    Decoding steady-state visual evoked potentials from electrocorticography

    We report on a unique electrocorticography (ECoG) experiment in which Steady-State Visual Evoked Potentials (SSVEPs) to frequency-and phase-tagged stimuli were recorded from a large subdural grid covering the entire right occipital cortex of a human subject. The paradigm is popular in EEG-based Brain Computer Interfacing where selectable targets are encoded by different frequency-and/or phase-tagged stimuli. We compare the performance of two state-of-the-art SSVEP decoders on both ECoG-and scalp-recorded EEG signals, and show that ECoG-based decoding is more accurate for very short stimulation lengths (i.e., less than 1 s). Furthermore, whereas the accuracy of scalp-EEG decoding bene fi ts from a multi-electrode approach, to address interfering EEG responses and noise, ECoG decoding enjoys only a marginal improvement as even a single electrode, placed over the posterior part of the primary visual cortex, seems to suf fi ce. This study shows, for the fi rst time, that EEG-based SSVEP decoders can in principle be applied to ECoG, and can be expected to yield faster decoding speeds using less electrodes

    Human Ipsilateral Motor Physiology and Neuroprosthetic Applications in Chronic Stroke

    Improving the recovery of lost motor function in hemiplegic chronic stroke survivors is a critical need to improve the lives of these patients. Over the last several decades, neuroprosthetic systems have emerged as novel tools with the potential to restore function in a variety of patient populations. While traditional neuroprosthetics have focused on using neural activity contralateral to a moving limb for device control, an alternative control signal may be necessary to develop brain-computer interface (BCI) systems in stroke survivors that suffer damage to the cortical hemisphere contralateral to the affected limb. While movement-related neural activity also occurs in the hemisphere ipsilateral to a moving limb, it is uncertain if these signals can be used within BCI systems. This dissertation examines the motor activity ipsilateral to a moving limb and the potential use of these signals for neuroprosthetic applications in chronic stroke survivors. Patients performed three-dimensional (3D) reaching movements with the arm ipsilateral to an electrocorticography (ECoG) array in order to assess the extent of kinematic information that can be decoded from the cortex ipsilateral to a moving limb. Additionally, patients performed the same task with the arm contralateral to the same ECoG arrays, allowing us to compare the neural representations of contralateral and ipsilateral limb movements. While spectral power changes related to ipsilateral arm movements begin later and are lower in amplitude than power changes related to contralateral arm movements, 3D kinematics from both contralateral and ipsilateral arm trajectories can be decoded with similar accuracies. The ability to decode movement kinematics from the ipsilateral cortical hemisphere demonstrates the potential to use these signals within BCI applications for controlling multiple degrees of freedom. Next we examined the relationship between electrode invasiveness and signal quality. The ability to decode movement kinematics from neural activity was significantly decreased in simulated electroencephalography (EEG) signals relative to ECoG signals, indicating that invasive signals would be necessary to implement BCI systems with multiple degrees of freedom. For ECoG signals, the human dura also causes a significant decrease in signal quality when electrodes with small spatial sizes are used. This tradeoff between signal quality and electrode invasiveness should therefore be taken into account when designing ECoG BCI systems. Finally, chronic stroke survivors used activity associated with affected hand motor intentions, recorded from their unaffected hemisphere using EEG, to control simple BCI systems. This demonstrates that motor signals from the ipsilateral hemisphere are viable for BCI applications, not only in motor-intact patients, but also in chronic stroke survivors. Taken together, these experiments provide initial demonstrations that it is possible to develop BCI systems using the unaffected hemisphere in stroke survivors with multiple degrees of freedom. Further development of these BCI systems may eventually lead to improving function for a significant population of patients

    dissertationRecording the neural activity of human subjects is indispensable for fundamental neuroscience research and clinical applications. Human studies range from examining the neural activity of large regions of the cortex using electroencephalography (EEG) or electrocorticography (ECoG) to single neurons or small populations of neurons using microelectrode arrays. In this dissertation, microscale recordings in the human cortex were analyzed during administration of propofol anesthesia and articulate movements such as speech, finger flexion, and arm reach. Recordings were performed on epilepsy patients who required long-term electrocorticographic monitoring and were implanted with penetrating or surface microelectrode arrays. We used penetrating microelectrode arrays to investigate the effects of propofol anesthesia on action potentials (APs) and local field potentials (LFPs). Increased propofol concentration correlated with decreased high-frequency power in LFP spectra and decreased AP firing rates, as well as the generation of large amplitude spike-like LFP activity; however, the temporal relationship between APs and LFPs remained relatively consistent at all levels of propofol anesthesia. The propofol-induced suppression of neocortical network activity allowed LFPs to be dominated by low-frequency spike-like activity, and correlated with sedation and unconsciousness. As the low-frequency spike-like activity increased, and the AP-LFP relationship became more predictable, firing rate encoding capacity was impaired. This suggests a mechanism for decreased information processing in the neocortex that accounts for propofol-induced unconsciousness. We also demonstrated that speech, finger, and arm movements can be decoded from LFPs recorded with dense grids of microelectrodes placed on the surface of human cerebral cortex for brain computer interface (BCI) applications using LFPs recorded over face-motor area, vocalized articulations of ten different words and silence were classified on a trial-by-trial basis with 82.4% accuracy. Using LFPs recorded over the hand area of motor cortex, three individual finger movements and rest were classified on a trial-by-trial basis with 62% accuracy. LFPs recorded over the arm area of motor cortex were used to continuously decode the arm trajectory with a maximum correlation coefficient of 0.82 in the x-direction and 0.76 in the y-direction. These findings demonstrate that LFPs recorded by micro-ECoG grids from the surface of the cerebral cortex contain sufficient information to provide rapid and intuitive control a BCI communication or motor prosthesis

    Shifts in Gamma Phase–Amplitude Coupling Frequency from Theta to Alpha Over Posterior Cortex During Visual Tasks

    The phase of ongoing theta (4–8 Hz) and alpha (8–12 Hz) electrophysiological oscillations is coupled to high gamma (80–150 Hz) amplitude, which suggests that low-frequency oscillations modulate local cortical activity. While this phase–amplitude coupling (PAC) has been demonstrated in a variety of tasks and cortical regions, it has not been shown whether task demands differentially affect the regional distribution of the preferred low-frequency coupling to high gamma. To address this issue we investigated multiple-rhythm theta/alpha to high gamma PAC in two subjects with implanted subdural electrocorticographic grids. We show that high gamma amplitude couples to the theta and alpha troughs and demonstrate that, during visual tasks, alpha/high gamma coupling preferentially increases in visual cortical regions. These results suggest that low-frequency phase to high-frequency amplitude coupling is modulated by behavioral task and may reflect a mechanism for selection between communicating neuronal networks

    Enhancing Nervous System Recovery through Neurobiologics, Neural Interface Training, and Neurorehabilitation.

    After an initial period of recovery, human neurological injury has long been thought to be static. In order to improve quality of life for those suffering from stroke, spinal cord injury, or traumatic brain injury, researchers have been working to restore the nervous system and reduce neurological deficits through a number of mechanisms. For example, neurobiologists have been identifying and manipulating components of the intra- and extracellular milieu to alter the regenerative potential of neurons, neuro-engineers have been producing brain-machine and neural interfaces that circumvent lesions to restore functionality, and neurorehabilitation experts have been developing new ways to revitalize the nervous system even in chronic disease. While each of these areas holds promise, their individual paths to clinical relevance remain difficult. Nonetheless, these methods are now able to synergistically enhance recovery of native motor function to levels which were previously believed to be impossible. Furthermore, such recovery can even persist after training, and for the first time there is evidence of functional axonal regrowth and rewiring in the central nervous system of animal models. To attain this type of regeneration, rehabilitation paradigms that pair cortically-based intent with activation of affected circuits and positive neurofeedback appear to be required-a phenomenon which raises new and far reaching questions about the underlying relationship between conscious action and neural repair. For this reason, we argue that multi-modal therapy will be necessary to facilitate a truly robust recovery, and that the success of investigational microscopic techniques may depend on their integration into macroscopic frameworks that include task-based neurorehabilitation. We further identify critical components of future neural repair strategies and explore the most updated knowledge, progress, and challenges in the fields of cellular neuronal repair, neural interfacing, and neurorehabilitation, all with the goal of better understanding neurological injury and how to improve recovery

    Non-Penetrating Microelectrode Interfaces for Cortical Neuroprosthetic Applications with a Focus on Sensory Encoding: Feasibility and Chronic Performance in Striate Cortex

    abstract: Growing understanding of the neural code and how to speak it has allowed for notable advancements in neural prosthetics. With commercially-available implantable systems with bi- directional neural communication on the horizon, there is an increasing imperative to develop high resolution interfaces that can survive the environment and be well tolerated by the nervous system under chronic use. The sensory encoding aspect optimally interfaces at a scale sufficient to evoke perception but focal in nature to maximize resolution and evoke more complex and nuanced sensations. Microelectrode arrays can maintain high spatial density, operating on the scale of cortical columns, and can be either penetrating or non-penetrating. The non-penetrating subset sits on the tissue surface without puncturing the parenchyma and is known to engender minimal tissue response and less damage than the penetrating counterpart, improving long term viability in vivo. Provided non-penetrating microelectrodes can consistently evoke perception and maintain a localized region of activation, non-penetrating micro-electrodes may provide an ideal platform for a high performing neural prosthesis; this dissertation explores their functional capacity. The scale at which non-penetrating electrode arrays can interface with cortex is evaluated in the context of extracting useful information. Articulate movements were decoded from surface microelectrode electrodes, and additional spatial analysis revealed unique signal content despite dense electrode spacing. With a basis for data extraction established, the focus shifts towards the information encoding half of neural interfaces. Finite element modeling was used to compare tissue recruitment under surface stimulation across electrode scales. Results indicated charge density-based metrics provide a reasonable approximation for current levels required to evoke a visual sensation and showed tissue recruitment increases exponentially with electrode diameter. Micro-scale electrodes (0.1 – 0.3 mm diameter) could sufficiently activate layers II/III in a model tuned to striate cortex while maintaining focal radii of activated tissue. In vivo testing proceeded in a nonhuman primate model. Stimulation consistently evoked visual percepts at safe current thresholds. Tracking perception thresholds across one year reflected stable values within minimal fluctuation. Modulating waveform parameters was found useful in reducing charge requirements to evoke perception. Pulse frequency and phase asymmetry were each used to reduce thresholds, improve charge efficiency, lower charge per phase – charge density metrics associated with tissue damage. No impairments to photic perception were observed during the course of the study, suggesting limited tissue damage from array implantation or electrically induced neurotoxicity. The subject consistently identified stimulation on closely spaced electrodes (2 mm center-to-center) as separate percepts, indicating sub-visual degree discrete resolution may be feasible with this platform. Although continued testing is necessary, preliminary results supports epicortical microelectrode arrays as a stable platform for interfacing with neural tissue and a viable option for bi-directional BCI applications.Dissertation/ThesisDoctoral Dissertation Biomedical Engineering 201

    Intrinsic and Extrinsic Biomechanical Factors in a Co-adaptive ECoG-based Brain Computer Interface

    Paralysis, due to spinal cord injury, amyotrophic lateral sclerosis (ALS), or stroke, is the result of severed communication between the brain and the motor periphery. Brain computer interfaces (BCIs) are neuroprosthetic devices that create novel communication pathways by measuring and transforming neural activity into operational commands. State of the art BCI systems measure brain activity using penetrating electrode arrays able to record from hundreds of individual cortical neurons simultaneously. Unfortunately, these systems are highly susceptible to signal degradation which limits their efficacy to 1-2 years. However, electrocorticography (ECoG) signals recorded from the surface of the brain deliver a more competitive balance between surgical risk, long-term stability, signal bandwidth, and signal-to-noise ratio when compared to both the aforementioned intracortical systems and the more common non-invasive electroencephalography (EEG) technologies. Historically, neural signals for controlling a computer cursor or robotic arm have been mapped to extrinsic, kinematic (i.e. position or velocity) variables. Although this strategy is adequate for use in simple environments, it may not be ideal for control of real-world prosthetic devices that are subject to external and unexpected forces. When reaching for an object, the trajectory of the hand through space can be defined in either extrinsic (e.g. Cartesian) or intrinsic (e.g. joint angles, muscle forces) frames of reference. During this movement, the brain has to perform a series of sensorimotor transformations that involve solving a complex, 2nd order differential equation (i.e. musculoskeletal biomechanics) in order to determine the appropriate muscle activations. Functional neuromuscular stimulation (FNS) is a desirable BCI application because it attempts to restore motor function to paralyzed limbs through electrical excitation of muscles. Rather than applying the conventional extrinsic kinematic control signals to such a system, it may be more appropriate to map neural activity to muscle activation directly and allow the brain to develop its own transfer function. This dissertation examines the application of intrinsic decoding schemes to control an upper limb using ECoG in non-human primates. ECoG electrode arrays were chronically implanted in rhesus monkeys over sensorimotor cortex. A novel multi-joint reaching task was developed to train the subjects to control a virtual arm simulating muscle and inertial forces. Utilizing a co-adaptive algorithm (where both the brain adapts via biofeedback and the decoding algorithm adapts to improve performance), new decoding models were initially built over the course of the first 3-5 minutes of each daily experimental session and then continually adapted throughout the day. Three subjects performed the task using neural control signals mapped to 1) joint angular velocity, 2) joint torque, and 3) muscle forces of the virtual arm. Performance exceeded 97%, 93%, and 89% accuracy for the three control paradigms respectively. Neural control features in the upper gamma frequency bands (70-115 and 130-175 Hz) were found to be directionally tuned in an ordered fashion, with preferred directions varying topographically in the mediolateral direction without distinction between motor and sensory areas. Long-term stability was demonstrated by all three monkeys, which maintained performance at 42, 55, and 57 months post-implantation. These results provide insights into the capabilities of sensorimotor cortex for control of non-linear multi-joint reaching dynamics and present a first step toward design of intrinsic, force-based BCI systems suitable for long-term FNS applications
