    Study of molecular oxygen titration into nitrogen post-discharge

    Za posledních padesát až šedesát let byla provedena celá řada experimentů v oblasti dohasínajícího dusíkového plazmatu a uveřejněno velké množství teoretických prací. Některé práce se zabývaly studiem dusíku v aktivním výboji, samotným dohasínáním nebo se zaměřily převážně na probíhající kinetické procesy v dohasínajícím plazmatu. Metodou uvedenou v práci bylo vyhodnoceno, jak ovlivní titrace kyslíku dohasínající dusíkové plazma v proudícím režimu. Experimentální výsledky byly získány za pomoci optické emisní spektroskopie. Získaná spektra byla změřena v rozsahu 300 - 700 nm. Všechna měření se prováděla za laboratorní teploty 300 K. Proud, byl udržován na konstantní hodnotě 120 mA, a to odpovídá výkonu 145 W. Tlak, byl po celou dobu měření také konstantní o hodnotě 1000 Pa. Průtok dusíku o čistotě 99,9999 %, dále dočišťovaného pomocí kolony Oxycler, aktivním výbojem byl 0,8 l/min. Průtok kyslíku čistoty 99,95 % titrační trubicí zaváděnou proti směru proudění dohasínajícího plazmatu byl udržován na hodnotě 4 ml/min. Oba průtoky byly nastaveny pomocí regulátorů hmotnostního průtoku. K detekci záření sloužil optický spektrometr Jobin Yvon TRIAX 550 s mřížkou 300 čar/mm a CCD detektorem chlazeným kapalným dusíkem. Při všech měřeních byla použita integrační doba 1s. Poloha titrační trubice zaváděné proti směru proudění plynu byla vzhledem ke konci aktivního výboje nastavována po 1 cm od 5 cm do 25 cm. Optická spektra pak byla snímána v polohách 3 - 29 cm od konce aktivního výboje. Ve spektrech byly identifikovány tyto spektrální systémy: 1. pozitivní, 1. negativní a 2. negativní systém dusíku. Kromě nich bylo nalezeno i několik spektrálních pásů odpovídajících spektrálnímu systému NO-beta .Z experimentálních dat pak byly stanoveny průběhy intenzit jednotlivých spektrálních pásů a změny ve vibračních rozděleních horních elektronových stavů výše uvedených spektrálních systémů.A huge number of experiments were carried out in the field of nitrogen post-discharges during the last 50 or 60 years and they were supported by many published theoretical works. Some papers were focused also on the nitrogen active discharge, post-discharge itself, or they focused mainly on the kinetic processes running during the post-discharge period. This experimental work shows how oxygen titration into post-discharge will influence nitrogen flowing post-discharge. Experimental data were obtained by optical emission spectrometry, Spectra were measured in the range 300 - 700 nm at laboratory temperature of 300K. Discharge current was kept constant at the value of 120 mA relating to the total discharge power of 145 W. Pressure was kept constant, too, at the value of 1000 Pa. The nitrogen of 99.9999 % purity (further purified by Oxiclear column) flow was adjusted at 0.8 l/min. Flow of oxygen (99.95 % purity) through he titration capillary introduced to post-discharge from down stream direction, was kept at 4 ml/min. Both gas flows were controlled by mass flow controllers. The optical emission spectrometer Jobin Yvon TRIAX 550 with 300 gr/mm grating equipped by liquid nitrogen cooled CCD detector was used for the spectra acquisition. The integration time of 1 s was used at all experiments. The position of titration tube end introduced into post discharge from the down stream side was set from 5 to 25 cm with respect to the end of the active discharge; the step of 1 cm was used. The optical emission spectra were measured at positions from 3 to 29 cm with respect to the active discharge end. The following nitrogen spectral systems were identified in the spectra: 1st positive, 1st negative and 2nd positive. Besides them, some bands of NO-beta system were found. The intensity profiles along the post discharge were obtained for selected vibrational spectral bands of these spectral systems and changes in the vibrational distributions of upper electronic states of these spectral systems were determined.

V teoretické části jsou obsaženy informace o geodetických a polohopisných základech, metody měření a náležitosti s přesností měření skutečného provedení přeložky potrubí. V praktické části se věnuji zaměřením bodů pro vyhotovení účelové mapy, jejiž součástí jsou měřické práce v terénu a zpracování v softwarech Groma 8.0 a AutoCAD 2022.The diploms thesis aim for the actual relocation of mine water pipilines at the cadastral area of Vítkovice. The theoretical part contains information about geodetic and topographical foundations, measurement methods and essentials with the accuracy of measuring the actual pipeline relocation.     Species of Ergasilus von Nordmann, 1832 (Copepoda: Ergasilidae) from cichlid fishes in Lake Tanganyika

    Ergasilus (von Nordmann, 1832) (Ergasilidae) is a species-rich group of parasitic copepods with a wide distribution in freshwater, marine and brackish environments. Up to now, 9 species of Ergasilus are known from cichlid fishes in Africa. In this study, 5 species, including 3 new, were collected from the gills of 12 cichlid species (11 genera: Bathybates, Ctenochromis, Eretmodus, Gnathochromis, Lamprologus, Neolamprologus, Ophthalmotilapia, Perissodus, Simochromis, Spathodus and Tanganicodus) of the northeastern shore of Lake Tanganyika in Burundi, namely E. macrodactylus (Sars, 1909), E. megacheir (Sars, 1909), E. caparti n. sp., E. parasarsi n. sp. and E. parvus n. sp. All species found were identified and described on the basis of adult female specimens using an integrative taxonomy approach mixing morphological characterization and molecular analyses of 2 ribosomal DNA markers (partial 18S and 28S rDNA sequences). An identification key for Ergasilus species from Lake Tanganyika is included. This study provides the first molecular data for Ergasilus species in Africa. The phylogenetic analyses suggest that the Ergasilus species parasitizing Lake Tanganyikan cichlids form a well-supported clade within the Ergasilidae. However, their phylogenetic relationships with other congeners still remain unclear due to a lack of molecular data for this diverse genus

    Nearctic Dactylogyrus species (Platyhelminthes, Monogenea) parasitizing cypriniform fishes in the context of morphology and phylogeny, with descriptions of seven new species

    Ribosomal DNA sequences are currently available for 32 morphologically recognized species of Dactylogyrus parasitizing Nearctic cypriniforms, but only 16 of them are assigned to nominal species. Herein, morphological data on 28 of the 32 species are provided, together with comments on their phylogenetic relationships in the context of the morphology of taxonomically important structures. Seven new species of Dactylogyrus are described from five species of leuciscids and one species of catostomids, as follows: D. aduncus n. sp. from Campostoma spadiceum, D. cloutmani n. sp. from Luxilus chrysocephalus isolepis, D. cornifrons n. sp. from Cyprinella venusta, D. fimbratus n. sp. from Rhinichthys cataractae, D. mcallisteri n. sp. from Semotilus atromaculatus, and D. chieni n. sp. and D. haneki n. sp. from Hypentelium nigricans. Four previously described species of Dactylogyrus, D. atromaculatus from S. atromaculatus, D. eos from C. neogaeus, D. parvicirrus from Notemigonus crysoleucas and D. perlus from L. c. isolepis, are redescribed and/or figured. As for the remaining 17 species, only the male copulatory organs (MCOs) taken from the respective hologenophores are illustrated. On the basis of phylogenetic analyses, two main clades of Nearctic Dactylogyrus were recognized and supported by the different morphology of the MCO. The first one included 22 strictly Nearctic species sharing the same MCO type with Dactylogyrus spp. parasitizing cyprinids likely of Asian origin. The second clade encompassed Dactylogyrus spp. with diverse MCO morphology and was placed in the sister position to Dactylogyrus spp. parasitizing European leuciscids and North-West African cyprinids

    Diversity, phylogeny and intraspecific variability of Paradiplozoon species (Monogenea: Diplozoidae) parasitizing endemic cyprinoids in the Middle East

    Diplozoidae are common monogenean ectoparasites of cyprinoid fish, with the genus Paradiplozoon being the most diversified. Despite recent studies on Diplozoidae from Europe, Africa and Asia, the diversity, distribution and phylogeny of this parasite group appears to be still underestimated in the Middle East. The objective of this study was to investigate the diversity, endemism and host specificity of diplozoids parasitizing cyprinoid fish from the Middle East, considering this region as an important historical interchange of fish fauna, and to elucidate the phylogenetic position of Middle Eastern Paradiplozoon species within Diplozoidae. Four Paradiplozoon species were collected from 48 out of 94 investigated cyprinoid species. Three known species, Paradiplozoon homoion, Paradiplozoon bliccae and Paradiplozoon bingolensis, were recorded on new cyprinoid host species, and a new species, Paradiplozoon koubkovae n. sp., was recorded on Luciobarbus capito and Capoeta capoeta from the Caspian Sea basin in Iran and Turkey. Paradiplozoon bliccae, exhibiting a wide host range in the Middle East, expressed both morphological and genetic intraspecific variabilities. The four Paradiplozoon species collected in the Middle East were placed in divergent clades, showing the rich evolutionary history of diplozoid parasites in the Middle East. Our study also revealed that two lineages of African diplozoids have a Middle Eastern origin. We stress the importance of applying an integrative approach combining morphological, ecological and molecular methods to reveal the real diversity of diplozoids
