    Comparative Study of Brain Activities in Immersive Visualization Environments: An Innovative Pedagogical Technique Abstract The primary objective of this work-in-progress is to investigate advanced and innovative pedagogical techniques in teaching and learning STEM concepts by utilizing Immersive Visualization Environments (IVE). Specifically, a comparative study of brain activities was performed to determine effectiveness of different Immersive Visualization Environments in pedagogy. Twelve randomly selected college students participated in this investigation using an electroencephalography (EEG) device for measuring brain waves of the subjects. The preliminary collected data supported the assertion that dome-shaped IVE elicited a more relaxed state than its counterpart, head-mounted IVE, yielding a higher performance.     Use of videotape learning packages : a marital enrichment field experiment with two delivery systems

    The purposes of this study were (a) to evaluate the effectiveness of two procedures (traditional group workshop and individual telephone conference/mail) for training home economics Extension agents to use videotape resources in working with married couples, and (b) to design, implement, and evaluate videotape learning packages for facilitating married couples' interpersonal competence skills in self-understanding, communication, and growth toward states of consensus and commitment to their relationships. Videotape learning packages were utilized by Extension agents acting as leaders with groups of married couples. The sample consisted of 50 married couples and 10 agent-leaders from 10 counties in two Agricultural Extension Service districts.     Revision and field test of a self-instructional program on the sewing machine

    The purposes of this study were to revise and field test the self-instructional program on the sowing machine developed by Moore. Revisions of the Sewing Machine Program, prepared from the recommendations of a preliminary field test, included the addition of (l) performance frames, (2) sections of frames for objectives not programed in the first edition, (3) colored frames for various models of sewing machines, (4) an introduction to programed instruction for the students, and (5) a number of illustrations. The revised program contained 340 frames of which 71 were "no response" frames and 123 were frames requiring performance at a sewing machine. Fifteen responses were teacher-reinforced

    An Ideology of IT Occupational Culture: The ASPIRE Values

    Many have conjectured that people in the IT occupation are different from non-IT business users and that such differences can lead to negative organizational outcomes. This study investigates how to measure these differences by developing a new, expanded theoretical framework of IT occupational culture (ITOC). This framework includes: Artifacts, Values, and Tacit Assumptions. Values form the core of any culture, and a cohesive set of cultural values is termed an "ideology." Using a mixed methodology, research was conducted in two parts to develop and measure an ideology of ITOC. A qualitative investigation based on interview data provided evidence of six core values in the ideology: Autonomy in Decision-Making, Structure in Environment, Precision in Communication, Innovation in Technology, Reverence for Technical Knowledge, and Enjoyment at the Workplace (ASPIRE). The quantitative investigation sought to validate the values and ultimately reduced the number of values to five based on factor analysis of survey data: Autonomy in Decision-Making, Structure/Precision, Innovation in Technology, Reverence for Technical Knowledge, and Enjoyment at the Workplace. IT respondents rated these values significantly higher than non-IT business personnel. Our findings have implications for practitioners and researchers and can provide a path to bridge the gap between IT and business users

    Molt-dependent transcriptomic analysis of cement proteins in the barnacle Amphibalanus amphitrite

    Abstract Background A complete understanding of barnacle adhesion remains elusive as the process occurs within and beneath the confines of a rigid calcified shell. Barnacle cement is mainly proteinaceous and several individual proteins have been identified in the hardened cement at the barnacle-substrate interface. Little is known about the molt- and tissue-specific expression of cement protein genes but could offer valuable insight into the complex multi-step processes of barnacle growth and adhesion. Methods The main body and sub-mantle tissue of the barnacle Amphibalanus amphitrite (basionym Balanus amphitrite) were collected in pre- and post-molt stages. RNA-seq technology was used to analyze the transcriptome for differential gene expression at these two stages and liquid chromatography-mass spectrometry/mass spectrometry (LC-MS/MS) was used to analyze the protein content of barnacle secretions. Results We report on the transcriptomic analysis of barnacle cement gland tissue in pre- and post-molt growth stages and proteomic investigation of barnacle secretions. While no significant difference was found in the expression of cement proteins genes at pre- and post-molting stages, expression levels were highly elevated in the sub-mantle tissue (where the cement glands are located) compared to the main barnacle body. We report the discovery of a novel 114kD cement protein, which is identified in material secreted onto various surfaces by adult barnacles and with the encoding gene highly expressed in the sub-mantle tissue. Further differential gene expression analysis of the sub-mantle tissue samples reveals a limited number of genes highly expressed in pre-molt samples with a range of functions including cuticular development, biominerialization, and proteolytic activity. Conclusions The expression of cement protein genes appears to remain constant through the molt cycle and is largely confined to the sub-mantle tissue. Our results reveal a novel and potentially prominent protein to the mix of cement-related components in A. amphitrite. Despite the lack of a complete genome, sample collection allowed for extended transcriptomic analysis of pre- and post-molt barnacle samples and identified a number of highly-expressed genes.     Original research: Trauma exposure and posttraumatic stress disorder among employees of New York City companies affected by the september 11, 2001 attacks on the World Trade Center

    OBJECTIVE: Several studies have provided prevalence estimates of posttraumatic stress disorder (PTSD) related to the September 11, 2001 (9/11) attacks in broadly affected populations, although without sufficiently addressing qualifying exposures required for assessing PTSD and estimating its prevalence. A premise that people throughout the New York City area were exposed to the attacks on the World Trade Center (WTC) towers and are thus at risk for developing PTSD has important implications for both prevalence estimates and service provision. This premise has not, however, been tested with respect to DSM-IV-TR criteria for PTSD. This study examined associations between geographic distance from the 9/11 attacks on the WTC and reported 9/11 trauma exposures, and the role of specific trauma exposures in the development of PTSD. METHODS: Approximately 3 years after the attacks, 379 surviving employees (102 with direct exposures, including 65 in the towers, and 277 with varied exposures) recruited from 8 affected organizations were interviewed using the Diagnostic Interview Schedule/Disaster Supplement and reassessed at 6 years. The estimated closest geographic distance from the WTC towers during the attacks and specific disaster exposures were compared with the development of 9/11–related PTSD as defined by the Diagnostic and Statistical Manual, Fourth Edition, Text Revision. RESULTS: The direct exposure zone was largely concentrated within a radius of 0.1 mi and completely contained within 0.75 mi of the towers. PTSD symptom criteria at any time after the disaster were met by 35% of people directly exposed to danger, 20% of those exposed only through witnessed experiences, and 35% of those exposed only through a close associate’s direct exposure. Outside these exposure groups, few possible sources of exposure were evident among the few who were symptomatic, most of whom had preexisting psychiatric illness. CONCLUSIONS: Exposures deserve careful consideration among widely affected populations after large terrorist attacks when conducting clinical assessments, estimating the magnitude of population PTSD burdens, and projecting needs for specific mental health interventions

    Older adults experience age-related physiological changes that increase sensitivity and decrease tolerance to alcohol and there are a number of age-related harms such as falls, social isolation and elder abuse, which are compounded by alcohol misuse. Despite this unique vulnerability and the fact that the number of older adults is increasing, the literature on drinking behaviour and alcohol-related harm in older adults is sparse. This article describes a secondary analysis of UK data to address this knowledge gap

    The Sodium Sialic Acid Symporter From Staphylococcus aureus Has Altered Substrate Specificity

    Mammalian cell surfaces are decorated with complex glycoconjugates that terminate with negatively charged sialic acids. Commensal and pathogenic bacteria can use host-derived sialic acids for a competitive advantage, but require a functional sialic acid transporter to import the sugar into the cell. This work investigates the sodium sialic acid symporter (SiaT) from Staphylococcus aureus (SaSiaT). We demonstrate that SaSiaT rescues an Escherichia coli strain lacking its endogenous sialic acid transporter when grown on the sialic acids N-acetylneuraminic acid (Neu5Ac) or N-glycolylneuraminic acid (Neu5Gc). We then develop an expression, purification and detergent solubilization system for SaSiaT and demonstrate that the protein is largely monodisperse in solution with a stable monomeric oligomeric state. Binding studies reveal that SaSiaT has a higher affinity for Neu5Gc over Neu5Ac, which was unexpected and is not seen in another SiaT homolog. We develop a homology model and use comparative sequence analyses to identify substitutions in the substrate-binding site of SaSiaT that may explain the altered specificity. SaSiaT is shown to be electrogenic, and transport is dependent upon more than one Na+ ion for every sialic acid molecule. A functional sialic acid transporter is essential for the uptake and utilization of sialic acid in a range of pathogenic bacteria, and developing new inhibitors that target these transporters is a valid mechanism for inhibiting bacterial growth. By demonstrating a route to functional recombinant SaSiaT, and developing the in vivo and in vitro assay systems, our work underpins the design of inhibitors to this transporter

    Hundreds of variants clustered in genomic loci and biological pathways affect human height

    Most common human traits and diseases have a polygenic pattern of inheritance: DNA sequence variants at many genetic loci influence the phenotype. Genome-wide association (GWA) studies have identified more than 600 variants associated with human traits, but these typically explain small fractions of phenotypic variation, raising questions about the use of further studies. Here, using 183,727 individuals, we show that hundreds of genetic variants, in at least 180 loci, influence adult height, a highly heritable and classic polygenic trait. The large number of loci reveals patterns with important implications for genetic studies of common human diseases and traits. First, the 180 loci are not random, but instead are enriched for genes that are connected in biological pathways (P = 0.016) and that underlie skeletal growth defects (P < 0.001). Second, the likely causal gene is often located near the most strongly associated variant: in 13 of 21 loci containing a known skeletal growth gene, that gene was closest to the associated variant. Third, at least 19 loci have multiple independently associated variants, suggesting that allelic heterogeneity is a frequent feature of polygenic traits, that comprehensive explorations of already-discovered loci should discover additional variants and that an appreciable fraction of associated loci may have been identified. Fourth, associated variants are enriched for likely functional effects on genes, being over-represented among variants that alter amino-acid structure of proteins and expression levels of nearby genes. Our data explain approximately 10% of the phenotypic variation in height, and we estimate that unidentified common variants of similar effect sizes would increase this figure to approximately 16% of phenotypic variation (approximately 20% of heritable variation). Although additional approaches are needed to dissect the genetic architecture of polygenic human traits fully, our findings indicate that GWA studies can identify large numbers of loci that implicate biologically relevant genes and pathways.
