Rule Extraction Method of Intuitionistic Fuzzy Information System
Rules acquisition is one of the difficult problems the intuitionistic fuzzy inference system encounters. For the problem of rules extraction in intuitionistic fuzzy environment, the definition of the intuitionistic fuzzy system is given at first, and the decision attributes and the condition attributes as well as the intuitionistic fuzzy logic relation implicated among the linguistic values corresponding to condition attributes are analysed as well, and the logic relation of information system is constructed in this account. Furthermore, by integrating the information with decision rules expressed by the data lines having same decision value in information system, the calculation method in raged to the reliability of intuitionistic fuzzy rules including confidence and non-confidence degrees is provided, and the rule screening method based on intuitionistic fuzzy reliability is also discussed in two situations. Based on that, the rule extraction algorithm of intuitionistic fuzzy information system is put forward. Finally, through specific numerical example the usability and validity of the algorithm are verified
Discovery of Potent, Selective Stem Cell Factor Receptor/Platelet Derived Growth Factor Receptor Alpha (c-KIT/PDGFRα) Dual Inhibitor for the Treatment of Imatinib-Resistant Gastrointestinal Stromal Tumors (GISTs)
Stem cell factor receptor (c-KIT)
and platelet derived growth factor receptor alpha (PDGFRα) kinases
play an important role in gastrointestinal stromal tumors (GISTs).
Here, we have discovered an c-KIT/PDGFRα dual inhibitor, compound <b>31</b>, with single-digit nanomolar potency against c-KIT and
PDGFRα. Compared to Imatinib (<b>1</b>), <b>31</b> showed better antiproliferative efficacy against various TEL-c-KIT/PDGFRα-BaF3
isogenic cells, including three <b>1</b>-resistant BaF3 cell
lines, as well as against GIST-T1 and GIST-882 cell lines. Furthermore,
compound <b>31</b> showed a good KinomeScan selectivity (468
kinases) (<i>S</i> score (1) = 0.01 at 1 μM concentration),
good metabolic stability in liver microsomes, and no hERG inhibitory
activity. It was worth noting that <b>31</b> inhibited GIST-T1
tumor growth (TGI = 81.5%) and even the BaF3-TEL-cKIT-T670I tumor
progression (TGI = 41.9%, <b>1</b>-resistant GISTs) at a dosage
of 100 mg/kg/day without exhibiting apparent toxicity

Impedance Detection and Modeling of Chemotherapeutic Agents by a Cancer Cell-Based Biosensor
<div><p>Impedance detection on adherently growing cells by micro-electrodes provides useful analytical methods for cancer research. Colon adenocarcinoma is one of the most common types of gastrointestinal cancer. To improve the survival rate of colon adenocarcinoma patients, chemotherapy becomes more and more important among the standard clinical treatments. In this study, a cell based-biosensor was developed to determine the cytotoxicity of therapy medicines to colorectal cancer using the LS180 cell line. By analyzing the varying parameters in the membrane and cell nucleus in an impedance circuit model, the anticancer mechanisms of the drugs were deduced by impedance sensing. By real-time monitoring of the biosensor, time- and concentration- dependent impedance changes were detected under treatment medicines, such as oxaliplatin, 5-fluorouracil, and the more effective combination regimen. The cell-based biosensor combined with circuit modeling provides a promising approach for drug screening and developing new therapeutic regimens with anticancer medicines.</p>
