 9 7 3 年毕业于中山大学药用植物学专业。现任湛江市药品检验所中药室主任, 主任中药师。擅长生药学、中药鉴定学和植物化学,连续参加三版中国药典部分药品标准的起草和修订工作, 对草豆范的薄层色谱鉴别进行了系统的研究。所建立的薄层色谱法被中国药典采用。编著著作1 本, 获广东药学院科技进步二等奖1 项, 发表科学论文28 篇, 取得“ 壮骨口服液” 发明专利1 项。为了鉴别金线莲的品种,探索同属植物之间的亲缘关系及同种 植物不同产地的遗传变异,并制定正品金线莲的 DNA 指纹图谱,采用随机扩增多态 DNA(RAPD)技术对兰科开唇兰属植物花叶开唇兰 Anoectochilus roxburghii(Wal-l.).Lindl.、台湾开唇兰 A.formosanus Hay. 进行了鉴定,结果选 择的 7 个引物对 3 个品种 共扩增出 98 个位点,其中 3 个引物为高特异性引物。结论 RAPD 技术不仅能鉴别种间差 异,而且能揭示同种不同产地植物的遗传变异。厦门市卫生局医学科学技术进步基金科研资助项

    目的:为获得能用于中药肉桂RAPD分析及其它分子生物学研究的高质量DNA,研究和改进几种DNA提取法。方法:先将样品与PVP和VitC共研碎,然后用Tris-HCI缓冲液洗涤样品,沉淀用改进的CTAB法、高盐低pH法和尿素法来提取DNA。通过电泳、紫外吸收A_(260)/A_(280)和PCR扩增检测所得DNA的产量和质量。结果:改进的CTAB法和高盐低pH法都能获得高质量的DNA进行PCR扩增,其中以改进的CTAB法效果最好。Objective: To extract high-quality DNA for RAPD and other molecular biology research from Chinese traditional medicine Cinnamomum cassia Presl.. Method: Improved CTAB method, low pH medium with high salt method(LPHS) and urea method were used for DNA extract. The yield and quality of DNA was analysed by electrophoresis, ratios of A_(260)/A_(280) and PCR amplification.
