    Nitric Oxide Regulates Neurogenesis in the Hippocampus following Seizures

    Hippocampal neurogenesis is changed by brain injury. When neuroinflammation accompanies injury, activation of resident microglial cells promotes the release of inflammatory cytokines and reactive oxygen/nitrogen species like nitric oxide (NO). In these conditions, NO promotes proliferation of neural stem cells (NSC) in the hippocampus. However, little is known about the role of NO in the survival and differentiation of newborn cells in the injured dentate gyrus. Here we investigated the role of NO following seizures in the regulation of proliferation, migration, differentiation, and survival of NSC in the hippocampus using the kainic acid (KA) induced seizuremouse model. We show that NO increased the proliferation of NSC and the number of neuroblasts following seizures but was detrimental to the survival of newborn neurons. NO was also required for the maintenance of long-term neuroinflammation.     LOAD DURING THE VERTICAL JUMP IN WATER: VALUES FOR PRESCRIPTION IN HYDROTHERAPY

    This study aimed to analyse the vertical component of the ground reaction force (GRF) in the vertical jump in water performed by men and women at two levels of immersion. 11 men and 11 women performed three vertical jumps on a water-proof force plate at hip and chest levels of immersion. No effect of gender was observed. No difference between levels of immersion was found for peak of propulsion [1.85 and 1.89 units of body weight (BW) at the hip and chest respectively]. During the landing phase, the force peak was significantly higher at the hip level (2.62 BW) than at the chest level (2.07 BW). The force during the propulsion phase was similar between the immersions; however the vertical load on landing needs to be considered when prescribing this exercise, even in water

    Electrochemical miRNA-34a-based biosensor for the diagnosis of Alzheimer’s disease

    Alzheimer's disease (AD) is the most common dementia type and a leading cause of death and disability in the elderly. Diagnosis is expensive and invasive, urging the development of new, affordable, and less invasive diagnostic tools. The identification of changes in the expression of non-coding RNAs prompts the development of diagnostic tools to detect disease-specific blood biomarkers. Building on this idea, this work reports a novel electrochemical microRNA (miRNA) biosensor for the diagnosis of AD, based on carbon screen-printed electrodes (C-SPEs) modified with two gold nanostructures and a complementary anti-miR-34a oligonucleotide probe. This biosensor showed good target affinity, reflected on a 100 pM to 1 M linearity range and a limit of detection (LOD) of 39 pM in buffer and 94 aM in serum. Moreover, the biosensors response was not affected by serum compounds, indicating selectivity for miR-34a. The biosensor also detected miR-34a in the cell culture medium of a common AD model, stimulated with a neurotoxin to increase miR-34a secretion.     Fatty acid characterization of cyanobacterial strains isolated from vela lake and mondego river rice fields (central-western, Portugal)

    It is know that cyanobacterial taxonomic groups are characterized by particular lipid patterns that can be used as their biological markers. The present study examined the fatty acid composition of nostocacean heterocystous cyanobacterial strains isolated from Central-western Portuguese freshwater shallow water bodies, namely Vela Lake and rice fields from Mondego River Basin. Morphological characterization showed that strains from Vela Lake belonged to Aphanizomenon gracile (strains UADFA16 and UADFA18), Aphanizomenon flos-aquae (strain UADFA15) and Anabaena cf. solitaria (strain UADFA14) species, whereas rice field strains belonged to Anabaena cylindrica (strain UTAD_A212) and Nostoc muscorum (strain UTAD_N213). Biochemical characterization inferred from lipid analysis showed that predominant fatty acid methyl esters (FAMEs) in the lipids of the strains were palmitic, oleic and α-linolenic, with trace amounts of myristic and C20 polyunsaturated FAMEs. To our knowledge, there is almost no information about lipid composition in freshwater cyanobacterial species living in different habitats in Portugal.     RELATIVE ERROR ANALYSIS DURING REPRODUCTION OF ISOMETRIC FORCE OF KNEE EXTENSORS IN YOUNG ADULTS

    This study compared the force reproduction of isometric knee extension at 60°, between the target forces at 30%, 50% and 70% of the maximum voluntary isometric force (MVIF) in young adults. Twenty young males took part in the study. The trials were performed on an instrumented chair developed for the study. Feedback was given to the participants during three trials prior the beginning of data collection in each analyzed target force. Participants performed 10 trials without any feedback for the target force at 30% and 50% of the MVIF and 3 trials for the target force at 70% of MVIF. The relative error decreased as the target force increased. The differences were significant between 30% and 70% (p=0.01) and between 50% and 70% (p=0.03). The reproduction of higher forces during isometric extension of the knee will produce lower relative errors for this specific task

    Determinants, Health Problems, and Food Insecurity in Urban Areas of the Largest City in Cape Verde.

    Urbanization processes are intertwined with nutritional transition because there is easier access to food of low nutritional quality at reduced prices, changing dietary patterns and leading to an increase of non-communicable chronic diseases. This study aims to understand the perceptions for high blood pressure, obesity, and alcoholism, describing some interactions of these dimensions in the problem of food security in the city of Praia. A qualitative study was carried out under the framework of the research project "UPHI-STAT: Urban Planning and Health Inequalities-moving from macro to micro statistics". Ten focus groups were conducted in three urban areas with distinct characteristics in the city of Praia, with a total of 48 participants. Participants reported frequent consumption of foods with poor nutritional quality, understanding the potential danger in terms of food security in the city of Praia. Easy access to and high levels of alcohol consumption, and poor quality of traditional drinks were mentioned by participants in the study areas. The impact of the economic situation on the possibility of access to safe and healthy options emerged as a differentiating factor.publishersversionpublishe

    BACKGROUND: Cape Verde presents a high rate of cardiovascular diseases. Low potassium and high sodium intakes are related to cardiovascular diseases. However, studies regarding these two micronutrients continue to be rare in African urban settings. This work aims to estimate potassium and sodium intakes and to analyse the self-reported salt intake by gender and by type of urban area in the city of Praia - the capital of Cape Verde. METHODS: In the first stage (n = 1912), an intra-urban study was designed in two types of urban areas (formal and informal), using a sampling strategy based on random selection of geographical coordinates, in order to apply a questionnaire. In a second stage, a 24-h dietary recall and anthropometric measurements were performed by local nutritionists. Potassium and sodium intakes were estimated for 599 participants (149 men and 450 women). Non-parametric methods (including quantile regression) were used in the statistical analysis. RESULTS: In informal areas, a higher percentage of women reported having hypertension (31.0%) compared to formal areas (19.7%). Based on 24-h dietary recall, median potassium intake for men was 2924.2 mg/day and for women and 2562.6 mg/day. Almost 70.0% of men and 80.0% of women ingested less than the recommended 3510 mg/day of potassium. In informal areas, men and women presented high medians of sodium intakes compared to formal areas (men: 4131.2 vs 3014.6 mg/day and women: 3243.4 vs 2522.4 mg/day). On the other hand, the percentage of participants exceeding 2000 mg/day for sodium was high (≥70.8%), even for participants that self-reported low-salt intake. Quantile regression models revealed effects of the type of urban area and gender in the potassium and sodium intakes, at least, in some quartiles, accounting for age, academic qualifications, and professional situation. CONCLUSIONS: A low potassium intake and a high sodium intake were found in Praia. Thus, efficient health education campaigns and health promotion are needed and should be tailored considering gender and urban areas.publishersversionpublishe

    Water hyacinth (Eichhornia crassipes) detection using coarse and high resolution multispectral data

    Efficient detection and monitoring procedures of invasive plant species are required. It is of crucial importance to deal with such plants in aquatic ecosystems, since they can affect biodiversity and, ultimately, ecosystem function and services. In this study, it is intended to detect water hyacinth (Eichhornia crassipes) using multispectral data with different spatial resolutions. For this purpose, high-resolution data (<0.1 m) acquired from an unmanned aerial vehicle (UAV) and coarse-resolution data (10 m) from Sentinel-2 MSI were used. Three areas with a high incidence of water hyacinth located in the Lower Mondego region (Portugal) were surveyed. Different classifiers were used to perform a pixel-based detection of this invasive species in both datasets. From the different classifiers used, the results were achieved by the random forest classifiers stand-out (overall accuracy (OA): 0.94). On the other hand, support vector machine performed worst (OA: 0.87), followed by Gaussian naive Bayes (OA: 0.88), k-nearest neighbours (OA: 0.90), and artificial neural networks (OA: 0.91). The higher spatial resolution from UAV-based data enabled us to detect small amounts of water hyacinth, which could not be detected in Sentinel-2 data. However, and despite the coarser resolution, satellite data analysis enabled us to identify water hyacinth coverage, compared well with a UAV-based survey. Combining both datasets and even considering the different resolutions, it was possible to observe the temporal and spatial evolution of water hyacinth. This approach proved to be an effective way to assess the effects of the mitigation/control measures taken in the study areas.     Enhanced 3T3-L1 differentiation into adipocytes by pioglitazone pharmacological activation of peroxisome proliferator activated receptor-gamma (PPAR-gamma)

    Despite the primary function of pioglitazone in antidiabetic treatment, this drug is a potent inducer of PPAR-γ, a crucial receptor that is involved in adipocyte differentiation. In this work, we propose an optimized methodology to enhance the differentiation of 3T3-L1 fibroblasts into adipocytes. This process is crucial for adipocyte secretome release, which is fundamental for understanding the molecular mechanisms that are involved in obesity for in vitro studies. To achieve this, a pioglitazone dose-response assay was determined over a range varying from 0 to 10 µM. Lipid accumulation was evaluated using Oil-Red-O. The results showed that 10 µM pioglitazone enhanced differentiation and increased secretome production. This secretome was then added into two cell lines: PC3 and RAW264.7. In the PC3 cells, an increase of aggressiveness was observed in terms of viability and proliferation, with the increase of anti-inflammatory cytokines. Conversely, in RAW264.7 cells, a reduction of viability and proliferation was observed, with a decrease in the overexpression of pro-inflammatory cytokines. Overall, the present work constitutes an improved method for adipocyte secretome production that is suitable for experimental biology studies and that could help with our understanding of the molecular mechanisms underlying adiposity influence in other cells.info:eu-repo/semantics/publishedVersio
