    Rifampicin: biotransformation study using the fungus Cunninghamella elegans and monitoring through UHPLC-MS

    Drug biotransformation studies appear as an alternative to pharmacological investigations of metabolites, development of new drug candidates with reduced investment and most efficient production. The objective of this study was to evaluate the capacity of biotransformation of Rifampicin (RIF) by the filamentous fungus Cunninghamella elegans as a microbial model of mammalian metabolism. In 120 h, C. elegans transformed the drug into the following two metabolites: rifampicin quinone and novel metabolite. The products of rifampicin formed in vitro were monitored by HPLC-PDA, being identified through UHPLC–QTOF/MS. Metabolites were characterized according to their chromatographic profile, mass fragments and UV spectral data. The major metabolic pathways of rifampicin transformed by the fungus were oxidation, demethylation and mono-oxidation. The microbial transformation of RIF showed the potential of Cunninghamella species to produce RIF metabolites. This process can be used for a cost effective method for both known and unknown metabolite production

    Rensch’s and Bergmann’s Rules in Cis-Andean South-American Howler Monkeys (Mammalia: Alouatta)

    Howler monkeys (genus Alouatta) are large folivorous primates living in South America. We tested for the application of both Rensch’s rule and Bergmann’s rule to body size variation in Alouatta. We found that Rensch’s rule does apply in howlers. In Alouatta, males exploit dominance rank competition, and take advantage from seasonal abundance of high nutritious fruit supply in their diet. This mating system and dietary charateristics suggest positive male selection for body size is responsible for Rensch’s rule. However, since folivory favors large body size in primates (to lower mass specific metabolic rate) and it is the primary dietary habitus in howlers, larger species do occur in the Amazon basin, originating a reversed Bergmann’s rule pattern for both males and females at the interspecific level. The spatial and phylogenetic components of such body size patterns of variation are both important, implying Alouatta ecomorphological differences to occur above the species level, justifying their non-overlapping geographic distribution

    Rat skin physiology is modified by age

    Animal models have been useful in the study of specific mechanisms affecting human skin. Such is the case with ageing and the micromechanical changes determining wrinkles in UV- irradiated mice. These models enabled us to understand that ageing encompasses many peculiar mechanical responses which cannot be explained by homogeneous deformation of the skin. Moreover, a major aspect to consider is that ageing processes differ among species and direct inferences are rarely possible. This project aimed to compare age-induced changes in skin properties by studying two Wistar rat groups of different ages – young adult rats (n=7, 20–24 week-old) and old adult rats (n=5, 48–72 week-old). Non-invasive measuring approaches involving transepidermal water loss (TEWL), superficial hydration (MoistureMeter), and biomechanics (Reviscometer, and Cutometer by MPA80) were taken in the animal dorsum (neck, bottom) over a period of five days, allowing statistical comparisons between groups. Results revealed no significant differences for TEWL, firmness or viscoelasticity between the two groups. However, a significant hydration decrease, of nearly 40%, was noted in the old adult rats group. These results, although preliminary, if translatable to humans could be useful to address age-related changes in processes such as healing, or patch dressing efficacy/safety where hydration is a critical determinant. Their potential use for the prediction of efficacy and or safety of topically applied products for the elderly as compared to young adults may warrant further investigation.

    Cárie dentária e fatores relacionados em crianças Brasileiras de região com água fluoretada e não fluoretada

    PURPOSE: To compare the caries prevalence, saliva buffering capacity (SBC), oral hygiene (OH), dietary habits, family income (FI) and frequency of visits to a dental office (Do) between Brazilian children living in areas with and without fluoridated public water supply. METHODS: Forty-six 5-7-year-old preschoolers were selected in Itatiba, SP, Brazil; 19 were from a fluoridated area, and 27 were from a non-fluoridated area. The caries index was determined according to the World Health Organization criteria, and the SBC was assessed by titration with hydrochloric acid. The FI, frequency of OH and visits to Do were estimated by questionnaire. The dietary habits were assessed with a diet chart. The differences between the groups were analyzed with Mann-Whitney-U tests (α=0.05). RESULTS: Children from the non-fluoridated area showed significantly higher dmft/DMFT than those from the fluoridated area, but they showed significantly lower SBC, OH frequency and FI. No significant differences were observed between the areas for dietary habits and visits to Do. CONCLUSION: Children from fluoridated areas showed higher salivary buffering capacity, family income and oral hygiene frequency as well as lower caries prevalence, supporting the beneficial effect of fluoride in the tap water for caries prevention.OBJETIVO: Comparar prevalência de cárie, capacidade tamponante da saliva (CTS), higiene bucal (HB), hábitos dietéticos, renda familiar (RF) e frequência de visita a consultórios odontológicos (Co) entre crianças brasileiras residentes em áreas de água de abastecimento público fluoretadas e não fluoretadas. METODOLOGIA: Quarenta e seis crianças entre 5-7 anos foram selecionadas em Itatiba-SP-Brasil, sendo 19 pertencentes à área fluoretada e 27 à área não fluoretada. O índice de cárie foi determinado de acordo com o critério da Organização Mundial de Saúde e a CTS foi medida por titulação com ácido clorídrico. A RF, frequência de HB e visita ao Co foram estimadas por questionário. Os hábitos dietéticos foram avaliados com diário de dieta. As diferenças entre as variáveis foram analisadas pelo teste de Mann Whitney (α=0,05). RESULTADOS: As crianças da área não fluoretada apresentaram significativamente maior ceod/CPOD que àquelas da área fluoretada, porém significativamente menor CTS, HB e RF. Com relação a hábitos dietéticos e visitas ao Co, não foi observada diferença significativa entre as áreas. CONCLUSÃO: As crianças da área fluoretada apresentaram maior capacidade tamponante da saliva, renda familiar e frequência de higiene bucal, assim como menor prevalência de cárie, reforçando o efeito benéfico do flúor nas águas de abastecimento para prevenir a cárie
