    Towards Systems Design for Supporting Enabling Communities

    Communities and community support systems should be designed to enable participants to work effectively towards conducting both collective and individual activities and achieving their goals. Such communities are called Enabling Communities. Our focus is the systems that support community members in doing whichever tasks or activities they need or want to. These systems include processes, technology, information and people. This paper provides a framework for Supporting Enabling Communities (SEComm) in two major components: (1) Participant Support Systems (PaSS) and (2) Community Participation Levels (CPaL). Three case studies apply this framework to an emerging virtual community and a community of practice

    A Transgenic Rat for Investigating the Anatomy and Function of Corticotrophin Releasing Factor Circuits.

    Corticotrophin-releasing factor (CRF) is a 41 amino acid neuropeptide that coordinates adaptive responses to stress. CRF projections from neurons in the central nucleus of the amygdala (CeA) to the brainstem are of particular interest for their role in motivated behavior. To directly examine the anatomy and function of CRF neurons, we generated a BAC transgenic Crh-Cre rat in which bacterial Cre recombinase is expressed from the Crh promoter. Using Cre-dependent reporters, we found that Cre expressing neurons in these rats are immunoreactive for CRF and are clustered in the lateral CeA (CeL) and the oval nucleus of the BNST. We detected major projections from CeA CRF neurons to parabrachial nuclei and the locus coeruleus, dorsal and ventral BNST, and more minor projections to lateral portions of the substantia nigra, ventral tegmental area, and lateral hypothalamus. Optogenetic stimulation of CeA CRF neurons evoked GABA-ergic responses in 11% of non-CRF neurons in the medial CeA (CeM) and 44% of non-CRF neurons in the CeL. Chemogenetic stimulation of CeA CRF neurons induced Fos in a similar proportion of non-CRF CeM neurons but a smaller proportion of non-CRF CeL neurons. The CRF1 receptor antagonist R121919 reduced this Fos induction by two-thirds in these regions. These results indicate that CeL CRF neurons provide both local inhibitory GABA and excitatory CRF signals to other CeA neurons, and demonstrate the value of the Crh-Cre rat as a tool for studying circuit function and physiology of CRF neurons

    Evaluation of delivery options for second-stage events

    Cesarean delivery in the second stage of labor is common, whereas the frequency of operative vaginal delivery has been declining. However, data comparing outcomes for attempted operative vaginal delivery in the second stage versus cesarean in the second stage are scant. Previous studies that examine operative vaginal delivery have compared it to a baseline risk of complications from a spontaneous vaginal delivery and cesarean delivery. However, when a woman has a need for intervention in the second stage, spontaneous vaginal delivery is not an option she or the provider can choose. Thus, the appropriate clinical comparison is cesarean versus operative vaginal delivery

    Soil sedimentology at Gusev Crater from Columbia Memorial Station to Winter Haven

    A total of 3140 individual particles were examined in 31 soils along Spirit’s traverse. Their size, shape, and texture were quantified and classified. They represent a unique record of 3 years of sedimentologic exploration from landing to sol 1085 covering the Plains Unit to Winter Haven where Spirit spent the Martian winter of 2006. Samples in the Plains Unit and Columbia Hills appear as reflecting contrasting textural domains. One is heterogeneous, with a continuum of angular-to-round particles of fine sand to pebble sizes that are generally dust covered and locally cemented in place. The second shows the effect of a dominant and ongoing dynamic aeolian process that redistributes a uniform population of medium-size sand. The texture of particles observed in the samples at Gusev Crater results from volcanic, aeolian, impact, and water-related processes

    Pan-Cancer Analysis of lncRNA Regulation Supports Their Targeting of Cancer Genes in Each Tumor Context

    Long noncoding RNAs (lncRNAs) are commonly dys-regulated in tumors, but only a handful are known toplay pathophysiological roles in cancer. We inferredlncRNAs that dysregulate cancer pathways, onco-genes, and tumor suppressors (cancer genes) bymodeling their effects on the activity of transcriptionfactors, RNA-binding proteins, and microRNAs in5,185 TCGA tumors and 1,019 ENCODE assays.Our predictions included hundreds of candidateonco- and tumor-suppressor lncRNAs (cancerlncRNAs) whose somatic alterations account for thedysregulation of dozens of cancer genes and path-ways in each of 14 tumor contexts. To demonstrateproof of concept, we showed that perturbations tar-geting OIP5-AS1 (an inferred tumor suppressor) andTUG1 and WT1-AS (inferred onco-lncRNAs) dysre-gulated cancer genes and altered proliferation ofbreast and gynecologic cancer cells. Our analysis in-dicates that, although most lncRNAs are dysregu-lated in a tumor-specific manner, some, includingOIP5-AS1, TUG1, NEAT1, MEG3, and TSIX, synergis-tically dysregulate cancer pathways in multiple tumorcontexts
