    Karakteristik Kerajinan Patung Loro Blonyo Wakiman di Dusun Kopen Desa Kenteng Kecamatan Purwantoro Kabupaten Wonogiri

    Wonogiri adalah daerah potensial yang menghasilkan berbagai barang kerajinan patung berbahan dasar kayu. Salah satu diantaranya kerajinan patung UD. Cantik milik Wakiman di Dusun Kopen Desa Kenteng Kecamatan Purwantoro. UD. Cantik memiliki karya khas yaitu patung Loro Blonyo. Produk tersebut telah mencapai pasar luar kota Wonogiri dan mampu bertahan di pasaran hingga saat ini. Hal ini dikarenakan UD. Cantik memiliki cara tersendiri dalam mengolah produknya agar tetap diminati. Proses finishing yang rumit telah menciptakan patung-patung yang unik serta karakteristik patung Loro Blonyo yang beragam. Berdasar latar belakang di atas, masalah yang dikaji dalam penelitian ini adalah: Proses pembuatan dan karakteristik patung Loro Blonyo karya Wakiman di Dusun Kopen Desa Kenteng Wonogiri. Pendekatan penelitian yang digunakan adalah kualitatif deskriptif, metode pengumpulan data observasi, wawancara, dan dokumentasi. Teknik analisis data yang digunakan adalah dengan menelaah hasil wawancara, observasi, dan pengamatan yang sudah dituliskan dalam catatan lapangan, dokumen resmi, dan gambar foto. Dari hasil penelitian dapat disimpulkan bahwa awal mula dikenalnya patung Loro Blonyo di Desa Kopen Kenteng yaitu dari seorang kolektor barang antik yang juga seorang perajin bernama Sono Karto. Pembeda patung Loro Blonyo milik Wakiman terdapat pada warna dan bentuk. Selain menggunakan warna merah, kuning, hijau, biru, putih, dan hitam, Wakiman juga menggunakan warna yang terkesan antik yang diperoleh dari proses finishing. Karakteristik bentuk patung Loro Blonyo berbeda dengan yang lain karena terdapat tekstur retak pada permukaan patung dan terbuat dari kayu utuh dan tidak terdapat unsur tempel pada bagian bagian tubuh sehingga lebih awet dari pada patung yang lain. The approach of the research used is qualitative descriptive , data collection method observation, interview, and documentation. Data analysis technique used is to analyse the results of interviews, observation, and the observation already written in a record field, official documents, and the photograph. The results can be inferred from the beginning of the familiar Loro Blonyo Kopen Kenteng in villages namely from a collector of antiques who was also named Sono Karto makers. Distinguishing a statue belonging to Loro Blonyo Wakiman there is in color and shape . In addition to using a red color, yellow, green, blue, white, and black, also use colors impressed Wakiman antique obtained from the process of finishing.     Discovery of Molecular DNA Methylation-Based Biomarkers through Genome-Wide Analysis of Response Patterns to BCG for Bladder Cancer

    Background: Bacillus Calmette-Guérin (BCG) immunotherapy, the standard adjuvant intravesical therapy for some intermediate and most high-risk non-muscle invasive bladder cancers (NMIBCs), suffers from a heterogenous response rate. Molecular markers to help guide responses are scarce and currently not used in the clinical setting. Methods: To identify novel biomarkers and pathways involved in response to BCG immunotherapy, we performed a genome-wide DNA methylation analysis of NMIBCs before BCG therapy. Genome-wide DNA methylation profiles of DNA isolated from tumors of 26 BCG responders and 27 failures were obtained using the Infinium MethylationEPIC BeadChip. Results: Distinct DNA methylation patterns were found by genome-wide analysis in the two groups. Differentially methylated CpG sites were predominantly located in gene promoters and gene bodies associated with bacterial invasion of epithelial cells, chemokine signaling, endocytosis, and focal adhesion. In total, 40 genomic regions with a significant difference in methylation between responders and failures were detected. The differential methylation state of six of these regions, localized in the promoters of the genes GPR158, KLF8, C12orf42, WDR44, FLT1, and CHST11, were internally validated by bisulfite-sequencing. GPR158 promoter hypermethylation was the best predictor of BCG failure with an AUC of 0.809 (p-value < 0.001). Conclusions: Tumors from BCG responders and BCG failures harbor distinct DNA methylation profiles. Differentially methylated DNA regions were detected in genes related to pathways involved in bacterial invasion of cells or focal adhesion. We identified candidate DNA methylation biomarkers that may help to predict patient prognosis after external validation in larger, well-designed cohorts

    Disease-Modifying Therapies and Coronavirus Disease 2019 Severity in Multiple Sclerosis

    Objective: This study was undertaken to assess the impact of immunosuppressive and immunomodulatory therapies on the severity of coronavirus disease 2019 (COVID-19) in people with multiple sclerosis (PwMS). Methods: We retrospectively collected data of PwMS with suspected or confirmed COVID-19. All the patients had complete follow-up to death or recovery. Severe COVID-19 was defined by a 3-level variable: mild disease not requiring hospitalization versus pneumonia or hospitalization versus intensive care unit (ICU) admission or death. We evaluated baseline characteristics and MS therapies associated with severe COVID-19 by multivariate and propensity score (PS)-weighted ordinal logistic models. Sensitivity analyses were run to confirm the results. Results: Of 844 PwMS with suspected (n = 565) or confirmed (n = 279) COVID-19, 13 (1.54%) died; 11 of them were in a progressive MS phase, and 8 were without any therapy. Thirty-eight (4.5%) were admitted to an ICU; 99 (11.7%) had radiologically documented pneumonia; 96 (11.4%) were hospitalized. After adjusting for region, age, sex, progressive MS course, Expanded Disability Status Scale, disease duration, body mass index, comorbidities, and recent methylprednisolone use, therapy with an anti-CD20 agent (ocrelizumab or rituximab) was significantly associated (odds ratio [OR] = 2.37, 95% confidence interval [CI] = 1.18\u20134.74, p = 0.015) with increased risk of severe COVID-19. Recent use (<1 month) of methylprednisolone was also associated with a worse outcome (OR = 5.24, 95% CI = 2.20\u201312.53, p = 0.001). Results were confirmed by the PS-weighted analysis and by all the sensitivity analyses. Interpretation: This study showed an acceptable level of safety of therapies with a broad array of mechanisms of action. However, some specific elements of risk emerged.     DMTs and Covid-19 severity in MS: a pooled analysis from Italy and France

    We evaluated the effect of DMTs on Covid-19 severity in patients with MS, with a pooled-analysis of two large cohorts from Italy and France. The association of baseline characteristics and DMTs with Covid-19 severity was assessed by multivariate ordinal-logistic models and pooled by a fixed-effect meta-analysis. 1066 patients with MS from Italy and 721 from France were included. In the multivariate model, anti-CD20 therapies were significantly associated (OR = 2.05, 95%CI = 1.39–3.02, p < 0.001) with Covid-19 severity, whereas interferon indicated a decreased risk (OR = 0.42, 95%CI = 0.18–0.99, p = 0.047). This pooled-analysis confirms an increased risk of severe Covid-19 in patients on anti-CD20 therapies and supports the protective role of interferon

    Suatu program komputer telah dibuat untuk membantu para rnahasiswa Fakultas Teknik Jurusan Sipil dalam mempelajari perilaku struktur akibat suatu sistim pembebanan dinamis. Program ini dibuat sebagai kelanjutan dari program lain yang dikembangkan untuk membantu pengajaran mata kuliah Gempa II di Universitas Kristen Petra Surabaya, Program ini dapat digunakan untuk struktur dimana pusat massa dan pusat kekakuan tidak berimpit yang menyebabkan terjadinya torsi. Program ini juga mampu menampilkan animasi tampak samping yang diidealisasikan sebagai lumped mass dan animasi tampak atas dari respons struktur akibat dibebani beban dinamis pada satu arah dan pengaruh terhadap arah tegak lurusnya. Penggunaan program ini diusahakan sesederhana mungkin sehingga dapat digunakan oleh pemakai yang mempunyai pengetahuan minimal tentang komputer. Dengan adanya prograrn komputer im diharapkan pemakai bisa memperoleh pengertian yang lebih baik tentang perilaku struktur akibat suatu sistim pembebanan dinamis, terutama mengenai pengaruh torsi akibat struktur bangunan yang tidak simetris
