    The Isolation,Identification and Activity Assays of the Myxobacteria

    通信作者:[email protected]近年来,粘细菌由于其次级代谢产物结构新颖、生物活性多样而备受关注,成为发展新药的重要来源.利用已灭活酵母菌或大肠杆菌(Escherichia coli)诱导法和滤纸片诱导法等方法进行粘细菌的分离,并采取多种方法对所分离出菌株进行纯化,从95份土样中共分离出118株粘细菌,纯化出54株.根据16SrDNA进行菌株鉴定,主要的粘细菌为粘球菌属(Myxococcus)、珊瑚菌属(Corallococcus)、多囊菌属(Polyangium)和堆囊菌属(Sorangium).对获得纯化的菌株进行了抗菌抗肿瘤活性测定,结果显示有46.3%的菌株具有抗菌作用,有66.7%的菌株具有抗肿瘤作用,表明粘细菌是抗菌抗肿瘤活性物质的重要来源.[英文文摘]Since many different types of  novel compounds with biological activities are discovered form Myxobacteria,Myxobacteria,become an important resource of medicine.In this experiment,through studying 95 soil and wood samples which were collectd from different regions of China,118Myxobacteria strains were isolated in which 54 isolates were purified.The morphological identification and 16 SrDNA sequencing of 54 Myxobacteria strains showed that they are distributed in 4 genera,and most of them belong to Myxococcus spp.and Sorangium spp ..Antibiotics and anti-tumor activities of the crude extracts of these strains were also tested.科技部国际合作项目(2007DFA30970);中央高校基本业务费(2010121092

    Screening of Epothilones Producing Strains Sorangium cellulosum

    通信作者:[email protected][中文文摘]为寻找潜在的产生埃博霉素(epothilone)的纤维堆囊菌(Sorangium cellulosum),以埃博霉素生物合成基因簇中的epoA,epoC和epoK基因为探针,分别对60株不同土壤样品来源的纤维堆囊菌的DNA样品进行PCR筛选.筛选结果得到了7株阳性菌株,进一步对其进行发酵和粗提物的HPLC-MS分析,检测结果显示3株菌XMU-Nc-03,XMU-So-64和XMU-So-112能产生埃博霉素B,其中XMU-So-112的产量最高.本实验在筛选产埃博霉素活性菌株的同时,还建立了一个快速可靠的发现埃博霉素产生菌的方法.[英文文摘]Epothilones are a new class of anticancer compounds produced by the Gram-negative myxobacterium Sorangium cellulosum.It shows high cytotoxicity for animal cells and mimic biological effect of taxol.In order to obtain epothilone producing strains,three pairs of primers were used to amplify frgments of epoA,epo C and epo K which encoding loading module,polyketide synthases (PKS) and P450 enzyme respectively in epothilone biosynthesis cluster from sixty S.cellulosumstrains,and the PCR products were sequenced to avoid the false-positive strains.Seven isolates XMU-So-58,XMU-So-64,XMU-So-70,XMU-So-72,XMU-So-112,XMU-So-264and XMU-Nc-03 were found to have at least one fragment of epoA,epo C and epo K.Then they were successfully purified and their secondary metabolites were analyzed using epothilone B as positive control by HPLC-Ms.The result showed that XMU-So-58,XMU-So-70,XMU-So-72 and XMU-So-264 didn't produce epothilone.While epothilone B were detected in the fermentation products of three strains,XMU-Nc-03,XMU-So-64 and XMU-So-112 with intensity of 2.7×105,6.2×105 and 7.0×106 respect vely,at the retention time of 9.8-9.9 min.Thus,XMU-So-112 was proved to be the best producer.In summary,the best producer of epothilone B not only was obtained from 60 Sorangiumstrains,but also it was established that an efficient method for screen ing antibiotic producersbased on PCR screening coupled with HPLC-MS analysis.科技部国际合作项目(2007DFA30970);中央高校基本科研业务费专项(2010121092,2011121037

    2002–2010年中国典型生态系统辐射及光能利用效率数据集

    辐射是陆地生态系统能量的主要来源,其利用效率表现为光能利用率,反映了生态系统转化光能、生成有机物质的能力。揭示典型生态系统的辐射及光能利用效率可以为评估区域光能资源及其利用效率提供参考,也为评估区域有机物质固定能力及碳吸收能力提供依据。基于中国陆地生态系统通量观测研究联盟(China FLUX)的长期观测结果及已发表文献的公开数据,构建了2002–2010年中国典型生态系统辐射及光能利用效率数据集,包含51个生态系统126个站点年辐射、光能利用效率及吸收光能利用效率的观测记录。另外,本数据集还包含生态系统代码、年份、经度、纬度、海拔、生态系统类型、年均气温、年总降水量、年均CO2质量浓度、年均叶面积指数、最大叶面积指数等生物气候信息。本数据集可以为评估生态系统生产能力、应对气候变化等方面的研究提供数据支持

    2000–2010年中国典型陆地生态系统实际蒸散量和水分利用效率数据集

    蒸散是陆地生态系统水分循环和能量平衡的关键过程,水分利用效率是反映生态系统碳水循环间耦合关系的重要指标,二者在生态学、农学、水文学、气候学等多个学科中均具有重要的应用价值。涡度相关法被认为是现今唯一能直接测量生物圈与大气间物质与能量交换通量的标准方法,已成为生态系统尺度碳水交换通量观测的主要方法。本文通过整合中国陆地生态系统通量观测联盟(China FLUX)的长期观测数据和中国区域其他观测站点基于涡度相关法发表的文献数据,构建了一套中国典型陆地生态系统实际蒸散量和水分利用效率数据集。本数据集共有实际蒸散量数据记录143条、水分利用效率数据记录96条,涉及5种生态系统类型45个生态系统,时间跨度为2000–2010年。本数据集可以为陆地生态系统碳水循环、生态系统管理和评估、全球变化等相关领域的研究提供数据支持
