    Hope as a Source of Resilience in Later Adulthood

    This research provided a preliminary investigation of how variations in trait and state hope are associated with positive adaptation to stress in later adulthood. Trait hope and neuroticism were measured by questionnaires and state hope, stress, and negative emotions were assessed daily for 45 days. Results from multilevel random coefficient modeling analyses suggested that daily hope provides protective benefits by keeping negative emotions low, while also contributing to adaptive recovery from stress. The dynamic linkages between daily hope, stress, and emotion were further moderated by individual differences in trait hope. Compared with those low in trait hope, high-hope individuals showed diminished stress reactivity and more effective emotional recovery

    How scientists and physicians use Twitter during a medical congress

    OBJECTIVES: During medical congresses Twitter allows discussions to disseminate beyond the congress hall and reach a wider audience. Insights into the dynamics of social media interactions during congresses, dissemination of scientific information and the determinants of a successful tweet may allow us to better understand social media's role in science communication. METHODS: We retrospectively extracted social media data during the European Congress of Clinical Microbiology and Infectious Diseases (ECCMID) 2017 and 2018 using NodeXL. We compared social media activity during these two congresses. Subsequently, we conducted in-depth analyses to identify the components of a successful tweet and multivariable analysis to assess independent factors associated with retweet activity. RESULTS: In 2018, approximately 13,000 delegates attended ECCMID, but only 591 Twitter accounts actively tweeted about the congress. Although fewer tweets were posted in 2018 compared to 2017 (4,213 vs 4,657, respectively), ECCMID2018 generated a 63% increase in the total number of retweets (p <0.001). According to multivariable logistic regression analysis, using multimedia, URL or hashtags and mentioning other Twitter account(s) were independently associated with retweet success. Mentioning of other users and use of multimedia were the only consistent predictors of retweets irrespective of the number of followers. CONCLUSIONS: A substantial increase in retweet activity and a modest increase in the number of influential Twitter accounts were observed between two successive congresses. Dissemination of scientific messages is more successful when connected accounts are actively involved in social media activity, and social media posts constitute the right combination of components.PostprintPeer reviewe

    A fluorescence in situ staining method for investigating spores and vegetative cells of Clostridia by confocal laser scanning microscopy and structured illuminated microscopy

    Non-pathogenic spore-forming Clostridia are of increasing interest due to their application in biogas production and their capability to spoil different food products. The life cycle for Clostridium includes a spore stage that can assist in survival under environmentally stressful conditions, such as extremes of temperature or pH. Due to their size, spores can be investigated by a range of microscopic techniques, many of which involve sample pretreatment. We have developed a quick, simple and non-destructive fluorescent staining procedure that allows a clear differentiation between spores and vegetative cells and effectively stains spores, allowing recovery and tracking in subsequent experiments. Hoechst 34580, Propidium iodide and wheat germ agglutinin WGA 488 were used in combination to stain four strains of Clostridia at different life cycle stages. Staining was conducted without drying the sample, preventing changes induced by dehydration and cells observed by confocal laser scanner microscopy or using a super-resolution microscope equipped with a 3D-structured illumination module. Dual staining with Hoechst/Propidium iodide differentiated spores from vegetative cells, provided information on the viability of cells and was successfully applied to follow spore production induced by heating. Superresolution microscopy of spores probed by Hoechst 34580 also allowed chromatin to be visualised. Direct staining of a cheese specimen using Nile Red and Fast Green allowed in situ observation of spores within the cheese and their position within the cheese matrix. The proposed staining method has broad applicability and can potentially be applied to follow Clostridium spore behaviour in a range of different environments

    Endogenous retinoids in rat epididymal tissue and rat and human spermatozoa

    Recent work has demonstrated high levels of retinoid binding proteins in rat epididymis, and a lumenal retinoic acid binding protein has been purified. These findings suggested that vitamin A may be involved in spermatozoal maturation in the epididymis. We further addressed this question by quantifying retinol, retinyl esters, and retinoic acid isomers from perfused epididymal tissue, from rat testicular and epididymal spermatozoa, and from human ejaculate sperm. HPLC showed vitamin A levels to be higher in caput than in corpus or cauda tissue. Retinoic acid and 9-cis-retinoic acid were found to be graded from lowest levels in caput to highest in cauda. Spermatozoa from caput epididymidis and enriched testicular spermatozoa were found to have higher levels of vitamin A than did spermatozoa from corpus or cauda epididymidis. Spermatozoal retinyl esters had acyl substituents similar to those seen in whole epididymis, and diminished in quantity in sperm from distal segments. Human ejaculate sperm were found to retain high levels of retinyl palmitate and stearate. Retinol and retinoic acid were only marginally detectable in human sperm. Retention of retinoids in mature spermatozoa suggests roles for vitamin A in spermatozoal reproductive physiology beyond the epididymal stage

    Mobile radio interferometric geodetic systems

    Operation of the Astronomical Radio Interferometric Earth Surveying (ARIES) in a proof of concept mode is discussed. Accuracy demonstrations over a short baseline, a 180 km baseline, and a 380 km baseline are documented. Use of ARIES in the Sea Slope Experiment of the National Geodetic Survey to study the apparent differences between oceanographic and geodetic leveling determinations of the sea surface along the Pacific Coast is described. Intergration of the NAVSTAR Global Positioning System and a concept called SERIES (Satellite Emission Radio Interferometric Earth Surveying) is briefly reviewed
