    Hierarchical workflow management system for life science applications

    In modern laboratories, an increasing number of automated stations and instruments are applied as standalone automated systems such as biological high throughput screening systems, chemical parallel reactors etc. At the same time, the mobile robot transportation solution becomes popular with the development of robotic technologies. In this dissertation, a new superordinate control system, called hierarchical workflow management system (HWMS) is presented to manage and to handle both, automated laboratory systems and logistics systems.In modernen Labors werden immer mehr automatisierte Stationen und Instrumente als eigenständige automatisierte Systeme eingesetzt, wie beispielsweise biologische High-Throughput-Screening-Systeme und chemische Parallelreaktoren.     An approach to inter-organizational workflow management in an electronic institution

    In a virtual organization, different business partners (individual organizations) cooperate in order to achieve a common goal. The coordination of the corresponding inter-organizational workflow is an important issue. In this paper an approach towards managing the operational embodiment of a manufacturing consortium is presented. This approach is conceptualized as a service within an Electronic Institution framework providing several agent-based services related with the formation and operation of virtual organizations. The behavior of the inter-organizational workflow management service is presented, modeled as an extension to a contract monitoring service.     EXPLOITING KASPAROV'S LAW: ENHANCED INFORMATION SYSTEMS INTEGRATION IN DOD SIMULATION-BASED TRAINING ENVIRONMENTS

    Despite recent advances in the representation of logistics considerations in DOD staff training and wargaming simulations, logistics information systems (IS) remain underrepresented. Unlike many command and control (C2) systems, which can be integrated with simulations through common protocols (e.g., OTH-Gold), many logistics ISs require manpower-intensive human-in-the-loop (HitL) processes for simulation-IS (sim-IS) integration. Where automated sim-IS integration has been achieved, it often does not simulate important sociotechnical system (STS) dynamics, such as information latency and human error, presenting decision-makers with an unrealistic representation of logistics C2 capabilities in context. This research seeks to overcome the limitations of conventional sim-IS interoperability approaches by developing and validating a new approach for sim-IS information exchange through robotic process automation (RPA). RPA software supports the automation of IS information exchange through ISs’ existing graphical user interfaces. This “outside-in” approach to IS integration mitigates the need for engineering changes in ISs (or simulations) for automated information exchange. In addition to validating the potential for an RPA-based approach to sim-IS integration, this research presents recommendations for a Distributed Simulation Engineering and Execution Process (DSEEP) overlay to guide the engineering and execution of sim-IS environments.Major, United States Marine CorpsApproved for public release.     Patterns-based Evaluation of Open Source BPM Systems: The Cases of jBPM, OpenWFE, and Enhydra Shark

    In keeping with the proliferation of free software development initiatives and the increased interest in the business process management domain, many open source workflow and business process management systems have appeared during the last few years and are now under active development. This upsurge gives rise to two important questions: what are the capabilities of these systems? and how do they compare to each other and to their closed source counterparts? i.e. in other words what is the state-of-the-art in the area?. To gain an insight into the area, we have conducted an in-depth analysis of three of the major open source workflow management systems - jBPM, OpenWFE and Enhydra Shark, the results of which are reported here. This analysis is based on the workflow patterns framework and provides a continuation of the series of evaluations performed using the same framework on closed source systems, business process modeling languages and web-service composition standards. The results from evaluations of the three open source systems are compared with each other and also with the results from evaluations of three representative closed source systems - Staffware, WebSphere MQ and Oracle BPEL PM, documented in earlier works. The overall conclusion is that open source systems are targeted more toward developers rather than business analysts. They generally provide less support for the patterns than closed source systems, particularly with respect to the resource perspective which describes the various ways in which work is distributed amongst business users and managed through to completion

    Advanced laboratory testing methods using real-time simulation and hardware-in-the-loop techniques : a survey of smart grid international research facility network activities

    The integration of smart grid technologies in interconnected power system networks presents multiple challenges for the power industry and the scientific community. To address these challenges, researchers are creating new methods for the validation of: control, interoperability, reliability of Internet of Things systems, distributed energy resources, modern power equipment for applications covering power system stability, operation, control, and cybersecurity. Novel methods for laboratory testing of electrical power systems incorporate novel simulation techniques spanning real-time simulation, Power Hardware-in-the-Loop, Controller Hardware-in-the-Loop, Power System-in-the-Loop, and co-simulation technologies. These methods directly support the acceleration of electrical systems and power electronics component research by validating technological solutions in high-fidelity environments.     Sustainable human-robot co-production for the bicycle industry

    Bicycle production has not changed much over the last 100 years, it is still performed mainly by manual labor in mass production. During the global pandemic, the demand for ecologically friendly and customized transport has increased. Hence, customers start to impose the same requirements on bikes as on cars: they want more customized products and short delivery time. This publication describes an approach to transform bicycle manufacturing towards human-robot co-production to enable smaller batch sizes and production on-shoring. We list the challenges of this transformation, our applied methods, and presents preliminary results of the cobot-driven prototypes

    Self-managed Workflows for Cyber-physical Systems

    Workflows are a well-established concept for describing business logics and processes in web-based applications and enterprise application integration scenarios on an abstract implementation-agnostic level. Applying Business Process Management (BPM) technologies to increase autonomy and automate sequences of activities in Cyber-physical Systems (CPS) promises various advantages including a higher flexibility and simplified programming, a more efficient resource usage, and an easier integration and orchestration of CPS devices. However, traditional BPM notations and engines have not been designed to be used in the context of CPS, which raises new research questions occurring with the close coupling of the virtual and physical worlds. Among these challenges are the interaction with complex compounds of heterogeneous sensors, actuators, things and humans; the detection and handling of errors in the physical world; and the synchronization of the cyber-physical process execution models. Novel factors related to the interaction with the physical world including real world obstacles, inconsistencies and inaccuracies may jeopardize the successful execution of workflows in CPS and may lead to unanticipated situations. This thesis investigates properties and requirements of CPS relevant for the introduction of BPM technologies into cyber-physical domains. We discuss existing BPM systems and related work regarding the integration of sensors and actuators into workflows, the development of a Workflow Management System (WfMS) for CPS, and the synchronization of the virtual and physical process execution as part of self-* capabilities for WfMSes. Based on the identified research gap, we present concepts and prototypes regarding the development of a CPS WFMS w.r.t. all phases of the BPM lifecycle. First, we introduce a CPS workflow notation that supports the modelling of the interaction of complex sensors, actuators, humans, dynamic services and WfMSes on the business process level. In addition, the effects of the workflow execution can be specified in the form of goals defining success and error criteria for the execution of individual process steps. Along with that, we introduce the notion of Cyber-physical Consistency. Following, we present a system architecture for a corresponding WfMS (PROtEUS) to execute the modelled processes-also in distributed execution settings and with a focus on interactive process management. Subsequently, the integration of a cyber-physical feedback loop to increase resilience of the process execution at runtime is discussed. Within this MAPE-K loop, sensor and context data are related to the effects of the process execution, deviations from expected behaviour are detected, and compensations are planned and executed. The execution of this feedback loop can be scaled depending on the required level of precision and consistency. Our implementation of the MAPE-K loop proves to be a general framework for adding self-* capabilities to WfMSes. The evaluation of our concepts within a smart home case study shows expected behaviour, reasonable execution times, reduced error rates and high coverage of the identified requirements, which makes our CPS~WfMS a suitable system for introducing workflows on top of systems, devices, things and applications of CPS.:1.     The Impact of Mild Traumatic Brain injury on Neuronal Networks and Neurobehavior

    Despite its enormous incidence, mild traumatic brain injury is not well understood. One aspect that needs more definition is how the mechanical energy during injury affects neural circuit function. Recent developments in cellular imaging probes provide an opportunity to assess the dynamic state of neural networks with single-cell resolution. In this dissertation, we developed imaging methods to assess the state of dissociated cortical networks exposed to mild injury. We probed the microarchitecture of an injured cortical circuit subject to two different injury levels, mild stretch (10% peak) and mild/moderate (35%). We found that mild injury produced a transient increase in calcium activity that dissipated within 1 h after injury. Alternatively, mild/moderate mechanical injury produced immediate disruption in network synchrony, loss in excitatory tone, and increased modular topology, suggesting a threshold for repair and degradation. The more significant changes in network behavior at moderate stretch are influenced by NMDA receptor activation and subsequent proteolytic changes in the neuronal populations. With the ability to analyze individual neurons in a circuit before and after injury, we identified several biomarkers that confer increased risk or protection from mechanical injury. We found that pre-injury connectivity and NMDA receptor subtype composition (NR2A and NR2B content) are important predictors of node loss and remodeling. Mechanistically, stretch injury caused a reduction in voltage-dependent Mg2+ block of the NR2B-cotaning NMDA receptors, resulting in increased uncorrelated activity both at the single channel and network level. The reduced coincidence detection of the NMDA receptor and overactivation of these receptors further impaired network function and plasticity. Given the demonstrated link between NR2B-NMDARs and mitochondrial dysfunction, we discovered that neuronal de-integration from the network is mediated through mitochondrial signaling. Finally, we bridged these network level studies with an investigation of changes in neurobehavior following blast-induced traumatic brain injury (bTBI), a form of mild TBI. We first developed and validated an open-source toolbox for automating the scoring of several common behavior tasks to study the deficits that occur following bTBI. We then specifically evaluated the role of neuronal transcription factor Elk-1 in mediating deficits following blast by exposing Elk-1 knockout mouse to equivalent blast pressure loading. Our systems-level behavior analysis showed that bTBI creates a complex change in behavior, with an increase in anxiety and loss of habituation in object recognition. Moreover, we found these behavioral deficits were eliminated in Elk-1 knockout animals exposed to blast loading. Together, we merged information from different perspectives (in silico, in vitro, and in vivo) and length scales (single channels, single-cells, networks, and animals) to study the impact of mild traumatic brain injury on neuronal networks and neurobehavior
