Holothurians have a reduced GPCR and odorant receptor-like repertoire compared to other echinoderms

Sea cucumbers lack vision and rely on chemical sensing to reproduce and survive. However, how they recognize and respond to environmental cues remains unknown. Possible candidates are the odorant receptors (ORs), a diverse family of G protein-coupled receptors (GPCRs) involved in olfaction. The present study aimed at characterizing the chemosensory GPCRs in sea cucumbers. At least 246 distinct GPCRs, of which ca. 20% putative ORs, were found in a transcriptome assembly of putative chemosensory (tentacles, oral cavity, calcareous ring, and papillae/tegument) and reproductive (ovary and testis) tissues from Holothuria arguinensis (57 ORs) and in the Apostichopus japonicus genome (79 ORs). The sea cucumber ORs clustered with those of sea urchin and starfish into four main clades of gene expansions sharing a common ancestor and evolving under purifying selection. However, the sea cucumber ORs repertoire was the smallest among the echinoderms and the olfactory receptor signature motif LxxPxYxxxxxLxxxDxxxxxxxxP was better conserved in cluster OR-l1 which also had more members. ORs were expressed in tentacles, oral cavity, calcareous ring, and papillae/tegument, supporting their potential role in chemosensing. Donepezil-based hybrids as multifunctional anti-Alzheimer's disease chelating agents: Effect of positional isomerization

The intricate and multifactorial nature of Alzheimer's disease (AD) requires the development of compounds able to hit different pathophysiological targets, such as cholinergic dysfunction, deposits of amyloid beta (A beta) peptide and metal dyshomeostasis. In order to continue the search for new anti-AD drugs, a design strategy was once more followed based on repositioning donepezil (DNP) drug, by ortho-attaching a benzylpiperidine mimetic of DNP moiety to a hydroxyphenyl-benzimidazole (BIM) chelating unit (compound 1). Herein, compound 1 and a positional isomer 2 are compared in terms of their potential multiple properties: both present good acetylcholinesterase (AChE) inhibition (low mu molar range) and are moderate/good inhibitors of A beta self- and Cu-mediated aggregation, the inhibition process being mainly due to ligand intercalation between the p-sheets of the fibrils; compound 1 has a higher chelating capacity towards Cu2+ and Zn2+ (pCu = 14.3, pZn = 6.4, pH 7.4, C-I/C-M = 10, C-M = 10(-6) M) than 2 (pCu = 10.7, pZn = 6.3), attributed to its ability to establish a tridentate (N,O,O) coordination to the metal ion. Both compounds are eligible as drug candidates for oral administration but compound 1 shows improved neuroprotective role by completely preventing Ali-induced cell toxicity

Avaliação da satisfação dos dadores do Instituto Português do Sangue - Centro Regional de Sangue do Porto

No contexto da qualidade o grau de satisfação do dador é um indicador importante, permitindo melhorar os serviços e promover o seu retorno. Com o objectivo de avaliar o grau de satisfação dos dadores do IPS-Centro Regional de Sangue do Porto realizou-se estudo descritivo transversal, numa amostra de 512 dadores. Verificou-se que os dadores encontram-se Satisfeitos e Muito Satisfeitos com a instituição e que existe associação entre o grau de satisfação e 5 variáveis independentes. Conclui-se que os recursos humanos IPS-CRSP desempenham a sua função adequadamente, ficando identificada como área para melhoria o alargamento do horário de atendimento.In the context of the quality the degree of donor’s satisfaction is an important indicator, thereby improving services and promoting their return. In order to evaluate the degree of donor’s satisfaction from IPS-Centro Regional de Sangue do Porto we held a cross-sectional survey with a sample of 512 donors. It was found that donors are Satisfied or Very Satisfied with the institution and an association exists between the degree of satisfaction and five independent variables. It is concluded that the human resources from IPS-CRSP perform their functions properly

Mitochondria at the Base of Neuronal Innate Immunity in Alzheimer’s and Parkinson’s Diseases

Mitochondria are exceptionally primed to play a key role in neuronal cell survival since they are involved in energy production and function as the metabolic center of cells. Several findings provide evidence for the role of mitochondria in neurodegeneration associated with Alzheimer’s and Parkinson’s diseases (AD and PD). Recent data highlight the role of mitochondrial proteins and mitochondrial reactive oxygen species in the intracellular signaling that regulates innate immunity and inflammation. In this chapter, we will discuss the relevance of the interplay between mitochondria and innate immunity, focusing on mitochondrial damage-associated molecular patterns (DAMPs) and how they can activate innate immunity and elicit AD and PD neurodegenerative process

PESQUISA DE ENTEROPARASITAS EM MEIOS DE TRANSPORTE PÚBLICO URBANO DA CIDADE DE PATOS DE MINAS MG – BRASIL

As infecções parasitárias possuem relação direta com os fatores socioeconômicos e hábitos culturais de uma população e são facilmente disseminadas de um local para outro. Consequentemente, o contágio de doenças infecciosas através da via indireta pelas mãos é de extrema importância, especialmente em locais públicos e de aglomeração humana como o transporte coletivo. Propõem-se pesquisar e identificar cistos de protozoários e ovos de helmintos em transportes coletivos urbanos da cidade de Patos de Minas-MG. A coleta de amostras foi realizada em catracas, corrimões e bancos de aproximadamente 30 ônibus coletivos de diferentes zonas da cidade. Para isso, utilizou-se o método de Graham, onde fitas adesivas tipo durex foram fixadas a diferentes superfícies do ônibus e posteriormente coladas em lâminas de microscopia, e analisadas em objetivas de 10x e 40x. Nas amostras foram observados raros cistos de protozoários e nenhum ovo de helminto. Apesar de não ter apresentado significado estatístico, os resultados indicaram a contaminação do meio de transporte e risco potencial para a população que utiliza este serviço

LISTERIOSE NA GESTAÇÃO: incidência de infecção em gestantes devido à baixa imunidade

Listeria monocytogenes é um bacilo que causa infecção em humanos com maior incidência durante a gestação, em pacientes imunodeprimidos ou com alguma doença de base grave, levando na maioria dos casos a meningite e bacteremia. A listeriose geralmente é assintomática, mas pode levantar suspeita em gestantes com sintomas de gripe, febre e dores no corpo. Com este trabalho foi possível concluir que a doença pode levar a graves comprometimentos como: derrames serosos, natimorto, aborto e até mesmo o óbito, podendo ser transmitida para o homem através da ingestão de alimentos contaminados pela bactéria. Supervision in continuous teacher training

The concept of Supervision is not new, however, in the XXI century, has been reframed and gained a more relevant status in Portugal, because it operates in a changing context of the exercise of the teaching profession, emerging associated with a new paradigm of teachers’ intervention and action. With regard, continuing education is assumed as a determining factor, and therefore the present study searched for i) evidence of implementation of supervising nature of supervision practices and iii) perceptions/opinions of those involved in the CFAEs, in order to establish the concept of supervision of its stakeholders. We followed a multiple case study design, selected and characterized five CFAEs geographically dispersed in the Portuguese mainland (North, Centre, Lisbon and South), we developed instruments for opinion collection, with a sample of directors, trainers and trainees with a mixed methodology: qualitative through interview with the directors of the CFAE’s and quantitative (the predominant approach) through a questionnaire applied to trainees and trainers (who do not accumulate those two functions in the period of analysis). Results of both the interviews and the questionnaires were presented by category. The relevance of pedagogical supervision in order to present a recognized and respected praxis is therefore assumed, with the purpose of making it essential to the efficient and effective balance of the Education and Training system(s) in Portugal.info:eu-repo/semantics/publishedVersio

MAPA OU TERRITÓRIO? A EXPERIÊNCIA DE CONSULTORES NA IDENTIFICAÇÃO DE ROTINAS ORGANIZACIONAIS

Problematizações envolvendo a dinâmica operativa de rotinas organizacionais vêm apresentando desafios crescentes para pesquisadores do constructo, ao considerar dificuldades e limitações circunstanciais da identificação do fenômeno em diferentes ambientes e contextos de ocorrência. Nessa perspectiva, por meio de uma abordagem fenomenográfica, esta investigação teve por objetivo coletar concepções de consultores externos sobre a identificação de rotinas organizacionais em suas experiências profissionais. Quatro diferentes modos de identificar o fenômeno emergiram dos relatos como resposta a esse problema de pesquisa: o fundamento em artefatos; o questionamento ao agente; a observação da prática; e a construção coletiva. O presente estudo sugere que a consistência de um entendimento pontual sobre a prática organizacional, entre consultores e agentes e entre os agentes da rotina, é influenciada por condições relacionais, refletidas em mecanismos de poder e confiança que envolvem esses sujeitos, sugerindo aos consultores o desenvolvimento de competências específicas para promover a identificação de rotinas nas organizações
