    Physical Activity and Life Quality of Part-Time Students

    Physical activities are important for everyone in order to keep the physical preparedness and support an adequate level of life quality. Physical activities of young people – students are basis for keeping health capacity, working ability, developing business and carrier, and providing economic and social stability. Sport activities are very popular nowadays. Anyone can choose various sport activities that are suitable and available, that favour physical fitness and increase health status. Physical activities usually are planned during leisure time and weekends. We have provided a questionnaire about physical activities of student groups in period from 2012 till 2016. Studies have indicated the number of smokers, assessed results of questionnaire about physical activity, the body mass, body heights, and anthropometric indices. Results reflected that general physical activity level (in scores) in examined groups – part time students – were low for female and moderate for male. Recent research suggests that a significant percentage of students didn’t get adequate physical activity. We fixed increased numbers of individuals of both genders who have no any sports activity in the last years. The mean value of body mass index (BMI) for male was above standard. The amount of physical activity of working people has dropped and the resulting poor physical condition is becoming a threat to their working ability, health and well-being. Contemporary life style is connected to full time job that reduces the time dedicated to physical activities. The consequences of physical inactivity are associated to multiple health related problems. Musculoskeletal pathology is one of the most common work–related health problem, as well as an important cause of morbidity for population. It is an inflammatory and degenerative ailment that affects all individuals in working age. Dále jsou zde popsány i marketingové aktivity v posledních letech a jejich současný stav.     Influence of high physical load endurance exercises on the anthropometric parameters and health status of military personnel

    Military personnel are in high-risk group for health disorders, musculoskeletal pathology and psychological overload. We provide evaluation of the anthropometric parameters and health status of military personnel, Cadets, who have participated in ten days duration Combat training course. Combat training course is associated with life-style change – social, environmental, climate and geographical conditions, biorhythms, nutrition etc., with an increase in physical and psych emotional loads. It is characterized with transition from daily routine of military study process in National Defence academy to military exercise in external environment. Cadets faced to feeding limitation, sleeping deprivation intensive and continuous training, strict discipline etc. We provided analysis of anthropometric parameters before, and in time of Combat training course, summarized data of health statement and health problems that were fixed in the medical cards. The proper body mass supports good health capacity and we provided assessment of it. Physical fitness and body composition have adverse effect on military performance. Body composition undergoes changes during higher physical load performance. Military personnel have high risk to health, concerning impact of external and internal factors. According the review of medical cards of cadets group, we indicated main problems, which are fixed after the Combat training course.publishersversionPeer reviewe

    A multiplexed bead assay for profiling glycosylation patterns on serum protein biomarkers of pancreatic cancer

    A multiplexed bead‐based immunoassay was developed to simultaneously profile glycosylation patterns of serum proteins to investigate their usefulness as biomarkers for pancreatic cancer. The multiplex assay utilized protein‐specific capture antibodies chemically coupled individually to beads labeled with specific amounts of fluorescent dye. Captured proteins were detected based on the extent and specific type of glycosylation as determined by successive binding of fluorescent lectin probes. Advantages to this technique include the fact that antibodies coupled to the beads had minimal nonspecific binding to the lectins ConA/SNA, avoiding the step of chemically blocking the antibody glycans and the bead assays were performed in a 96‐well filter plate enabling high‐throughput screening applications with improved reproducibility. The assay was tested with ConA and SNA lectins to examine the glycosylation patterns of α‐1‐β glycoprotein (A1BG) and serum amyloid p (SAP) component for use as potential biomarkers for the detection of pancreatic cancer based on the results from prior biomarker studies. The results showed that the SNA response on the captured A1BG protein could distinguish chronic pancreatitis samples from pancreatic cancer with a p ‐value of 0.035 and for the SAP protein with SNA, a p ‐value of 0.026 was found between the signal of normal controls and the pancreatic cancer samples. For the ConA response, a decline in the signal for both proteins in the serum samples was found to distinguish pancreatic cancer from normal controls and renal cell carnoma samples (A1BG, p <0.05; and SAP, p <0.0001).Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/87003/1/2028_ftp.pd

    Rituximab suppresses disease activity after natalizumab withdrawal: an exploratory study

    Background Natalizumab is highly effective in reducing multiple sclerosis disease activity; however it carries a risk of progressive multifocal leukoencephalopathy, that represents the main reason of drug discontinuation. After natalizumab withdrawal, reactivation of disease is soon observed and, until now, it is not known which treatment strategy should be followed after natalizumab discontinuation. Aim of this study is to evaluate rituximab efficacy in controlling disease activity after natalizumab withdrawal

    Glycoprotein analysis using protein microarrays and mass spectrometry

    Protein glycosylation plays an important role in a multitude of biological processes such as cell–cell recognition, growth, differentiation, and cell death. It has been shown that specific glycosylation changes are key in disease progression and can have diagnostic value for a variety of disease types such as cancer and inflammation. The complexity of carbohydrate structures and their derivatives makes their study a real challenge. Improving the isolation, separation, and characterization of carbohydrates and their glycoproteins is a subject of increasing scientific interest. With the development of new stationary phases and molecules that have affinity properties for glycoproteins, the isolation and separation of these compounds have advanced significantly. In addition to detection with mass spectrometry, the microarray platform has become an essential tool to characterize glycan structure and to study glycosylation-related biological interactions, by using probes as a means to interrogate the spotted or captured glycosylated molecules on the arrays. Furthermore, the high-throughput and reproducible nature of microarray platforms have been highlighted by its extensive applications in the field of biomarker validation, where a large number of samples must be analyzed multiple times. This review covers a brief survey of the other experimental methodologies that are currently being developed and used to study glycosylation and emphasizes methodologies that involve the use of microarray platforms. This review describes recent advances in several options of microarray platforms used in glycoprotein analysis, including glycoprotein arrays, glycan arrays, lectin arrays, and antibody/lectin arrays.     Pediatric multiple sclerosis: update on diagnostic criteria, imaging, histopathology and treatment choices

    Pediatric multiple sclerosis (MS) represents less than 5% of the MS population, but patients with pediatric-onset disease reach permanent disability at a younger age than adult onset patients. Accurate diagnosis at presentation and optimal long-term treatment is vital to mitigate ongoing neuroinflammation and irreversible neurodegeneration. However, it may be difficult to early differentiate pediatric MS from acute disseminated encephalomyelitis (ADEM) and neuromyelitis optica spectrum disorders (NMOSD) as they often have atypical presentation that differs from that of adult-onset MS.
