Diagnostic accuracy of liquid biopsy in endometrial cancer
Background: Liquid biopsy is a minimally invasive collection of a patient body fluid sample. In oncology, they offer several advantages compared to traditional tissue biopsies. However, the potential of this method in endometrial cancer (EC) remains poorly explored. We studied the utility of tumor educated platelets (TEPs) and circulating tumor DNA (ctDNA) for preoperative EC diagnosis, including histology determination. Methods: TEPs from 295 subjects (53 EC patients, 38 patients with benign gynecologic conditions, and 204 healthy women) were RNA-sequenced. DNA sequencing data were obtained for 519 primary tumor tissues and 16 plasma samples. Artificial intelligence was applied to sample classification. Results: Platelet-dedicated classifier yielded AUC of 97.5% in the test set when discriminating between healthy subjects and cancer patients. However, the discrimination between endometrial cancer and benign gynecologic conditions was more challenging, with AUC of 84.1%. ctDNA-dedicated classifier discriminated primary tumor tissue samples with AUC of 96% and ctDNA blood samples with AUC of 69.8%. Conclusions: Liquid biopsies show potential in EC diagnosis. Both TEPs and ctDNA profiles coupled with artificial intelligence constitute a source of useful information. Development of a New Polymeric Nanocarrier Dedicated to Controlled Clozapine Delivery at the Dopamine D2-Serotonin 5-HT1A Heteromers
Clozapine, the second generation antipsychotic drug, is one of the prominent compounds used for treatment of schizophrenia. Unfortunately, use of this drug is still limited due to serious side effects connected to its unspecific and non-selective action. Nevertheless, clozapine still remains the first-choice drug for the situation of drug-resistance schizophrenia. Development of the new strategy of clozapine delivery into well-defined parts of the brain has been a great challenge for modern science. In the present paper we focus on the presentation of a new nanocarrier for clozapine and its use for targeted transport, enabling its interaction with the dopamine D2 and serotonin 5-HT1A heteromers (D2-5-HT1A) in the brain tissue. Clozapine polymeric nanocapsules (CLO-NCs) were prepared using anionic surfactant AOT (sodium docusate) as an emulsifier, and bio-compatible polyelectrolytes such as: poly-l-glutamic acid (PGA) and poly-l-lysine (PLL). Outer layer of the carrier was grafted by polyethylene glycol (PEG). Several variants of nanocarriers containing the antipsychotic varying in physicochemical parameters were tested. This kind of approach may enable the availability and safety of the drug, improve the selectivity of its action, and finally increase effectiveness of schizophrenia therapy. Moreover, the purpose of the manuscript is to cover a wide scope of the issues, which should be considered while designing a novel means for drug delivery. It is important to determine the interactions of a new nanocarrier with many cell components on various cellular levels in order to be sure that the new nanocarrier will be safe and won’t cause undesired effects for a patient
In vitro biological activity of gold nanoparticles (Au@RD NPs) obtained using aqueous extract of Damask rose (Rosa damascene)
Synteza nanocząstek metali (NPs) oparta na chemii zrównoważonego rozwoju staje
się coraz popularniejszą ekologiczną metodą stosowaną zamiast syntezy chemicznej.
W szczególności ekstrakty roślinne jako czynniki redukujące i stabilizujące są
stopniowo wykorzystywane do syntezy metalicznych nanocząstek. Ekstrakty z róży
damasceńskiej (Rosa damascena), wykazujące właściwości antyoksydacyjne, antybakteryjne,
przeciwzapalne czy antynowotworowe, mogą być wykorzystane do syntezy
nanocząstek złota.
Celem badań była ekologiczna i kontrolowana synteza nanocząstek złota z zastosowaniem
ekstraktu wodnego z róży damasceńskiej (Au@RD NPs). Analizowano
wpływ Au@RD NPs na przeżywalność, poziom uszkodzeń DNA i apoptozę w komórkach
prawidłowych – ludzkich limfocytach, oraz nowotworowych: HL60 i A549.
W badaniach in vitro nie wykazano cyto- i genotoksyczności Au@RD NPs wobec komórek
prawidłowych, natomiast dla komórek nowotworowych wykazano selektywną cyto- i genotoksyczność oraz indukcję apoptozy. Uzyskane wyniki wskazują, że nanocząstki
złota otrzymane zgodnie z zasadami chemii zrównoważonego rozwoju są
atrakcyjnymi materiałami hybrydowymi dla badań biologicznych łączącymi dwa aktywne
składniki – nanocząstki metali i polifenole roślinne.Synthesis of metal nanoparticles (NPs) based on Sustainable Chemistry is an increasingly
popular ecological method, instead of chemical synthesis. In particular, plant
extracts as reducing and stabilizing agents are gradually used for the synthesis of metallic
nanoparticles. Extracts of Damask rose (Rosa damascena) have antioxidant, antibacterial,
anti-infl ammatory or anti-cancer properties that can be used to synthesize
gold nanoparticles.
The aim of the study was the ecological and controlled synthesis of gold nanoparticles
using Damask rose aqueous extract (Au@RD NPs). The effect of NPs Au@RD on
survival, DNA damage and apoptosis in normal cells: human lymphocytes and cancer
cells: HL60 and A549 was studied.
In vitro studies did not revealed cyto- and genotoxicity of Au@RD NPs in the
case of normal cells, while for cancer cells: selective cyto- and genotoxicity and induction
of apoptosis were demonstrated. The data show that gold nanoparticles obtained
in accordance with the principles of Sustainable Chemistry are attractive hybrid materials
for biological purposes combining two active components – metal nanoparticles
and plant polyphenols
