    Optimal Algorithms for Scheduling under Time-of-Use Tariffs

    We consider a natural generalization of classical scheduling problems in which using a time unit for processing a job causes some time-dependent cost which must be paid in addition to the standard scheduling cost. We study the scheduling objectives of minimizing the makespan and the sum of (weighted) completion times. It is not difficult to derive a polynomial-time algorithm for preemptive scheduling to minimize the makespan on unrelated machines. The problem of minimizing the total (weighted) completion time is considerably harder, even on a single machine. We present a polynomial-time algorithm that computes for any given sequence of jobs an optimal schedule, i.e., the optimal set of time-slots to be used for scheduling jobs according to the given sequence. This result is based on dynamic programming using a subtle analysis of the structure of optimal solutions and a potential function argument. With this algorithm, we solve the unweighted problem optimally in polynomial time. For the more general problem, in which jobs may have individual weights, we develop a polynomial-time approximation scheme (PTAS) based on a dual scheduling approach introduced for scheduling on a machine of varying speed. As the weighted problem is strongly NP-hard, our PTAS is the best possible approximation we can hope for

    We consider a natural generalization of classical scheduling problems to a setting in which using a time unit for processing a job causes some time-dependent cost, the time-of-use tariff, which must be paid in addition to the standard scheduling cost. We focus on preemptive single-machine scheduling and two classical scheduling cost functions, the sum of (weighted) completion times and the maximum completion time, that is, the makespan. While these problems are easy to solve in the classical scheduling setting, they are considerably more complex when time-of-use tariffs must be considered. We contribute optimal polynomial-time algorithms and best possible approximation algorithms. For the problem of minimizing the total (weighted) completion time on a single machine, we present a polynomial-time algorithm that computes for any given sequence of jobs an optimal schedule, i.e., the optimal set of time slots to be used for preemptively scheduling jobs according to the given sequence.     Strong LP formulations for scheduling splittable jobs on unrelated machines

    International audienceA natural extension of the makespan minimization problem on unrelated machines is to allow jobs to be partially processed by different machines while incurring an arbitrary setup time. In this paper we present increasingly stronger LP-relaxations for this problem and their implications on the approximability of the problem. First we show that the straightforward LP, extending the approach for the original problem, has an integrality gap of 3 and yields an approximation algorithm of the same factor. By applying a lift-and-project procedure, we are able to improve both the integrality gap and the implied approximation factor to 1+ϕ1+ϕ , where ϕϕ is the golden ratio. Since this bound remains tight for the seemingly stronger machine configuration LP, we propose a new job configuration LP that is based on an infinite continuum of fractional assignments of each job to the machines. We prove that this LP has a finite representation and can be solved in polynomial time up to any accuracy. Interestingly, we show that our problem cannot be approximated within a factor better than ee−1≈1.582(unless =)ee−1≈1.582(unless P=NP) , which is larger than the inapproximability bound of 1.5 for the original problem

    Exosomes released upon mitochondrial ASncmtRNA knockdown reduce tumorigenic properties of malignant breast cancer cells

    Indexación ScopusDuring intercellular communication, cells release extracellular vesicles such as exosomes, which contain proteins, ncRNAs and mRNAs that can influence proliferation and/or trigger apoptosis in recipient cells, and have been proposed to play an essential role in promoting invasion of tumor cells and in the preparation of metastatic niches. Our group proposed the antisense non-coding mitochondrial RNA (ASncmtRNA) as a new target for cancer therapy. ASncmtRNA knockdown using an antisense oligonucleotide (ASO-1537S) causes massive death of tumor cells but not normal cells and strongly reduces metastasis in mice. In this work, we report that exosomes derived from ASO-1537S-treated MDA-MB-231 breast cancer cells (Exo-1537S) inhibits tumorigenesis of recipient cells, in contrast to exosomes derived from control-ASO-treated cells (Exo-C) which, in contrast, enhance these properties. Furthermore, an in vivo murine peritoneal carcinomatosis model showed that Exo-1537S injection reduced tumorigenicity compared to controls. Proteomic analysis revealed the presence of Lactadherin and VE-Cadherin in exosomes derived from untreated cells (Exo-WT) and Exo-C but not in Exo-1537S, and the latter displayed enrichment of proteasomal subunits. These results suggest a role for these proteins in modulation of tumorigenic properties of exosome-recipient cells.     Minimal information for studies of extracellular vesicles (MISEV2023): From basic to advanced approaches

    Extracellular vesicles (EVs), through their complex cargo, can reflect the state of their cell of origin and change the functions and phenotypes of other cells. These features indicate strong biomarker and therapeutic potential and have generated broad interest, as evidenced by the steady year-on-year increase in the numbers of scientific publications about EVs. Important advances have been made in EV metrology and in understanding and applying EV biology. However, hurdles remain to realising the potential of EVs in domains ranging from basic biology to clinical applications due to challenges in EV nomenclature, separation from non-vesicular extracellular particles, characterisation and functional studies. To address the challenges and opportunities in this rapidly evolving field, the International Society for Extracellular Vesicles (ISEV) updates its 'Minimal Information for Studies of Extracellular Vesicles', which was first published in 2014 and then in 2018 as MISEV2014 and MISEV2018, respectively. The goal of the current document, MISEV2023, is to provide researchers with an updated snapshot of available approaches and their advantages and limitations for production, separation and characterisation of EVs from multiple sources, including cell culture, body fluids and solid tissues. In addition to presenting the latest state of the art in basic principles of EV research, this document also covers advanced techniques and approaches that are currently expanding the boundaries of the field. MISEV2023 also includes new sections on EV release and uptake and a brief discussion of in vivo approaches to study EVs. Compiling feedback from ISEV expert task forces and more than 1000 researchers, this document conveys the current state of EV research to facilitate robust scientific discoveries and move the field forward even more rapidly

    Skin detection is a low-level operation used in multiple application fields, e.g. video surveillance, human computer interface. The paper presents a multi-procedure methodology followed by a fu- sion stage that can be applied in the robust detec- tion of skin. Two well-known methodologies are fused with a novel one, which is presented here as well, through the application of the fuzzy integral. The automated parameterization of the resulting procedure and its performance on a benchmark database are finally described
