Multifunctional House
Diplomová práce je řešena jako kompletní dokumentace polyfunkčního domu. Polyfunkční dům je situován v obci Nová Paka a funkčně je navržen jako restaurace, garáže a 8 bytových jednotek. Zastavěná plocha objektu je 886,95 m2. Objekt je zastřešen plochou střechou. Nosná konstrukce je navržena z prefabrikovaného železobetonového skeletu. Výplňové zdivo je z keramických tvarovek Porotherm. Dům je zateplen kontaktním zateplovacím systémem ETICS. Diplomová práce obsahuje statické a dispoziční řešení, požární bezpečnost, úsporu energie a bezpečnost při užívání. Výkresy byly zpracovány v softwaru určeném k projektování - ArchiCAD.Master´s thesis is designed as a complete documentation of a multifunctional house. The multifunctional house is located in the town Nová Paka and functionally designed as a restaurant, garages and 8 flats. Built-up area is 886.95 square meters. The building is covered with a flat roof. The supporting structure is designed prefabricated reinforced concrete frame. Infill walls are made of clay blocks Porotherm. The house is insulated contact system ETICS. The thesis contains a static layout, fire safety, energy saving and safety in use. Drawings were processed with software for design - ArchiCAD.
Suggestion of a Financial Concept for the Development of a House
Diplomová práce analyzuje způsoby financování výstavby rodinného domu, s důrazem na možnost současné investice. Tato analýza je pak provedena na modelovém příkladu. Praktická část obsahuje výběr produktů z každé posuzované oblasti, se kterými bude prováděna následná analýza: vhodná hypotéka, investice do finančního trhu, investice do alternativního zdroje vytápění objektu a fotovoltaické elektrárny. Dále demonstruje možnosti, jak snížit celkovou cenu úvěru díky současné investici do finančních trhů nebo pomocí alternativní investice. Do úvahy je bráno i riziko spojené s investicí. Nakonec je provedena komparativní analýza všech zkoumaných možností, ze které vychází vyhodnocení a navržení nejvhodnějšího systému financování.The diploma thesis analyzes available methods of financing of a new housing construction with the focus on the possibilities of concurrent investment. The above analysis is demonstrated in a model case study. Practical part contents selection of the products to be analyzed from each evaluated field: suitable mortgage loan, finance market investment, the investment to the alternate heating equipment of the home and photovoltaic power plant. Than demonstrate the possibilities of decreasing the price of the loan with the assistance of concurrent investment into the financial markets or the alternative investment. The risk of investing has been taken into consideration as well. At the end are demonstrated comparative analysis and evaluation of all financing possibilities resulting in suggesting the most suitable method of home loan financing.

The Cause of Mechanical Vibration of Palasher Synchronous Motors and its Removal
The paper describes the procedure and the result of the analysis of mechanical vibrations of mining machines at the Palasher mine in the Russian Federation. The cause of these vibrations was the change in the magnitude of the magnetic flux in the air gap. The simulations showed the possibility of eliminating these changes in magnetic flux by injecting suitable voltages into the stator windings. The implementation of this injection is further described by torque variable component feed-direct compensation. Finally, oscillography records are presented before and after injection

Smartphone Voice Calls Provide Early Biomarkers of Parkinsonism in Rapid Eye Movement Sleep Behavior Disorder.
BACKGROUND
Speech dysfunction represents one of the initial motor manifestations to develop in Parkinson's disease (PD) and is measurable through smartphone.
OBJECTIVE
The aim was to develop a fully automated and noise-resistant smartphone-based system that can unobtrusively screen for prodromal parkinsonian speech disorder in subjects with isolated rapid eye movement sleep behavior disorder (iRBD) in a real-world scenario.
METHODS
This cross-sectional study assessed regular, everyday voice call data from individuals with iRBD compared to early PD patients and healthy controls via a developed smartphone application. The participants also performed an active, regular reading of a short passage on their smartphone. Smartphone data were continuously collected for up to 3 months after the standard in-person assessments at the clinic.
RESULTS
A total of 3525 calls that led to 5990 minutes of preprocessed speech were extracted from 72 participants, comprising 21 iRBD patients, 26 PD patients, and 25 controls. With a high area under the curve of 0.85 between iRBD patients and controls, the combination of passive and active smartphone data provided a comparable or even more sensitive evaluation than laboratory examination using a high-quality microphone. The most sensitive features to induce prodromal neurodegeneration in iRBD included imprecise vowel articulation during phone calls (P = 0.03) and monopitch in reading (P = 0.05). Eighteen minutes of speech corresponding to approximately nine calls was sufficient to obtain the best sensitivity for the screening.
CONCLUSION
We consider the developed tool widely applicable to deep longitudinal digital phenotyping data with future applications in neuroprotective trials, deep brain stimulation optimization, neuropsychiatry, speech therapy, population screening, and beyond. © 2024 The Author(s). High-resolution metagenomic reconstruction of the freshwater spring bloom
Background
The phytoplankton spring bloom in freshwater habitats is a complex, recurring, and dynamic ecological spectacle that unfolds at multiple biological scales. Although enormous taxonomic shifts in microbial assemblages during and after the bloom have been reported, genomic information on the microbial community of the spring bloom remains scarce.
Results
We performed a high-resolution spatio-temporal sampling of the spring bloom in a freshwater reservoir and describe a multitude of previously unknown taxa using metagenome-assembled genomes of eukaryotes, prokaryotes, and viruses in combination with a broad array of methodologies. The recovered genomes reveal multiple distributional dynamics for several bacterial groups with progressively increasing stratification. Analyses of abundances of metagenome-assembled genomes in concert with CARD-FISH revealed remarkably similar in situ doubling time estimates for dominant genome-streamlined microbial lineages. Discordance between quantitations of cryptophytes arising from sequence data and microscopic identification suggested the presence of hidden, yet extremely abundant aplastidic cryptophytes that were confirmed by CARD-FISH analyses. Aplastidic cryptophytes are prevalent throughout the water column but have never been considered in prior models of plankton dynamics. We also recovered the first metagenomic-assembled genomes of freshwater protists (a diatom and a haptophyte) along with thousands of giant viral genomic contigs, some of which appeared similar to viruses infecting haptophytes but owing to lack of known representatives, most remained without any indication of their hosts. The contrasting distribution of giant viruses that are present in the entire water column to that of parasitic perkinsids residing largely in deeper waters allows us to propose giant viruses as the biological agents of top-down control and bloom collapse, likely in combination with bottom-up factors like a nutrient limitation.
Conclusion
We reconstructed thousands of genomes of microbes and viruses from a freshwater spring bloom and show that such large-scale genome recovery allows tracking of planktonic succession in great detail. However, integration of metagenomic information with other methodologies (e.g., microscopy, CARD-FISH) remains critical to reveal diverse phenomena (e.g., distributional patterns, in situ doubling times) and novel participants (e.g., aplastidic cryptophytes) and to further refine existing ecological models (e.g., factors affecting bloom collapse). This work provides a genomic foundation for future approaches towards a fine-scale characterization of the organisms in relation to the rapidly changing environment during the course of the freshwater spring bloom
